| 4 | Bio-inspired polygonal skeleton structure for vibration isolation:Design,modelling,and experiment显示文摘Inspired by the safe landing of a cat falling from a high altitude,a bio-inspired polygonal skeleton(BIPS) structure is proposed,and its nonlinear characteristics are systematically studied to explore its potential application in the suppression of vibration. The polygon is formed by the skeleton structure of the cat’s entire body and the ground. The BIPS system consists of two symmetrical bionic legs with three robs(as skeleton) and four horizontal springs(as muscle). Two bionic legs are connected through the bearing platform(as spine),which could adjust the distance between the two bionic legs. A theoretical model is developed to characterize its stiffness nonlinearity through geometrical and mechanical analysis. Parameter analysis reveals that the BIPS structure has diverse stiffness,including nonlinear positive stiffness and negative stiffness. By imitating adjustment of leg posture and telescopic function of the spine(control the distance between legs),these flexible stiffness properties can be adjusted by structure parameters. In addition,the load capacity and working range can also be designed by the length of the bars,the initial angle,the mounting position,and the spring stiffness. The experimental setup is established,and the vibration isolation performance under various excitation is tested. The experimental results verify the accuracy of the dynamic model and also show that the proposed BIPS structure can suppress the vibration effectively under a variety of excitations. | 8 | Evaluation methods of heavy metal pollution in soils based on enzyme activities:A review显示文摘Soil enzyme activities have been suggested as suitable indicators for the evaluation of metal contamination because they are susceptible to microbial changes caused by heavy metal stress and are strictly related to soil nutrient cycles.However,there is a growing lack of recognition and summary of the historic advancements that use soil enzymology as the proposal of evaluation methods.Here,we review the most common methods of heavy metal pollution evaluation based on enzyme activities,which include single enzyme index,combined enzyme index,enzyme-based functional diversity index,microbiological stress index,and ecoenzymatic stoichiometry models.This review critically examines the advantages and disadvantages of these methods based on their execution complexity,performance,and ecological implications and gets a glimpse of avenues to come to improved future evaluation systems.Indices based on a single enzyme are variable and have no consistent response to soil heavy metals,and the following three composite indices are characterized by the loss of many critical microbial processes,which thus not conducive to reflect the effects of heavy metals on soil ecosystems.Considering the dexterity of ecoenzymatic stoichiometry methods in reflecting changes in soil functions under heavy metal stress,we propose that microbial metabolic limitations quantified by ecoenzymatic stoichiometry models could be promising indicators for enhancing the reality and acceptance of results and further improving the potential for actual utility in environmental decision-making. | Yongxing Cui Xia Wang Xiangxiang Wang Xingchang Zhang Linchuan Fang | 2021 | Soil Ecology Letters2021,3,3: | 3 |
| 9 | Preparation of Au nanoparticles modified TiO_2 nanotube array sensor and its application as chemical oxygen demand sensor显示文摘Au nanoparticles(AuNPs) were electrodeposited at the highly ordered anatase TiO_2 nanotube array(TiO_2 NA) electrode under sonicating, and the AuNP-TiO_2 NA sensor was characterized by scanning electron microscope(SEM), X-ray photoelectron spectroscopy(XPS), X-ray diffraction(XRD). The photoelectrochemical experiments indicate the AuNP-TiO_2 NA sensor has lower photoelectro-resistance,higher photoelectrocatalytical activity and stability than that of pure TiO_2 NA sensor under the same conditions. | 10 | A comparative proteomics approach to detect unintended effects in transgenic Arabidopsis显示文摘Recently, as part of biosafety assessments, unintended effects have been given much attention. In this study, we applied a proteomics approach to elucidate the unintended effects of random T-DNA insertion in transgenic plants. Separated proteins extracted from 12 transgenic Arabidopsis thaliana with different T-DNA insertion sites and from wild-type (ecotype Col-o) were analyzed. In the transgenic plants, 102 significantly altered protein spots were detected, in which 59 were up-regulated and 43 down-regulated. MALDI-TOF MS analysis showed that most of these expression