5Construction, detection and microarray analysis on the Shigella flexneri 2a sitC mutant显示文摘In order to overcome the defects of difficult gene operations in low-copy suicide plasmid pCVD442, Gateway technology was applied in the construction process of recombinant plasmid for gene knockout in this study. With this improved knockout system, we inactivated sitC gene, which is associated with iron transport in Shigella flexneri 2a strain 301, to yield the mutant, MTS. The functional detection of the mutant was performed at the level of culture medium, cell and animal experiment, respectively. The gene expression profiles were compared with DNA microarray between the mutant and the wild type under iron-restricted conditions. The results showed that MTS grew obviously less well than the wild-type strains in L broth containing 150 μmol/L iron chelator DIP(2,2′-dipyridyl). Addition of iron or manganese to the cultures stimulated the growth of MTS to wild-type levels in rich culture medium. In either the experiment on the ability of intracellular multiplication and cell-to-cell spread in HeLa and U937 cell lines, or the experiment on keratoconjunctivitis in guinea pigs, MTS showed no obvious changes in virulence compared with the parental strain Sf301. When 65 μmol/L DIP was added to the cultured HeLa cells, the ability of intracellular multiplication of MTS reduced about 51.6% as compared with that of Sf301. The analysis of expression profiles under iron-limited condition showed that MTS was more sensitive for the change of iron deficiency than Sf301. There are 106 more up-regulated genes in MTS than in wild-type strains, which are involved in membrane transportation, amino acid metabolism and uncategorized function genes, while down-regulated genes are mainly in- volved in energy and carbohydrate metabolism. Under low iron conditions, the expression levels of known iron-transport associated genes generally increased. Additionally, the number of these genes and their increase amplitude in MTS are more than those in Sf301. 9Distinct lung microbial community states in patients with pulmonary tuberculosis显示文摘An improved understanding of the lung microbiome may lead to better strategies to diagnose, treat, and prevent pulmonary tuberculosis (PTB). However, the characteristics of the lung microbiomes of patients with TB remain largely undefined. In this study, 163 bronchoalveolar lavage (BAL) samples were collected from 163 sputum-negative suspected PTB patients. Furthermore, 12 paired BAL samples were obtained from 12 Mycobacterium tuberculosis-positive (MTB+) patients before and after negative conversion following a two-month anti-TB treatment. The V3–V4 region of the 16S ribosomal RNA (rRNA) gene was used to characterize the microbial composition of the lungs. The results showed that the prevalence of MTB in the BAL samples was 42.9% (70/163) among the sputum-negative patients. The α-diversity of lung microbiota was significantly less diverse in MTB+ patients compared with Mycobacterium tuberculosis-negative (MTB–) patients. There was a significant difference in β-diversity between MTB+ and MTB– patients. MTB+ patients were enriched with Anoxybacillus, while MTB– patients were enriched with Prevotella, Alloprevotella, Veillonella, and Gemella. There was no significant difference between the Anoxybacillus detection rates of MTB+ and MTB– patients. The paired comparison between the BAL samples from MTB+ patients and their negative conversion showed that BAL negative-conversion microbiota had a higher α-diversity. In conclusion, distinct features of airway microbiota could be identified between samples from patients with and without MTB. 12Thiol-ene conjugation of a VEGF peptide to electrospun scaffolds for potential applications in angiogenesis显示文摘Vascular endothelial growth factor(VEGF)plays a vital role in promoting attachment and proliferation of endothelial cells,and induces angiogenesis.In recent years,much research has been conducted on the functionalization of tissue engineering scaffolds with VEGF or a VEGF-mimetic peptide to promote angiogenesis.However,most chemical reactions are nonspecific and require organic solvents,which can compromise control over functionalization and alter peptide/protein activity.An attractive alternative is the fabrication of functionalizable electrospun fibers,which can overcome these hurdles.In this study,we used thiol-ene chemistry for the conjugation of a VEGF-mimetic peptide to the surface of poly(ε-caprolactone)(PCL)fibrous scaffolds with varying amounts of a functional PCL-diacrylate(PCL-DA)polymer.30%PCL-DA was selected due to homogeneous fiber morphology.A VEGF-mimetic peptide was then immobilized on PCL-DA fibrous scaffolds by a light-initiated thiol-ene reaction.7-Mercapto-4-methylcoumarin,RGD-FITC peptide and VEGF-TAMRA mimetic peptide were used to validate the thiol-ene reaction on the fibrous scaffolds.Tensile strength and elastic modulus of the 30%PCL-DA fibrous scaffolds were significantly increased after the reaction.Conjugation of the 30%PCL-DA fibrous scaffolds with the VEGF peptide increased the surface water wettability of the scaffolds.Patterned structures could be obtained after using a photomask on the fibrous film.Moreover,in vitro studies indicated that scaffolds functionalized with the VEGF-mimetic peptide were able to induce phosphorylation of the VEGF receptor and enhanced HUVECs survival,proliferation and adhesion.A chick chorioallantoic membrane(CAM)assay further indicated that the VEGF peptide functionalized scaffolds were able to promote angiogenesis in vivo.These results show that scaffold functionalization can be controlled via a simple polymer mixing approach,and that the functionalized VEGF peptide-scaffolds have potential for vascular tissue regeneration.Tianyu Yao Honglin Chen Rong Wang Rebeca Rivero Fengyu Wang Lilian Kessels Stijn M.Agten Tilman M.Hackeng Tim G.A.M.Wolfs Daidi Fan Matthew B.Baker Lorenzo Moroni 2023Bioactive Materials2023,,2:1

