1Animal experiment and clinical study of effect of gamma-interferon on hepatic fibrosis显示文摘AIM To evaluate the antifibrotic effect ofdifferent doses of recombinant human Gamma-Interferon (IFN-γ) intwo rat models of hepaticfibrosis, and to observe its effect on moderatechronic hepatitis B virus fibrosis.METNODS Hepatic fibrosis was successfullyinduced in 150 and 196 rats by subcutaneousinjection of carbon tetrachloride (CCl4) andintraperitoneal injection of dimethylnitrosamine(DMN), respectively. Each of the two modeldose IFN-γ group (15 MU/kg per day, i.m. for 8group (1.67 MU/kg daily, i.m. for 8 weeks).Another group of 10 rats without any treatmentwas used as normal controls. At the end of theexperiment, semi-quantitative histopathologicalscores of inflammation and fibrosis, liver (αsmooth muscle actin (α-SMA) expression level,liver hydroxyl proline content and serumhyaluronic acid levels were compared. And 47medium chronic hepatitis B viral fibrosispatients were studied. They were given IFN-γtreatment, 100MU/day i.m. for the first threemonths and 100MU qod i.m. for the next sixmonths. Semi-quantitative pathological scoresof inflammation and fibrosis and serum hepaticfibrosis indices were compared within the 9months.RESULTS In animal experiment, thepathological fibrosis scores and liver hydroxylproline content were found to be significantlylower in rats treated with different doses of IFN-γ as compared with rats in fibrotic model groupinduced by either CCI4 or DMN, in a dose-dependent manner. For CCI4-induced model,pathological fibrosis scores in high, medium andIow doses IFN-γ groups were 5.10 ± 2.88, 7.70 ±3.53 and 8.00 ± 3.30, respectively, but the scorewas 14.60 ± 7.82 in fibrotic model group.Hydroxyl proline contents were 2.83 ± 1.18, 3.59± 1.22 and 4.80 ± 1.62, in the three IFN-γgroups, and 10.01 ± 3.23 in fibrotic model group.The difference was statistically significant(P<0.01). Similar results were found in DMN-induced model. Pathological fibrosis scoreswere 6.30±0.48, 8.10 ±2.72 and 8.30 ±2.58, inhigh, medium and Iow doses IFN-γ groups, and12.60 ± 3.57 in fibrotic model group. Hydroxylproline contents were 2.72 ± 0.58, 3.14 ± 0.71and 3.62 ± 1.02, in the three IFN-γ groups, and12.79 ± 1.54 in fibrotic model group. Thedifference was statistically significant(P<0.01). Serum hepatic fibrosis indicesdecreased significantly in the 47 patients afterIFN-γ treatment (HA: 433.38 ± 373.00 vs 281.57± 220.48; LN: 161.22± 41.02 vs 146.35 ± 44.67;PCⅢ: 192.59 ± 89.95 vs 156.98 ± 49.22; C-Ⅳ:156.30 ± 44.01 vs 139.14 ± 34.47) and thedifferences between the four indices weresignificant (P<0.05). Thirty-three patientsCONCLUSION All the three doses of IFN-γ areeffective in treating rat liver fibrosis induced byeither CCl4 or DMN, the higher the dose, thebetter the effect. 2Sleep and Cognitive Abnormalities in Acute Minor Thalamic Infarction显示文摘In order to characterize sleep and the cognitive patterns in patients with acute minor thalamic infarction(AMTI), we enrolled 27 patients with AMTI and 12 matched healthy individuals. Questionnaires about sleep and cognition as well as polysomnography(PSG) were performed on days 14 and 90 post-stroke. Compared to healthy controls, in patients with AMTI, hyposomnia was more prevalent; sleep architecture was disrupted as indicated by decreased sleep efficiency, increased sleep latency, and decreased non-rapid eye movement sleep stages 2 and 3;more sleep-related breathing disorders occurred; and cognitive functions were worse, especially memory. While sleep apnea and long-delay memory recovered to a large extent in the patients, other sleep and cognitive function deficit often persisted. Patients with AMTI are at an increased risk for hyposomnia, sleep structure disturbance,sleep apnea, and memory deficits. 5Biocompatibility and Immunotoxicology of the Preclinical Implantation of a Collagen-based Artificial Dermal Regeneration Matrix显示文摘Objective Graft rejection, with the possibility of a violent immune response, may be severe and life threatening. Our aim was to thoroughly investigate the biocompatibility and immunotoxicology of collagen-based dermal matrix(DM) before assessment in clinical trials. Methods DM was subcutaneously implanted in BALB/c mice in two doses to induce a potential immune response. The spleen and lymph nodes were assessed for shape, cell number, cell phenotype via flow cytometry, cell activation via CCK8 kit, Annexin V kit, and Ki67 immunostaining. Serum samples were used to measure antibody concentration by enzyme-linked immunosorbent assay. Local inflammation was analyzed by histology and immunohistochemistry staining. Data analysis was performed by one-way ANOVA and non-parametric tests. Results Our data illustrate that the spleen and lymph node sizes were similar between the negative control mice and mice implanted with DM. However, in the high-dose DM(DM-H) group, the total cell populations in the spleen and lymph nodes, T cells and B cells in the spleen had slight increases in prophase, and the low-dose DM(DM-L) group did not display gross abnormities. Moreover, DM-H initiated moderate cell activation and proliferation in the early phase post-immunization, whereas DM-L did not. Neither DM-H nor DM-L implantation noticeably increased IgM and IgG serum concentrations. Examination of the local cellular response revealed only benign cell infiltration and TNF-α expression in slides of DM in the early phase. Conclusion Overall, DM-H may have induced a benign temporary acute immune response post-implantation, whereas DM-L had quite low immunogenicity. 