| 1 | Characterization of drug responses of mini patient-derived xenografts in mice for predicting cancer patient clinical therapeutic response显示文摘Background:Patient-derived organoids and xenografts(PDXs)have emerged as powerful models in functional diag-nostics with high predictive power for anticancer drug response.However,limitations such as engraftment failure and time-consuming for establishing and expanding PDX models followed by testing drug efficacy,and inability to subject to systemic drug administration for ex vivo organoid culture hinder realistic and fast decision-making in selecting the right therapeutics in the clinic.The present study aimed to develop an advanced PDX model,namely MiniPDX,for rapidly testing drug efficacy to strengthen its value in personalized cancer treatment.Methods:We developed a rapid in vivo drug sensitivity assay,OncoVee®MiniPDX,for screening clinically relevant regimens for cancer.In this model,patient-derived tumor cells were arrayed within hollow fiber capsules,implanted subcutaneously into mice and cultured for 7 days.The cellular activity morphology and pharmacokinetics were systematically evaluated.MiniPDX performance(sensitivity,specificity,positive and negative predictive values)was examined using PDX as the reference.Drug responses were examined by tumor cell growth inhibition rate and tumor growth inhibition rate in PDX models and MiniPDX assays respectively.The results from MiniPDX were also used to evaluate its predictive power for clinical outcomes.Results:Morphological and histopathological features of tumor cells within the MiniPDX capsules matched those both in PDX models and in original tumors.Drug responses in the PDX tumor graft assays correlated well with those in the corresponding MiniPDX assays using 26 PDX models generated from patients,including 14 gastric cancer,10 lung cancer and 2 pancreatic cancer.The positive predictive value of MiniPDX was 92%,and the negative predictive value was 81%with a sensitivity of 80%and a specificity of 93%.Through expanding to clinical tumor samples,Min-iPDX assay showed potential of wide clinical application.Conclusions:Fast in vivo MiniPDX assay based on capsule implantation was developed-to assess drug responses of both PDX tumor grafts and clinical cancer specimens.The high correlation between drug responses of paired MiniPDX and PDX tumor graft assay,as well as translational data suggest that MiniPDX assay is an advanced tool for personalized cancer treatment. | Feifei Zhang Wenjie Wang Yuan Long Hui Liu Jijun Cheng Lin Guo Rongyu Li Chao Meng Shan Yu Qingchuan Zhao Shun Lu Lili Wang Haitao Wang Danyi Wen | 2018 | Cancer Communications2018,38,1: | 20 |
| 2 | Noninvasive Ultrasound Deep Brain Stimulation for the Treatment of Parkinson’s Disease Model Mouse显示文摘Modulating basal ganglia circuitry is of great signifcance in the improvement of motor function in Parkinson’s disease(PD).Here,for the frst time,we demonstrate that noninvasive ultrasound deep brain stimulation(UDBS)of the subthalamic nucleus(STN)or the globus pallidus(GP)improves motor behavior in a subacute mouse model of PD induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP).Immunohistochemical c-Fos protein expression confrms that there is a relatively high level of c-Fos expression in the STN-UDBS and GP-UDBS group compared with sham group(both p<0.05).Furthermore,STN-UDBS or GPUDBS signifcantly increases the latency to fall in the rotarod test on day 9(p<0.05)and decreases the time spent climbing down a vertical rod in the pole test on day 12(p<0.05).Moreover,our results reveal that STN-UDBS or GP-UDBS protects the dopamine(DA)neurons from MPTP neurotoxicity by downregulating Bax(p<0.001),upregulating Bcl-2(p<0.01),blocking cytochrome c(Cyt C)release from mitochondria(p<0.05),and reducing cleaved-caspase 3 activity(p<0.01)in the ipsilateral substantia nigra(SN).Additionally,the safety of ultrasound stimulation is characterized by hematoxylin and eosin(HE)and Nissl staining;no hemorrhage or tissue damage is detected.Tese data demonstrate that UDBS enables modulation of STN or GP neural activity and leads to neuroprotection in PD mice,potentially serving as a noninvasive strategy for the clinical treatment of PD. | Hui Zhou Lili Niu Long Meng Zhengrong Lin Junjie Zou Xiangxiang Xia Xiaowei Huang Wei Zhou Tianyuan Bian Hairong Zheng | 2019 | Research2019,,1: | 9 |
