2AMPKα1 regulates Idh2 transcription through H2B O-GlcNAcylation during brown adipogenesis显示文摘AMP-activated protein kinase (AMPK) is indispensable for the development and maintenance of brown adipose tissue (BAT), and its activity is inhibited due to obesity. The isocitrate dehydrogenase 2 (IDH2) is a mitochondrial enzyme responsible for the production of α-ketoglutarate, a key intermediate metabolite integrating multiple metabolic processes. We previously found that AMPKα1 ablation reduced cellular α-ketoglutarate concentration during brown adipocyte differentiation, but the effect of AMPKα1 on Idh2 expression remains undefined. In the present study, mouse C3H10T1/2 cells were transfected with Idh2-CRISPR/Cas9, and induced to brown adipogenesis. Our data suggested that brown adipogenesis was compromised due to IDH2 deficiency in vitro, which was accompanied by down-regulation of PR-domain containing 16. Importantly, the IDH2 content was reduced in brown stromal vascular cells (BSVs) separated from AMPKα1 knockout (KO) BAT, which was associated with lower contents of histone 2B (H2B) O-GlcNAcylation and monoubiquitination. Furthermore, both GlcNAcylated-H2B (S112) and ubiquityl-histone 2B (K120) contents in the Idh2 promoter were decreased in AMPKα1 KO BSVs. Meanwhile, ectopic O-linked N-acetylglucosamine transferase (OGT) expression was positively correlated with Idh2 expression, while OGT (T444A) mutation abolished the regulatory effect of AMPKα1 on Idh2. In vivo, reduced AMPKα1 activity and lower IDH2 abundance were observed in BAT of obese mice when compared with those in control mice. 3Paleogeomorphy evolution of the Ordovician weathering crust and its implication for reservoir development,eastern Ordos Basin显示文摘Gas reservoir development of the Ordovician weathering crust in the Ordos Basin is closely controlled by the pre-Carboniferous paleogeomorphy.Previous studies show that the paleogeomorphy is high in the west and low in the east,and the karst highland,karst slope and karst basin are developed from west to east.With further exploration in recent years,many karst breccia that represent strong karstification,are found in the east area which previously is considered to be the Ordovician karst basin.Thus,it is necessary to revaluate controlling factors of karst paleogeomorphy development from a viewpoint of the dynamic paleogeomorphy evolution,to investigate the paleogeomorphy evolution of the Ordovician weathering crust in geological history and guide further research and prediction of development law of reservoir spaces.In order to reconstruct the paleogeomorphy of the weathering crust in the top of Ordovician in the east Ordos Basin,paleogeography,thickness of residual strata and paleokarst characteristics are well studied.The result shows that a wide range of paleokarst highland occurred in the central to east part of Ordos Basin in the early period,and the karst reservoir spaces were formed;but in the late period,the east part of the basin gradually evolved into the paleokarst basin,the current pre-Carboniferous paleogeomorphy was thus formed,and the dissolution reservoir spaces formed in the early period were mostly filled,accordingly the reservoirs were tight.However fracture networks formed by cave collapse connect intercrystalline pores of dolomite,therefore,the reservoirs can still be well preserved locally and are worthy targets for hydrocarbon exploration.Xinshan Wei Junfeng Ren Junxing Zhao Daofeng Zhang Shunshe Luo Liubin Wei Juanping Chen 2018Petroleum Research2018,3,1:2
4A high-quality sponge gourd(Luffa cylindrica)genome显示文摘Sponge gourd(Luffa cylindrica)is an important cultivated vegetable and medicinal plant in the family Cucurbitaceae.In this study,a draft genome sequence of the sponge gourd inbred line P93075 was analyzed.Using Illumina,PacBio,and 10×Genomics sequencing techniques as well as new assembly techniques such as FALCON and chromatin interaction mapping(Hi-C),a chromosome-scale genome of approximately 656.19 Mb,with an N50 scaffold length of 48.76 Mb,was generated.From this assembly,25,508 protein-coding gene loci were identified,and 63.81%of the whole-genome consisted of transposable elements,which are major contributors to the expansion of the sponge gourd genome.According to a phylogenetic analysis of conserved genes,the sponge gourd lineage diverged from the bitter gourd lineage approximately 