7Using a support vector machine method to predict the development indices of very high water cut oilfields显示文摘Because the oilfields in eastern China are in the very high water cut development stage, accurate forecast of oilfield development indices is important for exploiting the oilfields efficiently. Regarding the problems of the small number of samples collected for oilfield development indices, a new support vector regression prediction method for development indices is proposed in this paper. This method uses the principle of functional simulation to determine the input-output of a support vector machine prediction system based on historical oilfield development data. It chooses the kernel function of the support vector machine by analyzing time series characteristics of the development index; trains and tests the support vector machine network with historical data to construct the support vector regression prediction model of oilfield development indices; and predicts the development index. 8Influence of DCM dye doping on the magnetic field dependent electroluminescence in organic light emitting diodes显示文摘The DCM dye doped organic electroluminescence devices with structure of ITO/NPB/Alq 3 : DCM/Alq 3 /LiF/Al were fabricated. From 15 K to room temperature, the magnetic field dependent of electroluminescence (MEL) of devices was investigated. Our observations indicated that the MEL is composed of two effects in different regimes: a low field (0≤B≤40 mT) effect and a high field (B > 40 mT) effect. For undoped devices, the low field effect exhibits a rapid rising with the increasing field, and the high field effect shows a slow increase and gradually saturates at room temperature. For doped devices, the low field rapid increase is also present, whereas the high field effect displays a decrease with the increasing field. The larger the injection current is, the more apparent the high field decrease is. In addition, the doped device demonstrates less temperature dependence of the high field effect than undoped device, although the undoped devices also present high field decrease of electroluminescence at low temperatures (T≤150 K). 9Biomarker Records of Phytoplankton Productivity and Community Structure Changes During the Last 14000 Years in the Mud Area Southwest off Cheju Island, East China Sea显示文摘The ecological environment in the East China Sea(ECS)and the Yellow Sea(YS)has changed significantly due to sea-level rising and the Kuroshio incursion since the last deglaciation.In this study,biomarker records of core F10B from the mud area southwest off Cheju Island(MSWCI)were generated to evaluate phytoplankton productivity and community structure changes in response to environmental evolution during the last 14 kyr.The contents of diatom,dinoflagellate and haptophyte biomarkers(brassicasterol,dinosterol and C37alkenones)display similar trends,with increasing phytoplankton productivity during the last 14kyr due to the increased influences of the Kuroshio,and especially due to the eddy-induced upwelling during the late Holocene.On the other hand,the contents of terrestrial biomarkers(C28+C30+C32n-alkanols)and terrestrial organic matter(TOM)proxies(TMBR′and BIT)all reveal decreasing TOM input into the area around the sampling site for the 14 kyr,mostly due to sea-level ris-ing.Phytoplankton biomarker ratios reveal a shift from a haptophyte-dominated community at 6.2 2.5 kyr BP to a diatom-dominated community at 2.5 1.45 kyr BP,likely caused by a stronger cold eddy circulation system at 2.5 1.45 kyr BP in the MSWCI.YUAN Zineng XING Lei LI Li ZHANG Hailong XIANG Rong ZHAO Meixun 2013Journal of Ocean University of China2013,12,4:10
