3The effect of collagen hydrogels on chondrocyte behaviors through restricting the contraction of cell/hydrogel constructs显示文摘Collagen is a promising material for tissue engineering,but the poor mechanical properties of collagen hydrogels,which tend to cause contraction under the action of cellular activity,make its application challengeable.In this study,the amino group of type I collagen(Col I)was modified with methacrylic anhydride(MA)and the photo-crosslinkable methacrylate anhydride modified type I collagen(CM)with three different degrees of substitution(DS)was prepared.The physical properties of CM and Col I hydrogels were tested,including micromorphology,mechanical properties and degradation properties.The results showed that the storage modulus and degradation rate of hydrogels could be adjusted by changing the DS of CM.In vitro,chondrocytes were seeded into these four groups of hydrogels and subjected to fluorescein diacetate/propidium iodide(FDA/PI)staining,cell counting kit-8(CCK-8)test,histological staining and cartilage-related gene expression analysis.In vivo,these hydrogels encapsulating chondrocytes were implanted subcutaneously into nude mice,then histological staining and sulfated glycosaminoglycan(sGAG)/DNA assays were performed.The results demonstrated that contraction of hydrogels affected behaviors of chondrocytes,and CM hydrogels with suitable DS could resist contraction of hydrogels and promote the secretion of cartilage-specific matrix in vitro and in vivo.Longpeng Dong Qingli Liu Yongli Gao Hengxing Jia Wenling Dai Likun Guo Hongsong Fan Yujiang Fan Xingdong Zhang 2021Regenerative Biomaterials2021,8,4:2
4Heat transfer characteristics in a narrow confined channel with discrete impingement cooling显示文摘Heat transfer characteristics in a narrow confined channel with discrete impingement cooling were investigated using thermal infrared camera. Detailed heat transfer distributions and comparisons on three surfaces with three impact diameters were experimentally studied in the range of Reynolds number of 3000 to 30000. The experimental results indicated that the strong impingement jet leaded to a high strength heat transfer zone in the ΔX=±2.5D;range of the impact center,which was 1.3–2.5 times of the average heat transfer value of the impingement wall. With the same coolant mass flow rate, small diameter case had lower heat transfer coefficient on both inner wall and outside wall, while the impingement wall was insensitive to the impact diameter. The surface averaged Nusselt number of inner wall was only 43%–57% of impingement wall, while the outside wall can reach up to 80%–90%. The larger the diameter, the higher heat transfer enhancement and the smaller the channel flow resistance was observed in term of Reynolds number. 5Locomotion Stability Analysis of Lower Extremity Augmentation Device显示文摘Stability is of great significance in the theoretical framework of biped locomotion.Real-time control and walking patterns planning are on the premise that the robot works in the stable condition.In this paper,we address the crucial issue of the locomotion stability based on the modified Poincare return map and the hybrid automata.Not akin to the traditional stability criteria,i.e.,the Zero Moment Point(ZMP)and the Center of Mass(CoM),the modified Poincare return map is more appropriate for both dynamic walking arid non-periodic walking.Moreover,a novel high-level reinforcement learning methodology,so-called active PI2 CMA-ES,is proposed in this paper to plan the exoskeleton locomotion.The proposed learning methodology demonstrates that the locomotion of the exoskeleton is asymptotically stable according to the modified Poincare return map criterion.Finally,the proposed learning methodology is tested by the Lower Extremity Augmentation Device(LEAD)and its effectiveness is verified by the experiments.Likun Wang Chaofeng Chen Wei Dong Zhijiang Du Yi Shen Guangyu Zhao 2019Journal of Bionic Engineering2019,16,1:1

