1Exome sequencing confirms molecular diagnoses in 38 Chinese families with hereditary spherocytosis显示文摘Hereditary spherocytosis(HS), the most common cause of congenital hemolytic anemia, is caused by deficiency of the erythrocyte membrane proteins. Five causative genes(ANK1, SPTB, SPTA1, SLC4 A1, and EPB42) have been identified. To date,molecular genetic studies have been performed in different populations, including the American, European, Brazilian, Japanese and Korean populations, whereas only a few studies have been described in the Chinese population. Here, by reanalysis of the exome data, we revealed causative mutations and established a definitive diagnosis of HS in all 38 Chinese families. We found 34 novel mutations and four reported mutations in three known HS-causing genes—17 in ANK1, 17 in SPTB and four in SLC4 A1,suggesting that ANK1 and SPTB are the major genes in Chinese patients with HS. All of the ANK1 or SPTB mutations, scattered throughout the entire genes, are non-recurrent; and most of them are null mutations, which might cause HS via a haploinsufficiency mechanism. De novo mutations in ANK1 or SPTB often occur with an unexpected high frequency(87.5% and64.2%, respectively). 2Amino-functionalized poloxamer 407 with both mucoadhesive and thermosensitive properties:preparation, characterization and application in a vaginal drug delivery system显示文摘Lack of mucoadhesive properties is the major drawback to poloxamer 407(F127)-based in situ hydrogels for mucosal administration. The objective of the present study was to construct a novel mucoadhesive and thermosensitive in situ hydrogel drug delivery system based on an aminofunctionalized poloxamer for vaginal administration. First, amino-functionalized poloxamer 407(F127-NH2) was synthesized and characterized with respect to its micellization behavior and interaction with mucin. Then using acetate gossypol(AG) as model drug, AG-loaded F127-NH2-based in situ hydrogels(NFGs) were evaluated with respect to rheology, drug release, ex vivo vaginal mucosal adhesion, in vivo intravaginal retention and local irritation after vaginal administration to healthy female mice. The results show that F127-NH2 is capable of forming a thermosensitive in situ hydrogel with sustained drug release properties. An interaction between positively charged F127-NH2 and negatively charged mucin wasrevealed by changes in the particle size and zeta potential of mucin particles as well as an increase in the complex modulus of NFG caused by mucin. Ex vivo and in vivo fluorescence imaging and quantitative analysis of the amount of AG remaining in mouse vaginal lavage all demonstrated greater intravaginal retention of NFG than that of an unmodified F127-based in situ hydrogel. 3Mutation profile and its correlation with clinicopathology in Chinese hepatocellular carcinoma patients显示文摘Background:Hepatocellular carcinoma(HCC)is one of the most common causes of cancer worldwide.Although many studies have focused on oncogene characteristics,the genomic landscape of Chinese HCC patients has not been fully clarified.Methods:A total of 165 HCC patients,including 146 males and 19 females,were enrolled.The median age was 55 years(range,27-78 years).Corresponding clinical and pathological information was collected for further analysis.A total of 168 tumor tissues from these patients were selected for next-generation sequencing(NGS)-based 450 panel gene sequencing.Genomic alterations including single nucleotide variations(SNV),short and long insertions and deletions(InDels),copy number variations,and gene rearrangements were analyzed.Tumor mutational burden(TMB)was measured by an algorithm developed in-house.The top quartile of HCC was classified as TMB high.Results:A total of 1,004 genomic alterations were detected from 258 genes in 168 HCC tissues.TMB values were identified in 160 HCC specimens,with a median TMB of 5.4 Muts/Mb(range,0-28.4 Muts/Mb)and a 75%TMB of 7.7 Muts/Mb.The most commonly mutated genes were