2Individualized prediction of perineural invasion in colorectal cancer: development and validation of a radiomics prediction model显示文摘Objective: To develop and validate a radiomics prediction model for individualized prediction of perineural invasion(PNI) in colorectal cancer(CRC).Methods: After computed tomography(CT) radiomics features extraction, a radiomics signature was constructed in derivation cohort(346 CRC patients). A prediction model was developed to integrate the radiomics signature and clinical candidate predictors [age, sex, tumor location, and carcinoembryonic antigen(CEA) level]. Apparent prediction performance was assessed. After internal validation, independent temporal validation(separate from the cohort used to build the model) was then conducted in 217 CRC patients. The final model was converted to an easy-to-use nomogram.Results: The developed radiomics nomogram that integrated the radiomics signature and CEA level showed good calibration and discrimination performance [Harrell's concordance index(c-index): 0.817; 95% confidence interval(95% CI): 0.811–0.823]. Application of the nomogram in validation cohort gave a comparable calibration and discrimination(c-index: 0.803; 95% CI: 0.794–0.812).Conclusions: Integrating the radiomics signature and CEA level into a radiomics prediction model enables easy and effective risk assessment of PNI in CRC. 3Effect of L-NAME on nitric oxide and gastrointestinal motility alterations in cirrhotic rats显示文摘AIM: To invsstigare the effect of L-NAME on nitric oxide andgastriubtestubal motility alterations in cirrhotic ratsMETHODS: Rats with cirrhosis induced by carbontetrachloride were randomly divided into two groups, one( n= 13) receiving 0. 5 mg@ kg-1 per clay of NG-nitro-L-argininemethyl ester (L-NAME), a nitric oxide synthase inhibitor,for 10 days, whereas the other group ( n = 13) and control( n = 10) rats were administrated the same volume of 9 g@ L-1saline.Half gastric emptying time and 2 h residual rate weremeasured by SPECT, using 99m Tc-DTPA-labeled bariumsuifate as test meal. Gastrointestinal transition time wasrecorded simultaneously. Serum concentration of nitrcoxide (NO) was determined by the kinetic cadmiunreduction and colorimetric methods. ImmunohistochemicalSABC method was used to observe the expression anddistribution of three types of nitric oxide synthase (NOS)isoforms in the mt gastrointestinal tract. Western blot wasused to detect expression of gastrointestinal NOS isoforms.RESULTS: Half gastric emptying time and trans-gastrointestinal time were significantly prolonged( 124.0 ± 26.4min; 33.7± 8.9min;72.1 ± 15.3 min; P<0.01), (12.4±0.5h; 9.5±0.3 h; 8.2±0.8 h; P<0.01), 2h residual rate wasraised in cirrhotic rots than in controls and cirrhotic ratstreated with L-NAME(54.9± 7.6 % ,13.7 ± 3.2 %, 34.9± 10.3%, P< 0.01). Serum concentration of NO was significantlyincreased in cirrhotic rots than in the other groups (8.20 ± 2.48)μmol@L-1, (5.94± 1.07) μmol@L-1 ,and control (5.66± 1.60) tμmol@L-1, P< 0.01. NOS staining intensities which weremainly located in the gastrointestinal tissues were markedlylower in cirrhotic rats than in the controls and cirrhotic ratsafter treated with L- NAME.CONCLUSION: Gastrointestinal motility was remarkablyinhibited in cirrhotic rats, which could he alleviated by L-NAME. 4Electroacupuncture reduces apoptotic index and inhibits p38 mitogen-activated protein kinase signaling pathway in the hippocampus of rats with cerebral ischemia/reperfusion injury显示文摘Electroacupuncture attenuates cerebral hypoxia and neuronal apoptosis induced by cerebral ischemia/reperfusion injury.To further identify the involved mechanisms,we assumed that electroacupuncture used to treat cerebral ischemia/reperfusion injury was associated with the p38 mitogen-activated protein kinase(MAPK) signaling pathway.We established rat models of cerebral ischemia/reperfusion injury using the modified Zea-Longa's method.At 30 minutes before model establishment,p38 MAPK blocker SB20358 was injected into the left lateral ventricles.At 1.5 hours after model establishment,electroacupuncture was administered at acupoints of Chize(LU5),Hegu(LI4),Zusanli(ST36),and Sanyinjiao(SP6) for 20 minutes in the affected side.Results showed that the combination of EA and SB20358 injection significantly decreased neurologic impairment scores,but no significant differences were determined among different interventional groups.Hematoxylin-eosin staining also showed reduced brain tissue injuries.Compared with the SB20358 group,the cells were regularly arranged,the structures were complete,and the number of viable neurons was higher in the SB20358 + electroacupuncture group.Terminal deoxynucleotidyl transferase(Td