7Design and mechanism insight on SiC quantum dots sensitized inverse opal TiO_(2) with superior photocatalytic activities under sunlight显示文摘The combination of SiC quantum dots sensitized inverse opal TiO_(2) photocatalyst is designed in this work and then applied in wastewater purification under simulated sunlight.From various spectroscopic techniques,it is found that electrons transfer directionally from SiC quantum dots to inverse opal TiO_(2),and the energy difference between their conduction/valence bands can reduce the recombination rate of photogenerated carriers and provide a pathway with low interfacial resistance for charge transfer inside the composite.As a result,a typical type-II mechanism is proved to dominate the photoinduced charge transfer process.Meanwhile,the composite achieves excellent photocatalytic performances(the highest apparent kinetic constant of 0.037 min^(-1)),which is 6.2 times(0.006 min^(-1))and 2.1 times(0.018 min^(-1))of the bare inverse opal TiO_(2) and commercial P25 photocatalysts.Therefore,the stability and non-toxicity of SiC quantum dots sensitized inverse opal TiO_(2) composite enables it with great potential in practical photocatalytic applications.Yingchao He Qiong Sun Likun Sun Zhixing Gan Liyan Yu Lifeng Dong 2023Frontiers of Chemical Science and Engineering2023,17,12:0
8Temporal variation of the underwater soundscape in Jiaotou Bay,an Indo-Pacific humpback dolphin(Sousa chinensis)habitat off Hainan Island,China显示文摘The underwater soundscape is an important ecological element affecting numerous aquatic animals,in particular dolphins,which must identify salient cues from ambient ocean noise.In this study,temporal variations in the soundscape of Jiaotou Bay were monitored from February 2016 to January 2017,where a population of Indo-Pacific humpback dolphins(Sousa chinensis)has recently been a regular sighting.An autonomous acoustic recorder was deployed in shallow waters,and 1/3-octave band sound pressure levels(SPLs)were calculated with central frequencies ranging from 25 Hz to 40 kHz,then were grouped into 3 subdivided bands via cluster analysis.SPLs at each major band showed significant differences on a diel,fishing-related period,seasonal,and tidal phase scale.Anthropogenic noise generated by passing ships and underwater explosions were recorded in the study area.The fish and dolphin acoustic activities both exhibited diel and seasonal variations,but no tidal cycle patterns.A negative significant relationship between anthropogenic sound detection rates and dolphin detection rates were observed,and fish detection rates showed no effect on dolphin detection rates,indicating anthropogenic activity avoidance and no forced foraging in dolphins in the study area.The results provide fundamental insight into the acoustic dynamics of an important Indo-Pacific humpback dolphin habitat within a coastal area affected by a rapid increase in human activity,and demonstrate the need to protect animal habitat from anthropogenic noises.Lijun DONG Jianchen DONG Francesco CARUSO Likun ZHAO Songhai LI 2021Integrative Zoology2021,16,4:0

10One-step synthesis of site-specific antibody-drug conjugates by reprograming IgG glycoengineering with LacNAc-based substrates显示文摘Glycosite-specific antibody-drug conjugatess(gsADCs), harnessing Asn297 N-glycan of IgG Fc as the conjugation site for drug payloads, usually require multi-step glycoengineering with two or more enzymes, which limits the substrate diversification and complicates the preparation process.Herein, we report a series of novel disaccharide-based substrates, which reprogram the IgG glycoengineering to one-step synthesis of gsADCs, catalyzed by an endo-N-acetylglucosaminidase(ENGase) of Endo-S2. IgG glycoengineering via ENGases usually has two steps: deglycosylation by wild-type(WT) ENGases and transglycosylation by mutated ENGases. But in the current method, we have found that disaccharide LacNAc oxazoline can be efficiently assembled onto IgG by WT Endo-S2 without hydrolysis of the product, which enables the one-step glycoengineering directly from native antibodies.Further studies on substrate specificity revealed that this approach has excellent tolerance on various modification of 6-Gal motif of LacNAc. Within 1 h, one-step synthesis of gsADC was achieved using the LacNAc-toxin substrates including structures free of bioorthogonal groups. These gsADCs demonstrated good homogeneity, buffer stability, in vitro and in vivo anti-tumor activity.
