| 6 | The landscape of investigator‐initiated oncology trials conducted in China's Mainland during the past decade(2010–2019)显示文摘The number of clinical trials conducted in China's Mainland,including investigator‐initiated trials(IITs),has increased rapidly in recent years.However,there are few data on the characteristics of cancer‐related IITs.We performed a comprehensive analysis of the landscape of cancer‐related IITs in China's Mainland in the past decade.All cancer‐related IITs registered on two clinical trial registries in the United States(www.clinicaltrials.gov,CT.gov)and China's Mainland(www.chictr.org.cn,ChiCTR)from 2010 to 2019 were identified.IITs were reviewed manually to validate classification,subcategorized by cancer type,and stratified by design characteristics to facilitate comparison across cancer types and with other specialties.A total of 8199 cancer‐related IITs were identified.The number of trials registered annually increased over time,especially in the last 5 years.Although interventional studies were predominant,randomized double‐blind studies accounted for only 8%of IITs.In the past decade,the trend for interventional studies conducted with different drugs increased year on year,although the increase in hormonal therapy IITs was not significant.Additionally,cancerrelated IITs were unevenly geographically distributed,with half concentrated in the economically developed cities Shanghai,Beijing,and Guangdong.We also found an increase in registration before participant enrollment(64.9%for trials in conducted in 2015–2019 vs.40.2%in 2010–2014,p<0.001)and data monitoring committee use(44.5%vs.40.0%,p=0.001)and a decrease in randomization(51.5%vs.62.7%,p<0.001)and funding(36.4%vs.56.3%,p<0.001)between these periods.We also observed changes in intervention type(decrease in cytotoxic drug therapy[34.8%vs.48.9%,p<0.001];increase in targeted therapy[17.8%vs.14.2%,p=0.004],immune checkpoint inhibitor therapy[6.6%vs.0.0%,p<0.001],and immune cell therapy[9.6%vs.4.5%,p<0.001]).Details of cancer‐related IITs conducted during the past decade illustrate the merits of oncology research in China's Mainland.Although the increased quantity of IITs is encouraging,limitations remain regarding the quality of clinical trials,regional imbalances,and funding allocation. | Ye Cao Lin‐Miao Ye Zhong Fan Wei Yang Li‐Ying Chen Yun Mei De‐Ying He Wen‐Jin Mo | 2023 | Cancer Innovation2023，2，1： | 0 |

| 13 | The pentatricopeptide repeat protein EMB1270 interacts with CFM2 to splice specific group Ⅱ introns in Arabidopsis chloroplasts显示文摘Chloroplast biogenesis requires the coordinated expression of chloroplast and nuclear genes.Here, we show that EMB1270, a plastid-localized pentatricopeptide repeat(PPR) protein, is required for chloroplast biogenesis in Arabidopsis thaliana.Knockout of EMB1270 led to embryo arrest,whereas a mild knockdown mutant of EMB1270 displayed a virescent phenotype. Almost no photosynthetic proteins accumulated in the albino emb1270 knockout mutant. By contrast, in the emb1270 knockdown mutant, the levels of ClpP1 and photosystem I(PSI) subunits were significantly reduced, whereas the levels of photosystem II(PSII) subunits were normal. Furthermore, the splicing efficiencies of the clpP1.2,ycf3.1, ndhA, and ndhB plastid introns were dramatically reduced in both emb1270 mutants. RNA immunoprecipitation revealed that EMB1270 associated with these introns in vivo. In an RNA electrophoretic mobility shift assay(REMSA), a truncated EMB1270 protein containing the 11 Nterminal PPR motifs bound to the predicted sequences of the clpP1.2, ycf3.1, and ndhA introns.In addition, EMB1270 specifically interacted with CRM Family Member 2(CFM2). | 19 | Development and validation of a nomogram to predict cardiac death after radiotherapy for esophageal cancer显示文摘Background:Patients frequently die from cardiac causes after radiotherapy for esophageal cancer.Early detection of cardiac death risk in these patients is crucial to improve clinical decision‐making and prognosis.Thus,we modeled the risk of cardiac death after irradiation for esophageal cancer.Methods:A retrospective analysis of 37,599 esophageal cancer cases treated with radiotherapy in the SEER database between 2000 and 2018 was performed.The selected cases were randomly assigned to the model development group(n=26,320)and model validation group(n=11,279)at a ratio of 7:3.We identified the risk factors most commonly associated with cardiac death by least absolute shrinkage and selection operator regression analysis(LASSO).The endpoints for model development and validation were 5‐and 10‐year survival rates.The net clinical benefit of the models was evaluated by decision curve analysis(DCA)and concordance index(C‐index).The performance of the models was further assessed by creating a receiver operating characteristic curve(ROC)and calculating the area under the curve(AUC).Kaplan‐Meier(K‐M)survival analysis was performed on the probability of death.Patients were classified according to death probability thresholds.Five‐and ten‐year survival rates for the two groups were shown using K‐M curves.Results:The major risk factors for cardiac death were age,surgery,year of diagnosis,sequence of surgery and radiotherapy,chemotherapy and a number of tumors,which were used to create the nomogram.The C‐indexes of the nomograms were 0.708 and 0.679 for the development and validation groups,respectively.DCA showed the good net clinical benefit of nomograms in predicting 5‐and 10‐year risk of cardiac death.The model exhibited moderate predictive power for 5‐and 10‐year cardiac mortality(AUC:0.833 and 0.854,respectively),and for the development and validation cohorts(AUC:0.76 and 0.813,respectively).Conclusions:Our nomogram may assist clinicians in making clinical decisions about patients undergoing radiotherapy for esophageal cancer based on early detection of cardiac death risk. | Xinfang Lv Xue Wu Kai Liu Xinke Zhao Chenliang Pan Jing Zhao Juan Chang Huan Guo Xiang Gao Xiaodong Zhi Chunzhen Ren Qilin Chen Hugang Jiang Chunling Wang Ying‐Dong Li | 2023 | Cancer Innovation2023，2，5： | 0 |