T)-mediated d UTP nick-end labeling assay showed a decreased apoptotic index in each group,with a significant decrease in the SB20358 + electroacupuncture group.Immunohistochemistry revealed reduced phosphorylated p38 expression at 3 days in the electroacupuncture group and SB20358 + electroacupuncture group compared with the ischemia/reperfusion group.There was no significant difference in phosphorylated p38 expression between the ischemia/reperfusion group and SB20358 group.These findings confirmed that the electroacupuncture effects on mitigating cerebral ischemia/reperfusion injury are possibly associated with the p38 MAPK signaling pathway.A time period of 3 days could promote the repair of ischemic cerebral nerves.Xiao Lan Xin Zhang Guo-ping Zhou Chun-xiao Wu Chun Li Xiu-hong Xu 2017Neural Regeneration Research2017,12,3:18

7Human avian influenza A (H5N1) virus infection in China显示文摘Highly pathogenic influenza A (H5N1) virus causes a widespread poultry deaths worldwide. The first human H5N1 infected case was reported in Hong Kong Special Administrative Region of China in 1997. Since then, the virus re-emerged in 2003 and continues to infect people worldwide. Currently, over 400 human infections have been reported in more than 15 countries and mortality rate is greater than 60%. H5N1 viruses still pose a potential pandemic threat in the future because of the continuing global spread and evolution. 9PD-L1 is a direct target of cancer-FOXP3 in pancreatic ductal adenocarcinoma(PDAC),and combined immunotherapy with antibodies against PD-L1 and CCL5 is effective in the treatment of PDAC显示文摘High expression of PD-L1 marks the poor prognosis of pancreatic ductal adenocarcinomas(PDAC).However,the regulatory mechanism of PD-L1 remains elusive.We recently reported that cancer Forkhead box protein 3(Cancer-FOXP3 or C-FOXP3)promoted immune evasion of PDAC by recruiting Treg cells into PDAC via upregulation of CCL5.In this study,we confirmed that PD-L1 was overexpressed in PDAC samples from two independent cohorts of patients with radical resection.Moreover,C-FOXP3 was colocalized and correlated with the expression of PD-L1 in tumor cells at the mRNA and protein levels,and this finding was confirmed by the The Cancer Genome Atlas(TCGA)database.Chromatin immunoprecipitation(ChIP)revealed that C-FOXP3 directly bound to the promoter region of PD-L1 in pancreatic cancer cells.Furthermore,overexpression of C-FOXP3 activated the luciferase reporter gene under the control of the PD-L1 promoter.However,mutation of the binding motif-a completely reversed the luciferase activity.In addition,C-FOXP3-induced upregulation of PD-L1 effectively inhibited the activity of CD8+T cells.Based on our recent finding that the CCL-5 antibody achieved a better response to PDAC models with high C-FOXP3 levels,we further demonstrated that the PD-L1 antibody strengthened the antitumor effect of CCL-5 blockade in xenograft and orthotopic mouse models with high C-FOXP3 levels.In conclusion,C-FOXP3 directly activates PD-L1 and represents a core transcription factor that mediates the immune escape of PDAC.Combined blockade of PD-L1 and CCL-5 may provide an effective therapy for patients with PDAC that have high C-FOXP3 levels.Xiuchao Wang Xin Li Xunbin Wei Haiping Jiang Chungen Lan Shengyu Yang Han Wang Yanhui Yang Caijuan Tian Zanmei Xu Jiangyan Zhang Jihui Hao He Ren 2020Signal Transduction and Targeted Therapy2020,5,1:11
10Rapid generation of genetic diversity by multiplex CRISPR/Cas9 genome editing in rice显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)-associated endonuclease 9(CRISPR/Cas9) system has emerged as a promising technology for specific genome editing in many species. Here we constructed one vector targeting eight agronomic genes in rice using the CRISPR/Cas9 multiplex genome editing system. By subsequent genetic transformation and DNA sequencing, we found that the eight target genes have high mutation efficiencies in the T_0 generation. Both heterozygous and homozygous mutations of all editing genes were obtained in T_0 plants. In addition, homozygous sextuple, septuple, and octuple mutants were identified. As the abundant genotypes in T_0 transgenic plants, various phenotypes related to the editing genes were observed. 