TP53,TERT,CTNNB1,AXIN1,RB1,TSC2,CCND1,ARID1A,and FGF19.SNV was the most common mutation type and C:G>T:A and guanine transformation were the most common SNVs.Compared to wild-type patients,the proportion of Edmondson grade III-IV and microvascular invasion was significantly higher in TP53 mutated patients(P<0.05).The proportion of tumors invading the hepatic capsule was significantly higher in TERT mutated patients(P<0.05).The proportion of Edmondson grade I-II,alpha fetoprotein(AFP)<25μmg/L,and those without a history of hepatitis B was significantly higher in CTNNB1 mutated patients(P<0.05).CTNNB1 mutations were associated with TMB high in HCC patients(P<0.05).Based on correlation analysis,the mutation of TP53 was independently correlated with microvascular invasion(P=0.002,OR=3.096)and Edmondson grade III-IV(P=0.008,OR=2.613).The mutation of TERT was independently correlated with tumor invasion of the liver capsule(P=0.001,OR=3.030),and the mutation of CTNNB1 was independently correlated with AFP(<25μmg/L)(P=0.009,OR=3.414).Conclusions:The most frequently mutated genes of HCC patients in China were TP53,TERT,and CTNNB1,which mainly lead to the occurrence and development of HCC by regulating the P53 pathway,Wnt pathway,and telomere repair pathway.There were more patients with microvascular invasion and Edmondson III-IV grade in TP53 mutated patients and more patients with hepatic capsule invasion in TERT mutated patients,while in CTNNB1 mutated patients,there were more patients with Edmondson I-II grade,AFP<25μmg/L,and a non-hepatitis B background.Also,the TMB values were significantly higher in CTNNB1 mutated patients than in wild type patients.Shuo Wang Huasheng Shi Tao Liu Manjiang Li Sanshun Zhou Xuan Qiu Zusen Wang Weiyu Hu Weidong Guo Xiaoqian Chen Honglin Guo Xiaoliang Shi Junping Shi Yunjin Zang Jingyu Cao Liqun Wu 2021Hepatobiliary Surgery and Nutrition2021,10,2:8

7Spatio-temporal analysis and simulation on shallow rainfall-induced landslides in China using landslide susceptibility dynamics and rainfall I-D thresholds显示文摘An empirical simulation method to simulate the possible position of shallow rainfall-induced landslides in China has been developed.This study shows that such a simulation may be operated in real-time to highlight those areas that are highly prone to rainfall-induced landslides on the basis of the landslide susceptibility index and the rainfall intensity-duration(I-D) thresholds.First,the study on landslide susceptibility in China is introduced.The entire territory has been classified into five categories,among which high-susceptibility regions(Zone 4-'High' and 5-'Very high') account for 4.15%of the total extension of China.Second,rainfall is considered as an external triggering factor that may induce landslide initiation.Real-time satellite-based TMPA3B42 products may provide real rainfall spatial and temporal patterns,which may be used to derive rainfall duration time and intensity.By using a historical record of 60 significant past landslides,the rainfall I-D equation has been calibrated.The rainfall duration time that may trigger a landslide has resulted between 3 hours and 45 hours.The combination of these two aspects can be exploited to simulate the spatiotemporal distribution of rainfall-induced landslide hazards when rainfall events exceed the rainfall I-D thresholds,where the susceptibility category is 'high' or 'very high'.This study shows a useful tool to be part of a systematic landslide simulation methodology,potentially providing useful information for a theoretical basis and practical guide for landslide prediction and mitigation throughout China.LI WeiYue LIU Chun Marco SCAIONI SUN WeiWei CHEN Yu YAO DongJing CHEN Sheng HONG Yang ZHANG KaiHang CHENG GuoDong 2017Science China Earth Sciences2017,60,4:5
