2Distribution,Frequency and Variation of Stripe Rust Resistance Loci Yr10,Lr34/Yr18 and Yr36 in Chinese Wheat Cultivars显示文摘Wheat stripe rust is a devastating disease in many regions of the world.In wheat,49 resistance genes for stripe rust have been officially documented,but only three genes are cloned,including the race-specific resistance Yr10 candidate gene(Yr10_(CG)) and slow-rusting genes Lr34/Yr18(hereafter designated as Yr18) and Yr36.In this study,we developed gene-specific markers for these genes and used them to screen a collection of 659 wheat accessions,including 485 Chinese cultivars.Thirteen percent and eleven percent of the tested Chinese cultivars were positive for the markers for Yr10_(CG) and Yr18_(RH)(the resistant haplotype of Yr18),respectively,but none were positive for the Yr36 marker.Since there is a limited use of the Yr10 gene in Chinese wheat,the relatively high frequency of wheat varieties with the Yr10_(CG) marker suggests that the identity of the Yr10 gene is unknown.With regards to the Yr18 gene,29%of the tested cultivars that are used in the Middle and Lower Yangtze Valleys' winter wheat zone were positive for Yr18_(RH) markers.A non-functional allele of Yr18_(RH) was identified in 'Mingxian 169',a commonly used susceptible check for studying stripe rust.The data presented here will provide useful information for marker-assisted selection for wheat stripe rust resistance.Cuiling Yuan Hui Jiang Honggang Wang Kun Li Heng Tang Xianbin Li Daolin Fu 2012Journal of Genetics and Genomics2012,39,11:12
3A method for the production and expedient screening of CRISPR/Cas9-mediated non-transgenic mutant plants显示文摘Developing CRISPR/Cas9-mediated non-transgenic mutants in asexually propagated perennial crop plants is challenging but highly desirable.Here,we report a highly useful method using an Agrobacterium-mediated transient CRISPR/Cas9 gene expression system to create non-transgenic mutant plants without the need for sexual segregation.We have also developed a rapid,cost-effective,and high-throughput mutant screening protocol based on Illumina sequencing followed by high-resolution melting(HRM)analysis.Using tetraploid tobacco as a model species and the phytoene desaturase(PDS)gene as a target,we successfully created and expediently identified mutant plants,which were verified as tetra-allelic mutants.We produced pds mutant shoots at a rate of 47.5%from tobacco leaf explants,without the use of antibiotic selection.Among these pds plants,17.2%were confirmed to be non-transgenic,for an overall non-transgenic mutation rate of 8.2%.Our method is reliable and effective in creating non-transgenic mutant plants without the need to segregate out transgenes through sexual reproduction.This method should be applicable to many economically important,heterozygous,perennial crop species that are more difficult to regenerate.Longzheng Chen Wei Li Lorenzo Katin-Grazzini Jing Ding Xianbin Gu Yanjun Li Tingting Gu Ren Wang Xinchun Lin Ziniu Deng Richard J.McAvoy Frederick G.Gmitter Jr. Zhanao Deng Yunde Zhao Yi Li 2018Horticulture Research2018,5,1:12

5Abiogenic hydrocarbons in commercial gases from the Songliao Basin,China显示文摘This paper discusses the kinetic fractionation, composition and distribution characteristics of carbon and hydrogen isotopes for various alkane gases formed in different environments, by different mecha- nisms and from different sources in nature. It is demonstrated that the biodegradation or thermode- gradation of complex high-molecule sedimentary organic material can form microbial gas or thermogenic gas. The δ 13C1 value ranges from -110‰ to -50‰ for microbial gases but from -50‰ to -35‰ (even heavier) for thermogenic gases. Controlled by the kinetic isotope fractionation, both microbial and thermogenic gases have δ 13C and δ D values characterized by normal distribution, i.e. δ 13C1< δ 13C2< δ 13C3< δ 13C4 and δ DCH4< δ DC2H6< δ DC3H8<δ DC4H10, and by a positive correlation between the δ 13C and δ D values. Simple carbonbearing