1Combined prednisone and levothyroxine improve treatment of severe thrombocytopenia in hepatitis B with compensatory cirrhosis accompanied by subclinical and overt hypothyroidism显示文摘The aim of the present study was to investigate the relationship between hypothyroidism and thrombocytopenia in hepatitis Brelated compensatory liver cirrhosis and to determine whether treatment with levothyroxine and prednisone is superior in a multicenter, open-label, observational study in China. In total, 125 consecutive hepatitis B-related compensated liver cirrhosis patients with severe thrombocytopenia accompanied by hypothyroidism were included. The patients were divided into four groups according to treatment strategy: a control group(n=29), a prednisone group(n=25), a levothyroxine group(n=32) and a prednisone plus levothyroxine group(n=39). Severe thrombocytopenia was more prevalent in hepatitis B-associated compensatory liver cirrhosis patients with hypothyroidism than in euthyroid patients(29.6% vs. 14.7%, P<0.05). Combination treatment with prednisone and levothyroxine decreased the risk of bleeding and improved platelet recovery compared to control treatment and treatment with either prednisone or levothyroxine alone. The platelet count before therapy, serum thyroid stimulating hormone and combination treatment with prednisone and levothyroxine were associated with bleeding events. Therefore, the present study suggests that hypothyroidism is associated with severe thrombocytopenia in hepatitis B-associated compensatory liver cirrhosis. 2Improved clinical outcomes of rhG-CSF-mobilized blood and marrow haploidentical transplantation compared to propensity score-matched rhG-CSF-primed peripheral blood stem cell haploidentical transplantation:a multicenter study显示文摘The effects of haploidentical rhG-CSF-mobilized blood and marrow transplantation(HBMT) on hematological malignances are well established. Previous prospective single-center studies have demonstrated better survival after HBMT versus haploidentical rhG-CSF-mobilized peripheral blood stem cell transplantation(HPBSCT) for acute leukemia(AL) not in remission(NR) or in more than the second complete remission(>CR2). To test the hypothesis that HBMT is still superior to HPBSCT for patients with AL, multiple myeloma(MM), or non-Hodgkin lymphoma(NHL) in CR1/CR2 and for patients with chronic myeloid leukemia in the first and second chronic phase lacking a matched donor, we designed a propensity score method-based multicenter study.Hematopoietic recovery, acute graft-versus-host disease(aGVHD), and chronic GVHD were comparable between the HBMT group(n=168) and the HPBSCT group(n=42). No significant differences were found in non-relapse mortality rate(20.17%±3.58%and 27.24%±7.16%, P=0.18) or relapse rate(19.96%±3.72% and 28.49%±8.25%, P=0.32) between the HBMT group and the HPBSCT group. HBMT recipients had better overall survival(65.0%±4.2% and 54.2%±8.3%, P=0.037) and disease-free survival(59.9%±4.6% and 44.3%±8.7%, P=0.051). Multivariate analysis showed that HPBSCT was associated with poorer DFS(HR(95%CI), 1.639(0.995–2.699), P=0.052). 3Accurate Determination of Zr Isotopic Ratio in Zircons by Femtosecond Laser Ablation MC-ICP-MS with “Wet” Plasma Technique显示文摘This work evaluates the use of femtosecond laser ablation multiple collector inductively coupled plasma mass spectrometry(fs-LA-MC-ICP-MS) for Zr isotopic analysis in zircons. The mass fractionation caused by instrumental mass discrimination was corrected by a combination of internal correction using Sr as an internal standard(coming from a NIST SRM 987 standard solution) and external correction using a matrix-matched standard. Several important instrument parameters were investigated, such as the effect of the addition of N;and 'wet' plasma condition, the mass fractionation behaviors between Zr isotopes and Sr isotopes, the position effect in laser ablation cell and the effect of laser ablation parameters(laser spot size and energy density). The Zr isotope compositions of seven zircons(GJ-1, 91500, Ple?ovice, Rak-17, Paki, Aus and Mala) were determined by the developed fs-LAMC-ICP-MS and thermal ionization mass spectrometry(TIMS). Our fs-LA-MC-ICP-MS results for Zr isotope compositions agreed with TIMS analyses within analytical uncertainties, indicating the presented method is a suitable tool to resolve isotopic zoning in natural zircons. 