1Risk factors of infected pancreatic necrosis secondary to severe acute pancreatitis显示文摘BACKGROUND: Severe acute pancreatitis(SAP) remains a clinical challenge with considerable morbidity and mortality.An early identification of infected pancreatic necrosis(IPN), a life-threatening evolution secondary to SAP, is obliged for a more preferable prognosis. Thus, the present study was conducted to identify the risk factors of IPN secondary to SAP. METHODS: The clinical data of patients with SAP were retrospectively analyzed. Univariate and multivariate logistic regression analyses were sequentially performed to assess the associations between the variables and the development of IPN secondary to SAP. A receiver operating characteristic(ROC) curve was created for each of the qualified independent risk factors. RESULTS: Of the 115 eligible patients, 39(33.9%) progressed to IPN, and the overall in-hospital mortality was 11.3%(13/115).The early enteral nutrition(EEN)(P=0.0092, OR=0.264), maximum intra-abdominal pressure(IAP)(P=0.0398, OR=1.131)and maximum D-dimer level(P=0.0001, OR=1.006) in the first three consecutive days were independent risk factors associated with IPN secondary to SAP. The area under ROC curve(AUC) was 0.774 for the maximum D-dimer level in the first three consecutive days and the sensitivity was 90% and the specificity was 58% at a cut-off value of 933.5 μg/L; the AUC was 0.831 for the maximum IAP in the first three consecutive days and the sensitivity was 95% and specificity was 58%at a cut-off value of 13.5 mm Hg. 2Minimally invasive surgery alone compared with intensity-modulated radiotherapy for primary stage I nasopharyngeal carcinoma显示文摘Background:The National Comprehensive Cancer Network guidelines recommend intensity-modulated radiotherapy(IMRT)as the primary curative treatment for newly diagnosed nasopharyngeal carcinoma(NPC),but the radiation-related complications and relatively high medical costs remain a consequential burden for the patients.Endoscopic nasopharyngectomy(ENPG)was successfully applied in recurrent NPC with radiation free and relatively low medical costs.In this study,we examined whether ENPG could be an effective treatment for localized stage I NPC.Methods:Ten newly diagnosed localized stage I NPC patients voluntarily received ENPG alone from June 2007 to September 2017 in Sun Yat-sen University Cancer Center.Simultaneously,the data of 329 stage I NPC patients treated with IMRT were collected and used as a reference cohort.The survival outcomes,quality of life(QOL),and medical costs between two groups were compared.Results:After a median follow-up of 59.0 months(95%CI 53.4-64.6),no death,locoregional recurrence,or distant metastasis was observed in the 10 patients treated with ENPG.The 5-year overall survival,local relapse-free survival,regional relapse-free survival,and distant metastasis-free survival among the ENPG-treated patients was similar to that among the IMRT-treated patients(100%vs.99.1%,100%vs.97.7%,100%vs.99.0%,100%vs.97.4%,respectively,P>0.05).In addition,compared with IMRT,ENPG was associated with decreased total medical costs($4090.42±1502.65 vs.$12620.88±4242.65,P<0.001)and improved QOL scores including dry mouth(3.3±10.5 vs.34.4±25.8,P<0.001)and sticky saliva(3.3±10.5 vs.32.6±23.3,P<0.001).Conclusions:ENPG alone was associated with promising long-term survival outcomes,low medical costs,and satisfactory QOL and might therefore be an alternative strategy for treating newly diagnosed localized stage I NPC patients who refused radiotherapy.However,the application of ENPG should be prudent,and prospective clinical tri-als were needed to further verify the results.You-Ping Liu Xing Lv Xiong Zou Yi-Jun Hua Rui You Qi Yang Le Xia Shao-Yan Guo Wen Hu Meng-Xia Zhang Si-Yuan Chen Mei Lin Yu-Long Xie Li-Zhi Liu Rui Sun Pei-Yu Huang Wei Fan Xiang Guo Ming-Huang Hong Ming-Yuan Chen 2019Cancer Communications2019,39,1:17