molecules (CH4, CO and CO2) can form abiogenic alkane gases via polymerization in the abiological chemical process in nature, with δ 13C1 heavier than -30‰. Moreover, controlled by the kinetic isotope fractionation, abiogenic alkane gases are characterized by a reverse distribution of δ 13C values and a normal trend of δ D values, namely δ 13C1> δ 13C2> δ 13C3> δ 13C4 and δ DCH4<δ DC2H6< δ DC3H8< δ DC4H10. The δ 13C values and δ D values are negatively correlated. Natural gases from 26 commercial gas wells distributed in the Xujiaweizi and Yingshan-Miaotaizi faulted depressions in the Songliao Basin, China, show δ13C1 values ranging from -30.5‰ to -16.7‰ with a very narrow δ D range between -203‰―-196‰. These gases are characterized by a reverse distribution of δ 13C values but a normal distribution of δ D values, and a negative correlation between their δ 13C and δ D values, indicating an abiological origin. The present study has revealed that abiogenic hydrocarbons not only exist in nature but also can make significant contribution to commercial gas reserviors. It is estimated that the reserve volume of alkane gases with abiogenic characteristics in these 26 gas wells in the Songliao Basin is over 500×108 m3. 8ROS-responsive cyclodextrin nanoplatform for combined photodynamic therapy and chemotherapy of cancer显示文摘Cyclodextrin(CD) has special spatial structure and well biological safety,so it has been widely used for constructing CD-based na noplatforms.Through functionalization,cyclodextrin can form various stimulusresponse nanoplatforms,such as pH,temperature,redox,light and magnetic fields.In this study,we designed a highly sensitive reactive oxygen species(ROS)-responsive polymer PCP which encapsulated doxorubicin(DOX) and purpurin 18(P18) to achieve the syne rgy of photodynamic and chemotherapy.The high content of reactive oxygen species(ROS) in the tumor microenvironment(TME) triggers the cleavage of the borate bond of MPEG-CD-PHB(PCP),thereby promoting the re lease of drugs.When irradiated with nea rinfrared laser,the photosensitizer P18 released by polymer micelles can produce reactive oxygen species to promote cell apoptosis.Compared with monotherapy,a series of experiments confirmed that our micelles had enhanced anti-cancer activity.This work was beneficial to the design of ROS-responsive materials and provides an effective strategy for the application of collaborative anti-tumor therapy.Die Jia Xianbin Ma Yi Lu Xinyi Li Shengxin Hou Yuan Gao Peng Xue Yuejun Kang Zhigang Xu 2021Chinese Chemical Letters2021,32,1:4

10Comparative metabolic profiling of Vitis amurensis and Vitis vinifera during cold acclimation显示文摘Vitis amurensis is a wild Vitis plant that can withstand extreme cold temperatures.However,the accumulation of metabolites during cold acclimation(CA)in V.amurensis remains largely unknown.In this study,plantlets of V.amurensis and V.vinifera cv.Muscat of Hamburg were treated at 4℃ for 24 and 72 h,and changes of metabolites in leaves were detected by gas chromatography coupled with time-of-flight mass spectrometry.Most of the identified metabolites,including carbohydrates,amino acids,and organic acids,accumulated in the two types of grape after CA.Galactinol,raffinose,fructose,mannose,glycine,and ascorbate were continuously induced by cold in V.amurensis,but not in Muscat of Hamburg.Twelve metabolites,including isoleucine,valine,proline,2-oxoglutarate,and putrescine,increased in V.amurensis during CA.More galactinol,ascorbate,2-oxoglutarate,and putrescine,accumulated in V.amurensis,but not in Muscat of Hamburg,during CA,which may be responsible for the excellent cold tolerance in V.amurensis.The expression levels of the genes encodingβ-amylase(BAMY),galactinol synthase(GolS),and raffinose synthase(RafS)were evaluated by quantitative reverse transcription-PCR.The expression BAMY(VIT_02s0012 g00170)and RafS(VIT_05s0077 g00840)were primarily responsible for the accumulation of maltose and raffinose,respectively.The accumulation of galactinol was attributed to different members of GolS in the two grapes.In conclusion,these results show the inherent differences in metabolites between V.amurensis and V.vinifera under CA.Fengmei Chai Wenwen Liu Yue Xiang Xianbin Meng Xiaoming Sun Cheng Cheng Guotian Liu Lixin Duan Haiping Xin Shaohua Li 2019Horticulture Research2019,6,1:3

