1Decreased osteogenesis of adult mesenchymal stem cel s by reactive oxygen species under cyclic stretch: a possible mechanism of age related osteoporosis显示文摘Age related defect of the osteogenic differentiation of mesenchymal stem cells(MSCs) plays a key role in osteoporosis. Mechanical loading is one of the most important physical stimuli for osteoblast differentiation.Here, we compared the osteogenic potential of MSCs from young and adult rats under three rounds of 2 h of cyclic stretch of 2.5% elongation at 1 Hz on 3 consecutive days. Cyclic stretch induced a significant osteogenic differentiation of MSCs from young rats, while a compromised osteogenesis in MSCs from the adult rats.Accordingly, there were much more reactive oxygen species(ROS) production in adult MSCs under cyclic stretch compared to young MSCs. Moreover, ROS scavenger N-acetylcysteine rescued the osteogenic differentiation of adult MSCs under cyclic stretch. Gene expression analysis revealed that superoxide dismutase 1(SOD1) was significantly downregulated in those MSCs from adult rats. 2Dysregulation of the miR-146a-Smad4 axis impairs osteogenesis of bone mesenchymal stem cells under inflammation显示文摘Osteoporosis is a common disease that affects patient quality of life, especially among the elderly population.Although inflammation contributes significantly to osteoporosis, the underlying mechanism is unclear. In this study, we found that tumor necrosis factor(TNF)-α, an inflammatory environment mimic, inhibits osteogenesis of bone mesenchymal stem cells(BMSCs), induces mi R-146 a and decreases Smad4. Moreover,overexpression of mi R-146 a inhibited the osteogenic ability of BMSCs, whereas blocking mi R-146 a partially rescued the osteogenesis deficiency under TNF-α treatment. Molecularly, mi R-146 a decreased Smad4 expression at the protein level by binding to an element located in the Smad4 30-untranslated region, and restoration of Smad4 reversed the inhibitory effects of mi R-146 a on osteogenesis. 7Optical tuning of exciton and trion emissions in monolayer phosphorene显示文摘Monolayer phosphorene provides a unique two-dimensional(2D)platform to investigate the fundamental dynamics of excitons and trions(charged excitons)in reduced dimensions.However,owing to its high instability,unambiguous identification of monolayer phosphorene has been elusive.Consequently,many important fundamental properties,such as exciton dynamics,remain underexplored.We report a rapid,noninvasive,and highly accurate approach based on optical interferometry to determine the layer number of phosphorene,and confirm the results with reliable photoluminescence measurements.Furthermore,we successfully probed the dynamics of excitons and trions in monolayer phosphorene by controlling the photo-carrier injection in a relatively low excitation power range.Based on our measured optical gap and the previously measured electronic energy gap,we determined the exciton binding energy to be~0.3 eV for the monolayer phosphorene on SiO_(2)/Si substrate,which agrees well with theoretical predictions.A huge trion binding energy of~100 meV was first observed in monolayer phosphorene,which is around five times higher than that in transition metal dichalcogenide(TMD)monolayer semiconductor,such as MoS_(2).The carrier lifetime of exciton emission in monolayer phosphorene was measured to be,220 ps,which is comparable to those in other 2D TMD semiconductors.Our results open new avenues for exploring fundamental phenomena and novel optoelectronic applications using monolayer phosphorene.Jiong Yang Renjing Xu Jiajie Pei Ye Win Myint Fan Wang Zhu Wang Shuang Zhang Zongfu Yu Yuerui Lu 2015Light(Science & Applications)2015,4,1:8

9Optimal b value of diffusion-weighted imaging on a 3.0T magnetic resonance scanner in Crohn's disease显示文摘AIM:To determine the optimal b value of diffusionweighted imaging for detecting active inflammation in Crohn's disease.METHODS:Thirty-one patients clinically diagnosed with active Crohn’s disease were referred for magnetic resonance examination.All patients were scanned on a3.0T magnetic resonance scanner using the same protocol involving four different b values(800,1500,2000and 2500 s/mm2).The diagnostic effect of diffusionweighted imaging was evaluated and compared with endoscopic findings.The diffusion-weighted image quality of four b value groups was evaluated and apparent diffusion coefficient was measured for both nor-mal and inflammatory intestinal segments.RESULTS:The contrast-to-noise ratio and signal-tonoise ratio were not satisfied when b value 2000 or2500 s/mm2 was adopted(36.52±14.95 vs 34.78±24.83,P>0.05;53.58±23.45 vs 47.58±29.67,P>0.05).The qualitative image quality was not enough to meet diagnostic requirement.No matter which b value was chosen,the apparent diffusion coefficient of inflammatory intestinal segments was significantly lower than that of normal intestinal segments(1.38±0.28 vs 2.00±0.38,P<0.01;1.09±0.20 vs 1.50±0.28,P<0.01;0.95±0.19 vs 1.34±0.28,P<0.01;0.88±0.14 vs 1.20±0.21,P<0.01).The lesion detection rate(90.32%),diagnostic sensitivity(81.18%)and specificity(95.10%)would be appropriate when b value 1500 s/mm2 was adopted.CONCLUSION:High b value is suitable for intestinal DW examination on a high field MR scanner.Qi Feng Yun-Qi Yan Jiong Zhu Jin-Lu Tong Jian-Rong Xu 2014World Journal of Gastroenterology2014,20,35:4