3Optimization of Agrobacterium tumefaciens-Mediated Transformation Systems in Tea Plant(Camellia sinensis)显示文摘In this study, an efficient plant regeneration protocol in vitro and transformation by Agrobacterium-mediated method of Camellia sinensis was achieved, which would lay the foundation for genetic improvement of tea plant by genetic engineering technology. The cotyledon callus of C.sinensis were used as the receptors for transformation by Agrobacterium tumefaciens EHA105 containing PS1aG-3. Some factors which affected the result of Agrobacterium-mediated transformation of C. sinensis were studied on the basis of GUS transient expression system. The optimum system of Agrobacterium-mediated transformation was that the cotyledon callus were pre-cultured for 3 d, and then infected by EHA105 for 15 min followed by 3 d co-culture in the dark on the YEB medium containing 150 μmol·L^(-1) acetosyringone(AS). The transient expression rate of GUS gene was 62.6%. After being delayed selective culture for 3 d, infected callus were transferred into the differentiation medium and the root induction medium both of which were supplemented with 100 mg·L^(-1) spectinomycin, and then resistant seedlings of C. sinensis were obtained. 4ZrC-ZrB_2-SiC ceramic nanocomposites derived from a novel single-source precursor with high ceramic yield显示文摘For the first time, ZrC-ZrB_2-SiC ceramic nanocomposites were successfully prepared by a single-source-precursor route, with allylhydridopolycarbosilane(AHPCS),triethylamine borane(TEAB),and bis(cyclopentadienyl) zirconium dichloride(Cp_2 ZrCl_2) as starting materials. The polymer-to-ceramic transformation and thermal behavior of obtained single-source precursor were characterized by means of Fourier transform infrared spectroscopy(FT-IR) and thermal gravimetric analysis(TGA). The results show that the precursor possesses a high ceramic yield about 85% at 1000 ℃.The phase composition and microstructure of formed ZrC-ZrB_2-SiC ceramics were investigated by means of X-ray diffraction(XRD) and high resolution transmission electron microscopy(HRTEM).Meanwhile, the weight loss and chemical composition of the resultant ZrC-ZrB_2-SiC nanocomposites were investigated after annealing at high temperature up to 1800 ℃. 6Long-term adenosine A1 receptor activation-induced sortilin expression promotes α-synuclein upregulation in dopaminergic neurons显示文摘Prolonged activation of adenosine A1 receptor likely leads to damage of dopaminergic neurons and subsequent development of neurodegenerative diseases.However,the pathogenesis underlying long-term adenosine A1 receptor activation-induced neurodegeneration remains unclear.In this study,rats were intraperitoneally injected with 5 mg/kg of the adenosine A1 receptor agonist N6-cyclopentyladenosine(CPA)for five weeks.The mobility of rats was evaluated by forced swimming test,while their cognitive capabilities were evaluated by Y-maze test.Expression of sortilin,α-synuclein,p-JUN,and c-JUN proteins in the substantia nigra were detected by western blot analysis.In addition,immunofluorescence staining of sortilin andα-synuclein was performed to detect expression in the substantia nigra.The results showed that,compared with adenosine A1 receptor antagonist 8-cyclopentyl-1,3-dipropylxanthine(5 mg/kg)+CPA co-treated rats,motor and memory abilities were reduced,surface expression of sortin andα-synuclein in dopaminergic neurons was reduced,and total sortilin and totalα-synuclein were increased in CPA-treated rats.MN9D cells were incubated with 500 nM CPA alone or in combination with 10μM SP600125(JNK inhibitor)for 48 hours.Quantitative real-time polymerase chain reaction analysis of sortilin andα-synuclein mRNA levels in MN9D cells revealed upregulated sortilin expression in MN9D cells cultured with CPA alone,but the combination of CPA and SP600125 could inhibit this expression.Predictions made using Jasper,PROMO,and Alibaba online databases identified a highly conserved sequence in the sortilin promoter that was predicted to bind JUN in