2Detection and characterization of diverse alpha-and betacoronaviruses from bats in China显示文摘Bats have been implicated as important reservoir hosts of alpha- and betacoronaviruses. In this study, diverse coronaviruses(CoVs) were detected in 50 of 951(positive rate 5.3%) intestinal specimens of eight bat species collected in four provinces and the Tibet Autonomous Region of China by pan-coronavirus RT-PCR screening. Based on 400-nt RNA-dependent RNA polymerase(Rd RP) sequence analysis, eight belonged to genus Alphacoronavirus and 42 to Betacoronavirus.Among the 50 positive specimens, thirteen gave rise to CoV full-length Rd RP gene amplification for further sequence comparison, of which three divergent sequences(two from a unreported province) were subjected to full genome sequencing. Two complete genomes of betacoronaviruses(JTMC15 and JPDB144) and one nearly-complete genome of alphacoronavirus(JTAC2) were sequenced and their genomic organization predicted. The present study has identified additional numbers of genetically diverse bat-borne coronaviruses with a wide distribution in China. 3Response to climate change of different tree species and NDVI variation since 1923 in the middle arid region of Ningxia,China显示文摘The normalized difference vegetation index(NDVI)is used extensively to describe vegetation cover and ecological environment change.The purpose of this study was to contrast the response of different tree species growing in the same habitat to climate change and retrieve past NDVI using tree-ring width data from tree cores collected from the transitional zone of Pinus tabulaeformis and Picea crassifolia in the Luoshan Mountains in the middle arid region of Ningxia.Correlation analysis indicated that radial growth of P.tabulaeformis is more sensitive to precipitation and temperature change than that of P.crassifolia.Natural factors such as water availability and heat at this elevation are more suited to the growth of P.crassifolia,and are more advantageous to its renewal and succession.P.crassifolia is probably the better of the two species for protecting the forest ecosystem and conserving water in the Luoshan desertification area.Ring width of P.crassifolia correlates significantly with average NDVI for April–May(r=0.641,p<0.01),and both of them are influenced positively by precipitation in April–May.The reconstructed NDVI for 1923–2007 shows the relatively low vegetation cover occurred in the 1920s–1930s,the1960s–1970s,and the early 21st century.The reconstructed NDVI better reflected the drought climate in the study area.YaJun Wang RuiJie Lu YuZhen Ma HongWei Meng ShangYu Gao 2014Research in Cold and Arid Regions2014,6,1:6
4Genome-wide Identification of microRNAs that Respond to Drought Stress in Seedlings of Tertiary Relict Ammopiptanthus mongolicus显示文摘microRNAs(miR NAs) are important post-transcriptional regulators of gene expression in plant abiotic stress responses. Ammopiptanthus mongolicus is a typical evergreen woody xerophyte, which makes it an ideal model system for studying drought tolerance in woody plants. The response of mi RNAs to drought stress is still unknown in this plant. In this research, we obtained 34 695 665 raw reads from two small RNA libraries constructed from control and drought-treated A. mongolicus seedlings by the Illumina deep sequencing technology. Length analysis revealed that reads of 20, 21, and 24 nucleotides accounted for the majority of the small RNAs in the two libraries. Sequence analyses identified 65 conserved mi RNA sequences from 190 members of 31 mi RNA families and 20 potential novel mi RNAs from 17 families that were differentially expressed between the two libraries. The expression patterns of 25 of these mi RNAs were significantly different in the two libraries, but only am-mi R4 a-c was downregulated during drought stress, the other 24 were upregulated. The expression trends determined by the Hi Seq sequencing and by q RT-PCR were similar. Furthermore, 35 target genes for the conserved mi RNA were markedly changed and 52 target genes for the potential novel mi RNAs were predicted and their functions were annotated by computational analysis. 