1Emergence and prevalence of naturally occurring lower virulent African swine fever viruses in domestic pigs in China in 2020显示文摘African swine fever virus(ASFV)has been circulating in China for more than two years,and it is not clear whether the biological properties of the virus have changed.Here,we report on our surveillance of ASFVs in seven provinces of China,from June to December,2020.A total of 22 viruses were isolated and characterized as genotype II ASFVs,with mutations,deletions,insertions,or short-fragment replacement occurring in all isolates compared with Pig/HLJ/2018(HLJ/18),the earliest isolate in China.Eleven isolates had four different types of natural mutations or deletion in the EP402R gene and displayed a nonhemadsorbing(non-HAD)phenotype.Four isolates were tested for virulence in pigs;two were found to be as highly lethal as HLJ/18.However,two non-HAD isolates showed lower virulence but were highly transmissible;infection with 106 TCID50 dose was partially lethal and caused acute or sub-acute disease,whereas 103 TCID50 dose caused non-lethal,sub-acute or chronic disease,and persistent infection.The emergence of lower virulent natural mutants brings greater difficulty to the early diagnosis of ASF and creates new challenges for ASFV control.Encheng Sun Zhenjiang Zhang Zilong Wang Xijun He Xianfeng Zhang Lulu Wang Wenqing Wang Lianyu Huang Fei Xi Haoyue Huangfu Ghebremedhin Tsegay Hong Huo Jianhong Sun Zhijun Tian Wei Xia Xuewu Yu Fang Li Renqiang Liu Yuntao Guan Dongming Zhao Zhigao Bu 2021Science China(Life Sciences)2021,64,5:46
2A multiplexed circulating tumor DNA detection platform engineered from 3D-coded interlocked DNA rings显示文摘Circulating tumor DNA(ctDNA)is a critical biomarker not only important for the early detection of tumors but also invaluable for personalized treatments.Currently ctDNA detection relies on sequencing.Here,a platform termed three-dimensional-coded interlocked DNA rings(3D-coded ID rings)was created for multiplexed ctDNA identification.The ID rings provide a ctDNA recognition ring that is physically interlocked with a reporter ring.The specific binding of ctDNA to the recognition ring initiates target-responsive cutting via a restriction endonuclease;the cutting then triggers rolling circle amplification on the reporter ring.The signals are further integrated with internal 3D codes for multiplexed readouts.ctDNAs from non-invasive clinical specimens including plasma,feces,and urine were detected and validated at a sensitivity much higher than those obtained through sequencing.This 3D-coded ID ring platform can detect any multiple DNA fragments simultaneously without sequencing.We envision that our platform will facilitate the implementation of future personalized/precision medicine.Sha Yang Xinyu Zhan Xiaoqi Tang Shuang Zhao Lianyu Yu Mingxuan Gao Dan Luo Yunxia Wang Kai Chang Ming Chen 2022Bioactive Materials2022,7,4:1

4In-process adaptive milling for large-scale assembly interfaces of a vertical tail driven by real-time vibration data显示文摘Assembly interfaces,the joint surfaces between the vertical tail and rear fuselage of a large aircraft,are thin-wall components.Their machining quality are seriously restricted by the machining vibration.To address this problem,an in-process adaptive milling method is proposed for the large-scale assembly interface driven by real-time machining vibration data.Within this context,the milling operation is first divided into several process steps,and the machining vibration data in each process step is separated into some data segments.Second,based on the real-time machining vibration data in each data segment,a finite-element-unit-force approach and an optimized space–time domain method are adopted to estimate the time-varying in-operation frequency response functions of the assembly interface.These FRFs are in turn employed to calculate stability lobe diagrams.Thus,the three-dimensional stability lobe diagram considering material removal is acquired via interpolation of all stability lobe diagrams.Third,to restrain milling chatter and resonance,the cutting parameters for next process step,e.g.,spindle speed and axial cutting depth,are optimized by genetic algorithm.Finally,the proposed method is validated by a milling test of the assembly interface on a vertical tail,and the experimental results demonstrate that the proposed method can improve the machining quality and efficiency of the assembly interface,i.e.,the surface roughness reduced from 3.2μm to 1.6μm and the machining efficiency improved by 33%.Xiong ZHAO Lianyu ZHENG Lu YU 2022Chinese Journal of Aeronautics2022,35,5:0
