1Human ACE2-Functionalized Gold“Virus-Trap”Nanostructures for Accurate Capture of SARS-CoV-2 and Single-Virus SERS Detection显示文摘The current COVID-19 pandemic urges the extremely sensitive and prompt detection of SARS-CoV-2 virus.Here,we present a Human Angiotensin-converting-enzyme 2(ACE2)-functionalized gold“virus traps”nanostructure as an extremely sensitive SERS biosensor,to selectively capture and rapidly detect S-protein expressed coronavirus,such as the current SARS-CoV-2 in the contaminated water,down to the single-virus level.Such a SERS sensor features extraordinary 106-fold virus enrichment originating from high-affinity of ACE2 with S protein as well as“virus-traps”composed of oblique gold nanoneedles,and 109-fold enhancement of Raman signals originating from multi-component SERS effects.Furthermore,the identification standard of virus signals is established by machine-learning and identification techniques,resulting in an especially low detection limit of 80 copies mL^(−1) for the simulated contaminated water by SARS-CoV-2 virus with complex circumstance as short as 5 min,which is of great significance for achieving real-time monitoring and early warning of coronavirus.Moreover,here-developed method can be used to establish the identification standard for future unknown coronavirus,and immediately enable extremely sensitive and rapid detection of novel virus.Yong Yang Yusi Peng Chenglong Lin Li Long Jingying Hu Jun He Hui Zeng Zhengren Huang Zhi-Yuan Li Masaki Tanemura Jianlin Shi John R.Lombardi Xiaoying Luo 2021Nano-Micro Letters2021,13,7:6
2Continuous desulfurization and bacterial community structure of an integrated bioreactor developed to treat SO_2 from a gas stream显示文摘Sulfide dioxide(SO2) is often released during the combustion processes of fossil fuels. An integrated bioreactor with two sections, namely, a suspended zone(SZ) and immobilized zone(IZ), was applied to treat SO2 for 6 months. Sampling ports were set in both sections to investigate the performance and microbial characteristics of the integrated bioreactor. SO2 was effectively removed by the synergistic effect of the SZ and IZ, and more than 85%removal efficiency was achieved at steady state. The average elimination capacity of SO2 in the bioreactor was 2.80 g/(m3·hr) for the SZ and 1.50 g/(m3· hr) for the IZ. Most SO2 was eliminated in the SZ. The liquid level of the SZ and the water content ratio of the packing material in the IZ affected SO2 removal efficiency. The SZ served a key function not only in SO2 elimination, but also in moisture maintenance for the IZ. The desired water content in IZ could be feasibly maintained without any additional pre-humidification facilities. Clone libraries of 16 S r DNA directly amplified from the DNA of each sample were constructed and sequenced to analyze the community composition and diversity in the individual zones.The desulfurization bacteria dominated both zones. Paenibacillus sp. was present in both zones, whereas Ralstonia sp. existed only in the SZ. 4PHYTOCHROME-INTERACTING FACTORS Interact with the ABA Receptors PYL8 and PYL9 to Orchestrate ABA Signaling in Darkness显示文摘PHYTOCHROME-INTERACTING FACTORS(PIFs)are a group of basic helix-loop-helix transcription factors that can physically interact with photoreceptors,including phytochromes and cryptochromes.It was previously demonstrated that PIFs accumulated in darkness and repressed seedling photomorphogenesis,and that PIFs linked different photosensory and hormonal pathways to control plant growth and development.In this study,we show that PIFs positively regulate the ABA signaling pathway during the seedling stage specifically in darkness.We found that PIFs positively regulate ABI5 transcript and protein levels in darkness in response to exogenous ABA treatment by binding directly to the G-box motifs in the ABI5 promoter.Consistently,PIFs and the G-box motifs in the ABI5 promoter determine ABI5 expression in darkness,and overexpression of ABI5 could rescue the ABA-insensitive phenotypes of pifq mutants in the dark.Moreover,we