| 1 | Mutation analysis of novel human liver-related putative tumor suppressor gene in hepatocellular carcinoma显示文摘AIM: To find the point mutations meaningful for inactivationof liver-related putative tumor suppressor gene (LPTS) gene,a human novel liver-related putative tumor suppressor geneand telomerase inhibitor in hepatocellular carcinoma.METHODS: The entire coding sequence of LPTS genewas examined for mutations by single strand conformationpolymorphism (SSCP) assay and PCR products directsequencing in 56 liver cancer cell lines, 7 ovarian cancerand 7 head & neck tumor cell lines and 70 pairs of HCCtissues samples. The cDNA fragment coding for the mostfrequent mutant protein was subcloned into GST fusionexpression vector. The product was expressed in E. coliand purified by glutathione-agarose column. Telomericrepeat amplification protocol (TRAP) assays wereperformed to study the effect of point mutation totelomerase inhibitory activity.RESULTS: SSCP gels showed the abnormal shifting bandsand DNA sequencing found that there were 5 differentmutations and/or polymorphisms in 12 tumor cell lineslocated at exon2, exon5 and exon7. The main alterationswere A(778)A/G and A(880)T in exon7. The change in siteof 778 could not be found in HCC tissue samples, while themutation in position 880 was seen in 7 (10 %) cases. Themutation in the site of 880 had no effect on telomeraseinhibitory activity.CONCLUSION: Alterations identified in this study arepolymorphisms of LPTS gene. LPTS mutations occur in HCCbut are infrequent and of little effect on the telomeraseinhibitory function of the protein. | 2 | Ghrelin protects myocardium from isoproterenol-induced injury in rats显示文摘AIM: To investigate the cardiac protective effects of ghrelin in rat with myocardial injury induced by isoproterenol (ISO). METHODS: Rats were subcutaneously injected ISO 40mg.kg^-1.d^-1 with or without ghrelin 1 or 10nmol.kg^-1.d^-1 for 2d. Hemodynamic parameters including mean arterial blood pressure and left ventricular pressure were measured at 12h after the last injection with ISO and/or ghrelin. Plasma lactate dehydrogenase (LDH) activity, plasma and myocardial contents of malondialdehyde (MDA), and conjugated diene were measured. Plasma ghrelin and endothelin-1 levels were assayed using radioimmunoassay methods. Endothelin-1 and ghrelin mRNA were determined using RT-PCR. RESULTS: About 45%(5/11) of rats after treatment with ISO alone died during experimental periods. However, no rats died after administration with ghrelin 10nmol.kg^-1.d^-1(0/11, P<0.05). Ghrelin also obviously ameliorated the hemodynamic disturbance in rats induced by ISO. The plasma LDH activity,contents of myocardial and plasma MDA, and conjugated diene level in plasma in ISO+G10nmol.kg^-1.d^-1 group were decreased by 28%, 34%, 73%, and 38% compared with those of ISO group (all P<0.01) respectively. ISO-induced endothelin-1 mRNA over-expression was inhibited and endothelin-1 level in plasma were inhibited by ghrelin 1 and 10nmol.kg^-1.d^-1. The ghrelin levels in plasma and ghrelin mRNA in myocardium were increased in the rats after injection of ISO. The plasma ghrelin level was further increased after ghrelin administration. | 3 | Blockade of L-type calcium channel in myocardium and calcium-induced contractions of vascular smooth muscle by CPU 86017显示文摘AIM: To assess the blockade by CPU 86017 on the L-type calcium channels in the myocardium and on the Ca2+- related contractions of vascular smooth muscle. METHODS: The whole-cell patch-clamp was applied to investi- gate the blocking effect of CPU 86017 on the L-type calcium current in isolated guinea pig myocytes and contrac- tions by KCl or phenylephrine (Phe) of the isolated rat tail arteries were measured. RESULTS: Suppression of the L-type current of the isolated myocytes by CPU 86017 was moderate, in time- and concentration-dependent man- ner and with no influence on the activation and inactivation curves. The IC50was 11.5 μmol/L. Suppressive effect of CPU 86017 on vaso-contractions induced by KCl 100 