1Integrated transcriptomic and metabolomic analyses of a wax deficient citrus mutant exhibiting jasmonic acid-mediated defense against fungal pathogens显示文摘Naturally,resistant crop germplasms are important resources for managing the issues of agricultural product safety and environment deterioration.We found a spontaneous mutant of‘Newhall’navel orange(Citrus sinensis Osbeck)(MT)with broad-spectrum protections against fungal pathogens in the orchard,postharvest-storage,and artificial inoculation conditions.To understand the defense mechanism of MT fruit,we constructed a genome-scale metabolic network that integrated metabolome and transcriptome datasets.The coordinated transcriptomic and metabolic data were enriched in two sub-networks,showing the decrease in very long chain fatty acid(by 41.53%)and cuticular wax synthesis(by 81.34%),and increase in the synthesis of jasmonic acid(JA)(by 95.23%)and JA-induced metabolites such as 5-dimethylnobietin(by 28.37%)in MT.Furthermore,cytological and biochemical analyses confirmed that the response to fungal infection in MT was independent of wax deficiency and was correlated with the levels of jasmonates,and the expression of plant defensin gene PDF1.2.Results of exogenous application of MeJA and JA inhibitors such as propyl gallate proved that JA-mediated defense contributes to the strong tolerance against pathogens in MT.Our results indicated that jasmonate biosynthesis and signaling are stimulated by the fatty acid redirection of MT,and participate in the tolerance of pathogenic fungi.Yizhong He Jingwen Han Runsheng Liu Yuduan Ding Jinqiu Wang Li Sun Xiaoming Yang Yunliu Zeng Weiwei Wen Juan Xu Hongming Zhang Xiang Yan Zhaoxing Chen Zuliang Gu Hong Chen Huanqing Tang Xiuxin Deng Yunjiang Cheng 2018Horticulture Research2018,5,1:8

3Repurposing econazole as a pharmacological autophagy inhibitor to treat pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma(PDAC)is characterized by the highest mortality among carcinomas.The pathogenesis of PDAC requires elevated autophagy,inhibition of which using hydroxychloroquine has shown promise.However,current realization is impeded by its suboptimal use and unpredictable toxicity.Attempts to identify novel autophagy-modulating agents from already approved drugs offer a rapid and accessible approach.Here,using a patient-derived organoid model,we performed a comparative analysis of therapeutic responses among various antimalarial/fungal/parasitic/viral agents,through which econazole(ECON),an antifungal compound,emerged as the top candidate.Further testing in cell-line and xenograft models of PDAC validated this activity,which occurred as a direct consequence of dysfunctional autophagy.More specifically,ECON boosted autophagy initiation but blocked lysosome biogenesis.RNA sequencing analysis revealed that this autophagic induction was largely attributed to the altered expression of activation transcription factor 3(ATF3).Increased nuclear import of ATF3 and its transcriptional repression of inhibitor of differentiation-1(ID-1)led to inactivation of the AKT/mammalian target of rapamycin(m TOR)pathway,thus giving rise to autophagosome accumulation in PDAC cells.The magnitude of the increase in autophagosomes was sufficient to elicit ER stress-mediated apoptosis.Furthermore,ECON,as an autophagy inhibitor,exhibited synergistic effects with trametinib on PDAC.This study provides direct preclinical and experimental evidence for the therapeutic efficacy of ECON in PDAC treatment and reveals a mechanism whereby ECON inhibits PDAC growth.Ningna Weng Siyuan Qin Jiayang Liu Xing Huang Jingwen Jiang Li Zhou Zhe Zhang Na Xie Kui Wang Ping Jin Maochao Luo Liyuan Peng Edouard C.Nice Ajay Goel Suxia Han Canhua Huang Qing Zhu 2022Acta Pharmaceutica Sinica B2022,12,7:2
