2Noninvasive assessment of hepatic fibrosis in Egyptian patients with chronic hepatitis C virus infection显示文摘AIM: To evaluate the accuracy of specific biochemical markers for the assessment of hepatic fibrosis in patients with chronic hepatitis C virus (HCV) infection. METHODS: One hundred and fifty-four patients with chronic HCV infection were included in this study; 124 patients were non-cirrhotic, and 30 were cirrhotic. The following measurements were obtained in all patients: serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), albumin, total bilirubin, prothrombin time and concentration, complete blood count, hepatitis B surface antigen (HBsAg), HCVAb, HCV-RNA by quantitative polymerase chain reaction, abdominal ultrasound and ultrasonic-guided liver biopsy. The following ratios, scores and indices were calculated and compared with the results of the histopathological examination: AST/ALT ratio (AAR), age platelet index (API), AST to platelet ratio index (APRI), cirrhosis discriminating score (CDS), Pohl score, G teborg University Cirrhosis Index (GUCI). RESULTS: AAR, APRI, API and GUCI demonstrated good diagnostic accuracy of liver cirrhosis (80.5%, 79.2%, 76.6% and 80.5%, respectively); P values were: < 0.01, < 0.05, < 0.001 and < 0.001, respectively. Among the studied parameters, AAR and GUCI gave the highest diagnostic accuracy (80.5%) with cutoff values of 1.2 and 1.5, respectively. APRI, API and GUCI were significantly correlated with the stage of fibrosis (P < 0.001) and the grade of activity (P < 0.001, < 0.001 and < 0.005, respectively), while CDS only correlated significantly with the stage of fibrosis (P < 0.001) and not with the degree of activity (P > 0.05). In addition, we found significant correlations for the AAR, APRI, API, GUCI and Pohl score between the non-cirrhotic (F0, F1, F2, F3) and cirrhotic (F4) groups (P values: < 0.001, < 0.05, < 0.001, < 0.001 and < 0.005, respectively; CDS did not demonstrate significant correlation (P > 0.05). 4Photopic negative response in diagnosis of glaucoma:an experimental study in glaucomatous rabbit model显示文摘AIM: To determine whether the photopic negative response(Ph NR) elicited by transient white flash on white background is characterizing for glaucoma model in rabbits.METHODS: Glaucoma was induced in twelve rabbits by subconjunctival injection of 0.05 m L of betamethasone in right eyes(each 1 m L contain betamethasone dipropionate5 mg and betamethasone sodium phosphate 2 mg).The intraocular pressure(IOP), electroretinogram(ERG) and visual evoked potential(VEP) were measured successively prior and on the 3, 7d, two weeks and four weeks postglaucoma induction. After four weeks, the animals were sacrificed and the globes were histopathologically examined.RESULTS: The IOP increased significantly after one week(P =0.0001), then it gradually returned to the control level. In ERG examination, the means of a and b wave amplitude and latency were not affected significantly.Ph NR amplitude decreased significantly within one week(P =0.0001), but its latency was not affected significantly(P =0.132). The means of VEP latency and amplitude were significantly affected after two weeks and four weeks of glaucoma induction(P =0.0001 and 0.02,respectively). The histopathologic examination of the globes showed reduced number of cells in the retinal ganglion cell layer with multiple vacuoles in the retinal nerve fibre layer.There was significant positive correlation between ganglion cell layer cells and Ph NR amplitude( r =0.8, P =0.002).CONCLUSION: The rise in IOP resulted in irreversible changes or incomplete recovery of VEP and Ph NR amplitude. 6Early therapeutic effect of platelet-rich fibrin combined with allogeneic bone marrow-derived stem cells on rats’ critical-sized mandibular defects显示文摘BACKGROUND Critically sized bone defects represent a significant challenge to orthopaedic surgeons worldwide.These defects generally result from severe trauma or resection of a whole large tumour.Autologous bone grafts are the current gold standard for the reconstruction of such defects.However,due to increased patient morbidity and the need for a second operative site,other lines of treatment should be introduced.To find alternative unconventional therapies to manage such defects,bone tissue engineering using a combination of suitable bioactive factors,cells,and biocompatible scaffolds offers a promising new approach for bone regeneration.AIM To evaluate the healing capacity of platelet-rich fibrin(PRF)membranes seeded with allogeneic mesenchymal bone marrow-derived stem cells(BMSCs)on critically sized mandibular defects in a rat model.METHODS