| 1 | Neutrophil extracellular traps mediate the crosstalk between glioma progression and the tumor microenvironment via the HMGB1/RAGE/IL-8 axis显示文摘Objective:Neutrophil extracellular traps(NETs)produced by tumor-infiltrating neutrophils(TINs)are associated with poor prognosis in patients with several types of cancer.However,the mechanisms underlying the involvement of NETs in glioma progression remain largely unknown.This study aimed to elucidate the roles of NETs in biological processes that drive the crosstalk between glioma progression and the tumor microenvironment.Methods:Neutrophil infiltration and NETs formation were investigated in glioma tissue through immunohistochemistry,and their relationships with clinicopathological features and outcomes were statistically evaluated.The effects of NETs on glioma cell progression were studied in a co-culture system.In vivo and in vitro experiments validated the reactive oxygen species activity and cytokine production of TINs,as well as the ERK signaling pathway activation and the metastasis of gliomas.Results:Neutrophil infiltration and NETs formation were induced in high-grade glioma compared with low-grade glioma.NETs induced by TINs were determined to be an oncogenic marker of high-grade gliomas and to be involved in cell proliferation and invasion.NETs overproduction promoted glioma cell proliferation,migration,and invasion.Furthermore,HMGB1 was found to bind to RAGE and activate the NF-κB signaling pathway in vitro.In addition,NETs stimulated the NF-κB signaling pathway,thus promoting IL-8 secretion in glioblastoma.Subsequently,IL-8 recruited neutrophils which in turn mediated NETs formation via the PI3 K/AKT/ROS axis in TINs.Conclusions:Our results suggest that NETs produced by TINs mediate the crosstalk between glioma progression and the tumor microenvironment by regulating the HMGB1/RAGE/IL-8 axis.Targeting NETs formation or IL-8 secretion may be an effective approach to inhibit glioma progression. | Caijun Zha Xiangqi Meng Lulu Li Shan Mi Da Qian Ziwei Li Pengfei Wu Shaoshan Hu Shihong Zhao Jinquan Cai Yanhong Liu | 2020 | Cancer Biology & Medicine2020,17,1: | 19 |
| 2 | Cytological mechanisms of interspecific incrossability and hybrid sterility between Oryza sativa L. and O. alta Swallen显示文摘Oryza sativa and O. alta belong to AA and CCDD genomes in Oryza, respectively. Interspecific repro-ductive isolation limits the transfer of favorable genes from O. alta into O. sativa. The cytological mechanisms of interspecific incrossability and hybrid sterility between O. sativa and O. alta were studied systematically in this paper. We indentified two cytological causes of interspecific incrossabil-ity. First, we observed embryo sac incompatibility that caused fertilization barriers of variable severity such as non-fertilization, fertilization stagnation and egg cell single-fertilization. Second, we observed hybrid inviability, the major cause for incrossability, apparent from hybrid embryo developmental stagnation and embryo abortion. Hybrid sterility included both embryo sac sterility and pollen sterility. The hybrid embryo sac was completely sterile and exhibited mainly embryo sac degeneration. Hybrid pollen was also sterile and mainly typical abortive. Hybrid sterility was mainly caused by severely ab-normal meioses of megasporocytes and pollen mother cells; it is the most important abnormality, being chromosome sterility. | 3 | A self-adaptive negative selection algorithm used for anomaly detection显示文摘A novel negative selection algorithm (NSA),which is referred to as ANSA,is presented. In many actual anomaly detection systems,the training data are just partially composed of the normal elements,and the self/nonself space often varies over time. Therefore,anom-aly detection system has to build the profile of the system based on a part of self elements and adjust itself to adapt those variables. However,previous NSAs need a large number of self elements to build the profile of the system,and lack adaptability. In order to over-come these limitations,the proposed approach uses a novel technique to adjust the self radius and evolve the nonself-covering detectors to build an appropriate profile of the system. To determine the performance of the approach,the experiments with the well-known data-set were performed. | 6 | Progress in the mechanism and targeted drug therapy for COPD显示文摘Chronic obstructive pulmonary disease(COPD)is emphysema and/or chronic bronchitis characterised by long-term breathing problems and poor airflow.The prevalence of COPD has increased over the last decade and the drugs most commonly used to treat it,such as glucocorticoids and bronchodilators,have significant therapeutic effects;however,they also cause side effects,including infection and immunosuppression.Here we reviewed the pathogenesis and progression of COPD and elaborated on the effects and mechanisms of newly developed molecular targeted COPD therapeutic drugs.Among these new drugs,we focussed on thioredoxin(Trx).Trx effectively prevents the progression of COPD by regulating redox status and protease/anti-protease balance,blocking the NF-κB and MAPK signalling pathways,suppressing the activation and migration of inflammatory cells and the production of cytokines,inhibiting the synthesis and the activation of adhesion factors and growth factors,and controlling the cAMP-PKA and PI3K/Akt signalling pathways.The mechanism by which Trx affects COPD is different from glucocorticoid-based mechanisms which regulate the inflammatory reaction in association with suppressing immune responses.In addition,Trx also improves the insensitivity of COPD to steroids by inhibiting the production and internalisation of macrophage migration inhibitory factor(MIF).Taken together,these findings suggest that Trx may be the ideal drug for treating COPD. | Cuixue Wang Jiedong Zhou Jinquan Wang Shujing Li Atsushi Fukunaga Junji Yodoi Hai Tian | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 8 |

