3Whole-Genome Resequencing of a Worldwide Collection of Rapeseed Accessions Reveals the Genetic Basis of Ecotype Divergence显示文摘Rapeseed (Brassica napus),an important oilseed crop,has adapted to diverse climate zones and latitudes by forming three main ecotype groups,namely winter,semiwinter,and spring types. However,genetic variations underlying the divergence of these ecotypes are largely unknown. Here,we report the global pattern of genetic polymorphisms in rapeseed determined by resequencing a worldwide collection of 991 germplasm accessions.A total of 5.56 and 5.53 million singlenucleotide polymorphisms (SNPs)as Well as 1.86 and 1.92 million InDels were identified by mapping reads to the reference genomes of 'Darmor-bzh'and 'Tapidor,'respectively.We generated a map of allelic drift paths that shows splits and mixtures of the main populations,and revealed an asymmetric evolution of the two subgenomes of B.napus by calculating the genetic diversity and linkage disequilibrium parameters.Selective-sweep analysis revealed genetic changes in genes orthologous to those regulating various aspects of plant development and response to stresses.A genome-wide association study identified SNPs in the promoter regions of FLOWERING LOCUS T and FLOWERING LOCUS C orthologs that corresponded to the different rapeseed ecotype groups. 7Neuroprotective effects of total saponins from Rubus parvifolius L. on cerebral ischemia/reperfusion injury in rats显示文摘This study examines the neuroprotective effects and mechanisms of action of total saponins from Rubus parvifolius L. (TSRP) on focal cerebral ischemia and reperfusion injury in rats. Focal cerebral ischemia and reperfusion injury was performed in rats using the suture method. The results indicate that intragastric injection of TSRP, at 5, 10 and 20 mg/kg, could decrease neurological impairment, reduce cerebral infarct volume, diminish pathological changes, and significantly inhibit the apoptosis of neurons surrounding the ischemic area. In addition, TSRP upregulated the expression of the anti-apoptotic factor Bcl-2, at the protein and mRNA levels, and it downregulated the expression of the pro-apoptotic factor Bax, at the protein and mRNA levels. 9The distribution of chlorophyll a in the tropical eastern Indian Ocean in austral summer显示文摘To study the effect of hydrographic factors on the spatial distributions of chlorophyll a (Chl a), an investigation was carried out in the tropical eastern Indian Ocean (80 -100 E along 7 S, and 7 -18 S along 80 E) in December 2010. The fluorescent method was used to obtain total Chl a and size-fractioned Chl a at the 26 stations. The results show that surface Chl a concentration averaged at (0.168 ± 0.095) mg/m 3 s.d. (range: 0.034-0.475 mg/m 3 ), concentrations appeared to be higher in the west for longitudinal variations, and higher in the north for latitudinal variations. Furthermore, the surface Chl a concentration was lower (0.034-0.066 mg/m 3 ) in the region to the south of 16 S. There was a strong subsurface Chl a maximum layer at all stations and the depth of the Chl a maximum increased towards to the east and south along with the respective nitracline. The spatial variation of Chl a was significant: correlation and regression analysis suggests that it was primarily affected by PO 3 4 , N(NO 3 -N+NO 2 -N) and temperature. Size-fractionated Chl a concentration clearly showed that the study area was a typical oligotrophic open ocean, in which picophytoplankton dominated, accounting for approximately 67.8% of total Chl a, followed by nanophytoplankton (24.5%) and microphytoplankton (7.6%). 10Generation of rat blood vasculature and hematopoietic cells in rat-mouse chimeras by blastocyst complementation显示文摘Interspecies chimera through blastocyst complementation could be an alternative approach to create human organs in animals by using human pluripotent stem cells.A mismatch of the major histocompatibility complex of vascular endothelial cells between the human and host animal will cause graft rejection in the transplanted organs.Therefore,to achieve a transplantable organ in animals without rejection,creation of vascular endothelial cells derived from humans within the organ is necessary.In this study,to explore whether donor xeno-pluripotent stem cells can compensate for blood vasculature in host animals,we generated rat-mouse chimeras by injection of rat embryonic stem