discovered that PIFs can physically interact with the ABA receptors PYL8 and PYL9,and that this interaction is not regulated by ABA.Further analyses showed that PYL8 and PYL9 promote PIF4 protein accumulation in the dark and enhance PIF4 binding to the ABI5 promoter,but negatively regulate PIF4-mediated ABI5 activation.Taken together,our data demonstrate that PIFs interact with ABA receptors to orchestrate ABA signaling in darkness by controlling ABI5 expression,providing new insights into the pivotal roles of PIFs as signal integrators in regulating plant growth and development.Lijuan Qi Shan Liu Cong Li Jingying Fu Yanjun Jing Jinkui Cheng Hong Li Dun Zhang Xiaoji Wang Xiaojing Dong Run Han Bosheng Li Yu Zhang Zhen Li William Terzaghi Chun-Peng Song Rongcheng Lin Zhizhong Gong Jigang Li 2020Molecular Plant2020,13,3:3
5Temporal variation of microbial population in a thermophilic biofilter for SO_2 removal显示文摘The performance of a biofilter relies on the activity of microorganisms during the gas contaminant treatment process. In this study, SO_2 was treated using a laboratory-scale biofilter packed with polyurethane foam cubes(PUFC), on which thermophilic desulfurization bacteria were attached. The thermophilic biofilter effectively reduced SO_2 within 10 months of operation time, with a maximum elimination capacity of 48.29 g/m^3/hr.Temporal shifts in the microbial population in the thermophilic biofilter were determined through polymerase chain reaction-denaturing gradient gel electrophoresis and deoxyribonucleic acid(DNA) sequence analysis. The substrate species and environmental conditions in the biofilter influenced the microbial population. Oxygen distribution in the PUFC was analyzed using a microelectrode. When the water-containing rate in PUFC was over 98%, the oxygen distribution presented aerobic–anoxic–aerobic states along the test route on the PUFC. 6Effects of oxygen and water content on microbial distribution in the polyurethane foam cubes of a biofilter for SO_2 removal显示文摘The performance of a biofilter for off-gas treatment relies on the activity of microorganisms and adequate O_2 and H_2O. In present study, a microelectrode was applied to analyze O_2 in polyurethane foam cubes(PUFCs) packed in a biofilter for SO_2 removal. The O_2 distribution varied with the density and water-containing rate(WCR) of PUFCs. The O_2 concentration dropped sharply from 10.2 to 0.8 mg/L from the surface to the center of a PUFC with 97.20%of WCR. The PUFCs with high WCR presented aerobic–anoxic–aerobic areas.Three-dimensional simulated images demonstrated that the structure of PUFCs with high WCR consisted of an aerobic 'shell' and an anoxic 'core', with high-density PUFCs featuring a larger anoxic area than low-density PUFCs. Moreover, the H_2O distribution in the PUFC was uneven and affected the O_2 concentration. Whereas aerobic bacteria were observed in the PUFC surface, facultative anaerobic microorganisms were found at the PUFC core, where the O_2 concentration was relatively low. 9Artificial evolution of OsEPSPS through an improved dual cytosine and adenine base editor generated a novel allele conferring rice glyphosate tolerance显示文摘Exploiting novel endogenous glyphosate-tolerant alleles is highly desirable and has promising potential for weed control in rice breeding. Here,through fusions of different effective cytosine and adenine deaminases with nCas9-NG, we engineered an effective surrogate two-component composite base editing system, STCBE-2, with improved C-to-T and A-to-G base editing efficiency and expanded the editing window. Furthermore,we targeted a rice endogenous OsEPSPS gene for artificial evolution through STCBE-2-mediated near-saturated mutagenesis. After hygromycin and glyphosate selection, we identified a novel OsEPSPS allele with an Asp-213-Asn(D213N)mutation(OsEPSPS-D213N) in the predicted glyphosate-binding domain, which conferred rice plants reliable glyphosate tolerance and had not been reported or applied in rice breeding. Collectively, we developed a novel dual base editor which will be valuable for artificial evolution of important genes in crops. 