| 3 | Vertically standing PtSe_2 film: a saturable absorber for a passively mode-locked Nd:LuVO_4 laser显示文摘The novel vertically standing Pt Se2 film on transparent quartz was prepared by selenization of platinum film deposited by the magnetron sputtering method, and an Nd:Lu VO4 passively mode-locked solid-state laser was realized by using the fabricated Pt Se2 film as a saturable absorber. The X-ray diffraction pattern and Raman spectrum of the film indicate its good crystallinity with a layered structure. The thickness of Pt Se2 film is measured to be 24 nm according to the cross-section height profile of the atomic force microscope image. Highresolution transmission electron microscopy images clearly demonstrate its vertically standing structure with an interlayer distance of 0.54 nm along the c-axis direction. The modulation depth(ΔT) and saturation fluence(Φs)of Pt Se2 film are measured to be 12.6% and 17.1 μJ∕cm2, respectively. The obtained mode-locked laser spectrum has a central wavelength of 1066.573 nm, with a 3 d B bandwidth of 0.106 nm. The transform limited pulse width of the mode-locked laser was calculated to be 15.8 ps. A maximum average output power of 180 m W with a working repetition rate of 61.3 MHz is obtained. | 4 | Effect of grain refinement and crystallographic texture produced by friction stir processing on the biodegradation behavior of a Mg-Nd-Zn alloy显示文摘The application of a single pass of friction stir processing(FSP) to Mg-Nd-Zn alloy resulted in grain refinement, texture evolution and redistribution of second phases, which improved corrosion resistance.In this work, an as-rolled Mg-Nd-Zn alloy was subjected to FSP. The microstructure in the processed zone of the FS-400 rpm alloy exhibited refined grains, a more homogenous grain size distribution, less second phases, and stronger basal plane texture. The corrosion behavior assessed using immersion tests and electrochemical tests in Hank’s solution indicated that the FS-400 rpm alloy had a lower corrosion rate, which was attributed to the increase of basal plane intensity and grain refinement. | 5 | Effect of lit-moxa stimulation of Guanyuan(CV 4) acupoint on levels of lactic acid and super-oxide dismutase in skeletal muscle of rats显示文摘OBJECTIVE:To observe the effect of lit-moxa stimulating acupoint Guanyuan(CV 4) on lactic acid and super-oxide dismutase(SOD) in skeletal muscle after exercise exhaustion.METHODS:Twenty-eight adult male Sprague-Dawley rats were randomly divided into normal control group,exhausted control group,exercise group and moxibustion group using exercise training and mild heating with lit-moxa stick as treatment methods.The exhausted control group,moxibustion group and exercise group received an exhaustive swimming after 20 days of intervention.Swimming exhausted times were recorded.Lactic acid and SOD concentration in soleus muscle were detected and compared between every two groups.RESULTS:The swimming exhausted times of the moxibustion group and the exercise group were significantly increased compare to the exhausted control group(P < 0.05).The lactic acid of the exhausted control group was significantly increased comparing with the normal control group(P <0.05),and the lactic acid of the moxibustion group and the exercise group were significantly lower than that of the exhausted control group(P < 0.05).The SOD level of the exhausted control group was significantly decreased comparing with the normal control group(P < 0.05),and the SOD level of the moxibustion group and the exercise group were significantly higher than that of the exhausted control group(both,P < 0.05).There was no statistical difference between the moxibustion group and the exercise group.CONCLUSION:With lit moxa stick,heat stimulating acupoint of Guanyuan(CV 4) decreased the levels of lactic acid and SOD in rat's skeletal muscle. | Mo Jie Wu Lili Sun Zhifang Wang Hongbin Ding Na He Long Zhang Lufen Liu Tonghua Li Xiaohong | 2015 | Journal of Traditional Chinese Medicine2015,35,2: | 6 |
| 6 | Effect of TiO_2@SiO_2 nanoparticles on the mechanical and UV-resistance properties of polyphenylene sulfide fibers显示文摘In order to avoid the inherent photo-catalysis and aggregation of TiO2 in PPS, TiO2 nanoparticles were coated with SiO2 layers, which were chosen as the UV absorbent to improve the UV stability of polyphenylene sul fi de(PPS) fi ber. The PPS–TiO2@SiO2 nanocomposites fi bers were prepared via melt spinning, and the nanocomposites fi bers displayed different crystallization behaviors on variation of the diameters of TiO2@SiO2 nanoparticles, as con fi rmed by Differential Scanning Calorimetry(DSC). The spinnability, breaking strength and UV-resistance properties of PPS nanocomposites fi bers, as measured by homemade melt spinning machine, Xenon-lamp Weather Resistance Test Chamber and Yarn Tensile Tester, manifested the dependence on the diameters. The addition of nanoparticles with the diameter of 25 nm improved the spinnability and the mechanical performance of PPS most, which is attributed to the heterogeneous nucleation effect of nanoparticles. The UVresistance properties of the PPS nanocomposites were improved by the addition of TiO2@SiO2 nanoparticles. | 7 | Ultraflat, broadband, and highly coherent supercontinuum generation in all-solid microstructured optical fibers with all-normal dispersion显示文摘High flatness, wide bandwidth, and high-coherence properties of supercontinuum(SC) generation in