| 1 | Elevated Neurosteroids in the Lateral Thalamus Relieve Neuropathic Pain in Rats with Spared Nerve Injury显示文摘Neurosteroids are synthesized in the nervous system from cholesterol or steroidal precursors imported from peripheral sources. These compounds are important allosteric modulators of c-aminobutyric acid A receptors(GABA_ARs), which play a vital role in pain modulation in the lateral thalamus, a main gate where somatosensory information enters the cerebral cortex. Using high-performance liquid chromatography/tandem mass spectrometry,we found increased levels of neurosteroids(pregnenolone,progesterone, deoxycorticosterone, allopregnanolone, and tetrahydrodeoxycorticosterone) in the chronic stage of neuropathic pain(28 days after spared nerve injury) in rats.The expression of the translocator protein TSPO, the upstream steroidogenesis rate-limiting enzyme, increased at the same time. In vivo stereotaxic microinjection of neurosteroids or the TSPO activator AC-5216 into the lateral thalamus(AP-3.0 mm, ML ±3.0 mm, DV 6.0 mm)alleviated the mechanical allodynia in neuropathic pain,while the TSPO inhibitor PK 11195 exacerbated it. The analgesic effects of AC-5216 and neurosteroids were significantly attenuated by the GABA_AR antagonist bicuculline. | 3 | Angiogenesis in hepatocellular carcinoma:mechanisms and anti-angiogenic therapies显示文摘Hepatocellular carcinoma(HCC)is the fourth leading cause of cancer-associated death worldwide.Angiogenesis,the process of formation of new blood vessels,is required for cancer cells to obtain nutrients and oxygen.HCC is a typical hypervascular solid tumor with an aberrant vascular network and angiogenesis that contribute to its growth,progression,invasion,and metastasis.Current anti-angiogenic therapies target mainly tyrosine kinases,vascular endothelial growth factor receptor(VEGFR),and plateletderived growth factor receptor(PDGFR),and are considered effective strategies for HCC,particularly advanced HCC.However,because the survival benefits conferred by these anti-angiogenic therapies are modest,new anti-angiogenic targets must be identified.Several recent studies have determined the underlying molecular mechanisms,including pro-angiogenic factors secreted by HCC cells,the tumor microenvironment,and cancer stem cells.In this review,we summarize the roles of pro-angiogenic factors;the involvement of endothelial cells,hepatic stellate cells,tumor-associated macrophages,and tumor-associated neutrophils present in the tumor microenvironment;and the regulatory influence of cancer stem cells on angiogenesis in HCC.Furthermore,we discuss some of the clinically approved anti-angiogenic therapies and potential novel therapeutic targets for angiogenesis in HCC.A better understanding of the mechanisms underlying angiogenesis may lead to the development of more optimized anti-angiogenic treatment modalities for HCC. | Changyu Yao Shilun Wu Jian Kong Yiwen Sun Yannan Bai Ruhang Zhu Zhuxin Li Wenbing Sun Lemin Zheng | 2023 | Cancer Biology & Medicine2023,20,1: | 3 |
| 4 | Biotinylated dextran amine anterograde tracing of the canine corticospinal tract显示文摘In this study, biotinylated dextran amine (BDA) was microinjected into the left cortical motor area of the canine brain. Fluorescence microscopy results showed that a large amount of BDA-labeled pyramidal cells were visible in the left cortical motor area after injection. In the left medulla oblongata, the BDA-labeled corticospinal tract was evenly distributed, with green fluorescence that had a clear boundary with the surrounding tissue. The BDA-positive corticospinal tract entered into the right lateral funiculus of the spinal cord and descended into the posterior part of the right lateral funiculus, close to the posterior horn, from cervical to sacral segments. There was a small amount of green fluorescence in the sacral segment. The distribution of BDA labeling in the canine central nervous system was consistent with the course of the corticospinal tract. Fluorescence labeling for BDA gradually diminished with time after injection. | 5 | Traffic-Aware VDC Embedding in Data Center: A Case Study of FatTree显示文摘Virtualization is a common technology for resource sharing in data center.To make efficient use of data center resources,the key challenge is to map customer demands(modeled as virtual data center,VDC) to the physical data center effectively.In this paper,we focus on this problem.Distinct with previous works,our study of VDC embedding problem is under the assumption that switch resource is the bottleneck of data center networks(DCNs).To this end,we not only propose relative cost to evaluate embedding strategy,decouple embedding problem into VM placement with marginal resource assignment and virtual link mapping with decided source-destination based on the property of fat-tree,but also design the traffic aware embedding algorithm(TAE) and first fit virtual link mapping(FFLM) to map virtual data center requests to a physical data center.Simulation results show that TAE+FFLM could increase acceptance rate and reduce network cost(about 49%in the case) at the same time.The traffic aware embedding algorithm reduces the load of core-link traffic and brings the optimization opportunity for data center network energy conservation. | LUO Shouxi YU Hongfang LI Lemin LIAO Dan SUN Gang | 2014 | China Communications2014,11,7: | 2 |