12Consumer Credit Risk Management in an Emerging Market: The Case of China显示文摘With the liberalization of the financial service sector mandated by China’s access to the WTO, China’s credit card market has received a great deal of attention from global financial institutions. This paper examines the enormous growth opportunities and key barriers facing the development of the credit card industry in China, and discusses the importance and tools of consumer credit risk management. In the process of rapid expansion of China’s consumer credit card industry, credit risk management should be treated as a top priority to avoid a pile up of bad debt in credit card receivables. This requires the development of an updated and comprehensive national consumer credit database and the use of credit risk modeling and scoring in predicting consumer behavior. 14CXCR4 antagonist AMD3100 attenuates colonic damage in mice with experimental colitis显示文摘AIM:To investigate the effects of the chemokine stromal cell-derived factor-1(CXCL12) receptor(CXCR4) antagonist AMD3100 on colonic inflammation and epithelial barrier in dextran sulfate sodium(DSS)-induced colitis in mice.METHODS:Experimental colitis was induced by administration of 5% DSS for 7 d,and assays performed on intestinal segments from the ileocecal valve to the anus.Colonic morphology was examined by hematoxylin and eosin staining.Colonic cytokines were determined by enzyme-linked immunosorbent assay.Myeloperoxidase(MPO) activity(indicator of inflammatory infiltration) was observed spectrophotometrically.Gut permeability was assessed by mucosal-to-serosal clearance of fluorescein isothiocyanate-conjugated dextran 4000(FD4) in everted gut sacs.The apoptosis of colonic epithelium was assessed by Hoechst-33342 staining.To further elucidate the role of CXCR4 in colonic inflammation,we also investigated the effect of AMD3100 on migration and cytokine production of isolated peripheral blood mononuclear cells(PBMCs).RESULTS:DSS-induced colitis was characterized by morphologic changes,as well as increased colonic cytokines,inflammatory infiltration,epithelial apoptosis,and intestinal permeability in mice.In AMD3100-treated mice,epithelial destruction,inflammatory infiltration,and submucosal edema were markedly reduced;colonic tumor necrosis factor-α(TNF-α),interleukin-6(IL-6) and interferon-γ(IFN-γ) levels,as well as MPO activity were significantly decreased.Increased intestinal permeability in DSS-treated mice was signif icantly reduced by AMD3100.The number of apoptotic cells in colitis mice was markedly increased after DSS administration,and decreased when treated with the CXCR4 antagonist AMD3100.In pre-activated PBMCs,CXCL12 stimulation signif icantly increased the migration of PBMCs,and was inhibited by AMD3100.Moderately increased TNF-α,IL-6,and IFN-γ from CXCL12-treated PBMCs were also reduced by AMD3100.CONCLUSION:The CXCR4 antagonist AMD3100 exerts therapeutic effects on experimental colitis by inhibiting colonic inflammation and enhancing epithelial barrier integrity.Xia, Xian-Ming Wang, Fang-Yu Xu, Wen-An Wang, Zhen-Kai Liu, Jiong Lu, You-Ke Jin, Xin-Xin Lu, Heng Shen, Yun-Zhu 2010World Journal of Gastroenterology2010,16,23:2

16Prognostic Nomogram for Patients with Hepatitis E Virus-related Acute Liver Failure:A Multicenter Study in China显示文摘Background and Aims:Timely and effective assessment scoring systems for predicting the mortality of patients with hepatitis E virus-related acute liver failure(HEV-ALF)are urgently needed.The present study aimed to establish an effective nomogram for predicting the mortality of HEV-ALF patients.Methods:The nomogram was based on a cross-sectional set of 404 HEV-ALF patients who were identified and enrolled from a cohort of 650 patients with liver failure.To compare the performance with that of the model for end-stage liver disease(MELD)scoring and CLIF-Consortiumacute-on-chronic liver failure score(CLIF-C-ACLFs)models,we assessed the predictive accuracy of the nomogram using the concordance index(C-index),and its discriminative ability using time-dependent receiver operating characteristics(td-ROC)analysis,respectively.Results:Multivariate logistic regression analysis of the development set carried out to predict mortality revealed that γ-glutamyl transpeptidase,albumin,total bilirubin,urea nitrogen,creatinine,international normalized ratio,and neutrophil-to-lymphocyte ratio were independent factors,all of which were incorporated into the new nomogram to predict the mortality of HEV-ALF patients.The area under the curve of this nomogram for mortality prediction was 0.671(95%confidence interval:0.602-0.740),which was higher than that of the MELD and CLIF-C-ACLFs models.Moreover,the td-ROC and decision curves analysis showed that both discriminative ability and threshold probabilities of the nomogram were superior to those of the MELD and CLIF-C-ACLFs models.A similar trend was observed in the validation set.Conclusions:The novel nomogram is an accurate and efficient mortality prediction method for HEV-ALF patients.Jian Wu Cuifen Shi Xinyu Sheng Yanping Xu Jinrong Zhang Xinguo Zhao Jiong Yu Xinhui Shi Gongqi Li Hongcui Cao Lanjuan Li 2021Journal of Clinical and Translational Hepatology2021,9,6:2

