1Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 2High-Resolution Genome-wide Association Study Identifies Genomic Regions and Candidate Genes for Important Agronomic Traits in Wheat显示文摘Wheat(Triticum aestivum)is a major staple food crop worldwide.Genetic dissection of important agronomic traits is essential for continuous improvement of wheat yield to meet the demand of the world's growing population.We conducted a large-scale genome-wide association study(GWAS)using a panel of 768 wheat cultivars that were genotyped with 327609 single-nucleotide polymorphisms generated by genotyping-by-sequencing and detected 395 quantitative trait loci(QTLs)for 12 traits under 7 environments.Among them,273 QTLs were delimited to≤1.0-Mb intervals and 7 of them are either known genes(Rht-D,Vrn-B1,and Vrn-D1)that have been cloned or known QTLs(TaGA2ox8,APO1,TaSus1-7B,and Rht12)that were previously mapped.Eight putative candidate genes were identified for three QTLs that enhance spike seed setting and grain size using gene expression data and were validated in three bi-parental populations.Protein sequence analysis identified 33 putative wheat orthologs that have high identity with rice genes in QTLs affecting similar traits.Large r^2 values for additive effects observed among the QTLs for most traits indicated that the phenotypes of these identified QTLs were highly predictable.Results from this study demonstrated that significantly increasing GWAS population size and marker density greatly improves detection and identification of candidate genes underlying a QTL,solidifying the foundation for large-scale QTL fine mapping,candidate gene validation,and developing functional markers for genomics-based breeding in wheat.Yunlong Pang Chunxia Liu Danfeng Wang Paul St.Amand Amy Bernardo Wenhui Li Fang He Linzhi Li Liming Wang Xiufang Yuan Lei Dong Yu Su Huirui Zhang Meng Zhao Yunlong Liangi Hongze Jia Xitong Shen Yue Lu Hongming Jiang Yuye Wu Anfei Li Honggang Wang Lingrang Kong Guihua Bai Shubing Liu 2020Molecular Plant2020,13,9:8

4Selective depression action of taurine in flotation separation of specularite and chlorite显示文摘Chlorite,as the most representative gangue mineral associated with specularite,of which the separation of these two minerals is difficult.This paper investigated the depression effect of taurine on specularite/chlorite separation via flotation experiments,adsorption tests,contact angle measurements,Zeta potential detection,FT-IR measurements,and XPS analyses.The results of single mineral flotation indicated that chlorite could be depressed selectively by taurine with the recovery of less than 30%,but the floatability of specularite remains high with recovery rate of 81.77%at pH 10.The artificial mixed mineral flotation results confirmed the effectiveness of taurine as a depressant.Surface adsorption,contact angle,and Zeta potential detection revealed taurine primarily adsorbs on the chlorite surface,which hampered the DDA’s subsequent adsorption and results in the chlorite’s poor floatability.The FT-IR spectra and XPS analyses provided further proof that taurine adsorbed on chlorite surface as an electron donor,and part of the electrons transferred from the sulfonic acid group of taurine to metal ions during the adsorption process.In addition,the hydrogen bond between amino-group of taurine and O ions in chlorite surface was also formed in the adsorption process.Finally,optimized adsorption configurations of taurine on chlorite surfaces were proposed.Mingyang Li Cheng Yang Zhaoyang Wu Xiangpeng Gao Xiong Tong Xiankun Yu Hongming Long 2022International Journal of Mining Science and Technology2022,32,3:4

6Current Density Distribution in Double-Sided GTAW Process显示文摘A three dimensional current density distribution in double-sided gas tungsten arc welding (GTAW) proces is performed. The current density distribution field is evaluated by numerically solving Maxwell equations in the domain of the workpiece. In the boundary condition, the current density on. the workpiece surface is assumed as Gaussian distribution. 7Interface Behavior of Copper and Steel by Plasma-MIG Hybrid Arc Welding显示文摘Plasma-MIG hybrid arc welding is a hybrid heating source welding method which is composed of plasma arc and MIG arc. During Plasma-MIG hybrid arc welding process, the interface behavior of copper and steel dissimilar alloy is investigated. The results show that electromagnetic stirring effect decreases and the heat input increases with the increase of outer plasma current in the hybrid arc welding process. The interface diffusion and interface thickness is controlled by the tradeoff of electromagnetic stirring effect degradation and heat input increase. 