level-altered proteins were involved in energy transfer, oxidative respiration and photosynthesis. However, none of these proteins was a toxic protein or allergen. Using plants with or without cold treatment, a natural environmental stress, as controls, we found that the number of the altered proteins was even less in those transgenic plants than those triggered by the cold treatment, suggesting that the transgenic events had a weaker impact on the plants than the environmental stresses. Interestingly, the phosphinothricin acetyl transferase (PAT), the BAR-encoded protein, was detected in nine out of twelve different T-DNA insertion lines at five different insertion sites. These data suggest that the most significant impact of transgenic events on the host plants is from the transgene itself, i.e., from the predictable intended effects, rather than unintended effects. | 13 | A Preliminary Urinary Metabolomics Study of Sprague-Dawley Rats after Short‑term Ketamine Administration by Proton Nuclear Magnetic Resonance Spectroscopy显示文摘Drug abuse has become a global problem.The mass spectrometry‑based metabolic consequences of ketamine administration in anesthesia and therapy have been well studied,but to the best of our knowledge,metabolomic studies of ketamine abuse based on nuclear magnetic resonance(NMR)spectroscopy are still lacking.In this study,twenty Sprague–Dawley rats were randomly assigned into two groups:a control group(n=10)and a ketamine group(n=10).The animals in the ketamine group received intraperitoneal injections of ketamine twice daily at 12‑h intervals at progressively increasing doses over a period of 9 days,while the control group received an equal volume of saline.The urine samples were collected for 24 h at days 0,1,3,5,7,and 9 for the metabolomics study.The metabolic changes in urine after short‑term ketamine administration were analyzed by proton NMR coupled with multivariate statistical analysis.The results indicated that short‑term ketamine exposure led to significant alterations of the metabolites in the urine of the rats.Specifically,1,3,7‑trimethyluric acid,1,3‑dimethyluric acid,acetoacetic acid,acetylglycine,creatine,sarcosine,dimethylglycine,glycine,and theobromine were significantly increased in the urine.Significant changes were also found in metabolites related to antioxidant and energy metabolism,including acetoacetic acid,succinate,1,3,7‑trimethyluric acid,1,3‑dimethyluric acid,creatine,and taurine.Our findings indicated that short‑term ketamine administration leads to disorder of energy metabolism and oxidative stress.In addition,the modified metabolites identified could serve as the new biological markers and potential biological indices reflecting the underlying mechanism of ketamine abuse. | Xiang Lu Qunxing Tang Yi Ye Run Guo Fan Chen Xinhua Dai Youyi Yan Linchuan Liao | 2016 | Journal of Forensic Science and Medicine2016,2,2: | 1 |
| 14 | Genome-wide profiling of histone H3 lysine 27 trimethylation and its modification in response to chilling stress in grapevine leaves显示文摘Histone H3 lysine 27 trimethylation(H3K27me3) is a histone modification associated with transcriptional repression. However, insights into the genome-wide pattern of H3K27me3 in grapevines are limited. Here, anti-H3K27 chromatin immunoprecipitation(ChIP), high-throughput sequencing, and transcriptome analysis were performed using leaves of Vitis amurensis. The leaves were treated at 4°C for 2 h and 24 h and used to investigate changes in H3K27me3 under chilling treatment. The results show that H3K27me3 is well-distributed both in gene regions(-50%) and in the intergenic region(-50%) in the grapevine genome(Vitis vinifera ‘Pinot Noir PN40024'). H3K27me3 was found to be localized in8 368 annotated gene regions in all detected samples(leaves at normal temperature and under chilling treatments) and mainly enriched in gene bodies with the adjacent promoter and downstream areas. The short-term chilling treatments(4°C for 2 h) induced 2 793 gains and 305losses in H3K27me3 modification. Subsequently, 97.3% of the alterations were restored to original levels after 24 h treatment. The ChIP-qPCR for five differential peaks showed similar results to the data for ChIP-seq, indicating that the chilling-induced H3K27me3 modification is reliable.Integrative analysis of transcriptome and ChIP-seq results showed that the expression of H3K27me3 target genes was significantly lower than those of non-target genes, indicating transcriptional repression of H3K27me3 in grapevine leaves. Furthermore, histone methylation alterations were detected in 82 genes and were related to either repression or activation of their expression during chilling stress.