14Reprogramming immunosuppressive myeloid cells by activated T cells promotes the response to anti-PD-1 therapy in colorectal cancer显示文摘Overcoming local immunosuppression is critical for immunotherapy to produce robust anti-tumor responses.Myeloid-derived suppressor cells(MDSCS)are key regulators of immunosuppressive networks and promote tumor progression.However,it remains unclear whether and how tumor-infltrating MDSCS are shaped in response to anti-PD-1 treatment and what their impact on therapeutic efficacy is in colorectal cancer(CRC).In this study,the levels of infltrating MDSCS were significantly higher in the non-responding organoids and were selectively reduced in the responding group,with MDSCS showing increased apoptosis and attenuated functional activity after anti-PD-1 treatment.A negative correlation between T-cell activation and MDSC function was also observed in fresh human CRC tissues.Mechanistic studies revealed that autocrine IFN-α/B upregulated TRAIL expression on activated T cells to elicit MDSC apoptosis via the TRAIL-DR5 interaction and acted synergistically with TNF-α to inhibit MDSC function of suppressing the T-cell response through the JNK NMDAR-ARG-1 pathway.Moreover,blockade of IFNa/βand TNF-α abolished the therapeutic efficacy of anti-PD-1 treatment by preserving the frequency and suppressive activity of infltrating MDSCS in a CRC mouse model.This result suggested that reprogramming MDSCS by IFN-α/βand TNF-α from activated T cells was necessary for succesful anti-PD-1 treatment and might serve as a novel strategy to improve the response and efficacy of anticancer therapy.Jing Chen Hong-Wei Sun Yan-Yan Yang Hai-Tian Chen Xing Juan Yu Wen Chao Wu Yi-Tuo Xu LiLian Jin Xiao-Jun Wu Jing Xu Limin Zheng 2021Signal Transduction and Targeted Therapy2021,6,2:0
15A chromosome-level genome assembly for Erianthus fulvus provides insights into its biofuel potential and facilitates breeding for improvement of sugarcane显示文摘Erianthus produces substantial biomass,exhibits a good Brix value,and shows wide environmental adaptability,making it a potential biofuel plant.In contrast to closely related sorghum and sugarcane,Erianthus can grow in degraded soils,thus releasing pressure on agricultural lands used for biofuel production.However,the lack of genomic resources for Erianthus hinders its genetic improvement,thus limiting its potential for biofuel production.In the present study,we generated a chromosome-scale reference genome for Erianthus fulvus Nees.The genome size estimated by flow cytometry was 937 Mb,and the assembled genome size was 902 Mb,covering 96.26%of the estimated genome size.A total of 35065 proteincoding genes were predicted,and 67.89%of the genome was found to be repetitive.A recent wholegenome duplication occurred approximately 74.10 million years ago in the E.fulvus genome.Phylogenetic analysis showed that E.fulvus is evolutionarily closer to S.spontaneum and diverged after S.bicolor.Three of the 10 chromosomes of E.fulvus formed through rearrangements of ancestral chromosomes.Phylogenetic reconstruction of the Saccharum complex revealed a polyphyletic origin of the complex and a sister relationship of E.fulvus with Saccharum sp.,excluding S.arundinaceum.On the basis of the four amino acid residues that provide substrate specificity,the E.fulvus SWEET proteins were classified as monoand disaccharide sugar transporters.Ortho-QTL genes identified for 10 biofuel-related traits may aid in the rapid screening of E.fulvus populations to enhance breeding programs for improved biofuel production.The results of this study provide valuable insights for breeding programs aimed at improving biofuel production in E.fulvus and enhancing sugarcane introgression programs.Ling Kui Aasim Majeed Xianhong Wang Zijiang Yang Jian Chen Lilian He Yining Di Xuzhen Li Zhenfeng Qian Yinming Jiao Guoyun Wang Lufeng Liu Rong Xu Shujie Gu Qinghui Yang Shuying Chen Hongbo Lou Yu Meng Linyan Xie Fu Xu Qingqing Shen Amit Singh Karl Gruber Yunbing Pan Tingting Hao Yang Dong Fusheng Li 2023Plant Communications2023,4,4:0
16Design and Algorithm Development of An Expert System for Continuous Health Monitoring of Sewer and Storm Water Pipes显示文摘Management and monitoring the health condition of underground utilities are essential in the process of sustainable urbanization development,as our daily necessities such as water,gas and electricity supplies are delivered through very complicated networks of pipes and cables underground.Regarding the management of water pipes,if their health condition is not regularly and properly monitored for preventive measures to be carried out in advance,the leakage of water pipes may cause road and slope collapse and hence lead to traffic jam and inconvenience to public,and more seriously fatality and property loss.However,there are no unified rules in Hong Kong on the interpretation of the health condition of utilities from pipe condition survey data.As the judgment is largely based on experience,it is unavoidable that subjective interpretation,particularly those cases with fuzzy results on for example site inspection,visual assessment based on photos and video recording may lead to different opinions.In this paper,the development of an expert system to enable collective decision making on the fuzzy results,and together with other assessment results to rank the overall health condition of drainage and sewage pipes based on the continuous tracking on the Utility Health Index(UHI)is introduced.The system is being tested in the Utility Info using real project data for further improvement.The long term tracking of the UHI and the site inspection results used for calculating the UHI can provide a fuller picture on the health condition of different sections of the pipe network.With the continuous monitoring of the pipe condition by the expert system,the potential problems of pipes can be continuously analyzed and identified,for the maintenance logistics and precautions to be planned and implemented more effectively.Esmond MOK King WONG Geoffrey SHEA Wu CHEN Lilian PUN 2014办公自动化2014,19,S1:0