| 9 | Identifying neutral allele Sb at pollen-sterility loci in cultivated rice with Oryza rufipogon origin显示文摘Pollen sterility is commonly found in the intra-specific hybrids of indica and japonica rice,which is one of the main constrains for the utilization of heterosis between indica and japonica.Six loci controlling the pollen sterility of F1 between indica and japonica have been identified from previous studies.Neutral alleles at each locus are potential to overcome the F1 pollen sterility associated with the locus.Therefore,exploitation and utilization of neutral alleles are of significant importance.The present research was based on fine mapping of the F1 pollen-sterility gene Sb and the abundant genetic diversity of Oryza rufipogon Griff.indigenous to Gaozhou,Guangdong Province(referred to as Gaozhou wild rice).Crosses were made using Taichung65(with the genotype of SbjSbj and referred to as E1) and its near-isogenic line of F1 pollen sterility gene Sb(with the genotype of SbiSbi,E2) as female parents,and 12 different accessions of Gaozhou wild rice as male parents.F1 pollen fertility was examined to identify the materials having the neutral alleles at the F1 pollen-sterility locus.Segregation of 4 molecular markers tightly linked with the Sb locus was analyzed in the F2 populations derived from the F1s carrying the neutral gene.The pollen fertility related to the 3 genotypes of the molecular markers was also checked by statistical test to determine whether it was consistent with the hypothesis.The results showed that the pollen fertility of two F1s from one accession of Gaozhou wild rice(GZW099) with E1 and E2 was(89.22±1.07)% and(85.65±1.05)%,respectively.Both of them were fertile and showed no significant difference by t-test.Segregation of the 3 genotypes of the 4 molecular markers followed the expected Mendelian ratio(1:2:1) in the F2 populations.There was no significant difference for the averaged pollen fertility of the plants related to the 3 genotypes,suggesting that no interaction exists between the alleles at the Sb locus in GZW099 and Taichung65 or E2.Evidentially,GZW099 carried the neutral gene(named SbnSbn) at the Sb locus,which provides valuable theoretical basis and resources for further studying and overcoming the sterility of indica-japonica hybrids. | SHI LeiGang LIU XiangDong LIU Bo ZHAO XingJuan WANG Lan LI JinQuan LU YongGen | 2009 | Chinese Science Bulletin2009,54,20: | 5 |
| 10 | Mining rice new germplasm containing S_5~n gene by functional molecular marker and sequencing显示文摘Inter-subspecific hybrids between indica and japonica varieties yield strong biological heterosis, but it is difficult to utilize the hybrids directly in commercial production due to sterility of F1. A special gene S5n may overcome the hybrids sterility, which is caused by the interaction between S5 loci. Recently, S5n had been cloned, and it was revealed containing a large DNA deletion sequence that made the gene nonfunctional, compared to S5i or S5j. We designed a pair of primers flanking the deletion sequence of S5n, and then applied to distinguish the varieties with S5n or non-S5n, convincing result suggested that the primers could be served as functional molecular marker to efficiently identify the new germplasm with S5n. Using the functional marker, we surveyed 197 varieties from China National Micro-core Rice Collection, and found ten of which represented S5n including following varieties: Haobuka, Sanbangqishiluo, Mubanggu, Xiaohonggu, Mowanggu’neiza, Laozaogu, Fanhaopi, Feie’nuo2, Baoxie-7B, Teqingxuanhui. Among them, two varieties Sanbangqishiluo and Laozaogu was previously reported to contain S5n gene. Further sequence analysis on the DNA sequence covering both sides of deletion in S5n of the 10 varieties confirmed that the detected sequences in above varieties was identical with those of varieties containing S5n, such as 02428 and Linglun. The temperature gradient and stress decreases significantly as the gas pressure increases. The convexity of the solid-liquid interface slightly increases when the gas pressure increases. | 13 | Safety,tolerability,and pharmacokinetics of BAT8001 in patients with HER2-positive breast cancer:An open-label,dose-escalation,phase I study显示文摘Background:The introductions of anti-human epidermal growth factor receptor-2(HER2)agents have significantly improved the treatment outcome of patients with HER2-positive breast cancer.BAT8001 is a novel antibodydrug conjugate targeting human epidermal growth factor receptor-2(HER2)-expressing cells composed of a trastuzumab biosimilar linked to the drug-linker Batansine.This dose-escalation,phase I study was designed to assess the safety,tolerability,pharmacokinetics,and