1Fas ligand expression in colon cancer:A possible mechanism of tumor immune privilege显示文摘AIM: To detect the expression of Fas ligand (FasL) in colon cancer tissues and cell lines and analyze the function of FasL-expressing colon cancer cells in inducing Fas-sensitive T lymphocyte apoptosis. METHODS: Ninety surgically resected colon cancer tissues and 15 hepatic metastasis specimens were investigatedby immunohistochemical method with normal colon mucosa and colon adenoma as control. The relationship between FasL expression and pathologic features was also analyzed.FasL expression of 4 colon cancer cell lines, SW620, Lovo, LS-174T and SW1116, were detected by Western blotting assay. The function of FasL expressed on colon cancer cells was determined by coculture assay with Jurkat T lymphocytes, the apoptotic rate of which was detectedby flow cytometry assay.RESULTS: Fifty-six (62.22%) cases of all the 90 colon cancer tissues and all (100%) the liver metastasis specimens expressed FasL, significantly higher than normal colon mucosa and colonic adenoma. Higher expression of FasL was found in more advanced stage of colon cancer and in cancer tissues with lymphatic or hepatic metastasis.All the colon cancer cell lines were found to express FasL.After coculture with the SW1116 cells for 24 h with an effector: target ratio 10:1, the rate of apoptosis of Jurkat cells rose from 1.9% to 21.0%.CONCLUSION: The expression of FasL is upregulated in colon cancer and the functionally expressed FasL can induce apoptosis of Fas-expressing T lymphocytes.WeiZhang Er-XunDing QiangWang Da-QiaoZhu JinHe Yu-LiLi Yuan-HeWang 2005World Journal of Gastroenterology2005,11,23:6
2Cloning and characterization of a mouse liver-specific gene mfrep-1,up-regulated in liver regeneration显示文摘Human fibrinogen-related protein-1/liver fibrinogen-related protein-1 (HFREP-1/LFIRE-1), a liver-specificprotein, is a member of fibrinogen superfamily that exerts various biological activities. However, the func-tion of HFREP-1/LFIRE-1 in liver remains unknown. Here we isolated its mouse ortholog gene-mousefibrinogen-related protein-1 (mfrep-1), which encoded 314 amino acids, exhibiting 80.4% similarity toHFREP-1/LFIRE-1. Northern blot analysis revealed that 1.2-kb mfrep-1 mRNA was detected selectivelyin mouse liver. To explore the function of MFREP-1, we examined the levels of mfrep-1 mRNA duringregeneration after 70% partial hepatectomy (PHx) in mice. mfrep-1 mRNA increased in the regeneratingliver and reached the first shoulder peak at 2-4 h after PHx. Cycloheximide pretreatment could suppress theinduction of mfrep-1, indicating the up-regulation of this gene need de novo protein synthesis. Its mRNAcontinued to elevate at 6 h thereafter and reached the second peak at 24 h. The enhanced expression ofmfrep-1 maintained high until 72 h and then declined slowly to the basal level. Immunohistochemistryassessment confirmed the up-regulated expression of MFREP-1 protein in parenchymal cells during liverregeneration. 4Transcriptome sequencing assisted discovery and computational analysis of novel SNPs associated with flowering in Raphanus sativus in-bred lines for marker-assisted backcross breeding显示文摘The sequencing of radish genome aids in the better understanding and tailoring of traits associated with economic importance.In order to accelerate the genomics assisted breeding and genetic selection,transcriptomes of 33 radish inbred lines with diverse traits were sequenced for the development of single nucleotide polymorphic(SNP)markers.The sequence reads ranged from 2,560,543,741 bp to 20,039,688,139 bp with the GC(%)of 47.80–49.34 and phred quality score(Q30)of 96.47–97.54%.A total of 4951 polymorphic SNPs were identified among the accessions after stringent filtering and 298 SNPs with efficient marker assisted backcross breeding(MAB)markers were generated from the polymorphic SNPs.Further,functional annotations of SNPs revealed the effects and importance of the SNPs identified in the flowering process.The SNPs were predominantly associated with the four major flowering related transcription factors such as MYB,MADS box(AG),AP2/EREB,and bHLH.In addition,SNPs in the vital flowering integrator gene(FT)and floral repressors(EMBRYONIC FLOWER 1,2,and FRIGIDA)were identified among the radish inbred lines.Further,50 SNPs were randomly selected from 298 SNPs and validated using Kompetitive Allele Specific PCR genotyping system(KASP)in 102 radish inbred lines.The homozygosity of the inbred lines varied from 56 to 96%and the phylogenetic analysis resulted in the clustering of inbred lines into three subgroups.Taken together,the SNP markers identified in the present study can be utilized for the discrimination,seed purity test,and adjusting parental combinations for breeding in radish.Jinhee Kim Abinaya Manivannan Do-Sun Kim Eun-Su Lee Hye-Eun Lee 2019Horticulture Research2019,6,1:2
