| 2 | Epac2-deficiency leads to more severe retinal swelling, glial reactivity and oxidative stress in transient middle cerebral artery occlusion induced ischemic retinopathy显示文摘Ischemia occurs in diabetic retinopathy with neuronal loss, edema, glial cell reactivity and oxidative stress. Epacs, consisting of Epac1 and Epac2, are c AMP mediators playing important roles in maintenance of endothelial barrier and neuronal functions. To investigate the roles of Epacs in the pathogenesis of ischemic retinopathy, transient middle cerebral artery occlusion(t MCAO) was performed on Epac1-deficient(Epac1-/-) mice, Epac2-deficient(Epac2-/-) mice, and their wild type counterparts(Epac1+/+ and Epac2+/+). Two-hour occlusion and 22-hour reperfusion were conducted to induce ischemia/reperfusion injury to the retina. After t MCAO, the contralateral retinae displayed similar morphology between different genotypes. Neuronal loss, retinal edema and increase in immunoreactivity for aquaporin 4(AQP4), glial fibrillary acidic protein(GFAP), peroxiredoxin 6(Prx6) were observed in ipsilateral retinae. Epac2-/- ipsilateral retinae showed more neuronal loss in retinal ganglion cell layer, increased retinal thickness and stronger immunostaining of AQP4, GFAP, and Prx6 than those of Epac2+/+. However, Epac1-/- ipsilateral retinae displayed similar pathology as those in Epac1+/+ mice. | 5 | Tp53 gene mediates distinct dopaminergic neuronal damage in different dopaminergic neurotoxicant models显示文摘Tp53, a stress response gene, is involved in diverse cell death pathways and its activation is implicated in the pathogenesis of Parkinson's disease. However, whether the neuronal Tp53 protein plays a direct role in regulating dopaminergic(DA) neuronal cell death or neuronal terminal damage in different neurotoxican models is unknown. In our recent studies, in contrast to the global inhibition of Tp53 function by phar macological inhibitors and in traditional Tp53 knock-out mice, we examined the effects of DA-specific Tp53 gene deletion after 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine and methamphetamine exposure Our data suggests that the Tp53 gene might be involved in both neuronal apoptosis and neuronal termi nal damage caused by different neurotoxicants. Additional results from other studies also suggest that as a master regulator of many pathways that regulate apoptosis and synaptic terminal damage, it is possible tha Tp53 may function as a signaling hub to integrate different signaling pathways to mediate distinctive targe pathways. Tp53 protein as a signaling hub might be able to evaluate the microenvironment of neurons assess the forms and severities of injury incurred, and determine whether apoptotic cell death or neuro nal terminal degeneration occurs. | 6 | Characteristics of annular impinging jets with/without swirling flow by short guide vanes显示文摘Annular jets impinging on a uniformly heated flat plate with or without swirling flow by short guide vanes are experimentally characterized. With the Reynolds number fixed at a relatively low value, the variation of jet flow structures with impinging distance is characterized using the technique of particle image velocimetry (PIV). Correspondingly, the distributions of wall pressure and heat transfer on the plate are measured. At sufficiently large impinging distances, without swirling flow, the obtained flow and wall pressure/heat transfer data are consistent with the classical observation for a conventional annular impinging jet, showing the transition from annular impinging jet flow to single circular impinging jet-like flow. In contrast, no such transition occurs in the presence of flow turning by short guide vanes. At short and intermediate impinging distances, flow turning causes more non-uniform distributions of wall pressure and heat transfer on the target plate and the local heat transfer rates higher than those of the conventional annular jet. This is attributed to the vortical flow structures shed and convected downstream from the short guide vanes. | 8 | Enhancement of Aggression Induced by Isolation Rearing is Associated with a Lack of Central Serotonin显示文摘Isolation rearing(IR) enhances aggressive behavior, and the central serotonin(5-hydroxytryptamine,5-HT) system has been linked to IR-induced aggression.However, whether the alteration of central serotonin is the cause or consequence of enhanced aggression is still unknown. In the present study, using mice deficient in central serotonin Tph2-/-and Lmx1 b-/-, we examined the association between central serotonin and aggression with or without social isolation. We demonstrated that central serotonergic neurons are critical for the enhanced aggression after IR. 5-HT depletion in wild-type mice increased aggression. On the other hand, application of 5-HT in Lmx1 b-/-mice inhibited the enhancement of aggression under social isolation conditions. Dopamine was downregulated in Lmx1 b-/-mice. Similar to 5-HT, L-DOPA decreased aggression in Lmx1 b-/-mice. | 11 | Hydrogen sulfide-induced enhancement of gastric fundus smooth muscle tone is mediated by voltagedependent potassium and calcium channels in mice显示文摘AIM:To investigate the effect of hydrogen sulfide(H2S)on smooth muscle motility in the gastric fundus.METHODS:The expression of cystathionineβ-synthase(CBS)and cystathionineγ-lyase(CSE)in cultured smooth muscle cells from the gastric fundus was examined by the immunocytochemistry technique.The tension of the