8Tuning the properties of Ni-based catalyst via La incorporation for efficient hydrogenation of petroleum resin显示文摘The hydrogenation of petroleum resin(PR)is an effective process to prepare high value-added hydrogenated PR(HPR).However,the preparation of non-noble metal-based catalysts with high catalytic activity for PR hydrogenation still remains a challenge.Herein,a La promoted Ni-based catalyst is reported through the thermal reduction of quaternary Ni La Mg Al-layered double hydroxides(Ni La Mg Al-LDHs).The incorporation of La is beneficial to the reduction and stability of Ni particles with reduced particle size,and the increased alkalinity effectively mitigates the breakage of molecular chains of PR.As a result,the La promoted Ni-based catalyst exhibits high catalytic activity and excellent stability for PR hydrogenation.A hydrogenation degree of 95.4%and 96.1%can be achieved for HC_(5)PR and HC_(9) PR with less reduced softening point,respectively.Notably,the hydrogenation degree still maintains at 92.7%even after 100 hours’reaction,much better than that without La incorporation or prepared using conventional impregnation method.Qunhong Liu Jiangtao Yang Hongwei Zhang Hongming Sun Shuzheng Wu Bingqing Ge Rong Wang Pei Yuan 2022Chinese Journal of Chemical Engineering2022,35,5:2
9Rice FLOURY ENDOSPERM 18 encodes a pentatricopeptide repeat protein required for 5′ processing of mitochondrial nad5 messenger RNA and endosperm development显示文摘Pentatricopeptide repeat(PPR) proteins, composing one of the largest protein families in plants,are involved in RNA binding and regulation of organelle RNA metabolism at the posttranscriptional level. Although several PPR proteins have been implicated in endosperm development in rice(Oryza sativa), the molecular functions of many PPRs remain obscure. Here, we identified a rice endosperm mutant named floury endosperm 18(flo18) with pleiotropic defects in both reproductive and vegetative development.Map-based cloning and complementation tests showed that FLO18 encodes a mitochondriontargeted P-type PPR protein with 15 PPR motifs.Mitochondrial function was disrupted in the flo18 mutant, as evidenced by decreased assembly of Complex I in the mitochondrial electron transport chain and altered mitochondrial morphology. Loss of FLO18 function resulted in defective 5′-end processing of mitochondrial nad5 transcripts encoding subunit 5 of nicotinamide adenine dinucleotide hydrogenase. 10Nutritional evaluation of whole soybean curd made from different soybean materials based on amino acid profiles显示文摘Objectives:Amino acid profiles of whole soybean curds(WSCs)and conventional soybean curds(CSCs)were comparatively studied to 1.,evaluate their nutritional value and 2.,study the suitable soybean material to fabricate WSCs.Materials and Methods:Nine types of soybean materials were selected to make WSCs and CSCs.Protein digestibility-corrected amino acid score(PDCAAS)and principal component analysis were selected to perform the comparative analysis of amino acid profiles of the products.Results:Total contents of amino acids in WSCs and CSCs were 41.24-54.87 mg/g and 27.77-36.82 mg/g.Content ranges of essential amino acids were 13.59-19.38 mg/g and 8.46-11.76 mg/g for WSCs and CSCs.The essential amino acid pattern of WSCs was basically close to amino acid pattern spectrum requirements except valine and sulfur amino acids.As a limiting amino acid,methionine represented less than 0.4 for PDCAAS in all soybean curds except WSCs made from relay-cropping Nandou12 and Nandou25.Conclusions:The overall quality of amino acids in WSCs was higher than that in CSCs,indicating that WSCs indeed exhibited higher nutritional value than CSCs from the viewpoint of amino acid profile.WSCs with the high nutritional value could be made using Nandou25 and relay-cropping Nandou12 as materials.Meili Li Hongming Dong Dingtao Wu Hong Chen Wen Qin Weiguo Liu Wenyu Yang Qing Zhang 2020Food Quality and Safety2020,4,1:2

12Current Density Distribution in Gas Tungsten Arc Welding Process显示文摘Heat transfer and fluid flow in a weld pool had been simulated for several decades, which underwent from theoretically analysis to numerical solutions. Because of the complexity of the welding process, many ideal assumptions were made to simplify the question. This makes the solution somewhat not complied with the practical conditions. In previous papers, the current distribution in the weld pool is assumed as axi-symmetrical, and so does the electromagnetic forces, which plays an important role in determining the fluid flow field. Actually the current distribution is different from the assumption. In this paper; a three dimensional current distribution in Gas Tungsten Arc Welding (GTAW) process is performed. 