10Expert consensus on the evaluation and diagnosis of combat injuries of the Chinese People's Liberation Army显示文摘The accurate assessment and diagnosis of combat injuries are the basis for triage and treatment of combat casualties. A consensus on the assessment and diagnosis of combat injuries was made and discussed at the second annual meeting of the Professional Committee on Disaster Medicine of the Chinese People's Liberation Army(PLA). In this consensus agreement, the massive hemorrhage, airway, respiration, circulation and hypothermia(MARCH) algorithm, which is a simple triage and rapid treatment and field triage score, was recommended to assess combat casualties during the first-aid stage, whereas the abbreviated scoring method for combat casualty and the MARCH algorithm were recommended to assess combat casualties in level Ⅱ facilities. In level Ⅲ facilities, combined measures, including a history inquiry, thorough physical examination, laboratory examination, X-ray, and ultrasound examination, were recommended for the diagnosis of combat casualties. 12Autophagy activation and the mechanism of retinal microvascular endothelial cells in hypoxia显示文摘AIM: To explore the state of autophagy and related mechanisms in the murine retinal microvascular endothelial cells(RMECs) under hypoxia stimulation.METHODS: The murine RMECs were primarily cultured and randomly divided into three groups: hypoxia group(cultured in 1% O_2 environment), hypoxia+autophagy inhibition group [pretreated with 5 mmol/L 3-methyladenine(3-MA) for 4 h followed by incubation in 1% O_2] and control group(cultured under normoxic condition). The state of autophagy in RMECs was examined by assaying the turnover of light chain 3 B(LC3BB) and expression of Beclin-1, Atg3 and Atg5 proteins with Western blotting, by detecting formation of autophagosomes with transmission electron microscopy(TEM) and by counting the number of GFP+ puncta in RMECs. The protein levels of AMPK, P-AMPK, Akt, P-Akt, m-TOR and P-m TOR were also assayed by Western blotting.RESULTS: Primary murine RMECs were successfully cultured. Under hypoxic conditions, the ratio of LC3BB-Ⅱ/Ⅰ and the expression of Beclin-1, Atg3 and Atg5 proteins were increased when compared with the control group. In addition, the numbers of autophagosome and the GFP+ puncta were also increased under hypoxia. However, pretreatment with 3-MA obviously attenuated these changes in autophagy in RMECs under hypoxia. Protein expression of P-Akt and P-AMPK was increased but P-m TOR level was decreased in cells exposed to hypoxia. 13Secretory/releasing proteome-based identification of plasma biomarkers in HBV-associated hepatocellular carcinoma显示文摘For successful therapy, hepatocellular carcinoma (HCC) must be detected at an early stage. Herein, we used a proteomic approach to analyze the secretory/releasing proteome of HCC tissues to identify plasma biomarkers. Serum-free conditioned media (CM) were collected from primary cultures of cancerous tissues and surrounding noncancerous tissues. Proteomic analysis of the CM proteins permitted the identification of 1365 proteins. The enriched molecular functions and biological processes of the CM proteins, such as hydrolase activity and catabolic processes, were consistent with the liver being the most important metabolic organ. Moreover, 19% of the proteins were characterized as extracellular or membrane-bound. For validation, secretory proteins involved in transforming growth factor-β signaling pathways were validated in plasma samples. Alphafetoprotein (AFP), metalloproteinase (MMP)1, osteopontin (OPN), and pregnancy-specific beta-1-glycoprotein (PSG)9 were significantly increased in HCC patients. The overall performance of MMP1 and OPN in the diagnosis of HCC remained greater than that of AFP. In addition, this study represents the first report of MMP1 as a biomarker with a higher sensitivity and specificity than AFP. Thus, this study provides a valuable resource of the HCC secretome with the potential to investigate serological biomarkers. 14Effects of lipid-lowering agents on diabetic retinopathy: a Meta-analysis and systematic review显示文摘AIM: To clarify this controversy and to provide evidence for application of lipid lowering agents in treatment of diabetic retinopathy(DR).METHODS: We searched the databases of Pub Med, Embase and Cochrane Library Central Register of Controlled Trials(CENTRAL) and abstracts from main annual meetings up to January 1, 2017. Google scholar and ClinicalT rials.gov were also searched for unpublished relevant studies. We included randomized controlled trials(RCTs) that studied lipid-lowering agents in type 1 or type 2 diabetes in this Meta-analysis. The primary endpoint was the progression of DR, and the secondary endpoints included vision loss, development of diabetic macular edema(DME) and aggravation of hard exudates. The pooled odds ratios(OR)with corresponding 95% confidence intervals(95%CIs) were calculated.RESULTS: After systemic and manual literature search by two independent investigators, we included 8 RCTs from 7 published articles with 13 454 participants in this Meta-analysis. The results revealed that lipid-lowering drugs were associated with reduced risk in DR progression [OR=0.77(95%CI: 0.62, 0.96), P=0.02]. Lipid-lowering agents might have protective effect on DME compared to placebo, although the difference was not statistically significant [OR=0.60(95%CI: 0.34, 1.08), P=0.09]. However, no significant differences in the worsening of vision acuity [OR=0.96(95%CI: 0.81,1.14), P=0.64] and hard exudates [OR=0.50(95%CI:0.15, 1.74), P=0.28] were found between the lipidlowering drugs and the placebo groups.CONCLUSION: In DR patients, lipid-lowering agents show a protective effect on DR progression and might be associated with reduced risk in the development of DME. However, lipid-lowering agents have no effects on vision loss and hard exudates aggravation. 