Sixty-three Sprague Dawley rats were subjected to bilateral bone defects of critical size in the mandibles created by a 5-mm diameter trephine bur.Rats were allocated to three equal groups of 21 rats each.Group I bone defects were irrigated with normal saline and designed as negative controls.Defects of group II were grafted with PRF membranes and served as positive controls,while defects of group III were grafted with PRF membranes seeded with allogeneic BMSCs.Seven rats from each group were killed at 1,2 and 4 wk.The mandibles were dissected and prepared for routine haematoxylin and eosin(HE)staining,Masson's trichrome staining and CD68 immunohistochemical staining.RESULTS Four weeks postoperatively,the percentage area of newly formed bone was significantly higher in group III(0.88±0.02)than in groups I(0.02±0.00)and II(0.60±0.02).The amount of granulation tissue formation was lower in group III(0.12±0.02)than in groups I(0.20±0.02)and II(0.40±0.02).The number of inflammatory cells was lower in group III(0.29±0.03)than in groups I(4.82±0.08)and II(3.09±0.07).CONCLUSION Bone regenerative quality of critically sized mandibular bone defects in rats was better promoted by PRF membranes seeded with BMSCs than with PRF membranes alone.Muhammad A Awadeen Fouad A Al-Belasy Laila E Ameen Mohamad E Helal Mohammed E Grawish 2020World Journal of Stem Cells2020,12,1:4

9Gas chromatography-mass spectrometry analysis of different organic crude extracts from the local medicinal plant of Thymus vulgaris L显示文摘Objective:To isolate and analyze the chemical composition in different crude extracts of from the leaves of locally grown of Thymus vulgaris L(T.vulgaris)by gas chromatography-mass spectrometry(GC-MS).Methods:The shade dried leaves powder was extracted with methanol by using Soxhlet extractor.Methanol crude extracts of T.vulgaris and the derived fractions of hexane,chloroform,ethyl acetate and butanol were obtained.Results:Qualitative analyses of various organic crude extracts of T.vulgaris by using GC-MS showed that there were different types of high and low molecular weight compounds.Most of the isolated and identified compounds by GC-MS in the crude extracts are basically biologically important.Further,the T.vulgaris leaf possessed certain characteristics that can be ascribed to cultivation on a domestic plantation.The crude extracts were prepared from the powder leaves of T.vulgaris for respective compounds can be chosen on the basis of above GC-MS analysis.Conclusions:All the major compounds were identified and characterized by spectroscopic method in different organic crude extracts of T.vulgaris are biologically active molecules.Thus the identification of a good number of compounds in various crude extracts of T.vulgaris might have some ecological role.Laila Salim Al Hashmi Mohammad Amzad Hossain Afaf Mohammed Weli Qasim Al-Riyami Jamal Nasser Al-Sabahi 2013Asian Pacific Journal of Tropical Biomedicine2013,3,1:4

11Effect of mesenchymal stem cells on anti-Thy1,1 induced kidney injury in albino rats显示文摘Objective:To evaluate the effect of mesenchymal stem cells(MSCs)in rats with anti-Thy 1,1nephritis.Methods:Female albino rats were divided into three groups,control group,anti-Thy 1,1group and treatment with i.v.MSCs group.MSCs were derived from bone marrow of male albino rats,Y—chromosome gene was detected by polymerase chain reaction in the kidney.Serum urea and creatinine were estimated for all groups.Kidney of all studied groups was examined histologically and histochemically(total carbohydrates and total proteins).DNA fragmentation and expression ofα-SMA were delected.Results:Kidney of animals injected with anti-Thy 1,1showed inflammatory leucocytic infiltration,hypertrophied glomeruli,tubular necrosis and congestion in the renal blood vessels.The kidney tissue also showed reduction of carbohydrates and total proteins together with increase in apoptosis and in expression ofα-SMA.Moreover,the levels of urea and creatinine were elevated.Treating animals with MSCs revealed that kidney tissue displayed an improvement in the histological and histochemical changes.Apoptosis andα-SMA expression were decreased,and the levels of urea and creatinine decreased.Conclusions:The obtained results demonstrated the potential of MSCs to ameliorate the structure and function of the kidney in rats with anti-Thy 1,1 nephritis possibly through the release of paracrine growth factor(s).Saber Sakr Laila Rashed Waheba Zarouk Rania El-Shamy 2013Asian Pacific Journal of Tropical Biomedicine2013,3,3:3
