1Saeng-Kankunbi-Tang(生肝健脾汤) Protects Liver against Oxidative Damage through Activation of ERK/Nrf2 Pathway显示文摘Objective: To investigate the cytoprotective effects of Saeng-kankunbi-tang(生肝健脾汤, SKT), a herbal prescription consisting of Artemisia capillaris and Alisma canaliculatum, and its underlying mechanism involved. Methods: In mice, blood biochemistry and histopathology were assessed in carbon tetrachloride(CCl4)-induced oxidative hepatic injury in vivo. The animal groups included vehicle-treated control, CCl4, SKT 500 mg/(kg·day) CCl4+SKT 200 or 500 mg/(kg·day). In Hep G2 cell, tert-butyl hydroperoxide(t BHP) induced severe oxidative stress and mitochondrial dysfunction in vitro. The cyto-protective effects of SKT were determined by 3-(4,5-dimethylthiazol)-2,5-diphenyltetrazolium bromide(MTT) assay, fluorescence activated cell sorting analysis and western blotting. Results: The administration of SKT prevented liver damage induced by CCl4 in mice, by inhibition of hepatocyte degeneration and inflammatory cell infiltration as well as plasma parameters such as alanine aminotransferase(P<0.01). Moreover, treatment with t BHP induced hepatocyte death and cellular reactive oxygen species production in hepatocyte cell line. However, SKT pretreatment(30–300 μg/m L) reduced this cell death and oxidative stress(P<0.01). More importantly, SKT inhibited the ability of t BHP to induce changes in mitochondrial membrane transition in cell stained with rhodamine 123(P<0.01). Furthermore, treatment with SKT induced extracellular signal-regulated kinases-mediated nuclear factor erythroid-2-related factor 2(Nrf2) activation as well as the expressions of heme oxygenase 1 and glutamate-cystein ligase catalytic, Nrf2 target genes. 5Surface plasmon excitation in semitransparent inverted polymer photovoltaic devices and their applications as label-free optical sensors显示文摘Herein,we report on surface plasmon(SP)-sensitive semitransparent inverted polymer photovoltaic(PV)devices that are based on multilayered material systems consisting of poly(3-hexylthiophene):fullerene-derivative bulk-heterojunction PV layers and thin gold or silver anodes.We demonstrate that these PV devices allow the simultaneous generation of both electrical power and SPs on their anodes for photoexcitation just above the optical absorption edge of the PV layers,resulting not only in attenuated total reflection,but also in attenuated photocurrent generation(APG)under the SP resonance(SPR)condition.Moreover,we also confirm that the biomolecular interaction of biotin–streptavidin on the PV devices can be precisely detected via apparent SPR angle shifts in the APG spectra,even without the need for complex attenuated total reflection configurations.We highlight our view that APG measurements made using these PV devices show great potential for the development of future generations of compact and highly sensitive SPR-based optical sensors.Byoungchoo Park Soo Hong Yun Chan Youn Cho Young Chan Kim Jung Chul Shin Hong Goo Jeon Yoon Ho Huh Inchan Hwang Ku Youn Baik Young In Lee Han Sup Uhm Guang Sup Cho Eun Ha Choi 2014Light(Science & Applications)2014,3,1:1

19Regulation of c-SMAC formation and AKT-mTOR signaling by the TSG101-IFT20 axis in CD4^(+)T cells显示文摘CD4^(+)T cells play major roles in the adaptive immune system,which requires antigen recognition,costimulation,and cytokines for its elaborate orchestration.Recent studies have provided new insight into the importance of the supramolecular activation cluster(SMAC),which comprises concentric circles and is involved in the amplification of CD4^(+)T cell activation.However,the underlying mechanism of SMAC formation remains poorly understood.Here,we performed single-cell RNA sequencing of CD4^(+)T cells left unstimulated and stimulated with anti-CD3 and anti-CD28 antibodies to identify novel proteins involved in their regulation.We found that intraflagellar transport 20(IFT20),previously known as cilia-forming protein,was upregulated in antibody-stimulated CD4^(+)T cells compared to unstimulated CD4^(+)T cells.We also found that IFT20 interacted with tumor susceptibility gene 101(TSG101),a protein that endocytoses ubiquitinated T-cell receptors.The interaction between IFT20 and TSG101 promoted SMAC formation,which led to amplification of AKT-mTOR signaling.However,IFT20-deficient CD4^(+)T cells showed SMAC malformation,resulting in reduced CD4^(+)T cell proliferation,aerobic glycolysis,and cellular respiration.Finally,mice with T-cell-specific IFT20 deficiency exhibited reduced allergen-induced airway inflammation.Thus,our data suggest that the IFT20-TSG101 axis regulates AKT-mTOR signaling via SMAC formation.Jiung Jeong In Kang Yumin Kim Keun Bon Ku Jang Hyun Park Hyun-Jin Kim Chae Won Kim Jeongwoo La Hi Eun Jung Hyeon Cheol Kim Young Joon Choi Jaeho Kim Joon Kim Heung Kyu Lee 2023Cellular & Molecular Immunology2023,20,5:0