4Gastrin attenuates sepsis-induced myocardial dysfunction by down-regulation of TLR4 expression in macrophages显示文摘Myocardial dysfunction is the most serious complication of sepsis.Sepsis-induced myocardial dysfunction(SMD)is often associated with gastrointestinal dysfunction,but its pathophysiological significance remains unclear.The present study found that patients with SMD had higher plasma gastrin concentrations than those without SMD.In mice,knockdown of the gastrin receptor,cholecystokinin B receptor(Cckbr),aggravated lipopolysaccharide(LPS)-induced cardiac dysfunction and increased inflammation in the heart,whereas the intravenous administration of gastrin ameliorated SMD and cardiac injury.Macrophage infiltration plays a significant role in SMD because depletion of macrophages by the intravenous injection of clodronate liposomes,48 h prior to LPS administration,alleviated LPSinduced cardiac injury in Cckbr-deficient mice.The intravenous injection of bone marrow macrophages(BMMs)overexpressing Cckbr reduced LPS-induced myocardial dysfunction.Furthermore,gastrin treatment inhibited toll-like receptor 4(TLR4)expression through the peroxisome proliferator-activated receptor a(PPAR-a)signaling pathway in BMMs.Thus,our findings provide insights into the mechanism of the protective role of gastrin/CCKBR in SMD,which could be used to develop new treatment modalities for SMD.Dandong Fang Yu Li Bo He Daqian Gu Mingming Zhang Jingwen Guo Hongmei Ren Xinyue Li Ziyue Zhang Ming Tang Xingbing Li Donghai Yang Chunmei Xu Yijie Hu Hongyong Wang Pedro AJose Yu Han Chunyu Zeng 2023Acta Pharmaceutica Sinica B2023,13,9:1

6Wear behavior of light-weight and high strength Fe-Mn-Ni-Al matrix self-lubricating steels显示文摘The good combination of mechanical and tribological properties for self-lubricating materials is crucial. In this work, novel self-lubricating Fe-16.4 Mn-4.8 Ni-9.9 Al-xC(wt%) steels containing graphite phase were fabricated using mechanical alloying and spark plasma sintering. The compositions of the steels were designed by using thermodynamic calculation, and the effect of carbon addition on the microstructure was further investigated. The steel possesses high hardness of 621 HV, high yield strength of 1437 MPa and good fracture toughness at room temperature. The yield strengths are still above 600 MPa at 600?C.The tribological behavior and mechanical properties from room temperature to 800?C were studied, and the wear mechanisms at elevated temperatures were discussed. The steel has a stable friction coefficient of 0.4 and wear rate in a magnitude of 10^(-6) mm^3/N·m below 600?C. 8Mineral Deposit Model of Cu-Fe-Au Skarn System in the Edongnan Region, Eastern China显示文摘Cu and Fe skarns are the world's most abundant and largest skarn type deposits,especially in China,and Au-rich skarn deposits have received much attention in the past two decades and yet there are few papers focused on schematic mineral deposit models of Cu-Fe-Au skarn systems.Three types of Au-rich deposits are recognized in the Edongnan region,Middle-Lower Yangtze River metallogenic belt:~140 Ma Cu-Au and Au-Cu skarn deposits and distal Au-Tl deposits.137-148 Ma Cu-Fe and 130-133 Ma Fe skarn deposits are recognized in the Edongnan region.The Cu-Fe skarn deposits have a greater contribution of mantle components than the Fe skarn deposits,and the hydrothermal fluids responsible for formation of the Fe skarn deposits involved a greater contribution from evaporitic sedimentary rocks compared to Cu-Fe skarn deposits.The carbonate-hosted Au-Tl deposits in the Edongnan region are interpreted as distal products of Cu-Au skarn mineralization.A new schematic mineral deposit model of the Cu-Fe-Au skarn system is proposed to illustrate the relationship between the Cu-Fe-Au skarn mineralization,the evaporitic sedimentary rocks,and distal Au-Tl deposits.This model has important implications for the exploration for carbonate-hosted Au-Tl deposits in the more distal parts of Cu-Au skarn systems,and Fe skarn deposits with the occurrence of gypsum-bearing host sedimentary rocks in the MLYRB,and possibly elsewhere.XIE Guiqing MAO Jingwen ZHU Qiaoqiao HAN Yingxiao LI Wei DUAN Chao YE Hui 2020Acta Geologica Sinica(English Edition)2020,94,6:0

10Performance of parasitoid Aphidius giftiensis Ashmead parasitizing Myzus persicae fed on artificial media显示文摘Aphidius gifuensis Ashmead is an economically important aphid parasitoid,particularly for pest Myzus persicae.The aim of the study was to feed M.persicae using a formulated artificial medium instead of tobacco to maintain parasitoid A.gifuensis in winter.The development,survival and reproduction of M.persicae fed with four artificial media(AM1-AM4),as well as the parasitism,emergence and longevity of A.gifuensis when reared on F3 M.persicae fed with AMI,were assessed.Over 15 days of the study,the survival rates of A.gifuensis for the AM1 