1Auranofin mitigates systemic iron overload and induces ferroptosis via distinct mechanisms显示文摘Iron homeostasis is essential for health;moreover,hepcidin-deficiency results in iron overload in both hereditary hemochromatosis and iron-loading anemia.Here,we identified iron modulators by functionally screening hepcidin agonists using a library of 640 FDA-approved drugs in human hepatic Huh7 cells.We validated the results in C57BL/6J mice and a mouse model of hemochromatosis(Hfe^(−/−)mice).Our screen revealed that the anti-rheumatoid arthritis drug auranofin(AUR)potently upregulates hepcidin expression.Interestingly,we found that canonical signaling pathways that regulate iron,including the Bmp/Smad and IL-6/Jak2/Stat3 pathways,play indispensable roles in mediating AUR’s effects.In addition,AUR induces IL-6 via the NF-κB pathway.In C57BL/6J mice,acute treatment with 5 mg/kg AUR activated hepatic IL-6/hepcidin signaling and decreased serum iron and transferrin saturation.Whereas chronically treating male Hfe^(−/−)mice with 5 mg/kg AUR activated hepatic IL-6/hepcidin signaling,decreasing systemic iron overload,but less effective in females.Further analyses revealed that estrogen reduced the ability of AUR to induce IL-6/hepcidin signaling in Huh7 cells,providing a mechanistic explanation for ineffectiveness of AUR in female Hfe^(−/−)mice.Notably,high-dose AUR(25 mg/kg)induces ferroptosis and causes lipid peroxidation through inhibition of thioredoxin reductase(TXNRD)activity.We demonstrate the ferroptosis inhibitor ferrostatin significantly protects liver toxicity induced by highdose AUR without comprising its beneficial effect on iron metabolism.In conclusion,our findings provide compelling evidence that TXNRD is a key regulator of ferroptosis,and AUR is a novel activator of hepcidin and ferroptosis via distinct mechanisms,suggesting a promising approach for treating hemochromatosis and hepcidin-deficiency related disorders.Lei Yang Hao Wang Xiang Yang Qian Wu Peng An Xi Jin Weiwei Liu Xin Huang Yuzhu Li Shiyu Yan Shuying Shen Tingbo Liang Junxia Min Fudi Wang 2020Signal Transduction and Targeted Therapy2020,5,1:5
2Transfer function based equivalent modeling method for wind farm显示文摘To effectively study the dynamics of power systems with large-scale wind farms(WFs), an equivalent model needs to be developed. It is well known that back-toback converters and their controllers are important for the dynamic responses of the wind turbine(WT) under disturbances. However, the detailed structure and parameters of the back-to-back converters and their controllers are usually unknown to power grid operators. Hence, it is difficult to build an accurate equivalent model for the WF using the component model-based equivalent modeling method. In this paper, a transfer function based equivalent modeling method for the WF is proposed. During modeling, the detailed structure and parameters of the WF are not required. The objective of the method is reproducing the output dynamics of the WF under the variation of the wind speed and power grid faults. A decoupled parameter-estimation strategy is also developed to estimate the parameters of the equivalent model. A WF that consists of 16 WTs is used to test the proposed equivalent model. Additionally,the proposed equivalent modeling method is applied to build the equivalent model for a real WF in Northwest China. 4Monitoring and Modeling the Effects of Groundwater Flow on Arsenic Transport in Datong Basin显示文摘Although arsenic-contaminated groundwater in the Datong Basin has been studied for more than 10 years, little has been known about the complex patterns of solute transport in the aquifer systems. Field monitoring and transient 3D unsaturated groundwater flow modeling studies were carried out on the riparian zone of the Sanggan River at the Datong Basin, northern China, to better understand the effects of groundwater flow on As mobilization and transport. The results indicate that irrigation is the primary factor in determining the groundwater flow paths. Irrigation can not only increase groundwater level and reduce horizontal groundwater velocity and thereby accelerate vertical and horizontal groundwater exchange among sand, silt and clay formations, but also change the HSconcentration, redox conditions of the shallow groundwater. Results of net groundwater flux estimation suggest that vertical infiltration is likely the primary control of As transport in the vadose zone, while horizontal water exchange is dominant in controlling As migration within the sand aquifers. Recharge water, including irrigation return water and flushed saltwater, travels downward from the ground surface to the aquifer and then nearly horizontally across the sand aquifer. The maximum value of As enriched in the riparian zone is roughly estimated to be 1 706.2 mg·d-1 for a horizontal water exchange of 8.98 m3·d-1 close to the river and an As concentration of 190 μg·L-1.Qian Yu Yanxin Wang Rui Ma Chunli Su Ya Wu Junxia Li 2014Journal of Earth Science2014,25,2:1
