| 2 | Mechanisms and Effects on HBV Replication of the Interaction between HBV Core Protein and Cellular Filamin B显示文摘Hepatitis B virus(HBV) infection is one of the major problems that threatens global health. There have been many studies on HBV, but the relationship between HBV and host factors is largely unexplored and more studies are needed to clarify these interactions. Filamin B is an actin-binding protein that acts as a cytoskeleton protein, and it is involved in cell development and several signaling pathways. In this study, we showed that filamin B interacted with HBV core protein,and the interaction promoted HBV replication. The interaction between filamin B and core protein was observed in HEK293T, Huh7 and HepG2 cell lines by co-immunoprecipitation and co-localization immnofluoresence. Overexpression of filamin B increased the levels of HBV total RNAs and pre-genome RNA(pg RNA), and improved the secretion level of hepatitis B surface antigen(HBsAg) and hepatitis B e antigen(HBeAg). In contrast, filamin B knockdown inhibited HBV replication, decreased the level of HBV total RNAs and pgRNA, and reduced the secretion level of HBsAg and HBeAg. In addition, we found that filamin B and core protein may interact with each other via four blocks of argentine residues at the C-terminus of core protein. In conclusion, we identify filamin B as a novel host factor that can interact with core protein to promote HBV replication in hepatocytes. | 3 | Spectrofluorimetric analysis of captopril based on its obstruction effect of the nanomaterial surface energy transfer between acridine orange and gold nanoparticles显示文摘A simple and sensitive method for detection of captopril was established based on its obstructive effect on nanomaterial surface energy transfer(NSET). It was found that the acridine orange(AO) could be adsorbed onto the surface of citrated-gold nanoparticles(Au NPs) through electrostatic interaction. Incidentally, the fluorescence of AO was quenched owing to the dipole-dipole interaction of NSET between AO fluorophore and the Au NPs. However, captopril could obstruct the occurrence of NSET between AO and Au NPs effectively with the formation of Au-S covalent bonds between it and the Au NPs. Consequently, AO molecules were moved away from the surface of Au NPs leading to a decline of the energy transfer efficiency. Moreover, the fluorescence of AO could be gradually restored with the addition of captopril. Under the optimal conditions, the recovered fluorescence intensity correlated linearly with the concentration of captopril in the range of 400 nmol/L–2.0 μmol/L with a detection limit of 71 nmol/L. Besides, the proposed method was successfully applied for the detection of captopril in troches with the recovery of 93%–102% and the RSD lower than 2.24%. | 4 | Rice FLOURY ENDOSPERM 18 encodes a pentatricopeptide repeat protein required for 5′ processing of mitochondrial nad5 messenger RNA and endosperm development显示文摘Pentatricopeptide repeat(PPR) proteins, composing one of the largest protein families in plants,are involved in RNA binding and regulation of organelle RNA metabolism at the posttranscriptional level. Although several PPR proteins have been implicated in endosperm development in rice(Oryza sativa), the molecular functions of many PPRs remain obscure. Here, we identified a rice endosperm mutant named floury endosperm 18(flo18) with pleiotropic defects in both reproductive and vegetative development.Map-based cloning and complementation tests showed that FLO18 encodes a mitochondriontargeted P-type PPR protein with 15 PPR motifs.Mitochondrial function was disrupted in the flo18 mutant, as evidenced by decreased assembly of Complex I in the mitochondrial electron transport chain and altered mitochondrial morphology. Loss of FLO18 function resulted in defective 5′-end processing of mitochondrial nad5 transcripts encoding subunit 5 of nicotinamide adenine dinucleotide hydrogenase. | 5 | Status of Stratigraphy Research in China显示文摘Scientific research and productive practice for earth history are inseparable from the accurate stratigraphic framework and time framework. Establishing the globally unified, precise and reliable chronostratigraphic series and geological time series is the major goal of the International Commission on Stratigraphy(ICS). Under the leadership of the ICS, the countries around the world have carried out research on the Global Standard Stratotype-section and Points(GSSPs) for the boundaries of