12Possible ameliorative effect of breastfeeding and the uptake of human colostrum against coeliac disease in autistic rats显示文摘AIM: To examine the possible ameliorative effect of breastfeeding and the uptake of human colostrum against coeliac disease in autistic rats. METHODS: Female rats were fed a standard diet and received a single intraperitoneal injection of 600 mg/kg sodium valproate on day 12.5 after conception. In study 1, neonatal rats were randomly subjected to blood tests to investigate autism. In study 2, the 1st group was fed by the mother after an injection of interferon-γ (IFN-γ) and administration of gliadin. The pups in the 2nd group were prevented from accessing maternal milk, injected IFN-γ, administered gliadin, and hand-fed human colostrum. The normal littermates fed by the table mothers were injected with physiological saline and served as normal controls in this study.RESULTS: The protein concentration was higher in group 2 than in group 1 in the duodenum (161.6 ± 9 and 135.4 ± 7 mg/g of tissue, respectively, P < 0.01). A significant increase (P < 0.001) in body weight was detected in human colostrum-treated pups on post natal day (PND) 7 and 21 vs suckling pups in group 1. A delay in eye opening was noticed in the treated rats in group 1 on PND 13 compared with the control group and group 2. Administration of a single intraperitoneal injection of 600 mg/kg sodium valproate on day 12.5 after conception resulted in significantly reduced calcium and vitamin D levels in study 1 compared with the control groups (P < 0.001). However, human colostrum uptake inhibited increases in the level of transglutaminase antibody in autistic pups with coeliac disease. 13Effect of the Si/Al ratio of HZSM-5 zeolite on the production of dimethyl ether before and after ultrasonication显示文摘A series of as-synthesized HZSM-5 zeolites with different Si/Al ratios(25,90,120,240 and 400)were post-treated by ultrasonication for an optimum time of 60 min.The morphology,acidity and textural properties of HZSM-5 were characterized with XRD,SEM,N_2 adsorption and NH3-TPD techniques.The catalytic performance was evaluated by dehydration of methanol to dimethyl ether(DME),which is a promising gaseous automotive fuel in future.It was found that the Si/Al ratio of HZSM-5 had considerable impacts on its catalytic performance for dehydration of methanol to DME.Its activity increased with decreasing Si/Al ratio from 400 to 25.Ultrasonication of HZSM-5 could significantly improve its catalytic performance.Sameh M.K.Aboul-Fotouh Laila I.Ali Mona A.Naghmash Noha A.K.Aboul-Gheit 2017燃料化学学报2017,45,5:3

15Dental pulp stem cells stimulate neuronal differentiation of PC12 cells显示文摘Dental pulp stem cells(DPSCs) secrete neurotrophic factors which may play an important therapeutic role in neural development, maintenance and repair. To test this hypothesis, DPSCs-conditioned medium(DPSCs-CM) was collected from 72 hours serum-free DPSCs cultures. The impact of DPSCs-derived factors on PC12 survival, growth, migration and differentiation was investigated. PC12 cells were treated with nerve growth factor(NGF), DPSCs-CM or co-cultured with DPSCs using Transwell inserts for 8 days. The number of surviving cells with neurite outgrowths and the length of neurites were measured by image analysis. Immunocytochemical staining was used to evaluate the expression of neuronal markers NeuN, microtubule associated protein 2(MAP-2) and cytoskeletal marker βIII-tubulin. Gene expression levels of axonal growth-associated protein 43 and synaptic protein Synapsin-I, NeuN, MAP-2 and βIII-tubulin were analysed by quantitative polymerase chain reaction(qRT-PCR). DPSCs-CM was analysed for the neurotrophic factors(NGF, brain-derived neurotrophic factor [BDNF], neurotrophin-3, and glial cell-derived neurotrophic factor [GDNF]) by specific ELISAs. Specific neutralizing antibodies against the detected neurotrophic factors were used to study their exact role on PC12 neuronal survival and neurite outgrowth extension. DPSCs-CM significantly promoted cell survival and induced the neurite outgrowth confirmed by NeuN, MAP-2 and βIII-tubulin immunostaining. Furthermore, DPSCsCM was significantly more effective in stimulating PC12 neurite outgrowths than live DPSCs/PC12 co-cultures over the time studied. The morphology of induced PC12 cells in DPSCs-CM was similar to NGF positive controls;however, DPSCs-CM stimulation of cell survival was significantly higher than what was seen in NGF-treated cultures. The number of surviving PC12 cells treated with DPSCs-CM was markedly reduced by the addition of anti-GDNF, whilst PC12 neurite outgrowth was significantly attenuated by anti-NGF, anti-GDNF and anti-BDNF antibodies. These findings demonstrated that DPSCs were able to promote PC12 survival and differentiation. DPSCs-derived NGF, BDNF and GDNF were involved in the stimulatory action on neurite outgrowth, whereas GDNF also had a significant role in promoting PC12 survival. DPSCs-derived factors may be harnessed as a cell-free therapy for peripheral nerve repair. All experiments were conducted on dead animals that were not sacrificed for the purpose of the study. 