both humans and rodents.A luciferase reporter assay of sortilin promoter plasmid-transfected HEK293T cells confirmed this prediction.After sortilin expression was inhibited by sh-SORT1,expression of p-JUN and c-JUN was detected by western blot analysis.Long-term adenosine A1 receptor activation levels upregulatedα-synuclein expression at the post-transcriptional level by affecting sortilin expression.The online tool Raptor-X-Binding and Discovery Studio 4.5 prediction software predicted that sortilin can bind toα-synuclein.Co-immunoprecipitation revealed an interaction between sortilin andα-synuclein in MN9D cells.Our findings indicate that suppression of prolonged adenosine A1 receptor activation potently inhibited sortilin expression andα-synuclein accumulation,and dramatically improved host cognition and kineticism.This study was approved by the University Committee of Animal Care and Supply at the University of Saskatchewan(approval No.AUP#20070090)in March 2007 and the Animals Ethics Committee of University of South China(approval No.LL0387-USC)in June 2017.Yun-Cheng Lv An-Bo Gao Jing Yang Li-Yuan Zhong Bo Jia Shu-Hui Ouyang Le Gui Tian-Hong Peng Sha Sun Francisco S.Cayabyab 2020Neural Regeneration Research2020,15,4:4
7Hot deformation behavior and microstructure evolution of a Mg–Gd–Nd–Y–Zn alloy显示文摘The hot deformation behavior of homogenized Mg–6.5Gd–1.3Nd–0.7Y–0.3Zn alloy was investigated during compression at temperatures of 250–400 ℃ and at strain rates ranging from 0.001 to 0.100 s^(-1). Microstructure analyses show that the flow behaviors are associated with the deformation mechanisms. At the lower temperatures(250–300 ℃), deformation twinning is triggered due to the difficult activation of dislocation cross-slip. Dynamic recrystallization(DRX) accompanied by dynamic precipitation occurs at the temperature of 350 ℃ and influences the softening behavior of the flow.DRX that develops extensively at original grain boundaries is the main softening mechanism at the high temperature of 400 ℃ and eventually brings a more homogeneous microstructure than that in other deformation conditions. 8The Relationship between Intrahepatic Distribution of Hepatitis B Virus Core Antigen and Serum ALT, HBV DNA Levels and HBeAg Status显示文摘Objective The clinical significance of differential distribution of hepatitis B virus(HBV)nucleocapsid antigen in hepatocytes remains unknown.The goal of this study is to determine the relationship between distinct HBV core antigen distribution pattern and alanine transaminase(ALT),liver histological inflammatory activity grades,serum HBeAg status and HBV DNA level.Methods Total of 958 cases with chronic hepatitis B were recruited into this study.Liver function tests,serum HBV DNA level,serological HBV markers and liver immunohistochemistry were examined according to the conventional instructions.Chi Square tests were performed to analyze the differences among these groups.Results It was found that 552(58%)cases were tested positive for HBV core antigen by immunohistochemical staining.Cytoplasmic hepatitis B core antigen(HBcAg)expression correlated with ALT level and serum HBV DNA and liver inflammatory activity scores,however,nuclear HBcAg expression in hepatocytes was associated with normal ALT level,lower liver inflammatory activity score and higher serum HBV DNA level and rate of HBeAg positivity.Both nuclear and cytoplasmic HBcAg expression in hepatocytes associated with a middle ALT level and liver inflammatory activity score,higher rate of serum detectable HBeAg and a higher HBV DNA level.However,undetectable core antigen was related to a lower ALT level and histological inflammatory activity grade,lower positive HBeAg rate and HBV DNA level.Conclusions Undetectable liver HBcAg is associated with HBV clearance,ALT normalization and hepatitis B e antigen(HBeAg)seroconversion,and cytoplasmic HBcAg expression associated with higher hepatic inflammatory activity.However,nuclear HBcAg expression correlates with immune tolerance characterized with normal ALT and lower liver inflammatory activity,higher HBV replication level and higher rate of HBeAg positivity.Qing He Qi-yuan Tang Xiao-hua Le De-liang Lv Xiang-mei Zhang Fei-jian Ao Yi-min Tang Shan Huang John Nunnari Gui-lin Yang 2012国际感染病学(电子版)2012,1,2:4