15Osthole decreases collagen Ⅰ/III contents and their ratio in TGF-β1-overexpressed mouse cardiac fibroblasts through regulating the TGF-β/Smad signaling pathway显示文摘The present study was designed to elucidate whether the mechanism by which osthole decreases collagenⅠ/III contents and their ratio is regulating the TGF-β/Smad signaling pathway in TGF-β1-overexpressed mouse cardiac fibroblasts(CFs). These CFs were cultured and treated with different concentrations of osthole. Our results showed that the TGF-β1 expression in the CFs transfected with that the recombinant expression plasmids pc DNA3.1(+)-TGF-β1 was significantly enhanced. After the CFs were treated with 1.25-5 μg·m L^(-1) of osthole for 24 h, the m RNA and protein expression levels of collagensⅠand III were reduced. The collagen Ⅰ/III ratio was also reduced. The m RNA and protein expression levels of TGF-β1, TβRⅠ, Smad2/3, P-Smad2/3, Smad4, and α-SMA were decreased, whereas the expression level of Smad7 was increased. These effects suggested that osthole could inhibit collagen Ⅰ and III expression and reduce their ratio via the TGF-β/Smad signaling pathway in TGF-β1 overexpressed CFs. 17MicroRNA-155 promotes the pathogenesis of experimental colitis by repressing SHIP-1 expression显示文摘AIM To explore the mechanism by which micro RNA-155(miR-155) regulates the pathogenesis of experimental colitis.METHODS A luciferase assay was performed to confirm the binding of miR-155 to the SHIP-1 3'-UTR. MiR-155 mimics, negative controls and SHIP-1 expression/knockdown vectors were established and then utilized in gain-and loss-of-function studies performed in raw264.7 cells and primary bone marrow-derived macrophages (BMDMs). Thereafter, dextran sulfate sodium(DSS)-induced colitis mouse model with or without antagomiR-155 treatment was established, and the levels of miR-155 and SHIP-1, as well as the pro-inflammatory capabilities, were measured by western blot, quantitative polymerase chain reaction, and immunohistochemistry.RESULTS MiR-155 directly bound to the 3'-UTR of SHIP-1 mRNA and induced a significant decrease in SHIP-1 expression in both raw264.7 cells and primary BMDMs. MiR-155 markedly promoted cell proliferation and proinflammatory secretions including IL-6, TNF-α, IL-1β, and IFN-γ, whereas these effects could be reversed by the restoration of SHIP-1 expression. In vivo studies showed that antagomiR-155 administration could alleviate DSS-induced intestinal inflammation in Balb/c mice. Moreover, significantly increased SHIP-1 expression, as well as decreased Akt activation and inflammatory response, were observed in the antagomiR-155-treated mice.CONCLUSION MiR-155 promotes experimental colitis by repressing SHIP-1 expression. 19Dynamically controlling terahertz wavefronts with cascaded metasurfaces显示文摘Dynamically controlling terahertz(THz)wavefronts in a designable fashion is highly desired in practice.However,available methods working at microwave frequencies do not work well in the THz regime due to lacking suitable tunable elements with submicrometer sizes.Here,instead of locally controlling individual meta-atoms in a THz metasurface,we show that rotating different layers(each exhibiting a particular phase profile)in a cascaded metadevice at different speeds can dynamically change the effective Jonesmatrix property of the whole device,thus enabling extraordinary manipulations on the wavefront and polarization characteristics of a THz beam impinging on the device.After illustrating our strategy based on model calculations,we experimentally demonstrate two proof-of-concept metadevices,each consisting of two carefully designed all-silicon transmissive metasurfaces exhibiting different phase profiles.Rotating two metasurfaces inside the fabricated devices at different speeds,we experimentally demonstrate that the first metadevice can efficiently redirect a normally incident THz beam to scan over a wide solid-angle range,while the second one can dynamically manipulate both the wavefront and polarization of a THz beam.Our results pave the way to achieving dynamic control of THz beams,which is useful in many applications,such as THz radar,and bio-and chemical sensing and imaging.Xiaodong Cai Rong Tang Haoyang Zhou Qiushi Li Shaojie Ma Dongyi Wang Tong Liu Xiaohui Ling Wei Tan Qiong He Shiyi Xiao Lei Zhou 2021Advanced Photonics2021,3,3:8