chronostratigraphic systems. In the current International Chronostratigraphic Chart(ICC), 65 GSSPs have been erected in the Phanerozoic Eonothem, and one has yet been erected in the Precambrian Eonothem. Based on the progress of research on stratigraphy especially that from its subcommissions, the ICS is constantly revising the ICC, and will publish a new International Stratigraphic Guide in 2020. After continual efforts and broad international cooperation of Chinese stratigraphers, 10 GSSPs within the Phanerozoic Eonothem have been approved and ratified to erect in China by the ICS and IUGS. To establish the standards for stratigraphic division and correlation of China, with the support from the Ministry of Science and Technology, the National Natural Science Foundation of China and the China Geological Survey, Chinese stratigraphers have carried out research on the establishment of Stages in China. A total of 102 stages have been defined in the 'Regional Chronostratigraphic Chart of China(geologic time)', in which 59 stages were studied in depth. In 2014, the 'Stratigraphic Chart of China' was compiled, with the essential contents as follows: the correlation between international chronostratigraphy and regional chronostratigraphy of China(geologic time), the distributive status of lithostratigraphy, the characteristics of geological ages, the biostratigraphic sequence, the magnetostratigraphy, the geological events and eustatic sea-level change during every geological stage. The 'Stratigraphical Guide of China and its Explanation(2014)' was also published. Chinese stratigraphers have paid much attention to stratigraphic research in south China, northeast China, north China and northwest China and they have made great achievements in special research on stratigraphy, based on the 1:1000000, 1:250000, 1:200000 and 1:50000 regional geological survey projects. Manifold new stratigraphic units were discovered and established by the regional geological surveys, which are helpful to improve the regional chronostratigraphic series of China. On the strength of the investigation in coastal and offshore areas, the status of marine strata in China has been expounded. According to the developing situation of international stratigraphy and the characteristics of Chinese stratigraphic work, the contrast relation between regional stratigraphic units of China and GSSPs will be established in the future, which will improve the application value of GSSPs and the standard of regional stratigraphic division and correlation. In addition, the study of stratigraphy of the Precambrian, terrestrial basins and orogenic belts will be strengthened, the Stratigraphic Chart of China will be improved, the typical stratigraphic sections in China will be protected and the applied study of stratigraphy in the fields of oil and gas, solid minerals, etc. will be promoted. | 6 | Solid-phase impregnation promotes Ce doping in TiO_(2) for boosted denitration of CeO_(2)/TiO_(2) catalysts显示文摘CeO_(2)/TiO_(2)(denoted as Ce Ti) catalysts obtained by solid-phase impregnation behaved better in lowtemperature selective catalytic reduction of NO_(x)with NH_(3)(NH_(3)-SCR) than that by conventional wet impregnation.To explore the main factors for activity distinction,the texture property,CeO_(2)dispersion and structure changes of TiO_(2)were comprehensively analyzed.It was found that surface changes of TiO_(2)had a significant impact on the improved activity.From results of inductively coupled plasma atomic emission spectrometer (ICP-AES),diffuse reflectance UV-vis spectroscopy (UV-vis-DRS) and Raman,it was inferred that Ce ions were partially incorporated into TiO_(2)lattice,accompanied with the formation of defects and vacancies during solid-phase impregnation.Accordingly,Ce Ti catalysts from solid-phase impregnation exhibited superiority in adsorption and activation of reactants.Further result from monitoring the preparation process indicated that the evolved NO played an important role in promoting Ce doping through depriving oxygen atoms on TiO_(2)surface.The interaction between Ce and Ti was enhanced.The catalyst performed better in NH_(3)-SCR,especially at low temperature,which testified the solid-phase impregnation could be an effective method to modulate interface structure for designing efficient catalyst. | Wang Song Jiawei Ji Kai Guo Xin Wang Xiaoqian Wei Yandi Cai Wei Tan Lulu Li Jingfang Sun Changjin Tang Lin Dong | 2022 | Chinese Chemical Letters2022,33,2: | 0 |
