| 1 | USP19 suppresses inflammation and promotes M2-like macrophage polarization by manipulating NLRP3 function via autophagy显示文摘Macrophage polarization to proinflammatory M1-like or anti-inflammatory M2-like cells is critical to mount a host defense or repair tissue.The exact molecular mechanisms controlling this process are still elusive.Here,we report that ubiquitin-specific protease 19(USP19)acts as an anti-inflammatory switch that inhibits inflammatory responses and promotes M2-like macrophage polarization.USP19 inhibited NLRP3 inflammasome activation by increasing autophagy flux and decreasing the generation of mitochondrial reactive oxygen species.In addition,USP19 inhibited the proteasomal degradation of inflammasome-independent NLRP3 by cleaving its polyubiquitin chains.USP19-stabilized NLRP3 promoted M2-like macrophage polarization by direct association with interferon regulatory factor 4,thereby preventing its p62-mediated selective autophagic degradation.Consistent with these observations,compared to wild-type mice,Usp19−/−mice had decreased M2-like macrophage polarization and increased interleukin-1βsecretion,in response to alum and chitin injections.Thus,we have uncovered an unexpected mechanism by which USP19 switches the proinflammatory function of NLRP3 into an anti-inflammatory function,and suggest that USP19 is a potential therapeutic target for inflammatory interventions. | Tao Liu Liqiu Wang Puping Liang Xiaojuan Wang Yukun Liu Jing Cai Yuanchu She Dan Wang Zhi Wang Zhiyong Guo Samuel Bates Xiaojun Xia Junjiu Huang Jun Cui | 2021 | Cellular & Molecular Immunology2021,18,10: | 15 |
| 2 | Effects of oxidized low density lipoprotein on the growth of human artery smooth muscle cells显示文摘Background Studies have shown that oxidized low density lipoprotein (ox-LDL) promotes the pathogenesis and development of atherosclerosis (AS), and that the proliferation, migration and phenotype alteration of vascular smooth muscle cells (vSMCs) into foam cells are critical changes in AS. It is proposed that ox-LDL might play a novel role in the pathologic process of vSMCs. The present study was performed ex vivo to investigate the effects of ox-LDL on the growth of cultured human vSMCs.Methods Using NaBr density gradient centrifugation, LDL from human plasma was isolated and purified. ox-LDL was produced from LDL after being incubated with CuSO4. ox-LDL was then added to the culture medium at different concentrations (25 μg/ml, 50 μg/ml, 75 μg/ml, 100 μg/ml, 125 μg/ml, and 150 μg/ml) for 7 days. The influence of ox-LDL on vSMC growth was observed from several aspects as growth curve, mitosis index, lipid staining, and in situ determination of apoptosis. The digital results were analyzed with SPSS 10.0.Results The ox-LDL produced ex vivo had a good purity and optimal oxidative degree, which was similar to the intrinsic ox-LDL in atherosclerotic plaque. ox-LDL at a concentration of 25 μg/ml demonstrated the strongest proliferation. At the concentration of 125 μg/ml, ox-LDL suppressed the growth of vSMCs. At concentrations of 25 μg/ml and 50 μg/ml, ox-LDL presented powerful mitotic trigger. When the concentration of ox-LDL increased, the mitotic index of vSMCs decreased gradually. ox-LDL induced more foam cells from vSMCs with rich intracellular lipid accumulation at concentrations of 25 μg/ml and 50 μg/ml. ox-LDL at higher concentrations induced more apoptotic vSMCs.Conclusions ox-LDL at lower concentrations may trigger proliferation and phenotype alteration into foam cells of vSMCs, and at higher concentrations it may induce apoptosis in vSMCs. ox-LDL plays an important role in the pathogenesis and development of atherosclerosis by its effect on vSMCs proliferation, phenotype alteration and apoptosis. | ZHAO Gao-feng SENG Jing jing ZHANG Hua SHE Ming-peng | 2005 | Chinese Medical Journal2005,,23: | 14 |