gastric fundus smooth muscle was recorded by an isometric force transducer under the condition of isometric contraction with each end of the smooth muscle strip tied with a silk thread.Intracellular recording was used to identify whether hydrogen sulfide affects the resting membrane potential of the gastric fundus in vitro.Cells were freshly separated from the gastric fundus of mice using a variety of enzyme digestion methods and whole-cell patch-clamp technique was used to find the effects of hydrogen sulfide on voltage-dependent potassium channel and calcium channel.Calcium imaging with fura-3AM loading was used to investigate the mechanism by which hydrogen sulfide regulates gastric fundus motility in cultured smooth muscle cells.RESULTS:We found that both CBS and CSE were expressed in the cul tured smooth muscle cel ls from the gastric fundus and that H2S increased the smooth muscle tension of the gastric fundus in mice at low concentrations.In addition,nicardipine and aminooxyacetic acid(AOAA),a CBS inhibitor,reduced the tension,whereas Nω-nitro-L-arginine methyl ester,a nonspecific nitric oxide synthase,increased the tension.The AOAA-induced relaxation was significantly recovered by H2S,and the Na HS-induced increase in tonic contraction was blocked by 5 mmol/L4-aminopyridine and 1μmol/L nicardipine.Na HS significantly depolarized the membrane potential and inhibited the voltage-dependent potassium currents.Moreover,Na HS increased L-type Ca2+currents and caused an elevation in intracellular calcium([Ca2+]i).CONCLUSION:These findings suggest that H2S may be an excitatory modulator in the gastric fundus in mice.The excitatory effect is mediated by voltagedependent potassium and L-type calcium channels. | Xiang-Min Meng Xu Huang Chun-Mei Zhang Dong-Hai Liu Hong-Li Lu Young-chul Kim Wen-Xie Xu | 2015 | World Journal of Gastroenterology2015,21,16: | 4 |

In this study, we aimed to determine whether GM1 alleviates neurotoxicity induced by moderate and high concentrations of propofol combined with remifentanil in the immature central nervous system. Hippocampal neural stem cells were isolated from newborn Sprague-Dawley rats and treated with remifentanil(5, 10, 20 ng/m L) and propofol(1.0, 2.5, 5.0 μg/m L), and/or GM1(12.5, 25, 50 μg/m L). GM1 reversed combined propofol and remifentanil-induced decreases in the percentage of 5-bromodeoxyuridine(+) cells and also reversed the increase in apoptotic cell percentage during neural stem cell proliferation and differentiation. However, GM1 with combined propofol and remifentanil did not affect β-tubulin(+) or glial fibrillary acidic protein(+) cell percentage during neural stem cell differentiation. | 13 | Analysis on Genetic Similarity of Japonica Rice Variety from Different Origins of Geography in the World显示文摘The genetic similarity and genetic difference among improved japonica rice varieties from different countries (or regions and organizations) were detected. The aim is to provide genetic basis to the breeding of japonica rice varieties. The genetic similarity and cluster of 313 improved japonica varieties from 20 countries (or regions and organizations) were analyzed using the SSR marker. With 34 SSR primers which were polymorphic and uniformly distributed in rice genome, totally 198 alleles were detected among these improved varieties with the average number of alleles per pair of primers of 5.8235. RM320, RM531, RM1, RM21, and RM336 located more alleles, which were 16, 13, 12, 10, and 10 respectively. RM320, RM336, RM286, RM531, and RM21 showed higher genetic diversity indexes, which were 2.3668, 2.0041, 1.9684, 1.9508, and 1.7203, respectively. The genetic similarity for improved japonica varieties among different countries (or regions and organizations) were ranged from 0.279 to 0.918, and the mean value was 0.653. The rice varieties from countries whose latitude and geography position were all nearer were clustered together with higher genetic similarity indexes. The rice varieties from countries who had more different latitude and far geography position were clustered separately with lower genetic similarity indexes. | 15 | Characterization and validation of somatic mutation spectrum to reveal heterogeneity in gastric cancer by single cell sequencing显示文摘Gastric cancer(GC) is a highly heterogeneous disease with multiple cellular types and poor prognosis.However, the cellular evolution and molecular basis of GC at the individual intra-tumor level has not been well demonstrated. We performed single-cell whole exome sequencing to detect somatic singlenucleotide variants(SNVs) and significantly mutated genes(SMGs) among 34 tumor cells and 9 normal cells from a patient with GC. The Complete Prediction for Protein Conformation(CPPC) approach directly predicting the folding conformation of the protein 3D structure with Protein Folding Shape Code, combined with functional experiments were used to confirm the characterization of mutated SMGs in GC cells. We identified 201 somatic SNVs, including 117 non-synonymous mutations in GC cells. Further analysis identified 24 significant mutated genes(SMGs) in single cells, for which a single amino acid change might affect protein conformation. Among them, two genes(CDC27 and FLG) that were mutated only in single cells but not in the corresponding tumor tissue, were recurrently present in another GC tissue cohort, and may play a potential role to promote carcinogenesis, as confirmed by functional characterization.