15Benefit analysis of multi-approach biomass energy utilization toward carbon neutrality显示文摘To reduce greenhouse gas(GHG)emissions,biomass has been increasingly developed as a renewable and clean alternative to fossil fuels because of its carbon-neutral characteristics.China has been investigating the rational development and use of bioenergy for developing its clean energy and achieving carbon neutrality.Substituting fossil fuels with multi-source and multi-approach utilized bioenergy and corresponding carbon reduction in China remain largely unexplored.Here,a comprehensive bioenergy accounting model with a multi-dimensional analysis was developed by combining spatial,life cycle,and multi-path analyses.Accordingly,the bioenergy production potential and GHG emission reduction for each distinct type of biomass feedstock through different conversion pathways were estimated.The sum of all available organic waste(21.55 EJ yr^(-1))and energy plants on marginal land(11.77 EJ yr^(-1))in China produced 23.30 EJ of bioenergy and reduced 2,535.32 Mt CO_(2)-eq emissions,accounting for 19.48%and 25.61%of China’s total energy production and carbon emissions in 2020,respectively.When focusing on the carbon emission mitigation potential of substituting bioenergy for conventional counterparts,bioelectricity was the most effective,and its potential was 4.45 and 8.58 times higher than that of gaseous and liquid fuel alternatives,respectively.In this study,life cycle emission reductions were maximized by a mix of bioenergy end uses based on biomass properties,with an optimal 78.56%bioenergy allocation from biodiesel,densified solid biofuel,biohydrogen,and biochar.The main regional bioenergy GHG mitigation focused on the Jiangsu,Sichuan,Guangxi,Henan,and Guangdong provinces,contributing to 31.32%of the total GHG mitigation potential.This study provides valuable guidance on exploiting untapped biomass resources in China to secure carbon neutrality by 2060.Jiaoyue Wang Jingying Fu Zhitong Zhao Longfei Bing Fengming Xi Feng Wang Jiang Dong Shiyun Wang Gang Lin Yan Yin Qinqin Hu 2023The Innovation2023,4,3:0

18Molecular characterization,expression patterns,and subcellular localization of a classical and a novel nonclassical MHC class Iαmolecules from Japanese eel Anguilla japonica显示文摘Major histocompatibility complex(MHC)molecules play a critical role in the immune response of vertebrate animals by presenting foreign antigens to T lymphocytes.In this study,we first cloned and identified a classical and a novel nonclassical MHC Iαmolecules from Japanese eel(Anguilla japonica),named as AjMHC I-UBA and AjMHC I-L,respectively.The full-length cDNA of AjMHC I-UBA contains an open reading frame(ORF)of 1047 bp encoding a predicted protein of 348 amino acids,while AjMHC I-L 1089 bp encodes 362 amino acids.The multiple alignment of the amino acid sequence showed that AjMHC I-UBA and AjMHC I-L consist of an N-terminal MHC I superfamily domain withinα1 andα2 helices,an IgC-MHC Iα3 domain,and a transmembrane region close to the C-terminal,which are similar to other fish and mammal species.Molecular polymorphism analysis showed that eight different major alleles of AjMHC I-UBA,named as AjMHC I-UBA*0101~1001,were identified from six Japanese eel individuals.Furthermore,a distinguishing signature of the nonclassical L-lineage genes specific motif“HINMTL”,including an N-glycosylation site(NXS/T),was present in theα3 domain of AjMHC I-L sequences.Although the predicted three-dimensional structures of AjMHC I-UBA and AjMHC I-L are similar to that of human MHC Iα,phylogenetic analysis showed that these two protein molecules belong to classical MHC I UBA gene of U-lineage and nonclassical MHC I gene of L-lineage,respectively.Quantitative real-time polymerase chain reaction(qRT-PCR)analysis revealed that the highest expression of AjMHC I-UBA and AjMHC I-L was found in the intestine and the expression level of AjMHC I-UBA in all of the examined tissues was