| 3 | Efficacy of Zhonglun'a-decoction-containing serum on fibroblastlike synoviocyte apoptosis in rats with collagen-induced arthritis via inhibiting Janus kinase/signal transducer and activator of transcription signaling pathway显示文摘OBJECTIVE: To investigate the efficacy of Zhonglun'a-decoction-containing serum(ZHONGL-CS)on the in vitro apoptosis of fibroblast-like synoviocytes(FLS) from rats with collagen-induced arthritis(CIA) by investigating the Janus kinase/signal transducer and activator of transcription(JAK/STAT) signaling pathway.METHODS: A CIA rat model was established using bovine type Ⅱ collagen. FLS were isolated, cultured and identified. A cell counting kit-8 was used to detect cell activity. The half maximal inhibitory concentration(IC50) was calculated. Experimental subjects were divided into control, CIA,ZHONGL-CS, JAK2 inhibitor AG490, and ZHONGLCS with AG490 groups. The in vitro cell cycle and apoptosis rate were detected in FLS by flow cytometry. Bcl-2-associated X protein(Bax), B-cell lymphoma 2(Bcl-2), caspase-3, cyclin D1, phosphorylated JAK2, STAT1, and STAT3 protein expressions in FLS were examined by Western blotting. JAK2, STAT1 and STAT3 mRNA levels were examined by quantitative real-time polymerase chain reaction.RESULTS: Compared with the CIA group, FLS proliferation was inhibited, the FLS G0/G1 cell cycle was arrested, and the rate of FLS apoptosis was increased in the ZHONGL-CS group. In the ZHONGLCS group, the protein levels of Bcl-2 and cyclin D1 were reduced compared with the CIA group and the levels of Bax and caspase-3 in FLS were increased. In the ZHONGL-CS group, the expressions of JAK2, STAT1, and STAT3 mRNA and the levels of phosphorylated JAK2, STAT1, and STAT3 proteins were reduced.CONCLUSION: ZHONGL-CS may induce FLS apoptosis in CIA rats. | 4 | In situ self-assembled organoid for osteochondral tissue regeneration with dual functional units显示文摘The regeneration of hierarchical osteochondral units is challenging due to difficulties in inducing spatial,directional and controllable differentiation of mesenchymal stem cells(MSCs)into cartilage and bone compartments.Emerging organoid technology offers new opportunities for osteochondral regeneration.In this study,we developed gelatin-based microcryogels customized using hyaluronic acid(HA)and hydroxyapatite(HYP),respectively for inducing cartilage and bone regeneration(denoted as CH-Microcryogels and OS-Microcryogels)through in vivo self-assembly into osteochondral organoids.The customized microcryogels showed good cytocompatibility and induced chondrogenic and osteogenic differentiation of MSCs,while also demonstrating the ability to self-assemble into osteochondral organoids with no delamination in the biphasic cartilage-bone structure.Analysis by mRNA-seq showed that CH-Microcryogels promoted chondrogenic differentiation and inhibited inflammation,while OS-Microcryogels facilitated osteogenic differentiation and suppressed the immune response,by regulating specific signaling pathways.Finally,the in vivo engraftment of pre-differentiated customized microcryogels into canine osteochondral defects resulted in the spontaneous assembly of an osteochondral unit,inducing simultaneous regeneration of both articular cartilage and subchondral bone.In conclusion,this novel approach for generating self-assembling osteochondral organoids utilizing tailor-made microcryogels presents a highly promising avenue for advancing the field of tissue engineering. | Zhen Yang Bin Wang Wei Liu Xiaoke Li Kaini Liang Zejun Fan Jiao Jiao Li Yudi Niu Zihao He Hui Li Du Wang Jianjing Lin Yanan Du Jianhao Lin Dan Xing | 2023 | Bioactive Materials2023,,9: | 2 |