5Reconstructing the annual precipitation variation since 1899 based on tree-ring width in the western Hedong sandy land of Ningxia显示文摘Based on the analysis of the correlation between the tree-ring width of Pinus tabulaeformis and the climate factors in the western Hedong sandy land of Ningxia, a conversion equation between the annual precipitation and the tree-ring width since 1899 was reconstructed. The results of cross verification indicated that the conversion equation is stable and the reconstructed results are reliable. The result of reconstructed annual precipitation showed the remarkable fluctuation of precipitation and dry-to-wet variation before the 1940s. The smaller fluctuation and high frequent changes of precipitation occurred during the period of 1940s-1980s and after the 1980s the change trend of the precipitation became high periodic extent and low frequent. The study found that there were some coincidences with the climate change in Changling Mountains, Helan Mountains and the east of Qilian Mountains. The relatively dry periods in the beginning of 20th century, 1920s to 1930s, the end of the 20th century and 2004 to 2006 in the western Hedong sandy land of Ningxia accelerated the desertification, while the relatively humid period during the periods of the 1910s-1920s, 1930s-1940s and 1990s is favorable to prevent and control the desertification, and to weaken the climate warming and drying. 8Reduced thermal boundary conductance in GaN-based electronic devices introduced by metal bonding layer显示文摘Achieving high interface thermal conductance is one of the biggest challenges in the nanoscale heat transport of GaN-based devices such as light emitting diodes(LEDs),and high electron mobility transistors(HEMTs).In this work,we experimentally measured thermal boundary conductance(TBC)at interfaces between GaN and the substrates with AuSn alloy as a commonly-used adhesive layer by time-domain thermoreflectance(TDTR).We find that the TBCs of GaN/Ti/AuSn/Ti/Si,GaN/Ti/AuSn/Ti/SiC,and GaN/Ti/AuSn/Ti/diamond,are 16.5,14.8,and 13.2 MW·m^(-2)·K^(-1)at room temperature,respectively.Our measured results show that the TBC of GaN/Ti/AuSn/Ti/SiC interface is inferior to the TBC of pristine GaN/SiC interface,due to the large mismatch of phonon modes between AuSn/Ti and substrates,shown as the difference of Debye temperature of two materials.Overall,we measured the TBC at interface between GaN and thermal conductive substrates,and provided a guideline for designing the interface between GaN and substrate at HEMT from a thermal management point of view.Susu Yang Houfu Song Yan Peng Lu Zhao Yuzhen Tong Feiyu Kang Mingsheng Xu Bo Sun Xinqiang Wang 2021Nano Research2021,14,10:1
9Extracellular-superoxide dismutase DNA methylation promotes oxidative stress in homocysteine-induced atherosclerosis显示文摘In the present study,we investigate the effect of homocysteine(Hcy)on extracellular-superoxide dismutase(ECSOD)DNA methylation in the aorta of mice,and explore the underlying mechanism in macrophages,trying to identify the key targets of Hcy-induced EC-SOD methylation changes.ApoE--mice are fed different diets for 15 weeks,EC-SODand DNAmethyltransferase1(DNMT1)expression levels aredetected byRT-PCRandwestern blot analysis.EC-SOD methylation levels are assessed by ntMS-PCR.After EC-SOD overexpression or knockdown in macrophages,following the transfection of macrophages with pEGFP-N1-DNMT1,the methylation levels of ECSOD are detected.Our data show that the concentrations of Hcy and the area of atherogenic lesions are significantly increased in ApoE--mice fed with a high-methionine diet,and have a positive correlation with the levels of superoxide anions,which indicates that Hcy-activated superoxide anions enhance the development of atherogenic lesions.EC-SOD expression is suppressed by Hcy,and the content of superoxide anion is increased when EC-SOD is silenced by RNAi in macrophages,suggesting that EC-SOD plays a major part in oxidative stress induced by Hcy.Furthermore,the promoter activity of EC-SOD is increased following transfection with the-1/-110o fragment,and EC-SOD methylation level is significantly suppressed by Hcy,and more significantly decreased upon DNMT1 overexpression.In conclusion,Hcy may alter the DNA methylation status and DNMT1 acts as the essential enzyme in the methyl transfer process to disturb the status of EC-SOD DNA methylation,leading to decreased expression of Ec-sODandincreasedoxidativestressandatherosclerosis.Shengchao Ma Guanjun Lu Qing Zhang Ning Ding Yuzhen Jie Hui Zhang Lingbo Xu Lin Xie Xiaoling Yang Huiping Zhang Yideng Jiang 2022Acta Biochimica et Biophysica Sinica2022,54,9:1
