12018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 4Application of regional citrate anticoagulation in patients at high risk of bleeding during intermittent hemodialysis:a prospective multicenter randomized controlled trial显示文摘Objective:Safe and effective anticoagulation is essential for hemodialysis patients who are at high risk of bleeding.The purpose of this trial is to evaluate the effectiveness and safety of two-stage regional citrate anticoagulation(RCA)combined with sequential anticoagulation and standard calcium-containing dialysate in intermittent hemodialysis(IHD)treatment.Methods:Patients at high risk of bleeding who underwent IHD from September 2019 to May 2021 were prospectively enrolled in 13 blood purification centers of nephrology departments,and were randomly divided into RCA group and saline flushing group.In the RCA group,0.04 g/mL sodium citrate was infused from the start of the dialysis line during blood draining and at the venous expansion chamber.The sodium citrate was stopped after 3 h of dialysis,which was changed to sequential dialysis without anticoagulant.The hazard ratios for coagulation were according to baseline.Results:A total of 159 patients and 208 sessions were enrolled,including RCA group(80 patients,110 sessions)and saline flushing group(79 patients,98 sessions).The incidence of severe coagulation events of extracorporeal circulation in the RCA group was significantly lower than that in the saline flushing group(3.64%vs.20.41%,P<0.001).The survival time of the filter pipeline in the RCA group was significantly longer than that in the saline flushing group((238.34±9.33)min vs.(221.73±34.10)min,P<0.001).The urea clearance index(Kt/V)in the RCA group was similar to that in the saline flushing group with no statistically significant difference(1.12±0.34 vs.1.08±0.34,P=0.41).Conclusions:Compared with saline flushing,the two-stage RCA combined with a sequential anticoagulation strategy significantly reduced extracorporeal circulation clotting events and prolonged the dialysis time without serious adverse events.Xiaoyan TANG Dezheng CHEN Ling ZHANG Ping FU Yanxia CHEN Zhou XIAO Xiangcheng XIAO Weisheng PENG Li CHENG Yanmin ZHANG Hongbo LI Kehui LI Bizhen GOU Xin WU Qian YU Lijun JIAN Zaizhi ZHU Yu WEN Cheng LIU Hen XUE Hongyu ZHANG Xin HE Bin YAN Liping ZHONG Bin HUANG Mingying MAO 2022Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,11:4

7Lactobacillus plantarum KLDS1.0386 with antioxidant capacity ameliorates the lipopolysaccharide-induced acute liver injury in mice by NF-κB and Nrf2 pathway显示文摘The Lactobacillus is of great interest as a natural antioxidant to ameliorate liver injury for its antioxidant capacity.In a previous experiment,Lactobacillus plantarum KLDS 1.0386 was excellent in adhesion and anti-inflammation.This research assessed the antioxidant capacity of L.plantarum KLDS 1.0386 and how it ameliorates the LPS-induced acute liver injury.In vitro results showed that L.plantarum KLDS 1.0386 had potential antioxidant capacity with 27 genes,and it had better capacities in hydroxyl radicals scavenging and lipid antioxidant than the reference strain L.plantarum WCFS1 (P < 0.05).In vivo results,L.plantarum KLDS 1.0386 significantly reduced hepatic tissue injury,pro-inflammatory cytokines (TNF-α,IL-6),myeloperoxidase (MPO) activities and biochemical indicators of hepatotoxicity (ALT,AST,ALP) (P < 0.05),compared with the LPS-treated mice.Moreover,L.plantarum KLDS 1.0386 increased the relative abundance of the genera,including Lachnospiraceae_NK4A136_group,Muribaculaceae and Alistipes,which significantly increased the level of acetic acid and butyric acid (P < 0.05).The results also showed L.plantarum KLDS 1.0386 significantly adjusted the expression of HO-1,NQO1,CAT,NF-κB,TLR4 and IκB-α in the liver (P < 0.05).Overall,these results indicated that L.plantarum KLDS 1.0386 had great functionality in reducing LPS-induced acute liver injury.Jiahuan Dong Lijun Ping Qinggang Xie Deyu Liu Li Zhao Smith Etareri Evivie Zhongjiang Wang Bailiang Li Guicheng Huo 2022Food Bioscience2022,47,3:2