12Instrument for insulation conditions detection of roof insulator on high-speed-train显示文摘With the rapid development of high-speed-railway,environment around high voltage device on train roof becomes very complicated. Most train accidents happened due to occurrence of flashover on roof insulator,but the insulation condition estimation of insulator in such environment is much difficult. To ensure the insulation property of electric equipment,and guarantee the operation safety of high-speed-train,here established an instrument with high reliability which can on-line monitor insulation condition of roof insulator and give out advanced alarm before the incipient insulator flashover. The instrument consists of three parts,Data Acquisition & Sensor,Data Processing and Back Processing. Anti-interference and protection methods are processed to Rogowski coil sensor for better leakage current signal. To avoid the fluctuation from railway power supply,four modules are set to filter the power supply waveform. Through laboratory measurement,it is shown that the leakage current and the impedance angle can be detected by the instrument accurately. 13Hybrid Dirac semimetal-based photodetector with efficient low-energy photon harvesting显示文摘Despite the considerable effort,fast and highly sensitive photodetection is not widely available at the low-photon-energy range(~meV)of the electromagnetic spectrum,owing to the challenging light funneling into small active areas with efficient conversion into an electrical signal.Here,we provide an alternative strategy by efficiently integrating and manipulating at the nanoscale the optoelectronic properties of topological Dirac semimetal PtSe_(2)and its van der Waals heterostructures.Explicitly,we realize strong plasmonic antenna coupling to semimetal states near the skin-depth regime(λ/10^(4)),featuring colossal photoresponse by in-plane symmetry breaking.The observed spontaneous and polarization-sensitive photocurrent are correlated to strong coupling with the nonequilibrium states in PtSe_(2)Dirac semimetal,yielding efficient light absorption in the photon range below 1.24 meV with responsivity exceeding∼0.2 A/W and noise-equivalent power(NEP)less than~38 pW/Hz^(0.5),as well as superb ambient stability.Present results pave the way to efficient engineering of a topological semimetal for high-speed and low-energy photon harvesting in areas such as biomedical imaging,remote sensing or security applications.Lin Wang Li Han Wanlong Guo Libo Zhang Chenyu Yao Zhiqingzi Chen Yulu Chen Cheng Guo Kaixuan Zhang Chia-Nung Kuo Chin Shan Lue Antonio Politano Huaizhong Xing Mengjie Jiang Xianbin Yu Xiaoshuang Chen Wei Lu 2022Light(Science & Applications)2022,11,3:2
14Unique Characteristics of Vertical Carbon Nanotube Field-effect Transistors on Silicon显示文摘A vertical carbon nanotube field-effect transistor(CNTFET) based on silicon(Si) substrate has been proposed and simulated using a semi-classical theory. A single-walled carbon nanotube(SWNT) and an n-type Si nanowire in series construct the channel of the transistor. The CNTFET presents ambipolar characteristics at positive drain voltage(Vd) and n-type characteristics at negative Vd. The current is significantly influenced by the doping level of n-Si and the SWNT band gap. The n-branch current of the ambipolar characteristics increases with increasing doping level of the n-Si while the p-branch current decreases. The SWNT band gap has the same influence on the p-branch current at a positive Vd and n-type characteristics at negative Vd. The lower the SWNT band gap, the higher the current. However, it has no impact on the n-branch current in the ambipolar characteristics.