41.6 million years ago.Additionally,many genes that respond to biotic and abiotic stresses were found to be lineage specific or expanded in the sponge gourd genome,as demonstrated by the presence of 462 NBS-LRR genes,a much greater number than are found in the genomes of other cucurbit species;these results are consistent with the high stress resistance of sponge gourd.Collectively,our study provides insights into genome evolution and serves as a valuable reference for the genetic improvement of sponge gourd.Haibin Wu Gangjun Zhao Hao Gong Junxing Li Caixia Luo Xiaoli He Shaobo Luo Xiaoming Zheng Xiaoxi Liu Jinju Guo Junqiu Chen Jianning Luo 2020Horticulture Research2020,7,1:2
5Astragalus polysaccharide regulates brown adipogenic differentiation through miR-1258-5p-modulated cut-like homeobox 1 expression显示文摘Astragalus polysaccharide(APS)is the major natural active component of Astragalus membranaceus,which has been recognized as one of the most popular herbal medicines worldwide.Enhancing the formation and function of brown adipose tissue increases energy expenditure and hence may potentially be used against obesity and type 2 diabetes.The aim of the present study was to explore the effect and mechanism of APS on brown adipocyte formation.Mouse C3H10T 1/2 cells were subject to APS,and both proliferation and brown adipogenic differentiation were determined.The results showed that APS exhibits a decreased proliferation ability,which is accompanied by downregulated proliferating cell nuclear antigen,cyclin D1,and cyclin-dependent kinase 4.APS promotes the differentiation of C3H10T 1/2 cells into brown adipocytes and induces the expressions of key brown adipogenic transcriptional factors,including CCAAT/enhancer-binding proteinβ,uncoupling protein 1,and PR domain-containing 16.Importantly,APS enables insulin sensitization in brown adipocytes,which may proceed through activation of the canonical phosphatidylinositol-3-kinase(PI3K)/protein kinase B(AKT)signaling pathway and AMP-activated protein kinase(AMPK).Furthermore,the level of cut-like homeobox 1(CUX1)is positively related to brown adipogenic differentiation,while APS regulates Cux1 expression through interaction with miR-1258-5p.Notably,the promotional effect of APS on brown adipogenic differentiation was abolished by Cux1 knockout.Collectively,our results suggest that APS enhances the differentiation of C3H10T 1/2 cells into brown adipocytes through regulating Cux1 via miR-1258-5p.Yuxin Cao Buhao Deng Shihe Zhang Hongmei Gao Pengkang Song Jianxin Zhang Junxing Zhao 2021Acta Biochimica et Biophysica Sinica2021,53,12:1

8Exp2 polymorphisms associated with variation for fiber quality properties in cotton(Gossypium spp.)显示文摘Plant expansins are a group of extracellular proteins thought to affect the quality of cotton fibers. Previous expression profile analysis revealed that six Expansin A genes are present in cotton, of which two(GhExp1 and GhExp2) produce transcripts that are specific to the developing cotton fiber. To identify the phenotypic function of Exp2, and to determine whether nucleotide variation among alleles of Exp2 affects fiber quality, candidate gene association mapping was conducted. Gene-specific primers were designed to amplify the Exp2 gene. By amplicon sequencing, the nucleotide diversity of Exp2 was investigated across92 accessions(including 7 Gossypium arboreum, 74 Gossypium hirsutum, and 11 Gossypium barbadense accessions) with different fiber qualities. Twenty-six SNPs and seven InDels including 14 from the coding region of Exp2 were detected, forming twelve distinct haplotypes in the cotton collection. Among the 14 SNPs in the coding region, five were missense mutations and nine were synonymous nucleotide changes. The average SNP/InDel per nucleotide ratio was 2.61%(one SNP per 39 bp), with 1.81 and 3.87% occurring in coding and non-coding regions, respectively. Nucleotide and haplotype diversity across the entire Exp2 region was 0.00603(π) and 0.844, respectively, and diversity in non-coding regions was higher than that in coding regions. For linkage disequilibrium(LD), the mean r2 value for all polymorphism loci pairs was 0.48, and LD did not decay over 748 bp. Based on132 simple sequence repeat(SSR) loci evenly covering 26 chromosomes, the population structure was estimated, and the accessions were divided into seven groups that agreed well with their genomic origin and evolutionary history. A general linear model was used to calculate the Exp2-wide diversity–trait associations of 5 fiber quality traits, considering population structure(Q). Four SNPs in Exp2 were associated with at least one of the fiber quality traits, but not with fiber elongation. The highest positive effect on UHML and STR was observed for haplotype Hap_6 of Exp2. There was a significant association of Exp2 with fiber quality traits. There were many haplotypes in the Exp2 region, of which the most favorable was Hap_6. 