significantly higher than that of AjMHC I-L.The expressions of AjMHC I-UBA and AjMHC I-L in liver,kidney and spleen were significantly induced following injection with the viral mimic poly I:C,LPS,and Aeromonas hydrophila infection.In vitro,the AjMHC I-UBA and AjMHC I-L transcripts of Japanese eel liver cells were significantly enhanced by the treatment of poly I:C or the stimulation of the high concentration of A.hydrophila.Subcellular localization showed that under natural conditions,AjMHC I-UBA and AjMHC I-L were uniformly distributed in the cytoplasm,and aggregated into spots or flakes in the cytoplasm after the stimulation of poly I:C or LPS.These results collectively suggested that AjMHC I-UBA and AjMHC I-L are important components possibly involved in Japanese eel defense against viral and bacterial infection.Taken together,these findings provide valuable insight into the immune function of MHC class I in the immune system of teleost.Jianjun Feng Xinwei Peng Peng Lin Yilei Wang Ziping Zhang Yuankai Xu Pengyun Chen Jingying Wu 2023Aquaculture and Fisheries2023,8,2:0
19Promoting Proton Migration Kinetics by Ni^(2+)Regulating Enables Improved Aqueous Zn-MnO_(2) Batteries显示文摘The energy storage behaviors of MnO_(2) for aqueous Zn-MnO_(2) batteries mainly depend on the Zn^(2+)/H^(+)intercalation but are limited by poor ion/electron migration dynamics and stability.Herein,a strategy is proposed that promoting proton migration kinetics ameliorates H^(+)storage activity by introducing Ni^(2+)intoγ-MnO_(2)(Ni-MnO_(2)).Ni^(2+)can lower the diffusion barrier of H^(+)and selectively induce the ion intercalation,thereby alleviating the electrostatic interaction with the lattice.Moreover,Ni^(2+)enables the adjacent[MnO6]octahedrons to have better electron conductivity.The Ni-MnO_(2) exhibits superior rate performance(nearly four times specific capacity compared with MnO_(2))and ultra-long-cycle stability(100%of capacity retention after 11000 cycles at 3.0 A g^(-1)).The calculation indicates that the Ni-MnO_(2) allows H^(+)migrate rapidly along the one-dimensional tunnel due to reduction of the activation energy caused by Ni^(2+)regulating,thus achieving excellent reaction kinetics.This work brings great potential for the development of high-performance aqueous Zn-MnO_(2) batteries.Jie Ji Jia Yao Yongchang Xu Houzhao Wan Bao Zhang Lin Lv Jingying Li Nengze Wang Zhaohan Zheng Jun Zhang Guokun Ma Li Tao Hanbin Wang Yi Wang Hao Wang 2023Energy & Environmental Materials2023,6,2:0
20Associations between LMO1 gene polymorphisms and central nervous system tumor susceptibility显示文摘Importance:LIM domain only 1(LMO1)gene polymorphisms were previously found to be implicated in the risk of several cancers.No available studies were performed regarding the predisposing effect of LMO1 gene single nucleotide polymorphisms(SNPs)on central nervous system(CNS)tumor risk.Objective:We aimed to determine whether the LMO1 gene SNPs were associated with the risk of CNS tumor by applying a case-control study with 191 cases and 248 controls in China.Methods:The contributions of LMO1 gene SNPs to the risk of CNS tumor was evaluated by multinomial logistic regression.Results:Based on the calculations of odds ratio(OR)and 95%confidence interval(CI),we failed to detect a significant relationship between each LMO1 gene SNP(rs110419 A>G,rs4758051 G>A,rs10840002 A>G,rs204938 A>G,and rs2168101 G>T)and CNS tumor risk,respectively.A negative association was also found in the combined effects on these five SNPs and CNS tumor risk.The stratification analysis further demonstrated the individuals with rs204938 AG/GG genotype confer to increased risk of CNS tumor compared with those with an AA genotype in males(OR:1.74,95%CI:1.01-2.98,P=0.046).Interpretation:We concluded that LMO1 gene SNPs may not strong enough to influence the risk of CNS tumor in Chinese children.More studies are required to verify this association.Huiran Lin Huitong Chen Ao Lin Xiaoping Liu Xiaokai Huang Jingying Zhou Li Yuan Zhenjian Zhuo 2021Pediatric Investigation2021,5,4:0