and AM2 treatments were significantly greater than those for the AM3 and AM4 treatments.The highest fecundity of F,A.gifuensis females was observed on AM1 feeding.No differences were noted in survival rate,development or nymph production between F:and F,M.persicae fed with AM1.No differences were recorded in the number of mummies,parasitism rate,adult Iongevity or proportion of A.gifuensis females between Treatment I(4.gifuensis parasitizing AM1-reared M.persicae)and the control.However,the emergence rate of Treatment I was lower than that of the control.The rearing of.A.gifuensis on M.persicae fed with an artificial medium was found to be a feasible proposition.This could save time,space and cost compared with traditional production methods.YE Jingwen CHAI Fanghua GU Xinghui HAN Shichou Lü Xin JI Sigui ZHANG Limeng LI Jun 2018烟草科技2018,51,A01:0
11Analysis on Air Quality Statuses in 31 Provincial Capital Cities in China--Taking 2014 as an Example显示文摘Air pollution is a hot issue of current research. In recent years,air quality of China shows the significantly declining tendency. To explore temporal-spatial distribution rule of air quality status of China not including Hongkong,Macao and Taiwan,air quality indexes( AQI) of 31 provincial capitals of China in 2014 were taken as research objects. By literature retrieval,data excavation,statistic analysis and logical reasoning,temporalspatial distribution rule of air quality status from 31 provincial capitals of China and its macroscopic origin were deeply studied. 12Revealing the roles of TLR7, a nucleic acid sensor for COVID-19 in pan-cancer显示文摘Recent studies suggested that cancer was a risk factor for coronavirus disease 2019 (COVID-19). Toll-like receptor 7 (TLR7), a severe acute respiratory syndrome 2 (SARS-CoV-2) virus’’s nucleic acid sensor, was discovered to be aberrantly expressed in many types of cancers. However, its expression pattern across cancers and association with COVID-19 has not been systematically studied. In this study, we proposed a computational framework to comprehensively study the roles of TLR7 in COVID-19 and pan-cancers at genetic, gene expression, protein, epigenetic, and single-cell levels. We found TLR7 mRNA expression was significantly up-regulated in 6 cancer types and down-regulated in 6 cancer types, further validated in the HPA database at the protein level. The genes significantly co-expressed with TLR7 were mainly enriched in the toll-like receptor signaling pathway, endolysosome, and signaling pattern recognition receptor activity. In addition, the abnormal TLR7 expression was associated with Mismatch repair (MMR), microsatellite instability (MSI), tumor mutational burden (TMB) in various cancers. Mined by the ESTIMATE algorithm, the expression of TLR7 was also closely linked to various immune infiltration patterns in pan-cancer, and TLR7 was mainly enriched in macrophages, as revealed by single-cell RNA sequencing. Finally, TLR7 expressions were very sensitive to a few targeted drugs, such as Alectinib and Imiquimod. 14A microenvironment sensitive pillar[5]arene-based fluorescent probe for cell imaging and drug delivery显示文摘Dansylamide(DNSA) is a typical ICT probe that has a favorable serum albumin sensitivity. Inspired by this, we designed a microenvironment sensitive fluorescent probe 4 C-G through introducing DNSA into pillar[5]arene. Unlike DNSA, 4 C-G displayed differentiated sensitivity to multiple proteins, which was benefit from pillar[5]arene assisted the probe to form complexes with proteins. 4 C-G could not only be applied in imaging of Hep G2, but also act as a favorable drug carrier for regorafenib(REG) encapsulation.The 4 C-G-REG complex would aggregate into high drug-loading fluorescent nanoparticles in a physiological environment(pH 7.4). Such nanoparticles exhibited p H-triggered enrichment ability, which rapidly enriched REG in the acidic environment(pH 6.0). 