mmol/L, phenylephrine 1 μmol/Lin KH solution (phase 1), Ca2+ free KH solution ( phase 2) , and by addition of CaCl2 into Ca2+-free KH solution (phase 3) were observed. The IC50 to suppress vaso-contractions by calcium entry via the receptor operated channel (ROC) and voltage-depen- dent channel (VDC) was 0.324 μmol/L and 16.3 μmol/L, respectively. The relative potency of CPU 86017 to suppress vascular tone by Ca2+ entry through ROC and VDC is 1/187 of prazosin and 1/37 of verapamil, respectively. CONCLUSION: The blocking effects of CPU 86017 on the L-type calcium channel of myocardium and vessel are moderate and non-selective. | 4 | Taurine antagonized oxidative stress injury induced by homocysteine in rat vascular smooth muscle cells显示文摘AIM: To observe protective effects of taurine on reactive oxygen species generation induced by homocysteine in rat vascular smooth muscle cells (VSMC). METHODS: Rat VSMC was incubated with various concentrations of homocysteine and taurine. The lactate dehydrogenase (LDH) activity which released into culture medium was elevated as an indicator for VSMC injury. The reactive oxygen species (ROS) — hydrogen peroxide (H2O2) and superoxide anion (O2ˉ· )were measured with luminol or lucigenin chemiluminescences method, and the mitochondria Mn-superoxide dismutase (Mn-SOD) and catalase (CAT) were also measured in treated VSMC. RESULTS: LDH leakage from cultured VSMC treated with homocystenie, was increased (P<0.01 vs control), and it was markedly inhibited when co-incubated with taurine (P<0.01). Homocysteine induced H2O2generation from VSMC in a con- centration dependent manner (P<0.01 vs control). However, taurine (5, 10, and 20 mmol/L) significantly antago- nized 0.5 mmol/L homocysteine-induced H2O2generation in VSMC in a concentration dependent manner (P<0.01 vs homocysteine alone group), although taurine itself did not alter the H2O2generation in VSMC (P>0.05 vs control). In this study, the superoxide anion in VSMC was not detectable by chemiluminent method. In addition, treatment of VSMC with taurine increased mitochondria Mn-SOD and CAT activity in a concentration dependent manner (P<0.05), but homocysteine decreased mitochondria Mn-SOD and CAT activity (P<0.01 vs control). In addition, co-administration of taurine markedly ameliorated homocysteine-induced inhibition of Mn-SOD and CAT activity in VSMC (P<0.01 vs homocysteine alone group). | 5 | Eocene fossil Alseodaphne from Hainan Island of China and its paleoclimatic implications显示文摘Alseodaphne changchangensis sp. nov. (Lauraceae), with a perfectly preserved fossil leaf, was found in the Eocene Changchang Formation from the Changchang Basin of Hainan Island, China. This represents the earliest occurrence at the lowest latitude for the genus Alseodaphne, and offers important fossil evidence for further investigation of the origin and evolution of this genus and the paleo-climate of Hainan Island in the Eocene. Based on leaf morphology and cuticle characteristics, our fossil specimen is closest to the living species A. hainanensis Merrill, which is distributed mainly in tropical lowland rain forests, tropical montane rain forests, and subtropical montane evergreen broad-leafed forests on Hainan Island, and also found on the Wanshan Islands of Zhuhai of Guangdong Province and in northern parts of Vietnam. In the above areas, the climate factors are similar, i.e., the mean annual temperature is 20-22.6℃, the mean annual range of temperature 12-12.6℃ and the mean annual precipitation 1784-2500 mm. | 6 | Expression of gonadotropin-releasing hormone receptor and effect of gonadotropin-releasing hormone analogue on proliferation of cultured gastric smooth muscle cells of rats显示文摘AIM: To investigate the expression of gonadotropin-releasing hormone (GnRH) receptor and the effects of GnRH analog(alarelin) on proliferation of cultured gastric smooth muscle cells (GSMC) of rats. METHODS: Immunohistochemical ABC methods and in situ hybridization methods were used to dectect protein and mRNA expression of GnRH receptor in