8Rapid screening detection of fluoroquinolone residues in milk based on turn-on fluorescence of terbium coordination polymer nanosheets显示文摘We report a facile, effective and rapid screening method for the determination of total fluoroquinolones(FQs) in milk using lanthanide coordination polymer nanomaterials as the sensing platform. The nonluminescent coordination polymer nanosheets(CPNSs) used in this work are composed of adenosine monophosphate(AMP) and terbium(Tb^(3+)) ions(AMP/Tb CPNSs). The as-prepared CPNSs were characterized by transmission electron microscopy, Fourier transform infrared spectroscopy and fluorescence techniques. Tb^(3+) acts as the center ion of the coordination polymer, with AMP assembled through phosphate and amine groups. However, the reaction between FQs and AMP/Tb CPNSs noticeably turns on the fluorescence due to the strong coordination interaction between FQs and Tb^(3+). Under optimized conditions, the fluorescence intensity of AMP/Tb at 545 nm shows a linear relationship with FQ concentrations between 0.04 μmol/L and 5.0 μmol/L. The relative standard deviation(RSD) of the intraday precision is below 1%, and detection limits are as low as 0.01 μmol/L. 12Evaluation of strategy for analyzing mouse liver plasma membrane proteome显示文摘Plasma membrane (PM) proteome is one of the major subproteomes present in the cell,and is very important in liver function. In the present work, C57 mouse liver PM was purified by density-gradient centrifugation. The purified PM was verified by electron microscope analysis and Western blotting. The results showed that the PM was enriched by more than 20-fold and the contamination of mitochondria was reduced by 2-fold compared with the homogenization fraction. Proteins were separated by 2DE and 1DE, trypsin-digested and submitted to ESI-Q-TOF and MALDI-TOF-TOF mass spectrometry or directly digested in solution and analyzed by LC-ESI ion trap mass spectrometry. In all, 547 non-redundant mouse liver PM proteins were identified, of which 34% contributed to plasma membrane or plasma membrane-related proteins. 13Effects of Sub-chronic Intoxication of 1,8-cineole on Blood Biochemical Indexes of Mice显示文摘[Objective] Effects of sub-chronic intoxication of 1,8-cineole on body weights,routine blood indexes and biochemical indexes of mice were investigated. [Method] One hundred and sixty mice with body weights of 15- 17 g were randomly divided into four groups( forty mice per group). Mice were injected to1,8- cineole with doses of 192. 45,64. 15 and 21. 38 mg / kg body weight( test groups) and the water solution of tween-80 with a volume fraction of 0. 5%( control group) respectively. Each mouse was administered orally at the dose of 0. 2 m L per 10 g body weight once a day consecutively for 90 d. The body weight,routine blood indexes and serum biochemical indexes of mice were determined on the 30 thd,60thd,90 thd and the 30 thd after stopping the administration of 1,8-cineole.[Result]The effects of 1,8-cineole on the body weight,routine blood indexes and serum biochemical indexes of mice with the doses of 64. 15 and 21. 38 mg/kg body weight had no statistically significant difference compared with the control group( P > 0. 05). 1,8-cineole with the dose of 192. 45 mg / kg body weight exhibited different influences on routine blood indexes and serum biochemical indexes of mice after the oral administration of 1,8-cineole for 60 d and 90 d,and statistically significant differences in many blood biochemical indexes were observed( P < 0. 05). However,the differences in routine blood indexes and serum biochemical indexes were not statistically significant between the test groups and the control group at the 30 thd after stopping the administration of 1,8-cineole( P > 0. 05).[Conclusion]1,8-cineole had sub-chronic oral toxicity to mice. The no observed adverse effect level( NOAEL) of 1,8-cineole was 64. 15 mg/kg body weight and the lowest observed adverse effect level( LOAEL) of 1,8-cineole was 192. 45 mg / kg body weight. 18Germ cell transplantation in infertility mouse显示文摘This work investigated the spermatogenesis in an infertility BALB/c-nu mouse model by reinfusing germline stem cells into seminiferous tubules. Donor germ cells were isolated from male FVB/NJ-GFP transgenic mice. Seminiferous tubule microinjection was applied to achieve intratubular germ cell transfer. The germ cells were injected into exposed testes of the infertility mice. We used green fluo-rescence and DNA analysis of donor cells from GFP transgenic mice as genetic marker. The natural mating and Southern blot methods were applied to analyze the effect of sperm cell transplantation and the sperm function after seminiferous tubule microinjection. The spermatogenesis was morphologi-cally observed from the seminiferous tubules in 41/60 (68.33%) of the injected recipient mice using allogeneic donor cells. In the colonized testes, matured spermatozoa were seen in the lumen of the seminiferous tubules. In this research, BALB/c-nu infertility mouse model, the recipient animal, was used to avoid immunological rejection of donor cells, and germ cell transplantation was applied to overcome infertility caused by busulfan treatment. These results demonstrate that this technique of germ cell transplantation is of great use.
