1Genomic variation,origin tracing,and vaccine development of SARS-CoV-2:A systematic review显示文摘COVID-19 has spread globally to over 200 countries with more than 40 million confirmed cases and one million deaths as of November 1,2020.The SARS-CoV-2 virus,leading to COVID-19,shows extremely high rates of infectivity and replication,and can result in pneumonia,acute respiratory distress,or even mortality.SARS-CoV-2 has been found to continue to rapidly evolve,with several genomic variants emerging in different regions throughout the world.In addition,despite intensive study of the spike protein,its origin,and molecular mechanisms in mediating host invasion are still only partially resolved.Finally,the repertoire of drugs for COVID-19 treatment is still limited,with several candidates still under clinical trial and no effective therapeutic yet reported.Although vaccines based on either DNA/mRNA or protein have been deployed,their efficacy against emerging variants requires ongoing study,with multivalent vaccines supplanting the first-generation vaccines due to their low efficacy against new strains.Here,we provide a systematic review of studies on the epidemiology,immunological pathogenesis,molecular mechanisms,and structural biology,as well as approaches for drug or vaccine development for SARSCoV-2.Tianbao Li Tao Huang Cheng Guo Ailan Wang Xiaoli Shi Xiaofei Mo Qingqing Lu Jing Sun Tingting Hui Geng Tian Leyi Wang Jialiang Yang 2021The Innovation2021,2,2:1
2Bilirubin inhibits the anticancer activity of sorafenib by blocking MCL-1 degradation in hepatocellular carcinoma cells显示文摘Objective:Sorafenib is a first-line drug for advanced hepatocellular carcinoma(HCC).Unfortunately,most patients with HCC do not respond to sorafenib,mainly because of the frequent development of drug resistance.Bilirubin is an end metabolite of heme catabolism and an indicator of liver function,but its direct role in regulating the anticancer activity of sorafenib in HCC cells is unclear.In the current study,we aimed to investigate the mechanism of action of bilirubin in sorafenib-mediated tumor suppression in HCC.Methods:A retrospective observational cohort of 100 patients receiving sorafenib was conducted to evaluate the potential role of bilirubin in predicting the prognosis of patients with HCC.Human HCC cell lines were treated with sorafenib in the absence or presence of bilirubin,and cell proliferation,apoptosis,and signaling pathways were assayed.The antagonistic effect of bilirubin toward sorafenib was assessed in nude mice bearing HCC xenografts.Results:Serum levels of bilirubin(including total,direct,and indirect bilirubin)negatively correlated with the overall survival of patients with HCC treated with sorafenib(P<0.05).Both in vitro and in vivo analyses demonstrated that bilirubin significantly abrogated sorafenib-mediated proliferation inhibition and apoptosis induction in HCC cells(P<0.05).Mechanically,bilirubin inhibited sorafenib-induced activation of GSK-3βand subsequent downstream MCL-1 degradation.Conclusions:Our study provides experimental evidence of the antagonistic effect of bilirubin toward sorafenib-mediated anticancer activity in HCC,and it suggests that bilirubin could be used to predict the efficacy of sorafenib treatment.Leyi Yao Qian Zhao Ding Yan Ziying Lei Yali Hao Jinghong Chen Qian Xue Xiaofen Li Qingtian Huang Daolin Tang QPing Dou Xin Chen Jinbao Liu 2022Cancer Biology & Medicine2022,19,7:0
3Characteristic metabolic and microbial profiles in acute ischemic stroke patients with phlegm-heat pattern显示文摘Objective:To explore the characteristics of plasma metabolites,feces gut microbiota and the crosstalk between gut microbiota and host metabolism in patients with acute ischemic stroke and phlegm-heat pattern(AIS-PHP).Methods:The metabolic and microbiome profiles of 20 AIS-PHP patients and 20 healthy controls(HCs)were analyzed using liquid chromatography-tandem mass spectrometry(LC-MS/MS)-based metabolomics and 16s rDNA sequencing,respectively.The covariation between LC-MS/MS-based metabolite data and 16s rDNA sequence data was presented.Results:Distinct alterations in the plasma metabolic phenotype of AIS-PHP patients were found,in which 16 metabolites differed significantly between the AIS-PHP patients and the HCs.These metabolites represented 17 different metabolic pathways,including amino acid metabolism,lipid metabolism,and nucleotide metabolism.Additionally,significant