| 4 | Epidemiological investigation of Helicobacter pylori infection in elderly people in Beijing显示文摘BACKGROUND The Helicobacter pylori(H.pylori)infection rate in China is approximately 50%.H.pylori is a pathogenic factor of peptic ulcer and chronic gastritis.In addition,H.pylori infection may also be associated with a variety of cardiovascular diseases in elderly people,such as arteriosclerosis,coronary heart disease,and cerebral infarction,having deleterious effect on their health.With the aging of the population,the disease characteristics of the elderly population have been increasingly valued by the whole society.We conducted an epidemiological survey of H.pylori infection among elderly people in Beijing to provide a basis for health management of H.pylori infection.AIM To understand the epidemiological characteristics of H.pylori infection in elderly people in Beijing.METHODS A total of 1090 elderly people aged more than 60 years from different parts of Beijing(urban and rural areas)were selected using the random cluster sampling method.Structured questionnaires were completed during home visits and the 13C-urea breath test was conducted for H.pylori detection.RESULTS The prevalence of H.pylori infection was 46.5%(507/1090).The infection rate in men was 51.8%,which was significantly higher than that in women(42.5%;P<0.05).The H.pylori infection rate in illiterate people was significantly higher than that in literate persons(53.5%vs 44.8%,P<0.05).The total infection rate of H.pylori gradually increased with age and the difference was statistically significant(P<0.01).The H.pylori infection rate in smokers was significantly higher than that in non-smokers and those who had quit smoking(P<0.05).CONCLUSION The prevalence of H.pylori infection among elderly people is 46.5%and the infection rate gradually increases with age.Sex,education level,age,and smoking were determined to be H.pylori infection risk factors.The relationship of H.pylori infection with region,occupation,drinking,and diet structure needs to be further studied. | Hong-Ming Zhu Bang-Yi Li Zhe Tang Jing She Xue-Ying Liang Li-Kou Dong Mei Zhang | 2020 | World Journal of Clinical Cases2020,8,11: | 11 |

| 8 | NADPH is superior to NADH or edaravone in ameliorating metabolic disturbance and brain injury in ischemic stroke显示文摘Our previous studies confirm that exogenous reduced nicotinamide adenine dinucleotide phosphate(NADPH)exerts a neuroprotective effect in animal models of ischemic stroke,and its primary mechanism is related to anti-oxidative stress and improved energy metabolism.However,it is unknown whether nicotinamide adenine dinucleotide(NADH)also plays a neuroprotective role and whether NADPH is superior to NADH against ischemic stroke?In this study we compared the efficacy of NADH,NADPH,and edaravone in ameliorating brain injury and metabolic stress in ischemic stroke.Transient middle cerebral artery occlusion/reperfusion(t-MCAO/R)mouse model and in vitro oxygen glucose deprivation/reoxygenation(OGD/R)model were established.The mice were intravenously administered the optimal dose of NADPH(7.5 mg/kg),NADH(22.5 mg/kg),or edaravone(3 mg/kg)immediately after reperfusion.We showed that the overall efficacy of NADPH in ameliorating ischemic injury was superior to NADH and edaravone.NADPH had a longer therapeutic time window(within 5 h)after reperfusion than NADH and edaravone(within 2 h)for ischemic stroke.In addition,NADPH and edaravone were better in alleviating the brain atrophy,while NADH and NADPH were better in increasing the long-term survival rate.NADPH showed stronger antioxidant effects than NADH and edaravone;but NADH was the best in terms of maintaining energy metabolism.Taken together,this study demonstrates that NADPH exerts better neuroprotective effects against ischemic stroke than NADH and edaravone. | Xin-xin Wang Fan Wang Guang-hui Mao Jun-chao Wu Mei Li Rong Han Jing She Rong Zhang Rui Sheng Zhong Chen Zheng-hong Qin | 2022 | Acta Pharmacologica Sinica2022,43,3: | 4 |

| 11 | Room temperature preparation of highly stable cesium lead halide perovskite nanocrystals by ligand modification for white lightemitting diodes显示文摘The poor stability of halide perovskite nanocrystals(NCs)has severely hindered future practical application.Herein,we proposed a facile and effective ligand modification route to synthesize stable CsPbBr_(3) nanocrystals by introducing a double-terminal ligand,namely 4,4'-Azobis(4-cyanovalericacid)(CA),to replace the conventional oleic acid(OA)ligand at room temperature.The as-synthesized CsPbBr_(3)-CA not only possesses high photoluminescence quantum yield(72%)related to the reduced trap defects,but also shows significantly improved stability exposure to water,ethanol,light,and/or heat benefiting from the CA ligand anchored to NC surfaces tightly.The photoluminescence intensity of CsPbBr_(3)-CA maintains about 80%and 75%of its initial emission intensity after immersed in water or ethanol for 360 min,respectively,whereas that of the CsPbBr_(3)-OA was quenched completely within a few minutes.Moreover,an all-inorganic white light-emitting diode(LED)covered 126%National Television System Committee(NTSC)standard and 92%Rec.2020 standard was fabricated by combining the green CsPbBr_(3)-CA and commercial red-emitting K2SiF6:Mn4+(KSF)phosphors onto a blue LED chip.Thus,the presented work initiates the development of the room temperature preparation of high quality CsPbBr_(3) and shows prospect for next-generation displays. | Yu Zhang Guishun Li Changkun She Shaohua Liu Fangyu Yue Chengbin Jing Ya Cheng Junhao Chu | 2021 | Nano Research2021,14,8: | 3 |