GSMC, respectively. Techniques of cell culture, OD value of MTT test, measure of ^3H-TdR incorporation, average fluorescent values of proliferating cell nuclear antigen (PCNA) and flow cytometric DNA analysis were used in the experiment. RESULTS: The cultured GSMC of rats showed immunoreactivity for GnRH receptor; positive staining was located in cytoplasm. GnRH receptor mRNA hybridized signals were also detected in cytoplasm. When alarelin (10^-9, 10^-7, 10^-5 mol/L) was administered into the medium and incubated for 24 h, OD value of MTT, ^3H-TdR incorporation and average fluorescent values of PCNA all decreased significantly as compared with the control group (P<0.05). The maximum inhibitory effect on cell proliferation was achieved a concentration of 10s mol/L and it acted in a dose-dependent manner. Flow cytometric DNA analysis revealed that alarelin could significantly enhance ratio of G1 phase and decrease ratio of S phase of GSMC of rats (P<0.05).The maximum inhibitory effect on ratio of S phase was at the concentration of 10-s mol/L and also acted in a dose-dependent manner. CONCLUSION: Our data suggest that GnRH receptor can be expressed by GSMC of rats. | 9 | Taurine inhibits ischemia/reperfusion-induced compartment syndrome in rabbits显示文摘Aim: To investigate effects of taurine on ischemia/reperfusion (I/R)-induced compartment syndrome in rabbit hind limbs. Methods: Rabbits underwent femoral artery occlusion after ligation of branches from terminal aorta to femoral artery. After a 7-h ischemia, reperfusion was established with the use of heparinized polyethylene shunts. Rabbits received taurine (1 g/kg) or normal saline (control) by iv infusion 10 min before shunt placement. During reperfusion, anterior compartment pressure (ACP) was monitored continuously in the left lower extremity. Gastrocnemius muscle triphenyltetrazolium chloride (TTC) level, taurine content and myeloperoxidase activity were assayed. Oxidative stress was induced in the in vitro gastrocnemius muscle slices by free radical generating systems (FRGS), and the malondialdehyde content was measured in presence or absence of taurine. Results: After 7 h of ischemia, none of the parameters that we measured were different from those before ischemia, except that TTC reduction decreased by 80%. In the control group, after 2 h of reperfusion, ACP increased 4.5-fold, and gastrocnemius muscle taurine content was reduced by 33%. In taurine-treated animals, at 2 h reperfusion, the mean arterial blood pressure and heart rate were increased, by 6% and 10%. ACP decreased by 39%, muscle edema decreased by 16%, TTC reduction increased by 150%, and lactate dehydrogenase decreased by 36% compared to control group. Plasma and muscle taurine content increased by 70% and 88%, respectively. In the taurine-treated group, at 2 h reperfusion, plasma malondialdehyde and conjugated diene content were decreased by 38% and 23%, respectively, and muscle malondialdehyde and conjugated diene content decreased by 22% and 30%, respectively compared to the control group. At 2 h reperfusion, myeloperoxidase activity was increased 3.5-fold in control animals. In the in vitro study, taurine decreased malondialdehyde content in muscle slices incubate dwith hypochlorous acid in a dose-dependent manner, but there was no change when incubated with hydrogen peroxide and xanthine oxidase. For realistic DSSs with clusters, while repairing a failed node, downloading more data from intra-cluster nodes than from cross-cluster nodes is effective. Therefore, differentiating the repair bandwidth from intra-cluster and cross-cluster is useful. For cluster DSSs, the tradeoff is considered with special repair assumptions where all alive nodes are used for repairing a failed node. In this paper, we investigate the optimal tradeoff for the cluster DSSs under more general storage/repair parameters. Furthermore, we propose a regenerating code construction strategy that achieves the points in the optimal tradeoff curve for the cluster DSSs with specific parameters as a numerical example.
