| 1 | Ghrelin protects myocardium from isoproterenol-induced injury in rats显示文摘AIM: To investigate the cardiac protective effects of ghrelin in rat with myocardial injury induced by isoproterenol (ISO). METHODS: Rats were subcutaneously injected ISO 40mg.kg^-1.d^-1 with or without ghrelin 1 or 10nmol.kg^-1.d^-1 for 2d. Hemodynamic parameters including mean arterial blood pressure and left ventricular pressure were measured at 12h after the last injection with ISO and/or ghrelin. Plasma lactate dehydrogenase (LDH) activity, plasma and myocardial contents of malondialdehyde (MDA), and conjugated diene were measured. Plasma ghrelin and endothelin-1 levels were assayed using radioimmunoassay methods. Endothelin-1 and ghrelin mRNA were determined using RT-PCR. RESULTS: About 45%(5/11) of rats after treatment with ISO alone died during experimental periods. However, no rats died after administration with ghrelin 10nmol.kg^-1.d^-1(0/11, P<0.05). Ghrelin also obviously ameliorated the hemodynamic disturbance in rats induced by ISO. The plasma LDH activity,contents of myocardial and plasma MDA, and conjugated diene level in plasma in ISO+G10nmol.kg^-1.d^-1 group were decreased by 28%, 34%, 73%, and 38% compared with those of ISO group (all P<0.01) respectively. ISO-induced endothelin-1 mRNA over-expression was inhibited and endothelin-1 level in plasma were inhibited by ghrelin 1 and 10nmol.kg^-1.d^-1. The ghrelin levels in plasma and ghrelin mRNA in myocardium were increased in the rats after injection of ISO. The plasma ghrelin level was further increased after ghrelin administration. | 2 | Taurine antagonized oxidative stress injury induced by homocysteine in rat vascular smooth muscle cells显示文摘AIM: To observe protective effects of taurine on reactive oxygen species generation induced by homocysteine in rat vascular smooth muscle cells (VSMC). METHODS: Rat VSMC was incubated with various concentrations of homocysteine and taurine. The lactate dehydrogenase (LDH) activity which released into culture medium was elevated as an indicator for VSMC injury. The reactive oxygen species (ROS) — hydrogen peroxide (H2O2) and superoxide anion (O2ˉ· )were measured with luminol or lucigenin chemiluminescences method, and the mitochondria Mn-superoxide dismutase (Mn-SOD) and catalase (CAT) were also measured in treated VSMC. RESULTS: LDH leakage from cultured VSMC treated with homocystenie, was increased (P<0.01 vs control), and it was markedly inhibited when co-incubated with taurine (P<0.01). Homocysteine induced H2O2generation from VSMC in a con- centration dependent manner (P<0.01 vs control). However, taurine (5, 10, and 20 mmol/L) significantly antago- nized 0.5 mmol/L homocysteine-induced H2O2generation in VSMC in a concentration dependent manner (P<0.01 vs homocysteine alone group), although taurine itself did not alter the H2O2generation in VSMC (P>0.05 vs control). In this study, the superoxide anion in VSMC was not detectable by chemiluminent method. In addition, treatment of VSMC with taurine increased mitochondria Mn-SOD and CAT activity in a concentration dependent manner (P<0.05), but homocysteine decreased mitochondria Mn-SOD and CAT activity (P<0.01 vs control). In addition, co-administration of taurine markedly ameliorated homocysteine-induced inhibition of Mn-SOD and CAT activity in VSMC (P<0.01 vs homocysteine alone group). | 4 | Protective role of metallothionein in stress-induced gastric ulcer in rats显示文摘AIM: To illustrate the pathophysiological role of metallothionein (MT) in gastric ulcer induced by stress. METHODS: Wistar rats underwent water-immersionrestraint (WIR) stress, ZnSO4 (an MT inducer) treatment, WIR+ZnSO4 or WIR+MT, and the ulcer index (UI) was estimated in excised stomach and liver tissues. The mRNA level of gastric MT was determined by semi-quantitative RT-PCR. The MT content in gastric and hepatic tissues was determined by Cd/hemoglobin affinity assay. The lipid peroxidation products malondialdehyde (MDA) and conjugated dienes (CD) were estimated by use of thiobarbituric acid reactive species and ultraviolet spectrophotometry. RESULTS: WIR stress induced severe