fibers are crucial in many applications. It is challenging to achieve SC spectra in a combination of the properties, since special dispersion profiles are required, especially when pump pulses with duration over 100 fs are employed. We propose an all-solid microstructured fiber composed only of hexagonal glass elements. The optimized fiber possesses an ultraflat all-normal dispersion profile, covering a wide wavelength interval of approximately 1.55 μm. An SC spectrum spanning from approximately 1030 to 2030 nm(corresponding to nearly one octave) with flatness<3 dB is numerically generated in the fiber with 200 fs pump pulses at 1.55 μm. The results indicate that the broadband ultraflat SC sources can be all-fiber and miniaturized due to commercially achievable 200-fs fiber lasers. Moreover, the SC pulses feature high coherence and a single pulse in the time domain, which can be compressed to 13.9-fs pulses with high quality even for simple linear chirp compensation. | 11 | Noninvasive ultrasound deep brain stimulation of nucleus accumbens induces behavioral avoidance显示文摘Ultrasound stimulation is an emerging noninvasive option in treating neuropsychiatric disorders. The present study investigates the behavioral alterations resulting from ultrasound stimulation on the nucleus accumbens(NAc) in freely moving mice. Our results show that an acute ultrasound stimulation on the NAc, rather than the visual cortex or auditory cortex, led to a pronounced avoidance behavior, while repeated NAc ultrasound stimulation resulted in an obvious conditioned place aversion with changes in synaptic protein(Glu A1/2 subunit) expression. Notably, NAc ultrasound stimulation suppressed the morphine-induced conditioned place preference. | 13 | The molecular conformation of silk fibroin regulates osteogenic cell behavior by modulating the stability of the adsorbed protein-material interface显示文摘Silk fibroin(SF)can be used to construct various stiff material interfaces to support bone formation.An essential preparatory step is to partially transform SF molecules from random coils toβ-sheets to render the material water insoluble.However,the influence of the SF conformation on osteogenic cell behavior at the material interface remains unknown.Herein,three stiff SF substrates were prepared by varying theβ-sheet content(high,medium,and low).The substrates had a comparable chemical composition,surface topography,and wettability.When adsorbed fibronectin was used as a model cellular adhesive protein,the stability of the adsorbed protein-material interface,in terms of the surface stability of the SF substrates and the accompanying fibronectin detachment resistance,increased with the increasingβ-sheet content of the SF substrates.Furthermore,(i)larger areas of cytoskeleton-associated focal adhesions,(ii)higher orders of cytoskeletal organization and(iii)more elongated cell spreading were observed for bone marrow-derived mesenchymal stromal cells(BMSCs)cultured on SF substrates with high vs.lowβ-sheet contents,along with enhanced nuclear translocation and activation of YAP/TAZ and RUNX2.Consequently,osteogenic differentiation of BMSCs was stimulated on highβ-sheet substrates.These results indicated that theβ-sheet content influences osteogenic differentiation of BMSCs on SF materials in vitro by modulating the stability of the adsorbed protein-material interface,which proceeds via protein-focal adhesion-cytoskeleton links and subsequent intracellular mechanotransduction.Our findings emphasize the role of the stability of the adsorbed protein-material interface in cellular mechanotransduction and the perception of stiff SF substrates with differentβ-sheet contents,which should not be overlooked when engineering stiff biomaterials. | Yanlin Long Xian Cheng John A.Jansen Sander G.C.Leeuwenburgh Jing Mao Fang Yang Lili Chen | 2021 | Bone Research2021,9,2: | 1 |

| 20 | High efficient oxygen reduction performance of Fe/Fe3C nanoparticles in situ encapsulated in nitrogen-doped carbon via a novel microwave-assisted carbon bath method显示文摘Fe-based carbon materials are widely considered promising to replace Pt/C as next-generation electrocatalysts towards oxygen reduction reaction (ORR). However, the preparation of Fe-based carbon materials is still carried out by conventional heating method (CHM). Herein, a novel microwave-assisted carbon bath method (MW-CBM) was proposed, which only took 35 min to synthesize Fe/Fe3C nanoparticles encapsulated in N-doped carbon layers derived from Prussian blue (PB). The catalyst contained large specific surface area and mesoporous structure, abundant Fe-Nx and C–N active sites, unique core-shell structure. Due to the synergistic effects of these features, the as-prepared Fe/Fe3C@NC-2 displayed outstanding ORR activity with onset potential of 0.98 VRHE and halfwave potential of 0.87 VRHE, which were more positive than 20 wt.% Pt/C (0.93 VRHE and 0.82 VRHE).