5Seasonal hydrological dynamics govern lifestyle preference of aquatic antibiotic resistome显示文摘Antibiotic resistance genes(ARGs)are a well-known environmental concern.Yet,limited knowledge exists on the fate and transport of ARGs in deep freshwater reservoirs experiencing seasonal hydrological changes,especially in the context of particle-attached(PA)and free-living(FL)lifestyles.Here,the ARG profiles were examined using high-throughput quantitative PCR in PA and FL lifestyles during four seasons representing two hydrological phenomena(vertical mixing and thermal stratification)in the Shuikou Reservoir(SR),Southern China.The results indicated that seasonal hydrological dynamics were critical for influencing the ARGs in PA and FL and the transition of ARGs between the two lifestyles.ARG profiles both in PA and FL were likely to be shaped by horizontal gene transfer.However,they exhibited distinct responses to the physicochemical(e.g.,nutrients and dissolved oxygen)changes under seasonal hydrological dynamics.The particle-association niche(PAN)index revealed 94 non-conservative ARGs(i.e.,no preferences for PA and FL)and 23 and 16 conservative ARGs preferring PA and FL lifestyles,respectively.A sharp decline in conservative ARGs under stratified hydrologic suggested seasonal influence on the ARGs transition between PA and FL lifestyles.Remarkably,the conservative ARGs(in PA or FL lifestyle)were more closely related to bacterial OTUs in their preferred lifestyle than their counterparts,indicating lifestyle-dependent ARG enrichment.Altogether,these findings enhanced our understanding of the ARG lifestyles and the role of seasonal hydrological changes in governing the ARG transition between the lifestyles in a typical deep freshwater ecosystem.Bob Adyari Liyuan Hou Lanping Zhang Nengwang Chen Feng Ju Longji Zhu Chang-Ping Yu Anyi Hu 2023Environmental Science and Ecotechnology2023,,1:0
6Tyrosine phosphatase SHP2 in ovarian granulosa cells balances follicular development by inhibiting PI3K/AKT signaling显示文摘In mammals,the growth and maturation of oocytes within growing follicles largely depends on ovarian granulosa cells(GCs)in response to gonadotropin stimulation.Many signals have been shown to regulate GC proliferation and apoptosis.However,whether the tyrosine phosphatase SHP2 is involved remains unclear.In this study,we identified the crucial roles of SHP2 in modulating GC proliferation and apoptosis.The production of both mature oocytes and pups was increased in mice with Shp2 specifically deleted in ovarian GCs via Fshr-Cre.Shp2 deletion simultaneously promoted GC proliferation and inhibited GC apoptosis.Furthermore,Shp2 deficiency promoted,while Shp2 overexpression inhibited,the proliferation of cultured primary mouse ovarian GCs and the human ovarian granulosa-like tumor cell line KGN in vitro.Shp2 deficiency promoted follicule-stimulating hormone(FSH)-activated phosphorylation of AKT in vivo.SHP2 deficiency reversed the inhibitory effect of hydrogen peroxide(H_(2)O_(2))on AKT activation in KGN cells.H_(2)O_(2) treatment promoted the interaction between SHP2 and the p85 subunit of PI3K in KGN cells.Therefore,SHP2 in GCs may act as a negative modulator to balance follicular development by suppressing PI3K/AKT signaling.The novel function of SHP2 in modulating proliferation and apoptosis of GCs provides a potential therapeutic target for the clinical treatment of follicle developmental dysfunction.Xiaoli Wei Lanping Zheng Yingpu Tian Haibin Wang Youqiang Su Gensheng Feng Chao Wang Zhongxian Lu 2022Journal of Molecular Cell Biology2022,14,7:0