16Low-density lipoprotein receptor genetic polymorphism in chronic hepatitis C virus Egyptian patients affects treatment response显示文摘AIM: To correlate a genetic polymorphism of the low-density lipoprotein(LDL) receptor with antiviral responses in Egyptian chronic hepatitis C virus(HCV) patients.METHODS: Our study included 657 HCV-infected patients with genotype 4 who received interferonbased combination therapy. Patients were divided into two groups based on their response to therapy: 356 were responders, and 301 were non-responders. Patients were compared to 160 healthy controls. All patients and controls underwent a thorough physical examination, measurement of body mass index(BMI) and the following laboratory tests: serum alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, albumin, total bilirubin, direct bilirubin, prothrombin time, prothrombin concentration, INR, complete blood count, serum creatinine, fasting blood sugar, HCV antibody, and hepatitis B surface antigen. All HCV patients were further subjected to the following laboratory tests: HCV-RNA using quantitative polymerase chain reaction(PCR), antinuclear antibodies, thyroid-stimulating hormone, an LDL receptor(LDLR) genotype study of LDLR exon8 c.1171G>A and exon-10 c.1413G>A using real-time PCR-based assays, abdominal ultrasonography, ultrasonographic-guided liver biopsy, and histopathological examination of liver biopsies. Correlations of LDL receptor polymorphisms with HAI, METAVIR score, presence of steatosis, and BMI were performed in all cases.RESULTS: There were no statistically significant differences in response rates between the different types of interferon used or LDLR exon10 c.1413G>A. However, there was a significant difference in the frequency of the LDL receptor exon8 c.1171G>A genotype between cases(AA: 25.9%, GA: 22.2%, GG: 51.9%) and controls(AA: 3.8%, GA: 53.1% and GG: 43.1%)(P < 0.001). There was a statistically significant difference in the frequency of the LDLR exon 8C:1171 G>A polymorphism between responders(AA: 3.6%, GA: 15.2%, GG: 81.2%) and nonresponders(AA: 52.2%, GA: 30.6%, GG: 17.2%)(P < 0.001). The G allele of LDL receptor exon8 c.1171G>A predominated in cases and controls over the A allele, and a statistically significant association with response to interferon was observed. The frequency of the LDLR exon8 c.1171G>A allele in non-responders was: A: 67.4% and G: 32.6 vs A: 11.2% and G: 88.8% in responders(P < 0.001). Therefore, carriers of the A allele exhibited a 16.4 times greater risk for nonresponse. There was a significant association between LDL receptors exon8 c.1171G>A and HAI(P < 0.011). There was a significant association between LDL receptors exon8 c.1171G>A and BMI. The mean BMI level was highest in patients carrying the AA genotype(28.7 ± 4.7 kg/m2) followed by the GA genotype(28.1 ± 4.8 kg/m2). The lowest BMI was the GG genotype(26.6 ± 4.3 kg/m2)(P < 0.001). 19Persian medicine non-pharmacological therapies for headache:phlebotomy and wet cupping显示文摘OBJECTIVE: To describe the common causes of headache as well as the different methods of Phlebotomy(Fasd) and wet Cupping(Hijamat) based on Persian Medicine.METHODS: In this study, we searched Google Scholar, Science Direct, Pub Med and Scopus to get any in vitro or clinical evidence of the efficacy and possible mechanisms of phlebotomy or wet cupping on different types of headaches. Also different types of headaches, according to PM, under the title of 'Sodaa'; extracted from famous Persian medical text books with key words including 'Sodaa','Bayze' and 'Shaqiqe'.RESULTS: According to PM, diseases(dystemperament) are created by abnormal alteration in temperament. Temperament is a monolithic quality yielded by interaction between opposing qualities of four elements that are coldness, hotness, dryness and wetness. Headaches are caused by dystemperament(sue-Mizaj) in brain or in total body that are divided into simple and corporal. Common forms of the latter type are Vaporal and Migrant headache(Bokhari and Rihi). Each of these headaches is treated either through phlebotomy or wet cupping.CONCLUSION: In Persian Medicine, Fasd and Hijamat are easy and economical methods for treatment of headache with reasonable results.