gastric mucosal lesions in rats. Compared with control rats, stressed rats had increased lipid peroxide content in serum and stomach and liver tissues. MDA content was increased by 34%, 21% and 29% and CD level by 270%, 83% and 28%, respectively. MT content in the stomach and liver was increased by 0.74- and 1.8-fold, and the MT-mRNA level in the stomach was increased by 26%. Pretreatment with ZnSO4 prevented gastric lesion development (the UI was 87% lower than that without pretreatment), and the MDA and CD content in serum and tissues was lower. The MT content in the liver was double in rats that were not pretreated, and the MT mRNA level in the stomach was 35% higher. MT administration 1 h before the WIR stress prevented gastric lesion development (the UI decreased by 47% compared with that in rats not pretreated), and the MDA and CD content in serum and tissues was significantly lower. CONCLUSION: In WIR-stressed rats, the MT level was increased in serum and in stomach and liver tissues. Pre-administration of exogenous MT or pre-induction of endogenous MT can protect the gastric mucosa against stress-induced ulcers and inhibits the formation of stressinduced lipid peroxide. | 6 | Optical and Radiative Properties of Aerosols during a Severe Haze Episode over the North China Plain in December 2016显示文摘The optical and radiative properties of aerosols during a severe haze episode from 15 to 22 December 2016 over Beijing, Shijiazhuang, and Jiaozuo in the North China Plain were analyzed based on the ground-based and satellite data, meteorological observations, and atmospheric environmental monitoring data. The aerosol optical depth at 500 nm was < 0.30 and increased to > 1.4 as the haze pollution developed. The ?ngstr?m exponent was > 0.80 for most of the study period. The daily single-scattering albedo was > 0.85 over all of the North China Plain on the most polluted days and was > 0.97 on some particular days. The volumes of fine and coarse mode particles during the haze event were approximately 0.05–0.21 and 0.01–0.43 μm3, respectively—that is, larger than those in the time without haze. The daily absorption aerosol optical depth was about 0.01–0.11 in Beijing, 0.01–0.13 in Shijiazhuang, and0.01–0.04 in Jiaozuo, and the average absorption ?ngstr?m exponent varied between 0.6 and 2.0. The aerosol radiative forcing at the bottom of the atmosphere varied from –23 to –227, –34 to –199, and –29 to –191 W m–2 for the whole haze period, while the aerosol radiative forcing at the top of the atmosphere varied from –4 to –98, –10 to –51,and –21 to –143 W m–2 in Beijing, Shijiazhuang, and Jiaozuo, respectively. Satellite observations showed that smoke,polluted dust, and polluted continental components of aerosols may aggravate air pollution during haze episodes. | 10 | Spatial and temporal distribution of the cloud optical depth over China based on MODIS satellite data during 2003–2016显示文摘The cloud optical depth(COD) is one of the important parameters used to characterize atmospheric clouds. We analyzed the seasonal variations in the COD over East Asia in 2011 using cloud mode data from the AERONET(Aerosol Robotic Network) ground-based observational network. The applicability of the MODIS(Moderate Resolution Imaging Spectroradiometer) COD product was verified and compared with the AERONET cloud mode dataset. There was a good correlation between the AERONET and the MODIS. The spatial and temporal distribution and trends in the COD over China were then analyzed using MODIS satellite data from 2003 to 2016. The seasonal changes in the AERONET data and the time sequence variation of the satellite data suggest that the seasonal variations in the COD are significant. The result shows that the COD first decreases and then increases with the season in northern China, and reaches the maximum in summer and minimum in winter.However, the spatial distribution change is just the opposite in southern China. The spatial variation trend shows the COD in China decreases first with time and gradually increases after 2014. And the trend of COD in the western and central China is consistent with that in China.