10Effect of TiO2 Nanoparticles on Charge Transportation in Mineral Oil and Natural Ester Based Nanofluid显示文摘TiO2 semiconductive nanoparticles are added into mineral and ester based transformer oil to form semiconductive nanofluids (SNFs) with the aim of enhancing the oil’s insulating performance. Charge accumulation and decay characteristics of both pure oils and SNFs are measured by pulse electroacoustic (PEA) technique. The result reveals that compared with pure oil, SNFs have more uniform in- ternal electric fields with voltage applied and higher charge decay rate after removing the applied voltage. This is caused by the increase of shallow trap density in SNFs, due to the test results of thermally stimulated current (TSC). 15Role of mucilage during achene germination and sprout growth of the endangered Tibetan medicinal herb Mirabilis himalaica (Nyctaginaceae) exposed to abiotic stresses显示文摘Aim Mirabilis himalaica(Nyctaginaceae)is an endangered medicinal plant mainly distributed in the plateau region of northern Tibet,China.The outer surface of M.himalaica achenes is covered by a pectinaceous mucilaginous layer upon hydration.However,the role of the achene mucilage is poorly understood.in this study,we inves-tigated the effects of mucilage on achene germination and sprout growth under abiotic stress to explain how M.himalaica survive the alpine environment.Methods We investigated the effect of mucilage on achenes germination by contrast the capacity of water absorption,dehydration and respira-tion of intact achene and the achene with mucilage removal.We performed abiotic stresses experiments including drought stress,salt stress,cold stress and high temperature stress,and quantified the effects of mucilage removal on achene germination rate,root and shoot lengths of seedlings.Important Findings Mucilage is extremely hydrophilic,and the mass of intact achenes can be 9-fold greater than that of demucilaged achenes.The removal of the mucilaginous layer did not significantly change final germination percentages under ideal conditions,but intact achenes(i.e.with muci-lage)took longer to germinate.The mucilage significantly decreased seed respiration rates by acting as a physical barrier that prevented oxygen diffusion.Germination rates,shoot and root growth of intact achenes were higher than those of demucilaged ones during exposures to cold,heat,osmotic and salt stresses.Achene mucilage presumably plays an ecologically important role in the life cycle of M.himalaica by aiding the critical achene germination and early seedling growth in the stressful habitats of the plateau region of northern Tibet.Yuzhen Chen Lu Zhang Xiao Lu Xiaozhong Lan Man Shen Cunfu Lu 2018Journal of Plant Ecology2018,11,2:1
16QTL mapping for plant height and fruit branch number based on RIL population of upland cotton显示文摘Background:Plant height(PH)and fruit branch number(FBN)are important traits for improving yield and mechanical harvesting of cotton.In order to identify genes of PH and FBN in cotton germplasms to develop superior cultivars,quantitative trait loci(QTLs)for these traits were detected based on the phenotypic evaluation data in nine environments across four locations and 4 years and a previously reported genetic linkage map of an recombinant inbred line(RIL)population of upland cotton.Results:In total,53 QTLs of PH and FBN,were identified on 21 chromosomes of the cotton genome except chromosomes c02,c09-c11,and c22.For PH,27 QTLs explaining 3.81%–8.54%proportions of phenotypic variance were identified on 18 chromosomes except c02,c08-c12,c15,and c22.For FBN,26 QTLs explaining 3.23%–11.00%proportions of phenotypic variance were identified on 16 chromosomes except c02-c03,c06,c09-c11,c17,c22-c23,and c25.Eight QTLs were simultaneously identified in at least two environments.Three QTL clusters containing seven QTLs were identified on three chromosomes(c01,c18 and c21).Eleven QTLs were the same as previously reported ones,while the rest were newly identified.Conclusions:The QTLs and QTL clusters identified in the current study will be helpful to further understand the genetic mechanism of PH and FBN development of cotton and will enhance the development of excellent cultivars for mechanical managements in cotton production.LIU Ruixian XIAO Xianghui GONG Juwu LI Junwen ZHANG Zhen LIU Aiying LU Quanwei SHANG Haihong SHI Yuzhen GE Qun IQBAL Muhammad Sajid CHEN Quanjia YUAN Youlu GONG Wankui 2020Journal of Cotton Research2020,3,1:1