9Leveraging proficiency and preference for online Karaoke recommendation显示文摘Recently,many online Karaoke(KTV)platforms have been released,where music lovers sing songs on these platforms.In the meantime,the system automatically evaluates user proficiency according to their singing behavior.Recommending approximate songs to users can initialize singers5 participation and improve users,loyalty to these platforms.However,this is not an easy task due to the unique characteristics of these platforms.First,since users may be not achieving high scores evaluated by the system on their favorite songs,how to balance user preferences with user proficiency on singing for song recommendation is still open.Second,the sparsity of the user-song interaction behavior may greatly impact the recommendation task.To solve the above two challenges,in this paper,we propose an informationfused song recommendation model by considering the unique characteristics of the singing data.Specifically,we first devise a pseudo-rating matrix by combing users’singing behavior and the system evaluations,thus users'preferences and proficiency are leveraged.Then we mitigate the data sparsity problem by fusing users*and songs'rich information in the matrix factorization process of the pseudo-rating matrix.Finally,extensive experimental results on a real-world dataset show the effectiveness of our proposed model.Ming HE Hao GUO Guangyi LV Le WU Yong GE Enhong CHEN Haiping MA 2020Frontiers of Computer Science2020,14,2:3
10Exploring the underlying mechanism of action of a traditional Chinese medicine formula,Youdujing ointment,for cervical cancer treatment显示文摘Background:A traditional Chinese medicine formula,Youdujing(YDJ)ointment,is widely used for treatment of human papilloma virus-related diseases,such as cervical cancer.However,the underlying mechanisms by which active compounds of YDJ alleviates cervical cancer are still unclear.Methods:We applied a comprehensive network pharmacology approach to explore the key mechanisms of YDJ by integrating potential target identification,network analysis,and enrichment analysis into classical molecular docking procedures.First,we used network and enrichment analyses to identify potential therapeutic targets.Second,we performed molecular docking to investigate the potential active compounds of YDJ.Finally,we carried out a network-based analysis to unravel potentially effective drug combinations.Results:Network analysis yielded four potential therapeutic targets:ESRI,NFKB1,TNF,and AKT1.Molecular docking highlighted that these proteins may interact with four potential active compounds of YDJ:E4,Y2,Y20,and Y21.Finally,we found that Y2 or Y21 can act alone or together with E4 to trigger apoptotic cascades via the mitochondrial apoptotic pathway and estrogen receptors.Conclusion:Our study not only explained why YDJ is effective for cervical cancer treatment,but also lays a strong foundation for future clinical studies based on this traditional medicine.Lei Zhang Ji Lv Ming Xiao Li Yang Le Zhang 2021Quantitative Biology2021,9,3:2

12Inhibition of metabolic disorders in vivo and in vitro by main constituent of Coreopsis tinctoria显示文摘Objective:To investigate the effects of the ethyl acetate extract of Coreopsis tinctoria(EAEC)on insulin resistance(IR)in rats fed a high-fat diet.Methods:Male Sprague-Dawley(SD)rats were fed a HFD(60%fat)supplemented with EAEC for 8 weeks.The administration of EAEC to the rats with HFD-induced insulin resistance reduced hyperglycemia,plasma levels of insulin,and steatosis in the liver.Metabolomic study was used to analyze the metabolic levels of the high glucose-treated cells,control cells and