| 2 | Secretory/releasing proteome-based identification of plasma biomarkers in HBV-associated hepatocellular carcinoma显示文摘For successful therapy, hepatocellular carcinoma (HCC) must be detected at an early stage. Herein, we used a proteomic approach to analyze the secretory/releasing proteome of HCC tissues to identify plasma biomarkers. Serum-free conditioned media (CM) were collected from primary cultures of cancerous tissues and surrounding noncancerous tissues. Proteomic analysis of the CM proteins permitted the identification of 1365 proteins. The enriched molecular functions and biological processes of the CM proteins, such as hydrolase activity and catabolic processes, were consistent with the liver being the most important metabolic organ. Moreover, 19% of the proteins were characterized as extracellular or membrane-bound. For validation, secretory proteins involved in transforming growth factor-β signaling pathways were validated in plasma samples. Alphafetoprotein (AFP), metalloproteinase (MMP)1, osteopontin (OPN), and pregnancy-specific beta-1-glycoprotein (PSG)9 were significantly increased in HCC patients. The overall performance of MMP1 and OPN in the diagnosis of HCC remained greater than that of AFP. In addition, this study represents the first report of MMP1 as a biomarker with a higher sensitivity and specificity than AFP. Thus, this study provides a valuable resource of the HCC secretome with the potential to investigate serological biomarkers. | 6 | Effect of W addition on phase transformation and microstructure of powder metallurgic Ti-22Al-25Nb alloys during quenching and furnace cooling显示文摘Powder metallurgic Ti2 AlNb alloys with W addition are sintered at 900, 1000, 1070 °C,and 1150 °C(i.e., in the O + B2, a_2+ B2 + O, a_2+ B2, and single B2 phase regions, respectively)for 12 h, followed by water quenching and furnace cooling. Comparisons of phase and microstructure between quenched and furnace-cooled W-modified alloys are carried out to illustrate the phase transformation and microstructure evolution during the cooling process. Furthermore, a comparison is also made between W-modified and W-free alloys, to reveal the function of the W alloying.W addition accelerates the solutions of a_2 and O phases during the high-temperature holding, and a Widmannsta¨tten B2 + O structure, which contributes to the properties, is induced by furnace cooling from all the phase regions. The Widmannsta¨tten structure includes a B2 matrix, primary O, and secondary O precipitates. However, W alloying refines the Widmannsta¨tten structure only when the alloys are solution-treated and then cooled from the single B2 phase. | 11 | Prediction of the coupled heat radiation and conduction parameters and boundary condition using the unscented Kalman filter显示文摘The boundary emissivity, thermal conductivity, and boundary time-varying heat flux in the participating media are retrieved in this work. In the forward model, the coupled radiation-conduction heat transfer in the participating medium is resolved by the finite volume method combined with discrete ordinates method. The inverse model utilizes the temperature signals at the appropriate position of the medium as the known information and uses the unscented Kalman filter(UKF) as an optimization tool to reconstruct the boundary emissivity, thermal conductivity, and boundary time-varying heat flux. It is found that when the emissivity, thermal conductivity, and boundary time-varying heat flux are reconstructed simultaneously, only the temperature information of two locations is required. The influence of the measurement noise, sampling interval, absorption coefficient,process noise covariance, measurement noise covariance on the accuracy and stability of the retrieval results is investigated in detail. | 13 | Comparison of gut microbiota in autism spectrum disorders and neurotypical boys in China:A case-control study显示文摘Background:Autism spectrum disorders(ASDs)are a set of complex neurobiological disorders.Growing evidence has shown that the microbiota that resides in the gut can modulate brain development via the gut–brain axis.However,direct clinical evidence of the role of the microbiota–gut–brain axis in ASD is relatively limited.Methods:A case-control study of 71 boys with ASD and 18 neurotypical controls was conducted at China-Japan Friendship Hospital.Demographic information and fecal samples were collected,and the gut microbiome was evaluated and compared by 16S ribosomal RNA gene sequencing and metagenomic sequencing.Results:A higher abundance of operational taxonomic units(OTUs)based on fecal bacterial profiling was observed in the ASD group.Significantly different microbiome profiles were observed between the two groups.At the genus level,we observed a decrease in the relative abundance of Escherichia,Shigella,Veillonella,Akkermansia,Provindencia,Dialister,Bifidobacterium,Streptococcus,Ruminococcaceae UCG_002,Megasphaera,Eubacterium_coprostanol,Citrobacter,Ruminiclostridium_5,and Ruminiclostridium_6 in the ASD cohort,while Eisenbergiella,Klebsiella,Faecalibacterium,and Blautia were significantly increased.Ten bacterial strains were selected for clinical discrimination between those with ASD and the neurotypical controls.The highest AUC value of the model was 0.947.Conclusion:Significant differences were observed in the composition of the gut microbiome between boys with ASD and neurotypical controls.These findings contribute to the knowledge of the alteration of the gut microbiome in ASD patients,which opens the possibility for early identification of this disease. | Fang Ye Xinying Gao Zhiyi Wang Shuman Cao Guangcai Liang Danni He Zhitang Lv Liming Wang Pengfei Xu Qi Zhang | 2021 | Synthetic and Systems Biotechnology2021,6,2: | 2 |
| 14 | Dynamic Simulation of Pelagic Longline Retrieval显示文摘To improve fishing gear efficiency, it is important to understand the interactions among sea current, fishing vessel, line hauler, and catches during pelagic longline gear retrieval. In this study, fishing gear configuration parameters, operational parameters, and 3 D ocean current data were collected from Indian Ocean. Dynamic models of pelagic longline gear retrieval were built using the lumped mass method and solved using the Euler-Trapezoidal method. From the results, the pulling force of line hauler exerted on the gear was 2800–3600 N. There were no significant differences(P > 0.05) between the time of the hook retrieval measured at sea and that obtained from the simulation. The absolute values of the movement velocity at representative nodes along the X, Y, and Z axes were 0.01–25.5 m s^(-1). These results suggest that the dynamic model of longline fishing gear retrieval can be used to analyze the interactions among sea current, fishing vessel, line hauler, longline gear, and catches, and to acquire the basic data for optimizing the design of the line hauler.
