| 2 | Expression of human soluble TRAIL in Chlamydomonas reinhardtii chloroplast显示文摘Tumor necrosis factor-related apop-tosis-inducing ligand (TRAIL) induces selectively apoptosis in various tumor cells and virus-infected cells, but rarely in normal cells. A chloroplast expres-sion vector, p64TRAIL, containing the cDNA coding for the soluble TRAIL (sTRAIL), was constructed with clpP-trnL-petB-chlL-rpl23-rpl2 as Chlamydomonas reinhardtii plastid homologous recombinant frag-ments and spectinomycin-resistant aadA gene as a select marker. The plasmid p64TRAIL was trans-ferred into the chloroplast genome of C. reinhardtii by the biolistic method. Three independently trans-formed lines were obtained by 100 mg/L spectino-mycin selection. PCR amplification, Southern blot analysis of the sTRAIL coding region DNA and culti-vation cells in the dark all showed that the exogenous DNA had been integrated into chloroplast genome of C. reinhardtii. Western blot analysis showed that human soluble TRAIL was expressed in C. reinhardtii chloroplast. The densitometric analysis of Western blot indicated that the expressed human sTRAIL protein in the chloroplasts of C. reinhardtii accounted for about 0.43%―0.67% of the total soluble proteins. | 3 | Atacamite and Nantokite in Kaerqueka Copper Deposit of Qimantag Area:Evidence for Cenozoic Climate Evolution of the Qaidam Basin显示文摘Using a mineralogy method to reflect climate evolution is a new approach to research Cenozoic environmental progression of the Qaidam Basin. In this paper, we present the chemical composition and crystallographic parameter of atacamite, nantokite, and tenorite in cryptocrystalline aggregates from the Kaerqueka copper deposit of Qimantag metallogenic belt by means of electron microprobe and in-situ X-ray diffraction analyses. Atacamite and nantokite occur in an intimate intergrowth as the secondary precipitation of chalcopyrite and bornite filling in the interstitial space of andradite in the Kaerqueka copper deposit, with an average composition of Cl: 12.38 wt.%, Cu: 63.76 wt.%, O: 21.46 wt.%. X-ray microdiffraction shows that the intergrowth contains nantokite with a cubic unit cell a=5.403(2) ? and atacamite with an orthorhombic unit cell a=6.030(3), b=6.883(2), c=9.114(1) ?. XRD quantitative calculation shows that the nanometric aggregate contains 36.07 wt.% tenorite, 18.41 wt.% atacamite, and 45.52 wt.% nantokite. The presence of nantokite and atacamite requires saline solutions for their formation and hyper-arid climate conditions for their preservation. Combined with the data of salt lakes and the pollen sequence of western China, we suggest that during the uplift of the Tibetan Plateau and the retreat of the Paratethys, saline water was forced to the surface through a basal fracture zone. In the hyper-arid climate of the Qaidam Basin, the recharge of groundwater by direct precipitation is negligible, and groundwater is derived from inflow from the salt lakes. Thus, atacamite is preserved. | 4 | Influences of the Three Gorges Dam in China on Precipitation over Surrounding Regions显示文摘Impacts of the Three Gorges Dam(TGD) in China on the regional pattern and annual amount of precipitation around the Three Gorges Reservoir(TGR) are examined by comparing observations before and after the operation of TGD(1984–2003 and 2004–13). Empirical orthogonal function(EOF) analysis of the annual precipitation anomalies clearly indicates that the land-use change associated with the construction of TGD has not significantly changed the precipitation pattern. To investigate the impacts of TGD on the rainfall amount, we compare the relative variations of atmospheric variables related to precipitation formation in three spatial bands: over TGR, near TGR, and far from TGR. It is found that the differences in annual rainfall over TGD between the two periods before and after the operation of TGD are small, suggesting a weak impact of TGD on the rainfall amount. | 15 | Cloning and identification of a novel binding protein of CD2 cytoplasmic domain显示文摘In order to understand why CD2 has a dual action of transduction of activation or apoptosis signals in T cells under different experimental conditions, we employed a yeast two-hybrid system to look for a binding protein of the cytoplasmic domain of CD2 which may be involved in this issue. A human T cell cDNA library was screened by a cDNA encoding the cytoplasmic domain of CD2 (Thr211-Gln336). The specificity of protein-protein interaction was verified by co-immunoprecipitation. The binding protein obtained, designated CD2cBP, was found to be homologous to v-fos transformation effector protein (Fte-1). | 18 | 5-Acetyl-6,7,8,4′-tetramethylnortangeretin induces apoptosis in multiple myeloma U266 cells显示文摘Multiple myeloma(MM)is one of the most common hematological malignancies and to date,it remains an incurable disease.In this study,we evaluated the inhibitory effect of 5-acetyloxy-6,7,8,4′-tetramethoxyflavone(5-ATMF),a compound from aged citrus peel extracts,on the MM U266 cell line.We found that the compound inhibited cell growth and induced cell apoptosis in multiple apoptotic assays.The apoptotic proteins caspase-3,caspase-9,and PARP(poly ADP-ribose polymerase)were cleaved when cells were treated with 5-ATMF.Along with the apoptosis process,the anti-apoptotic protein,Bcl-2(B-cell lymphoma-2),was significantly downregulated and the pro-apoptotic protein,Bax(Bcl-2 associated X protein),upregulated along with the release of cytochrome C(Cyt c)and the reduction of mitochondrial membrane potential(MMP).Notably,we found that the phosphorylation of Bad(Bcl-2/Bcl-XL-associated death promoter)was decreased but Bad remained unchanged in this process.On adding 5-ATMF to U266 cells,we observed that 5-ATMF repressed the phosphorylation of Akt(PKB Protein Kinase B PKB),thereby increasing the release of Cyt c and inhibiting the phosphorylation of Bad.These effects were enhanced by Ly294002,an inhibitor of Akt.These results suggested that 5-ATMF exerts a pro-apoptotic effect on U266 cells,possibly associated with the mitochondrial apoptotic pathway induced by the PI3K/Akt/Bad pathway. | Dexian Zhi Su Liu Li Lin Liwen Wang Jinhan Wang Jing Ma Suying Wang Hui Zhao Chi-Tang Ho Yafei Wang Qiang Liu | 2014 | Food Science and Human Wellness2014,3,3: | 0 |
| 19 | Evolution of the Mid-tropospheric Vortex during the Formation of Super Typhoon Megi(2010)显示文摘As a follow-up of a previously published article on the contribution of tropical waves,this study explores the evolution of the mid-tropospheric mesoscale cyclonic vortex(MV)during the formation of Typhoon Megi(2010)with a successful cloud-resolving simulation.It is found that the formation and intensification of the MV were related to the deep convection and subsequent stratiform precipitation,while the weakening of the MV was related to the shallow convection.Both the upward transport of vorticity related to the deep convection and the horizontal convergence associated with the stratiform precipitation contributed to the formation and intensification of the MV.Even though the latter was dominant,the former could not be ignored,especially in the early stage of the MV.The MV played dual roles in the formation of Megi.On the one hand,the formation and intensification of MV were primarily associated with the stratiform precipitation,which induced the low-level divergence inhibiting the spin-up of the near-surface cyclonic circulation.On the other hand,the coupled low-level cold core under the MV benefited the accumulation of the convective available potential energy(CAPE),which was favorable for the convective activity.A sensitivity experiment with the evaporative cooling turned off indicated that the development of the MV retarded the genesis process of Megi. | Li ZHUO Dexian FANG Zheng WU Juan FANG | 2020 | Advances in Atmospheric Sciences2020,37,7: | 0 |