| 6 | Preparation of a Pt-Ni_(2)P/NF catalyst for highly efficient hydrogen evolution using a magnetic field to promote Ni-Pt galvanic replacement显示文摘The noble metal Pt is an ideal catalyst for promoting the hydrogen evolution reaction(HER)during the electrolysis of water.However,Pt is also expensive and suffers from low utilization rates.In this work,a Pt-Ni_(2)P/NF nanorod catalyst with a low Pt loading was synthesized under different magnetic fields,and it was found that the application of a magnetic field can increase the rate of the galvanic replacement reaction.When the magnetic field strength increases from 0 to 600 mT,the chemical reaction rate increases gradually,and the utilization rate of Pt increased by 2.3 times under 600 mT.The mechanism of the magnetic field-induced magnetohydrodynamic(MHD)effect on the galvanic replacement reaction was revealed.In a 1 M KOH solution and at a current density of 10 mA cm^(-2),the overpotential of Pt-Ni_(2)P/NF prepared by applying a 600 mT magnetic field was as low as 15 mV and the Tafel slope was 37 mV dec^(-1),compared with values of 82 mV and 70 mV dec^(-1) for a specimen prepared without a magnetic field.Additionally,at an overpotential of 90 mV,the mass-based Pt activity of the former material was 12 times greater while its turnover frequency was 19 times greater.This work provides theoretical and technical knowledge expected to assist in the controllable preparation of materials in magnetic fields and the efficient utilization of metallic resources. | Zhong Wang Shuang Yuan Tuo Zang Tengxiao Li Yudi Zhou Jiaqi Liu Tie Liu Kai Wang Qiang Wang | 2023 | Journal of Materials Science & Technology2023,,11: | 1 |

| 17 | Effect of a high strength chemical industry wastewater on microbial community dynamics and mesophilic methane generation显示文摘A high strength chemical industry wastewater was assessed for its impact on anaerobic microbial community dynamics and consequently mesophilic methane generation. Cumulative methane production was 251 mL/g total chemical oxygen demand removed at standard temperature and pressure at the end of 30 days experimental period with a highest recorded methane percentage of 80.6% of total biogas volume. Volatile fatty acids(VFAs) analysis revealed that acetic acid was the major intermediate VFAs produced with propionic acid accumulating over the experimental period. Quantitative analysis of microbial communities in the test and control groups with quantitative real time polymerase chain reaction highlighted that in the test group, Eubacteria(96.3%) was dominant in comparison with methanogens(3.7%). The latter were dominated by Methanomicrobiales and Methanobacteriales while Methanosarcinaceae in test groups increased over the experimental period, reaching a maximum on day 30. Denaturing gradient gel electrophoresis profile was performed, targeting the 16S rRNA gene of Eubacteria and Archaea, with the DNA samples extracted at 3 different time points from the test groups. A phylogenetic tree was constructed for the sequences using the neighborhood joining method. The analysis revealed that the presence of organisms resembling Syntrophomonadaceae could have contributed to increased production of acetic and propionic acid intermediates while decrease of organisms resembling Pelotomaculum sp. could have most likely contributed to accumulation of propionic acid. | 19 | Extracting the phase distribution of the electron wave packet ionized by an elliptically polarized laser pulse显示文摘We use an interferometic scheme to extract the phase distribution of the electron wave packet from above-threshold ionization in elliptically polarized laser fields.In this scheme,an electron wave packet released from a circularly polarized laser pulse acts as a reference wave and interferes with the electron wave packet ionized by a time-delayed counter-rotating elliptically polarized laser field.The generated vortex-shaped interference pattern in the photoelectron momentum distribution enables us to extract the phase distribution of the electron wave packet in the elliptically polarized laser pulse with high precision.By artificially screening the ionic potential at different ranges when solving the time-dependent Schördinger equation,we find that the angle-dependent phase distribution of the electron wave packet in the elliptically polarized laser field shows an obvious angular shift as compared to the strong-field approximation,whose value is the same as the attoclock shift.We also show that the amplitude of the angle-dependent phase distribution is sensitive to the ellipticity of the laser pulse,providing an alternative way to precisely calibrate the laser ellipticity in the attoclock measurement. | Ya-Nan Qin Min Li Yudi Feng Siqiang Luo Yueming Zhou Peixiang Lu | 2021 | Frontiers of physics2021,16,3: | 0 |