10Vitamin A injection at birth improves muscle growth in lambs显示文摘Vitamin A and its metabolite,retinoic acid(RA)play important roles in regulating skeletal muscle development.This study was conducted to investigate the effects of early intramuscular vitamin A injection on the muscle growth of lambs.A total of 16 newborn lambs were given weekly intramuscular injections of corn oil(control group,n=8)or 7,500 IU vitamin A palmitate(vitamin A group,n=8)from birth to 3 wk of age(4 shots in total).At 3 wk of age and weaning,biceps femoris muscle samples were taken to analyze the effects of vitamin A on the myogenic capacity of skeletal muscle cells.All lambs were slaughtered at 8 months of age.The results suggest that vitamin A treatment accelerated the growth rate of lambs and increased the loin eye area(P<0.05).Consistently,vitamin A increased the diameter of myofibers in longissimus thoracis muscle(P<0.01)and increased the final body weight of lambs(P<0.05).Vitamin A injection did not change the protein kinase B/mammalian target of rapamycin and myostatin signaling(P>0.05).Moreover,vitamin A upregulated the expression of PAX7(P<0.05)and the myogenic marker genes including MYOD and MYOG(P<0.01).The skeletal muscle-derived mononuclear cells from vitamin A-treated lambs showed higher expression of myogenic genes(P<0.05)and formed more myotubes(P<0.01)when myogenic differentiation was induced in vitro.In addition,in vitro analysis showed that RA promoted myogenic differentiation of the skeletal muscle-derived mononuclear cells in the first 3 d(P<0.05)but not at the later stage(P>0.05)as evidenced by myogenic gene expression and fusion index.Taken together,neonatal intramuscular vitamin A injection promotes lamb muscle growth by promoting the myogenic potential of satellite cells.Pengkang Song Xiaoyou Chen Jiamin Zhao Qiang Li Xinrui Li Yu Wang Bo Wang Junxing Zhao 2023Animal Nutrition2023,,3:0
11S-wave velocity inversion and prediction using a deep hybrid neural network显示文摘The S-wave velocity is a critical petrophysical parameter in reservoir description,prestack seismic inversion,and geomechanical analysis.However,obtaining the S-wave velocity from field measurements is difficult.When no measured Swave data are available,petrophysical modelling provides the most accurate S-wave velocity prediction.However,because of the complexity of underground geological structures and diversity of rock minerals,the prediction results of petrophysical modelling are easily affected by factors such as the cognition and experience of the modeller.Therefore,the development of novel robust and simple S-wave velocity inversion and prediction methods independent of the modeller is critical.Inspired by ensemble learning and based on the geologic sedimentation law of reservoirs and their characteristics in logging response,an Swave velocity inversion and prediction method based on deep hybrid neural network was developed by combining the classical convolution neural network(CNN)with the long short-term memory(LSTM)network.Considering the conventional logging data such as acoustic and density as the input in the proposed method,the CNN was used to establish the nonlinear mapping relationship between the input data and S-wave velocity,and the LSTM network was used to integrate the vertical variation trend of the stratum.Thus,intelligent data-driven inversion and prediction of the S-wave velocity were realised.The experimental results revealed that the proposed method exhibited a strong generalisation ability and could obtain prediction results comparable to those of petrophysical modelling with a single-well data set for training.Thus,a novel methodology for robust and convenient S-wave velocity prediction was devised.The proposed method has considerable academic and application implications.Jun WANG Junxing CAO Shuang ZHAO Qiaomu QI 2022Science China Earth Sciences2022,65,4:0