8In vivo retention of poloxamer-based in situhydrogels for vaginal application in mouse and rat models显示文摘The purpose of this study is to evaluate the in vivo retention capabilities of poloxamer-based in situ hydrogels for vaginal application with nonoxinol-9 as the model drug. Two in situ hydrogel formulations, which contained 18% poloxamer 407 plus 1% poloxamer 188(GEL1, relative hydrophobic)or 6% poloxamer 188(GEL2, relative hydrophilic), were compared with respect to the rheological properties, in vitro hydrogel erosion and drug release. The vaginal retention capabilities of these hydrogel formulations were further determined in two small animal models, including drug quantitation of vaginal rinsing fluid in mice and isotope tracing with99 m Tc in rats. The two formulations exhibited similar phase transition temperatures ranging from 27 to 32 1C. Increasing the content of poloxamer 188 resulted in higher rheological moduli under body temperature, but slightly accelerated hydrogel erosion and drug release. When compared in vivo, GEL1 was eliminated significantly slower in rat vagina than GEL2,while the vaginal retention of these two hydrogel formulations behaved similarly in mice. In conclusion,increases in the hydrophilic content of formulations led to faster hydrogel erosion, drug release andintravaginal elimination. 9Efficacy of inverso isomer of CendR peptide on tumor tissue penetration显示文摘The dense extracellular matrix and high interstitial fluid pressure of tumor tissues prevent the ability of anti-tumor agents to penetrate deep into the tumor parenchyma for treatment effects. C-end rule(CendR) peptides can enhance the permeability of tumor blood vessels and tumor tissues via binding to neuropilin-1(NRP-1), thus aiding in drug delivery. In this study, we selected one of the CendR peptides(sequence RGERPPR) as the parent L-peptide and substituted D-amino acids for the L-amino acids to synthesize its inverso peptide D(RGERPPR). We investigated the NRP-1 binding activity and tumorpenetrating ability of D(RGERPPR). We found that the binding affinity of D(RGERPPR) with NRP-1 and the cellular uptake was significantly higher than that of RGERPPR. Evans Blue tests revealed that D(RGERPPR) exhibited improved tumor-penetrating ability in C6, U87 and A549 tumor-bearing nude mice. Using nude mice bearing A549 xenograft tumors as a model, we found that the rate of tumor growth in the group co-administered with D(RGERPPR) and gemcitabine(Gem) was significantly lower than the gemcitabine-treated group with a tumor suppression rate(TSR%) of 55.4%. 10SPDEF suppresses head and neck squamous cell carcinoma progression by transcriptionally activating NR4A1显示文摘SAM pointed domain containing E26 transformation-specific transcription factor(SPDEF)plays dual roles in the initiation and development of human malignancies.However,the biological role of SPDEF in head and neck squamous cell carcinoma(HNSCC)remains unclear.In this study,the expression level of SPDEF and its correlation with the clinical parameters of patients with HNSCC were determined using TCGA-HNSC,GSE65858,and our own clinical cohorts.CCK8,colony formation,cell cycle analysis,and a xenograft tumor growth model were used to determine the molecular functions of SPDEF in HNSCC.ChIP-qPCR,dual luciferase reporter assay,and rescue experiments were conducted to explore the potential molecular mechanism of SPDEF in HNSCC.Compared with normal epithelial tissues,SPDEF was significantly downregulated in HNSCC tissues.Patients with HNSCC with low SPDEF mRNA levels exhibited poor clinical outcomes.Restoring SPDEF inhibited HNSCC cell viability and colony formation and induced G0/G1 cell cycle arrest,while silencing SPDEF promoted cell proliferation in vitro.The xenograft tumor growth model showed that tumors with SPDEF overexpression had slower growth rates,smaller volumes,and lower weights.SPDEF could directly bind to the promoter region of NR4A1 and promoted its transcription,inducing the suppression of AKT,MAPK,and NF-κB signaling pathways.Moreover,silencing NR4A1 blocked the suppressive effect of SPDEF in HNSCC cells.Here,we demonstrate that SPDEF acts as a tumor suppressor by transcriptionally activating NR4A1 in HNSCC.Our findings provide novel insights into the molecular mechanism of SPDEF in tumorigenesis and a novel potential therapeutic target for HNSCC.Yanting Wang Xianyue Ren Weiyu Li Ruoyan Cao Suyang Liu Laibo Jiang Bin Cheng Juan Xia 2021International Journal of Oral Science2021,13,4:3