15Sandwich structured ultra-strong-heat-shielding aerogel/copper composite insulation board for safe lithium-ion batteries modules显示文摘The fire hazard of lithium-ion batteries(LIBs)modules is extremely serious due to their high capacity.Moreover,once a battery catches fire,it can easily result in a fire of the entire LIBs modules.In this work,a sandwich structure composite thermal insulation(STI)board(copper//silica dioxide aerogel//copper)with the advantages of low thermal conductivity(0.031 W m-1K-1),low surface radiation emissivity(0.1)and good thermal convection inhibition effect has been designed.The thermal runaway(TR)occurrence time of adjacent LIBs increases from 1384 s to more than 6 h+due to the protection of STI board.No TR propagation occurs within LIBs modules with protect of a STI board when a battery catches fire.The ultra-strong-heat-shielding mechanism of STI board has been revealed.The TR propagation of LIBs modules has been insulated effectively by STI board through reducing the heat transfer of convection,conduction and radiation.The air flow rate between the heater and LIBs and radiant heat absorbed by LIBs decrease by 63.5%and 35.1%with protection of STI board,respectively.A high temperature difference inside the STI board is also formed.This work provides direction for the designing of safe thermal insulation board for LIBs modules.Heng Yu Xiaowei Mu Yulu Zhu Can Liao Longfei Han Jingwen Wang Wei Cai Yongchun Kan Lei Song Yuan Hu 2023Journal of Energy Chemistry2023,,1:0
16Screening and identification of HLA-A2-restricted neoepitopes for immunotherapy of non-microsatellite instability-high colorectal cancer显示文摘Colorectal cancer has one of the highest mortality rates among malignant tumors,and most patients with non-microsatellite instability-high (MSI-H) colorectal cancer do not benefit from targeted therapy or immune checkpoint inhibitors.Identification of immunogenic neoantigens is a promising strategy for inducing specific antitumor T cells for cancer immunotherapy.Here,we screened potential high-frequency neoepitopes from non-MSI-H colorectal cancer and tested their abilities to induce tumorspecific cytotoxic T cell responses.Three HLA-A2-restricted neoepitopes (P31,P50,and P52) were immunogenic and could induce cytotoxic T lymphocytes in peripheral blood mononuclear cells from healthy donors and colorectal cancer patients.Cytotoxic T lymphocytes induced in HLA-A2.1/K^(b) transgenic mice could recognize and lyse mutant neoepitope-transfected HLA-A2^(+) cancer cells.Adoptive transfer of cytotoxic T lymphocytes induced by the peptide pool of these three neoepitopes effectively inhibited tumor growth and increased the therapeutic effects of anti-PD-1 antibody.These results revealed the potential of high-frequency mutation-specific peptide-based immunotherapy as a personalized treatment approach for patients with non-MSI-H colorectal cancer.The combination of adoptive T cell therapy based on these neoepitopes with immune checkpoint inhibitors,such as anti-PD-1,could provide a promising treatment strategy for non-MSI-H colorectal cancer.Ranran Shi Yubing Li Ling Ran Yu Dong Xiuman Zhou Jingwen Tang Lu Han Mingshuang Wang Liwei Pang Yuanming Qi Yahong Wu Yanfeng Gao 2022Science China(Life Sciences)2022,65,3:0
17IAGNES algorithm for protocol recognition显示文摘In the process of protected protocol recognition,an improved AGglomerative NESting algorithm( IAGNES) with high adaptability is proposed,which is based on the AGglomerative NESting algorithm( AGNES),for the challenging issue of how to obtain single protocol data frames from multiprotocol data frames. It can improve accuracy and efficiency by similarity between bit-stream data frames and clusters,extract clusters in the process of clustering. Every cluster obtained contains similarity evaluation index which is helpful to evaluation. More importantly,IAGNES algorithm can automatically recognize the number of cluster. 20Heating of multi-species upflowing ion beams observed by Cluster on March 28, 2001显示文摘Cluster satellites observed three successive outflowing ion beams on 28 March, 2001. It is generally accepted that these ion beams, composed of H^+, He^+, and O^+ ions, with three inverted-V structures in their energy spectra, are produced by acceleration through U-shaped potential structures. By eliminating the background ion population and employing Maxwelling fitting, we find that ions coming from the center of the potential structure have higher temperature than those from the flanks. Higher temperature of O^+ and He+compared to that of H^+ indicates that heavy ions are preferentially heated; we further infer that the heating efficiencies of O^+ and He^+ ions differ between the center and edges of the U-shaped potential structures.