5Replication and gene expression of mutant hepatitis B virus in a transgenic mouse containing the complete viral genome with mutant sgene显示文摘AIM:To establish the transgenic mouse line harbouring complete hepatitis B virus (HBV) genome with mutant s gene (adr subtype).METHODS: Transgenic mice were generated by microinjecting HBV genome into fertilized eggs. Integration, expression, replication of HBV gene and histological changes in transgenic mice were estimated by genomic DNA PCR, serum DNA PCR, Southern blot, ELISA, HE staining, immunohistochemistry and transmission electron microscopy. Transgenic mice with HBsAg positive in serum were bred and analyzed.RESULTS: A total of 288 eggs survived from microinjections were transplanted into the oviducts of 13 pseudopregnant mice and 49 pups were produced. Twenty-six mice were identified to have the integrated HBV gene. Serum HBsAg and HBeAg were detected in 2 of 43 mice. HBsAg and HBcAg in cytoplasm or nuclei of hepatocytes were detected in 10 mice. Founders with HBsAg in serum were named lineages G145R-15 and G145R-18. Of the 16 F1 offsprings generated by G145R-15 founder, 12 were positive for HBV genome with PCR, 10 were positive for HBsAg and HBcAg with immunohistochemistry and 7 were positive for HBsAg and HBeAg with ELISA. Only 1 of 8 F1 offsprings generated by G145R-18 founder was survived and it was detected positive for HBV genome, HBsAg, HBcAg and HBeAg. Both of the two lineages had some pathological characteristics of mild chronic hepatitis B in the liver, such as swelling of hepatocytes and focal hepatocellular necrosis and parenchymal lymphomononuclear cell infiltrate.CONCLUSION: Transgenic mice harbouring HBV with mutant s gene can be generated. The HBV genes are integrated in the transgenic mice genome and can be expressed, replicated, packaged and excreted. 6Melatonin combined with exercise cannot alleviate cerebral injury in a rat model of focal cerebral ischemia/reperfusion injury显示文摘Previous studies have demonstrated that melatonin combined with exercise can alleviate secondary damage after spinal cord injury in rats.Therefore,it is hypothesized that melatonin combined with exercise can also alleviate ischemic brain damage.In this study,adult rats were subjected to right middle cerebral artery occlusion after receiving 10 mg/kg melatonin or vehicle subcutaneously twice daily for 14 days.Forced exercise using an animal treadmill was performed at 20 m/min for 30 minutes per day for 6 days prior to middle cerebral artery occlusion.After middle cerebral artery occlusion,each rat received melatonin combined with exercise,melatonin or exercise alone equally for 7 days until sacrifice.Interestingly,rats receiving melatonin combined with exercise exhibited more severe neurological deficits than those receiving melatonin or exercise alone.Hypoxia-inducible factor 1α mRNA in the brain tissue was upregulated in rats receiving melatonin combined with exercise.Similarly,microtubule associated protein-2 mRNA expression was significantly upregulated in rats receiving melatonin alone.Chondroitin sulfate proteoglycan 4 (NG2) mRNA expression was significantly decreased in rats receiving melatonin combined with exercise as well as in rats receiving exercise alone.Furthermore,neural cell loss in the primary motor cortex was significantly reduced in rats receiving melatonin or exercise alone,but the change was not observed in rats receiving melatonin combined with exercise.These findings suggest that excessive intervention with melatonin,exercise or their combination may lead to negative effects on ischemia/reperfusion-induced brain damage.Seunghoon Lee Jinhee Shin Minkyung Lee Yunkyung Hong Sang-Kil Lee Youngjeon Lee Tserentogtokh Lkhagvasuren Dong-Wook Kim Young-Ae Yang Kyu-Tae Chang Yonggeun Hong 2012Neural Regeneration Research2012,7,13:2