11Increasing prevalence of antibiotic resistance genes in manured agricultural soils in northern China显示文摘Land application of manure tends to result in the dissemination of antibiotic resistance in the environment.In this study,the influence of long-term manure application on the enrichment of antibiotic resistance genes(ARGs)and mobile genetic elements(MGEs)in agricultural soils was investigated.All the analyzed eight ARGs(tetA,tetW,tetX,sull,sulll,ermF,aac(6')-Ib-cr and blaTEM)and two MGEs(intJJ and Tn916/1545)were detected in both the manured and control soils,with relative abundances ranging from 10^-6 to 10^-2 Compared with the control soil,the relative abundances of ARGs and MGEs in manured soils were enriched 1.0-18.1 fold and 0.6-69.1 fold,respectively.High-throughput sequencing analysis suggested that at the phylum level,the bacteria carrying intll and ermF might be mainly affiliated with Proteobacteria and Bacteroides,respectively.The dominant genera carrying intll and ermF cow\d be Pseudomonas and Bacteroides,independent of manure application.Correlation analysis revealed that ARGs had strong links with soil physicochemical properties(TC,TN,and OM),heavy metals(Cu,Zn and Pb)and MGEs,indicating that the profile and spread of ARGs might be driven by the combined impacts of multiple factors.In contrast,soil pH and C/N exhibited no significant relationships with ARGs.Our findings provide evidence that long-term manure application could enhance the prevalence and stimulate the propagation of antibiotic resistance in agricultural soils.Nan Wu Weiyu Zhang Shiyu Xie Ming Zeng Haixue Liu Jinghui Yang Xinyuan Liu Fan Yang 2020Frontiers of Environmental Science & Engineering2020,14,1:3
12The effects of two free-floating plants (Eichhornia crassipes and Pistia stratiotes) on the burrow morphology and water quality characteristics of pond loach (Misgurnus anguillicaudatus) habitat显示文摘Loach exhibit conspicuous drilling behaviors in the mud of shallow waters,yet their burrow morphology and the factors affecting this behavior have received little attention.We characterized the burrow morphology and water quality of the pond loach Misgurnus anguillicaudatus in three scenarios:in tanks without plants,tanks with the free-floating plant water hyacinth Eichhornia crassipes,and tanks with water lettuce Pistia stratiotes.Water hyacinth effectively removed water TN,COD,NO3-N and NH4-N,and water lettuce removed water TP and NH4-N.Water hyacinth and water lettuce markedly reduced water turbidity and DO,increased TOC and EC.Water hyacinth purified water more effectively than water lettuce,providing a suitable habitat for loach feeding,living and burrowing.The burrow structures were V-shaped,Y-shaped,inverted L-shaped,or complicated dendritic networks composed of multiple V shapes.The hyacinth treatment was characterized by the greatest burrow volume,length,depth,and structural complexity,but the opening size was reduced by dense root mat coverage.Burrows in the water lettuce treatment were characterized by intermediate volume,length,branches and sinuosity,but they had the largest opening and pit size.The control treatment had a flat bottom with the smallest,shortest burrows.This study indicates that free-floating plants improve habitat suitability and change burrow morphology and may be used to improve loach breeding methods。Jinqing Wang Guihua Fu Weiyue Li Ying Shi Jicai Pang Qiang Wang Weiguang Lü Change Liu Jiansheng Liu 2018Aquaculture and Fisheries2018,3,1:2