11Sealing properties and structure optimization of packer rubber under high pressure and high temperature显示文摘During hydraulic fracturing operations of low-permeability reservoirs,packers are the key component to ensure the success of multistage fracturing.Packers enable sections of the wellbore to be sealed off and separately fractured by hydraulic pressure,one at a time,while the remainder of the wellbore is not affected.However,reliable sealing properties of the packer rubber are required to meet the high-pressure and high-temperature(HPHT)conditions of reservoirs(such as 70 MPa and 170 ℃).In this study,the structures of the packer rubber with two different materials are optimized numerically by ABAQUS and validated by experiments.The optimization process starts from the packer rubber with a conventional structure,and then,the weakest spots are identified by ABAQUS and improved by slightly varying its structure.This process is iterative,and the final optimized structure of a single rubber barrel with expanding back-up rings is achieved.For the structure of three rubber barrels with metallic protective covers,both HNBR and AFLAS fail under HPHT conditions.For the final optimized structure,the packer rubber made of AFLAS can work better under HPHT than that made of HNBR which ruptures after setting.The results show that the optimized structure of a single rubber barrel with expanding back-up rings and the material AFLAS are a good combination for the packer rubber playing an excellent sealing performance in multistage fracturing in horizontal wells.Wen-Jian Lan Han-Xiang Wang Xin Zhang Sheng-Shan Chen 2019Petroleum Science2019,16,3:11

18Prediction of Coke Yield of FCC Unit Using Different Artificial Neural Network Models显示文摘In fluid catalytic cracking(FCC) unit, it is greatly important to control the coke yield, since the increase of coke yield not only leads to the reduction of total liquid yield, but also affects the heat balance and operation of FCC unit. Consequently, it is significant to predict the coke yield accurately. The coke formation and burning reactions are affected by many parameters which influence each other, so it is difficult to establish a prediction model using traditional models. This paper combines the industrial production data and establishes a generalized regression neural network(GRNN) model and a back propagation(BP) neural network model to predict the coke yield respectively. The comparison and analysis results show that the accuracy and stability of the BP neural network prediction results are better than that of the GRNN. Then, the particle swarm optimization to optimize BP neural network(PSO-BP) and genetic algorithm to optimize the BP neural network(GA-BP) were further used to improve the prediction precision. The comparison of these models shows that they can improve the prediction precision. 20World’s first spaceflight on-orbit demonstration of a flexible solar array system based on shape memory polymer composites显示文摘With a 10%reversible compressive strain in more than 10 deformation cycles,the shape memory polymer composites(SMPCs)could be used for deployable structure and releasing mechanism.In this paper,without traditional electro-explosive devices or motors/controllers,the deployable SMPC flexible solar array system(SMPC-FSAS)is studied,developed,ground-based tested,and finally on-orbit validated.The epoxy-based SMPC is used for the rolling-out variable-stiffness beams as a structural frame as well as an actuator for the flexible blanket solar array.The releasing mechanism is primarily made of the cyanate-based SMPC,which has a high locking stiffness to withstand 50 g gravitational acceleration and a large unlocking displacement of 10 mm.The systematical mechanical and thermal qualification tests of the SMPC-FSAS flight hardware were performed,including sinusoidal sweeping vibration,shocking,acceleration,thermal equilibrium,thermal vacuum cycling,and thermal cycling test.The locking function of the SMPC releasing mechanisms was in normal when launching aboard the SJ20 Geostationary Satellite on 27 Dec.,2019.The SMPC-FSAS flight hardware successfully unlocked and deployed on 5 Jan.,2020 on geostationary orbit.The triggering signal of limit switches returned to ground at the 139 s upon heating,which indicated the successful unlocking function of SMPC releasing mechanisms.A pair of epoxy-based SMPC rolled variable-stiffness tubes,which clapped the flexible blanket solar array,slowly deployed and finally approached an approximate 100%shape recovery ratio within 60 s upon heating.The study and on-orbit successful validation of the SMPC-FSAS flight hardware could accelerate the related study and associated productions to be used for the next-generation releasing mechanisms as well as space deployable structures,such as new releasing mechanisms with low-shocking,testability and reusability,and ultra-large space deployable solar arrays.LAN Xin LIU LiWu ZHANG FengHua LIU ZhengXian WANG LinLin LI QiFeng PENG Fan HAO SiDa DAI WenXu WAN Xue TANG Yong WANG Mian HAO YanYan YANG Yang YANG Cheng LIU YanJu LENG JinSong 2020Science China(Technological Sciences)2020,63,8:7