cells(rESCs)into mouse blastocysts with deficiency of Flk-1 protein,which is associated with endothelial and hematopoietic cell development.We found that rESCs could differentiate into vascular endothelial and hematopoietic cells in the rat-mouse chimeras.The whole yolk sac(YS)of Flk-1^EGFP/ECFP rat-mouse chimera was full of rat blood vasculature.Rat genes related to vascular endothelial cells,arteries,and veins,blood vessels formation process,as well as hematopoietic cells,were highly expressed in the YS.Our results suggested that rat vascular endothelial cells could undergo proliferation,migration,and self-assembly to form blood vasculature and that hematopoietic cells could differentiate into B cells,T cells,and myeloid cells in rat-mouse chimeras,which was able to rescue early embryonic lethality caused by Flk-1 deficiency in mouse.Xiaomin Wang Hui Shi Juanjuan Zhou Qingjian Zou Quanjun Zhang Shixue Gou Pengfei Chen Lisha Mou Nana Fan Yangyang Suo Zhen Ouyang Chengdan Lai Quanmei Yan Liangxue Lai 2020Journal of Genetics and Genomics2020,47,5:2

12The dichotomous and incomplete adaptive immunity in COVID-19 patients with different disease severity显示文摘The adaptive immunity that protects patients from coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is not well characterized.In particular,the asymptomatic patients have been found to induce weak and transient SARS-CoV-2 antibody responses,but the underlying mechanisms remain unknown;meanwhile,the protective immunity that guide the recovery of these asymptomatic patients is elusive.Here,we characterized SARS-CoV-2-specific B-cell and T-cell responses in 10 asymptomatic patients and 64 patients with other disease severity(mild,n=10,moderate,n=32,severe,n=12)and found that asymptomatic or mild symptomatic patients failed to mount virus-specific germinal center(GC)B cell responses that result in robust and prolonged humoral immunity,assessed by GC response indicators including follicular helper T(TFH)cell and memory B cell responses as well as serum CXCL13 levels.Alternatively,these patients mounted potent virus-specific TH1 and CD8+T cell responses.In sharp contrast,patients of moderate or severe disease induced vigorous virus-specific GC B cell responses and associated TFH responses;however,the virus-specific TH1 and CD8+T cells were minimally induced in these patients.These results,therefore,uncovered the protective immunity in asymptomatic patients and also revealed the strikingly dichotomous and incomplete humoral and cellular immune responses in COVID-19 patients with different disease severity,providing important insights into rational design of effective COVID-19 vaccines.Leiqiong Gao Jing Zhou Sen Yang Lisha Wang Xiangyu Chen Yang Yang Ren Li Zhiwei Pan Jing Zhao Zhirong Li Qizhao Huang Jianfang Tang Li Hu Pinghuang Liu Guozhong Zhang Yaokai Chen Lilin Ye 2021Signal Transduction and Targeted Therapy2021,6,4:2

18Nano Fe_(2)O_(3)embedded in montmorillonite with citric acid enhanced photocatalytic activity of nanoparticles towards diethyl phthalate显示文摘Nano-Fe_(2)O_(3)embedded in montmorillonite particles(Fe-Mt)were prepared to degrade diethyl phthalate(DEP)with citric acid(CA)under xenon light irradiation.Compared to pristine montmorillonite(Na-Mt),the embedding process increased 14.5-fold of iron content and 1.8-fold of specific surface area.The synthesized Fe-Mt have more oxygen vacancies than Fe_(2)O_(3)nanoparticles(nFe_(2)O_(3)),which could induce more reactive oxygen species(ROSs)generation in the presence of CA under xenon lamp irradiation.Fe-Mt with CA enhanced photo-assisted degradation of DEP 2.5 times as compared to nFe_(2)O_(3)with CA.Quenching experiments,electron paramagnetic resonance(EPR)spectroscopy and identification of products confirmed that surface-bound·OH was the main radical to degrade DEP.Common anions(i.e.,NO_(3)-,CO_(3)^(2-),Cl^-)and humic acid could compete·OH with DEP and cause slower degradation of DEP.The removal efficiency of DEP was more than 56%with Fe-Mt after three recycles,and the dissolved Fe concentration from Fe-Mt was below 75μmol/L,indicating Fe-Mt had a good stability as a catalyst.Fe-Mt together with CA appeared to be a promising strategy to remove organic pollutants in surface water,or topsoil under solar irradiation.Zhaoyue Sun Lisha Feng Guodong Fang Longgang Chu Dongmei Zhou Juan Gao 2021Journal of Environmental Sciences2021,33,3:1