12Vitamin A regulates mitochondrial biogenesis and function through p38 MAPK‑PGC‑1α signaling pathway and alters the muscle fiber composition of sheep显示文摘Background Vitamin A(VA)and its metabolite,retinoic acid(RA),are of great interest for their wide range of physiological functions.However,the regulatory contribution of VA to mitochondrial and muscle fiber composition in sheep has not been reported.Method Lambs were injected with 0(control)or 7,500 IU VA palmitate into the biceps femoris muscle on d 2 after birth.At the age of 3 and 32 weeks,longissimus dorsi(LD)muscle samples were obtained to explore the effect of VA on myofiber type composition.In vitro,we investigated the effects of RA on myofiber type composition and intrinsic mechanisms.Results The proportion of type I myofiber was greatly increased in VA-treated sheep in LD muscle at harvest.VA greatly promoted mitochondrial biogenesis and function in LD muscle of sheep.Further exploration revealed that VA elevated PGC-1αmRNA and protein contents,and enhanced the level of p38 MAPK phosphorylation in LD muscle of sheep.In addition,the number of type I myofibers with RA treatment was significantly increased,and type IIx myofibers was significantly decreased in primary myoblasts.Consistent with in vivo experiment,RA significantly improved mitochondrial biogenesis and function in primary myoblasts of sheep.We then used si-PGC-1αto inhibit PGC-1αexpression and found that si-PGC-1αsignificantly abrogated RA-induced the formation of type I myofibers,mitochondrial biogenesis,MitoTracker staining intensity,UQCRC1 and ATP5A1 expression,SDH activity,and enhanced the level of type IIx muscle fibers.These data suggested that RA improved mitochondrial biogenesis and function by promoting PGC-1αexpression,and increased type I myofibers.In order to prove that the effect of RA on the level of PGC-1αis caused by p38 MAPK signaling,we inhibited the p38 MAPK signaling using a p38 MAPK inhibitor,which significantly reduced RA-induced PGC-1αand MyHC I levels.Conclusion VA promoted PGC-1αexpression through the p38 MAPK signaling pathway,improved mitochondrial biogenesis,and altered the composition of muscle fiber type.Pengkang Song Jiamin Zhao Fanqinyu Li Xiaoyi Zhao Jinxin Feng Yuan Su Bo Wang Junxing Zhao 2024Journal of Animal Science and Biotechnology2024,15,2:0
13Distribution and depositional model of microbial carbonates in the Ordovician middle assemblage,Ordos Basin,NW China显示文摘Based on outcrop profiles,drilling cores,cast thin sections etc.,the types,microfacies combinations and distribution pattern of microbial carbonates in the Ordovician middle assemblage of the mid-eastern Ordos Basin have been systematically analyzed.The middle assemblage of Ordovician in the mid-eastern Ordos Basin has microbial carbonates formed by the calci-fication of cyanobacteria,including microbial biostromes and microbial mounds made of stromatolites,thrombolites,and on-colites.The distribution of the carbonates shows obvious“stratum-control”and“regional”characteristics.The microbial bio-stromes 2–3 m thick each are controlled by sequence cycles and sedimentary facies changes,and were mainly formed in the tidal flat environment during the depositional stages of the Ma56 and Ma55 sub-members.The microbial biostrome in the Ma55 sub-member occurring near the carbonate-evaporite transition interface in the early stage of the transgression is distributed mainly in the Mizhi subsag in the eastern part of the basin;the microbial biostrome in the Ma56 sub-member turns up near the carbonate-evoporite transition zone in ring shape in the east of the central uplift.The ancient landform had noticeable control on the distribution of microbial mounds.The microbial mounds or mound-shoal complexes developing mainly during the de-positional stages of Ma57_Ma510 sub-members are about 15–25 m thick in single layer and distributed largely in the Wushenqi-Jingbian paleouplift.The development model of the microbial carbonate rocks shows that the carbonate-evaporite lithologic transition zone and the Wushenqi-Jingbian paleouplift are favorable exploration zones of microbial carbonates in the Ordovician middle assemblages.WEI Liubin ZHAO Junxing SU Zhongtang WEI Xinshan REN Junfeng HUANG Zhengliang WU Chunying 2021Petroleum Exploration and Development2021,48,6:0