| 13 | The CBP/p300 histone acetyltransferases function as plant-specific MEDIATOR subunits in Arabidopsis显示文摘In eukaryotes,MEDIATOR is a conserved multisubunit complex that links transcription factors and RNA polymerase II and that thereby facilitates transcriptional initiation.Although the composition of MEDIATOR has been well studied in yeast and mammals,relatively little is known about the composition of MEDIATOR in plants.By affinity purification followed by mass spectrometry,we identified 28 conserved MEDIATOR subunits in Arabidopsis thaliana,including putative MEDIATOR subunits that were not previously validated.Our results indicated that MED34,MED35,MED36,and MED37 are not Arabidopsis MEDIATOR subunits,as previously proposed.Our results also revealed that two homologous CBP/p300 histone acetyltransferases,HAC1 and HAC5(HAC1/5)are in fact plant-specific MEDIATOR subunits.The MEDIATOR subunits MED8 and MED25(MED8/25)are partially responsible for the association of MEDIATOR with HAC1/5,MED8/25 and HAC1/5 co-regulate gene expression and thereby affect flowering time and floral development.Our in vitro observations indicated that MED8 and HAC1 form liquid-like droplets by phase separation,and our in vivo observations indicated that these droplets co-localize in the nuclear bodies at a subset of nuclei.The formation of liquid-like droplets is required for MED8 to interact with RNA polymerase II.In summary,we have identified all of the components of Arabidopsis MEDIATOR and revealed the mechanism underlying the link of histone acetylation and transcriptional regulation. | Jing Guo Long Wei Shan-Shan Chen Xue-Wei Cai Yin-Na Su Lin Li She Chen Xin-Jian He | 2021 | Journal of Integrative Plant Biology2021,63,4: | 3 |

| 20 | Prophylactic juxtapapillary laser photocoagulation in pediatric morning glory syndrome显示文摘AIM:To determine the anatomic and visual outcomes of prophylactic juxtapapillary laser photocoagulation treatment alone in the prevention of retinal detachment(RD)in a cohort of pediatric patients diagnosed with morning glory syndrome(MGS).METHODS:A total of 24 eyes of 22 consecutive patients aged 0-15 y diagnosed with MGS treated with prophylactic juxtapapillary laser photocoagulation alone were reviewed.Data including demographics,ocular examination,anatomic and visual outcomes,following treatment and complications were collected.RESULTS:Two patients had bilateral laser treatment and 20 had monocular laser treatment.The age at treatment of 13(59.1%)patients was less than 12 mo.The presenting symptoms included strabismus(6/22,27.3%),decreased vision(2/22,9.1%),and routine fundus screening(14/22,63.6%).Fifteen(68.2%)patients underwent cranial magnetic resonance imaging(MRI)examinations,and 3 of those 15(20.0%)had abnormal findings in the nervous system.Based on preoperative wide-field fundus photography and B-scan echography,all(100.0%)eyes had no obvious RD.On postoperative 1 mo and 6 mo and the following follow-ups,the anatomic outcomes of all eyes remained stable.The mean follow-up duration was 27.7±17.5 mo.No severe complications were found.Preoperative visual acuity acquired from 2(9.1%)patients ranged from light perception to 20/200.Postoperative acuity acquired from 11(50.0%)patients ranged from light perception to 20/125.CONCLUSION:The preliminary anatomic and visual outcomes of prophylactic juxtapapillary laser treatment alone in pediatric MGS patients are relatively stable in a short-term follow-up.Further long-term clinical observation will be needed to confirm its efficacy and safety. | Yi-Hua Zou Kai-Qin She Jia-Ning Ren Ting-Yi Liang Ping Fei Yu Xu Jing Li Xiang Zhang Jie Peng Pei-Quan Zhao | 2022 | International Journal of Ophthalmology(English edition)2022,15,5: | 1 |