marein-treated cells.Results:High glucose and high fat conditions caused a significant increase in blood glucose,insulin,serum TC,TG and LDL-C levels,leading to abnormal IR in rats.However,treatment with EAEC protects against HFD-induced IR by improving the fasting serum glucose homeostasis and lipid homeostasis.The high glucose conditions significantly decreased glycogen synthesis and increased PEPCK,G6Pase and Krebs cycle-related enzyme protein levels,leading to an abnormal metabolic state in Hep G2 cells.However,treatment with marein improved IR by increasing glucose uptake and glycogen synthesis and by downregulating PEPCK and G6Pase protein levels.The statistical analysis of the HPLC/MS data demonstrated that marein could restore the normal metabolic state.Conclusion:The results revealed that EAEC ameliorates IR in rats,and marein has the potential to improve IR by ameliorating glucose metabolism disorders.Bao-ping Jiang Qiu-yue Lv Jia-mei Xiang Liang Le Ke-ping Hu Li-jia Xu Pei-gen Xiao 2018Chinese Herbal Medicines2018,10,2:2
13A distinct D1-MSN subpopulation down-regulates dopamine to promote negative emotional state显示文摘Dopamine(DA)level in the nucleus accumbens(NAc)is critical for reward and aversion encoding.DA released from the ventral mesencephalon(VM)DAergic neurons increases the excitability of VM-projecting D1-dopamine receptor-expressing medium spiny neurons(D1-MSNs)in the NAc to enhance DA release and augment rewards.However,how such a DA positive feedback loop is regulated to maintain DA homeostasis and reward-aversion balance remains elusive.Here we report that the ventral pallidum(VP)projection of NAc D1-MSNs(D1^(NAc-VP))is inhibited by rewarding stimuli and activated by aversive stimuli.In contrast to the VM projection of D1-MSN(D1^(NAc-VM)),activation of D1^(NAc-VP) projection induces aversion,but not reward.D1^(NAc-VP) MSNs are distinct from the D1^(NAc-VM) MSNs,which exhibit conventional functions of D1-MSNs.Activation of D1^(NAc-VP) projection stimulates VM GABAergic transmission,inhibits VM DAergic neurons,and reduces DA release into the NAc.Thus,D1^(NAc-VP) and D1^(NAc-VM) MSNs cooperatively control NAc dopamine balance and reward-aversion states.Zhiyuan Liu Qiumin Le Yanbo Lv Xi Chen Jian Cui Yiming Zhou Deqin Cheng Chaonan Ma Xiujuan Su Lei Xiao Ruyi Yang Jiayi Zhang Lan Ma Xing Liu 2022Cell Research2022,32,2:2

15Surfactant-assisted fabrication of graphene frameworks endowing epoxy composites with superior thermal conductivity显示文摘With the rapid growth in electronic device performance,there has been an increasing demand for thermally conductive polymer composites to handle the thermal management issue,thus contributing to the great importance to develop the graphene framework,which is evaluated as the most promising reinforcements for enhancing the thermal conductivity of polymer.Vacuum filtration is a common method to fabricate graphene framework,whereas,it is available to prepare a framework with centimeter-scale thickness by filtrating the graphene-water dispersion,due to the fact of sample cracking caused by the mismatch of surface tension between graphene and water.In this work,a surfactantassisted strategy was proposed by adjusting the surface tension of the water close to that of graphene first,then performing a conventional filtration process,to fabricate graphene framework.As a result,a thick graphene framework(thickness:3 cm)was successfully prepared,and after embedding into epoxy,the framework endows the composite(13.6 wt%)with a high in-plane thermal conductivities of12.4 W/mK,which is equivalent to≈64 times higher than that of neat epoxy.Our method is simple and compatible with the conventional filtration process,suggesting great potential for the mass-production of graphene framework to meet the practical application requirements.Fan Zhang Chen Ye Wen Dai Le Lv Qilong Yuan Kuan W.A.Chee Ke Yang Nan Jiang Cheng-Te Lin Zhaolin Zhan Dan Dai He Li 2020Chinese Chemical Letters2020,31,1:2