alterations of gut microbiota composition and taxon were revealed at the phylum level between the AIS-PHP patients and the HCs.In AIS-PHP,Bacteroidetes,Firmicutes,and Proteobacteria dominated.Moreover,some microbes that differed between the 2 groups manifested a sole association with certain metabolites,such as the connection between Bacteroides and inosine and between Lachnospiraceae_unclassified and hypoxanthine.Conclusion:The present study preliminarily investigated the metabolomic and gut microbiome characteristics of AIS-PHP patient indicators.The link between metabolic and microbial dysbiosis in AIS-PHP sheds new light on the function of gut microbiota and associated metabolomics in the pathogenesis of the disease.Xing Huang Yun Cao Leyi Zhang Shuren Li Xin Shu Xiyan Xin Kazuo Sugimoto Jia Liu Ying Gao Jingling Chang 2023Journal of Traditional Chinese Medical Sciences2023,10,3:0
4Metabotropic glutamate receptors(mGluRs)in epileptogenesis:an update on abnormal mGluRs signaling and its therapeutic implications显示文摘Epilepsy is a neurological disorder characterized by high morbidity,high recurrence,and drug resistance.Enhanced signaling through the excitatory neurotransmitter glutamate is intricately associated with epilepsy.Metabotropic glutamate receptors(mGluRs)are G protein-coupled receptors activated by glutamate and are key regulators of neuronal and synaptic plasticity.Dysregulated mGluR signaling has been associated with various neurological disorders,and numerous studies have shown a close relationship between mGluRs expression/activity and the development of epilepsy.In this review,we first introduce the three groups of mGluRs and their associated signaling pathways.Then,we detail how these receptors influence epilepsy by describing the signaling cascades triggered by their activation and their neuroprotective or detrimental roles in epileptogenesis.In addition,strategies for pharmacological manipulation of these receptors during the treatment of epilepsy in experimental studies is also summarized.We hope that this review will provide a foundation for future studies on the development of mGluR-targeted antiepileptic drugs.Leyi Huang Wenjie Xiao Yan Wang Juan Li Jiaoe Gong Ewen Tu Lili Long Bo Xiao Xiaoxin Yan Lily Wan 2024Neural Regeneration Research2024,19,2:0
5Retrieval of Winter Wheat Canopy Carotenoid Content with Ground-and Airborne-Based Hyperspectral Data显示文摘Accurate assessment of canopy carotenoid content(CC_(x+c)C)in crops is central to monitor physiological conditions in plants and vegetation stress,and consequently supporting agronomic decisions.However,due to the overlap of absorption peaks of carotenoid(C_(x+c))and chlorophyll(C_(a+b)),accurate estimation of carotenoid using reflectance where carotenoid absorb is challenging.The objective of present study was to assess CC_(x+c)C in winter wheat(Triticum aestivum L.)with ground-and aircraft-based hyperspectral measurements in the visible and near-infrared spectrum.In-situ hyperspectral reflectance were measured and airborne hyperspectral data were acquired during major growth stages of winter wheat in five consecutive field experiments.At the canopy level,a remarkable linear relationship(R^(2)=0.95,p<0.001)existed between C_(x+c) and Ca+b,and correlation between CC_(x+c)C and wavelengths within 400 to 1000 nm range indicated that CC_(x+c)C could be estimated using reflectance ranging from visible to near-infrared wavebands.Results of Cx+c assessment based on chlorophyll and carotenoid indices showed that red edge chlorophyll index(CI red edge)performed with the highest accuracy(R^(2)=0.77,RMSE=22.27μg/cm^(2),MAE=4.97μg/cm^(2)).Applying partial least square regression(PLSR)in CC_(x+c)C retrieval emphasized the significance of reflectance within 700 to 750 nm range in CC_(x+c)C assessment.Based on CI red edge index,use of airborne hyperspectral imagery achieved satisfactory results in mapping the spatial distribution of CC_(x+c)C.This study demonstrates that it is feasible to accurately assess CC_(x+c)C in winter wheat with red edge chlorophyll index provided that C_(x+c) correlated well with C_(a+b) at the canopy scale.it is therefore a promising method for CC_(x+c)C retrieval at regional scale from aerial hyperspectral imagery.Ting Cui Xianfeng Zhou Yufeng Huang Yanting Guo Yunrui Lin Leyi Song Jingcheng Zhang 2023Phyton-International Journal of Experimental Botany2023,92,9:0