13Emerging systemic delivery strategies of oncolytic viruses:A key step toward cancer immunotherapy显示文摘Oncolytic virotherapy(OVT)is a novel type of immunotherapy that induces anti-tumor responses through selective self-replication within cancer cells and oncolytic virus(OV)-mediated immunostimulation.Notably,talimogene laherparepvec(T-Vec)developed by the Amgen company in 2015,is the first FDA-approved OV product to be administered via intratumoral injection and has been the most successful OVT treatment.However,the systemic administration of OVs still faces huge challenges,including in vivo pre-existing neutralizing antibodies and poor targeting delivery efficacy.Recently,state-of-the-art progress has been made in the development of systemic delivery of OVs,which demonstrates a promising step toward broadening the scope of cancer immunotherapy and improving the clinical efficacy of OV delivery.Herein,this review describes the general characteristics of OVs,focusing on the action mechanisms of OVs as well as the advantages and disadvantages of OVT.The emerging multiple systemic administration approaches of OVs are summarized in the past five years.In addition,the combination treatments between OVT and traditional therapies(chemotherapy,thermotherapy,immunotherapy,and radiotherapy,etc.)are highlighted.Last but not least,the future prospects and challenges of OVT are also discussed,with the aim of facilitating medical researchers to extensively apply the OVT in the cancer therapy.Weiyue Ban Jianhuan Guan Hanwei Huang Zhonggui He Mengchi Sun Funan Liu Jin Sun 2022Nano Research2022,15,5:2
14Developmental toxicity of TCBPA on the nervous and cardiovascular systems of zebrafish (Danio rerio): A combination of transcriptomic and metabolomics显示文摘Tetrachlorobisphenol A(TCBPA),a widely used halogenated flame retardant,is frequently detected in environmental compartments and human samples.However,unknown developmental toxicity and mechanisms limit the entire understanding of its effects.In this study,zebrafish(Danio rerio)embryos were exposed to various concentrations of TCBPA while a combination of transcriptomics,behavioral and biochemical analyzes as well as metabolomics were applied to decipher its toxic effects and the potential mechanisms.We found that TCBPA could interfere with nervous and cardiovascular development through focal adhesion and extracellular matrix-receptor(ECM-receptor)interaction pathways through transcriptomic analysis.Behavioral and biochemical analysis results indicated abnormal swimming behavior of zebrafish larvae.Morphological observations revealed that TCBPA could cause the loss of head blood vessels.Metabolomic analysis showed that arginine-related metabolic pathways were one of the main pathways leading to TCBPA developmental toxicity.Our study demonstrated that by using omics,TCBPA was shown to have neurological and cardiovascular developmental toxicity and the underlying mechanisms were uncovered and major pathways identified.Wentao Liu Yifan Pan Lu Yang Yun Xie Xuanyue Chen Jing Chang Weiyu Hao Lifei Zhu Bin Wan 2023Journal of Environmental Sciences2023,,5:2

17Preliminary investigations into surface molecularly imprinted nanoparticles for Helicobacter pylori eradication显示文摘This paper reports investigations into the preparation and characterization of surface molecularly imprinted nanoparticles(SMINs) designed to adhere to Helicobacter pylori(H.pylori).Imprinted nanoparticles were prepared by the inverse microemulsion polymerization method.A fraction of Lpp20,an outer membrane protein of H.pylori known as NQA,was chosen as template and modified with myristic acid to facilitate its localization on the surface of the nanoparticles.The interaction between these SMINs with the template NQA were evaluated using surface plasmon resonance(SPR),change in zeta potential and fluorescence polarization(FP).The results were highly consistent in demonstrating a preferential recognition of the template NQA for SMINs compared with the control nanoparticles.In vitro experiments also indicate that such SMINs are able to adhere to H.pylori and may be useful for H.pylori eradication.Jiaying Han Yinjing Sun Jiapeng Hou Yuyan Wang Yu Liu Cao Xie Weiyue Lu Jun Pan 2015Acta Pharmaceutica Sinica B2015,5,6:2