preliminary anti-tumor activity of BAT8001 in patients with HER2-positive locally advanced or metastatic breast cancer.Methods:This trial was conducted in subjects with histologically confirmed HER2-positive breast cancer(having evaluable lesions and an Eastern Cooperative Oncology Group performance status of 0 or 1)using a 3+3 design of escalating BAT8001 doses.Patients received BAT8001 intravenously in a 21-day cycle,with dose escalation in 5 cohorts:1.2,2.4,3.6,4.8,and 6.0 mg/kg.The primary objective was to evaluate the safety and tolerability of BAT8001.Preliminary activity of BAT8001 was also assessed as a secondary objective.Results:Between March 2017 to May 2018,29 HER2-positive breast cancer patients were enrolled.The observed dose-limiting toxicities were grade 4 thrombocytopenia and grade 3 elevated transaminase.The maximum tolerated dose was determined to be 3.6 mg/kg.Grade 3 or greater adverse events(AEs)occurred in 14(48.3%)of 29 patients,including thrombocytopenia in 12(41.4%)patients,aspartate aminotransferase increased in 4(13.8%)patients,γ-glutamyl transferase increased in 2(6.9%)patients,alanine aminotransferase increased in 2(6.9%)patients,diarrhea in 2(6.9%)patients.Objective response was observed in 12(41.4%,95%confidence interval[CI]=23.5%-61.1%)and disease control(including patients achieving objective response and stable disease)was observed in 24(82.8%,95%CI=64.2%-94.2%)patients.Conclusions:BAT8001 demonstrated favorable safety profiles,with promising anti-tumor activity in patients with HER2-positive locally advanced or metastatic breast cancer.BAT8001 has the potential to provide a new therapeutic option in patients with metastatic HER2-positive breast cancer. | Ruoxi Hong Wen Xia Liye Wang Kaping Lee Qianyi Lu Kuikui Jiang Shengfeng Li Jinquan Yu Jin Wei Weijia Tang Danyang Zhou Xin An Jiajia Huang Cong Xue Xiwen Bi Yanxia Shi Zhongyu Yuan Fei Xu Shusen Wang | 2021 | Cancer Communications2021,41,2: | 2 |

| 15 | Molecular evolution of rice S_5~n and functional comparison among different sequences显示文摘The wide compatibility gene, S 5 n , can overcome embryo sac sterility between indica and japonica subspecies of rice. Therefore, it is very important to characterize the features of the S 5 n sequence to reveal the origin and evolution of S 5 n . In this paper, 26 cultivated rice haplotypes and 22 wild rice accessions harboring S 5 n were used to sequence S 5 n . The results showed that 15 genotypes among the 48 materials were fully consistent with control cultivar 02428 (CK). The other 33 accessions had different degrees of variation in the S 5 n sequence. Variations in the coding region mainly occurred in the second exon and eight materials showed a 10-bp deletion at 1710–1719 bp, including wild (O. nivara) and cultivated rice, such as IRW501 and Yuetai B. S 5 n sequences were not biased and evolved neutrally. The 48 materials could be divided into 4 categories using a phylogenetic tree of the amino acid sequences. Most of the wild rice clustered together, and the cultivated rice clustered into another group. Eight cultivated rice and O. nivara (wild rice) clustered in another group, which were found to lack 10 consecutive bases in exon 2. Eight rice varieties with high numbers of differences in their S 5 n coding regions were crossed with testers (typically indica and japonica) to produced test cross F 1 populations. The F 1 s were examined for their ability to overcome indica-japonica hybrid sterility. | 16 | Kinectin 1 promotes the growth of triple-negative breast cancer via directly co-activating NF-kappaB/p65 and enhancing its transcriptional activity显示文摘Triple-negative breast cancer(TNBC)is the most challenging subtype of breast cancer.Various endeavor has been made to explore the molecular biology basis of TNBC.Herein,we reported a novel function of factor Kinectin 1(KTN1)as a carcinogenic promoter in TNBC.KTN1 expression in TNBC was increased compared with adjacent tissues or luminal or Her2 subtypes of breast cancer,and TNBC patients with high KTN1 expression have poor prognosis.In functional studies,knockdown of KTN1 inhibited the proliferation and invasiveness of TNBC both in vitro and in vivo,while overexpression of KTN1 promoted cancer cell proliferation and invasiveness.RNA-seq analysis revealed that the interaction of cytokine-cytokine receptor,particularly CXCL8 gene,was upregulated by KTN1,which was supported by the further experiments.CXCL8 depletion inhibited the tumorigenesis and progression of TNBC.Additionally,rescue experiments validated that KTN1-mediated cell growth acceleration in TNBC was dependent on CXCL8 both in vitro and in vivo.Furthermore,it was found that KTN1 enhanced the phosphorylation of NF-κB/p65 protein at Ser536 site,and specifically bound to NF-κB/p65 protein in the nucleus and cytoplasm of cells.Moreover,the transcription of CXCL8 gene was directly upregulated by the complex of KTN1 and NF-κB/p65 protein.Taken together,our results elucidated a novel mechanism of KTN1 gene in TNBC tumorigenesis and progression.KTN1 may be a potential molecular target for the development of TNBC treatment. | Lin Gao Shanze Chen Malin Hong Wenbin Zhou Bilan Wang Junying Qiu Jinquan Xia Pan Zhao Li Fu Jigang Wang Yong Dai Ni Xie Qinhe Yang Hsien-Da Huang Xiang Gao Chang Zou | 2021 | Signal Transduction and Targeted Therapy2021,6,8: | 2 |