18XAFS study of Cu^(2+) in aqueous solution of CuBr_2显示文摘Copper ion is the essential microelement to many organisms. In this paper, the local structure of Cu2+ in CuBr2 aqueous solutions with different concentrations are investigated by using X-ray absorption fine structure (XAFS) technique. XANES (X-ray Absorption Near Edge Structure) spectra indicate that charge transfer from Br- to Cu2+ decreases with the solution concentration, which lead to a shift of the absorption edge. The shoulder appearing at the rising edge proves to be characteristic of a tetragonal distortion. The Fourier transform magnitudes of EXAFS (Extended X-ray absorption fine structure) data of Cu species suggest that more Cu-Br bonds may exist in high concentrations. A fivefold coordination configuration like a pyramid is used as the fitting parameters. From the analysis of the coordination numbers, the proportion of Cu-O and Cu-Br is 4:1 in the saturated solution. The Br atom is on the equatorial plane of the model. 19Characteristics of β-oxidative and reductive metabolism on the acyl side chain of cinnamic acid and its analogues in rats显示文摘Cinnamic acid and its analogues (pyragrel and ozagrel) undergo chain-shortened (β-oxidative) and reductive metabolism on acyl side chain. In this study, we characterized the β-oxidative and reductive metabolism on acyl side chain of cinnamic acid and its analogues using primary rat hepatocytes, hepatic mitochondrial, and microsomal systems. A compartmental model including parent compounds and metabolites was developed to characterize in vivo β-oxidative and reductive metabolism following an intravenous dose of parent compounds to rats. The ?tted total in vivo clearance values were further compared with the in vitro values predicted by the well-stirred model. We showed that hepatic microsomal CYP450s did not catalyze β-oxidative or reductive metabolism of the three compounds. Similar to β-oxidation of fatty acids,β-oxidative metabolism on their acyl side chain occurred mainly in mitochondria, which was highly dependent on ATP, CoA and NAD+. Fatty acids and NADH inhibited the β-oxidative metabolism. Reductive metabolism occurred in both mitochondria and microsomes. Reduction in mitochondria was ATP-, CoA-, and NAD(P)H-dependent and reversible, which was suppressed by enoyl reductase inhibitor triclosan. Reduction in microsomes was ATP-, CoA-, and NADPH-dependent but little affected by triclosan. Both plasma concentrations of β-oxidative metabolites and reductive metabolites were successfully ?tted using the compartmental model. The estimated total in vivo clearance values were consistent with those predicted from hepatocytes and organelles, implicating signi?cance of in vitro kinetics. 20Aberrant functional connectivity network in subjective memory complaint individuals relates to pathological biomarkers显示文摘Background:Individuals with subjective memory complaints(SMC)feature a higher risk of cognitive decline and clinical progression of Alzheimer’s disease(AD).However,the pathological mechanism underlying SMC remains unclear.We aimed to assess the intrinsic connectivity network and its relationship with AD-related pathologies in SMC individuals.Methods:We included 44 SMC individuals and 40 normal controls who underwent both resting-state functional MRI and positron emission tomography(PET).Based on graph theory approaches,we detected local and global functional connectivity across the whole brain by using degree centrality(DC)and eigenvector centrality(EC)respectively.Additionally,we analyzed amyloid deposition and tauopathy via florbetapir-PET imaging and cerebrospinal fluid(CSF)data.The voxel-wise two-sample T-test analysis was used to examine between-group differences in the intrinsic functional network and cerebral amyloid deposition.Then,we correlated these network metrics with pathological results.Results:The SMC individuals showed higher DC in the bilateral hippocampus(HP)and left fusiform gyrus and lower DC in the inferior parietal region than controls.Across all subjects,the DC of the bilateral HP and left fusiform gyrus was positively associated with total tau and phosphorylated tau181.However,no significant between-group difference existed in EC and cerebral amyloid deposition.Conclusion:We found impaired local,but not global,intrinsic connectivity networks in SMC individuals.Given the relationships between DC value and tau level,we hypothesized that functional changes in SMC individuals might relate to pathological biomarkers.Kaicheng Li Xiao Luo Qingze Zeng Yeerfan Jiaerken Xiaojun Xu Peiyu Huang Zhujing Shen Jingjing Xu Chao Wang Jiong Zhou Min-Ming Zhang the Alzheimer’s Disease Neuroimaging Initiative 2018Translational Neurodegeneration2018,7,1:2