13Experimental study on enhancement of the metastatic potential of portal vein tumor thrombus-originated hepatocellular carcinoma cells using portal vein serum显示文摘Objective: Portal vein metastasis of hepatocellular carcinoma(HCC) results in a poor prognosis and seriously affects the survival rate of patients. The mechanism underlying the formation of portal vein tumor thrombus(PVTT) is complex and is not yet fully understood. This study was conducted to investigate the impact of portal vein blood on the proliferation, metastasis, invasion and apoptosis of PVTT cells and to explore its possible mechanisms, which was expected to lay a foundation for ascertaining the mechanism underlying the portal vein metastasis of HCC.Methods: Peripheral blood and portal vein blood were collected from patients with HCC, and the sera from these two sources were used to culture the PVTT-originated HCC cell line CSQT-2. The cells were collected after 24 h, and flow cytometry was performed to detect cell proliferation, cell cycle stages and apoptosis. Transwell migration and invasion assays were applied to detect the metastasis and invasion of the cells in each group. The changes in the expression of MMP-2 and MMP-9 in cells were detected via Western blotting. The contents of IL-12, IFN-γ, IL-1β, IL-2 and TNF-α in the two groups of sera were quantified using corresponding kits. Results: Compared with the group of cells cultured with peripheral serum, the cells cultured with portal vein serum showed significantly lower apoptosis(P<0.01), significantly enhanced cell metastasis and invasion(P<0.01), whereas cell proliferation and the stages of the cell cycle did not differ significantly(P>0.05). A significantly increased expression level of MMP-2 has been observed in tumor cells treated portal vein serum. In addition, compared with peripheral serum, the content of IL-12 was significantly decreased in portal vein serum(P<0.05), while the contents of IFN-γ, IL-1β, IL-2, and TNF-α did not differ significantly(P>0.05). Conclusions: Portal vein serum from HCC patients could inhibit the apoptosis of PVTT-originated HCC cells and promote cell metastasis and invasion. 14Preparation of Nano-structured LiFe_xMn_(1-x)PO_4(x=0,0.2,0.4) by Reflux Method and Research on the Influences of Fe(Ⅱ) Substitution显示文摘Nano structured LiFexMn1-xPO4(x=0,0.2,0.4) materials were successfully prepared by one-step reflux method in a water/PEG400 mixed solvent, and were coated by carbon using glucose as the precursor. The materials were characterized by X-ray diffraction (XRD), field emission scanning electron microscopy (FESEM) and transmission electron microscopy (TEM). The electrochemical properties of the materials were investigated by galvanostatic cycling, cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). It was found that the materials consisted of nanorods with a diameter of 50 nm and a length of 500 nm. Galvanostatic cycling showed that the capacity of LiMnPO 4 could be largely increased by Fe2+ substitution. At a current rate of C/20, the capacity of the three samples (x=0,0.2,0.4) were 47, 107 and 150 mA·h·g-1 , respectively. CV result showed that the Fe2+ substitution could decrease the polarization during charging/discharging, accelerating the electrochemical process. 17Behavior of Corrosion-Repaired Concrete Beams Reinforced by Epoxy Mortar显示文摘The performance of concrete beams repaired with epoxy mortar was investigated by constructing twelve beam specimens.All the beam specimens were subjected to a constant current for accelerated corrosion.Six specimens were corroded without subsequent reparation as a control group,and the other six beam specimens were corroded and repaired utilizing epoxy mortar.All specimens were tested to failure.During test process,we focused on the failure pattern of beam specimens,structural cracks,mid-deflections,bearing capacity,and probed into the influence of corrosion degree and repair of epoxy mortar on the performance of beam specimens.It was observed that corrosion-repaired beams in the loading test were in a bending failure pattern.It is obvious that cracking loads and bending stiffness of repaired beams and corrosion-repaired beams were larger than those of unrepaired beams and secondly-corroded beams.When the mass loss of main steel bars was smaller than 10%,the bearing capacity of the repaired beams was similar to that of the unrepaired beams.When the mass loss of main steel bars was larger than 10%,the bearing capacity of the repaired beams increased significantly compared with that of the unrepaired beams.Wu Jin Tang Jinzhi Li Hongming Shang Baokang Wang Zhe 2016Transactions of Nanjing University of Aeronautics and Astronautics2016,33,3:1
