| 5 | Targeted PERK inhibition with biomimetic nanoclusters confers preventative and interventional benefits to elastase-induced abdominal aortic aneurysms显示文摘Abdominal aortic aneurysm(AAA)is a progressive aortic dilatation,causing~80%mortality upon rupture.Currently,there is no approved drug therapy for AAA.Surgical repairs are invasive and risky and thus not recommended to patients with small AAAs which,however,account for~90%of the newly diagnosed cases.It is therefore a compelling unmet clinical need to discover effective non-invasive strategies to prevent or slow down AAA progression.We contend that the first AAA drug therapy will only arise through discoveries of both effective drug targets and innovative delivery methods.There is substantial evidence that degenerative smooth muscle cells(SMCs)orchestrate AAA pathogenesis and progression.In this study,we made an exciting finding that PERK,the endoplasmic reticulum(ER)stress Protein Kinase R-like ER Kinase,is a potent driver of SMC degeneration and hence a potential therapeutic target.Indeed,local knockdown of PERK in elastase-challenged aorta significantly attenuated AAA lesions in vivo.In parallel,we also conceived a biomimetic nanocluster(NC)design uniquely tailored to AAA-targeting drug delivery.This NC demonstrated excellent AAA homing via a platelet-derived biomembrane coating;and when loaded with a selective PERK inhibitor(PERKi,GSK2656157),the NC therapy conferred remarkable benefits in both preventing aneurysm development and halting the progression of pre-existing aneurysmal lesions in two distinct rodent models of AAA.In summary,our current study not only establishes a new intervention target for mitigating SMC degeneration and aneurysmal pathogenesis,but also provides a powerful tool to facilitate the development of effective drug therapy of AAA. | Nisakorn Yodsanit Takuro Shirasu Yitao Huang Li Yin Zain Husain Islam Alexander Christopher Gregg Alessandra Marie Riccio Runze Tang Eric William Kent Yuyuan Wang Ruosen Xie Yi Zhao Mingzhou Ye Jingcheng Zhu Yi Huang Nicholas Hoyt Mengxue Zhang John A.Hossack Morgan Salmon K.Craig Kent Lian-Wang Guo Shaoqin Gong Bowen Wang | 2023 | Bioactive Materials2023,,8: | 0 |
| 6 | Water-in-Water Emulsions,Ultralow Interfacial Tension,and Biolubrication显示文摘A water-in-water(W/W)emulsion consists of droplets formed by the spontaneous liquid-liquid separation of two immiscible aqueous phases.The inherent properties of the W/W interfaces,low or ultralowinterfacial tension(γ_(W/W)=1-1000μN/m)and a large thickness of several nanometers,beget the poor inherent stability of emulsions.Herein,we report a nanofibril emulsifier having Schiff base reactivity to generate a W/W emulsion.The W/W emulsion has superior stability(stable>6 months)because collagen nanofibrils,acting as a stabilizer of W/W emulsions,can simultaneously satisfy the requirements of sizeandoverall coverage ratio of the phase interfaces.W/Wemulsions having γ_(W/W)∼10μN/m were used as synthetic synovial fluids,showing superior lubrication performance with a coefficient of friction in the range of 0.003-0.011,which has been demonstrated to be suitable for joint lubrication.An intraarticular injection assessment further confirmed this protective effect on articular cartilage in vivo.Our study reveals the mechanism of emulsion stabilization and opens up the possibility of osteoarthritis(OA)treatment using the biolubrication effects of W/W emulsions for lubricated artificial implant surfaces. | Yitong Wang Jin Yuan Yunpeng Zhao Ling Wang Luxuan Guo Lei Feng Jiwei Cui Shuli Dong Shanhong Wan Weimin Liu Heinz Hoffmann Kiet Tieu Jingcheng Hao | 2022 | CCS Chemistry2022,4,6: | 0 |
| 7 | Retrieval of Winter Wheat Canopy Carotenoid Content with Ground-and Airborne-Based Hyperspectral Data显示文摘Accurate assessment of canopy carotenoid content(CC_(x+c)C)in crops is central to monitor physiological conditions in plants and vegetation stress,and consequently supporting agronomic decisions.However,due to the overlap of absorption peaks of carotenoid(C_(x+c))and chlorophyll(C_(a+b)),accurate estimation of carotenoid using reflectance where carotenoid absorb is challenging.The objective of present study was to assess CC_(x+c)C in winter wheat(Triticum aestivum L.)with ground-and aircraft-based hyperspectral measurements in the visible and near-infrared spectrum.In-situ hyperspectral reflectance were measured and airborne hyperspectral data were acquired during major growth stages of winter wheat in five consecutive field experiments.At the canopy level,a remarkable linear relationship(R^(2)=0.95,p<0.001)existed between C_(x+c) and Ca+b,and correlation between CC_(x+c)C and wavelengths within 400 to 1000 nm range indicated that CC_(x+c)C could be estimated using reflectance ranging from visible to near-infrared wavebands.Results of Cx+c assessment based on chlorophyll and carotenoid indices showed that red edge chlorophyll index(CI red edge)performed with the highest accuracy(R^(2)=0.77,RMSE=22.27μg/cm^(2),MAE=4.97μg/cm^(2)).Applying partial least square regression(PLSR)in CC_(x+c)C retrieval emphasized the significance of reflectance within 700 to 750 nm range in CC_(x+c)C assessment.Based on CI red edge index,use of airborne hyperspectral imagery achieved satisfactory results in mapping the spatial distribution of CC_(x+c)C.This study demonstrates that it is feasible to accurately assess CC_(x+c)C in winter wheat with red edge chlorophyll index provided that C_(x+c) correlated well with C_(a+b) at the canopy scale.it is therefore a promising method for CC_(x+c)C retrieval at regional scale from aerial hyperspectral imagery. | Ting Cui Xianfeng Zhou Yufeng Huang Yanting Guo Yunrui Lin Leyi Song Jingcheng Zhang | 2023 | Phyton-International Journal of Experimental Botany2023,92,9: | 0 |
| 8 | Uncovering the magnetic response of open-shell graphene nanostructures on metallic surfaces at different doping levels显示文摘Open-shell graphene nanostructures(GNs)are promising candidates for future spintronics and quantum technologies.Recent progress based on on-surface synthetic approach has successfully created such GNs on metallic surfaces.Meanwhile,the doping effect of metallic surfaces is inevitably present and can significantly tune their electronic and magnetic properties.Here,we investigate the zigzag end states of open-shell 7-armchair graphene nanoribbons(7-AGNRs)on Au(111),Au(100)and Ag(111)surfaces.Combined with the manipulation of a scanning tunneling microscope,we demonstrate that the end states can be tuned from empty states to singly occupied states and to doubly occupied states by substrate doping.Furthermore,the singly occupied states can be finely tuned,with the occupancy number of the states and related magnetic behaviors uncovered by experiments at different temperatures and magnetic fields.Our results provide a comprehensive study of the magnetic response of open-shell GNs on metallic surfaces at different doping levels. | Zengfu Ou Jun Wang Jihai Zhang Yukang Ding Shenwei Chen Wenya Zhai Jingcheng Li Dingyong Zhong Donghui Guo | 2024 | Science China(Physics,Mechanics & Astronomy)2024,67,2: | 0 |
