2Flow Field Investigation in a Trapezoidal Duct with Swirl Flow Induced by Impingement Jets显示文摘An enlarged model of trapezoidal duct near the leading-edge in the blade is built up. The effects of impingement jets, swirl flow, cross flow and effusion flow are considered. Experiments are performed to measure flow fields in this confined passage and exit holes on one of its side walls. Cross flow and effusion flow are induced in the channel by the outflow of side exit hole (SEH) and film cooling hole (FCH), which are oriented on one end wall and bottom wall of the passage. Detailed flow structures are measured for two impingement angles of 35° and 45° with 6 combinations of outflow ratios. Results show that the small jets impinge the target wall effectively while the large jets contribute to inducing and impelling a strong counter-clockwise vortex in the upper part of the passage. Cross flow plays a dominate role for the flow structures in the passage and exit holes. It deflects jets, enhances swirl and deteriorates side exit conditions. Impingement angle is another significant factor for the flow characteristics. Its effect reveals more evidently with cross flow. 3CD38 deficiency alleviates Ang II-induced vascular remodeling by inhibiting small extracellular vesicle-mediated vascular smooth muscle cell senescence in mice显示文摘CD38 is the main enzyme for nicotinamide adenine dinucleotide(NAD)degradation in mammalian cells.Decreased NAD levels are closely related to metabolic syndromes and aging-related diseases.Our study showed that CD38 deficiency significantly alleviated angiotensin II(Ang ll)-induced vascular remodeling in mice,as shown by decreased blood pressures;reduced vascular media thickness,media-to-lumen ratio,and collagen deposition;and restored elastin expression.However,our bone marrow transplantation assay showed that CD38 deficiency in lymphocytes led to lack of protection against Ang ll-induced vascular remodeling,suggesting that the effects of CD38 on Ang ll-induced vascular remodeling might rely primarily on vascular smooth muscle cells(VSMCs),not lymphocytes.In addition,we observed that CD38 deficiency or NAD supplementation remarkably mitigated Ang ll-induced vascular senescence by suppressing the biogenesis,secretion,and internalization of senescence-associated small extracellular vesicles(SA-sEVs),which facilitated the senescence of neighboring non-damaged VSMCs.Furthermore,we found that the protective effects of CD38 deficiency on VSMC senescence were related to restoration of lysosome dysfunction,particularly with respect to the maintenance of sirtuin-mediated mitochondrial homeostasis and activation of the mitochondria-lysosomal axis in VSMCs.In conclusion,our findings demonstrated that CD38 and its associated intracellular NAD decline are critical for Ang ll-induced VSMC senescence and vascular remodeling.Lu Gan Demin Liu Jing Liu Erya Chen Chan Chen Lian Liu Hang Hu Xiaohui Guan Wen Ma Yanzi Zhang Yarong He Bofu Liu Songling Tang Wei Jiang Jianxin Xue Hongbo Xin 2021Signal Transduction and Targeted Therapy2021,6,7:2

7Allometric models for aboveground biomass of six common subtropical shrubs and small trees显示文摘The aboveground biomass(AGB)of shrubs and small trees is the main component for the productivity and carbon storage of understory vegetation in subtropical secondary forests.However,few allometric models exist to accurately evaluate understory biomass.To estimate the AGB of five common shrub(diameter at base<5 cm,<5 m high)and one small tree species(<8 m high,trees’s seedling),206 individuals were harvested and species-specific and multi-species allometric models developed based on four predictors,height(H),stem diameter(D),crown area(Ca),and wood density(ρ).As expected,the six species possessed greater biomass in their stems compared with branches,with the lowest biomass in the leaves.Species-specific allometric models that employed stem diameter and the combined variables of D~2H andρDH as predictors accurately estimated the components and total AGB,with R^(2) values from 0.602 and 0.971.A multi-species shrub allometric model revealed that wood density×diameter×height(ρDH)was the best predictor,with R^(2) values ranging from between 0.81 and 0.89 for the components and total AGB,respectively.These results indicated that height(H)and diameter(D)were effective predictors for the models to estimate the AGB of the six species,and the introduction of wood density(ρ)improved their accuracy.The optimal models selected in this study could be applied to estimate the biomass of shrubs and small trees in subtropical regions.Cheng Huang Chun Feng Yuhua Ma Hua Liu Zhaocheng Wang Shaobo Yang Wenjing Wang Songling Fu Han Y.H.Chen 2022Journal of Forestry Research2022,33,4:1

9Anti-PD-L1 antibody enhances curative effect of cryoablation via antibody-dependent cell-mediated cytotoxicity mediating PD-L1^(high)CD11b^(+)cells elimination in hepatocellular carcinoma显示文摘Cryoablation(CRA)and microwave ablation(MWA)are two main local treatments for hepatocellular carcinoma(HCC).However,which one is more curative and suitable for combining with immunotherapy is still controversial.Herein,CRA induced higher tumoral PD-L1 expression and more T cells infiltration,but less PD-L1^(high)CD11b^(+)myeloid cells infiltration than MWA in HCC.Furthermore,CRA had better curative effect than MWA for anti-PD-L1 combination therapy in mouse models.Mechanistically,anti-PD-L1 antibody facilitated infiltration of CD8^(+)T cells by enhancing the secretion of CXCL9 from cDC1 cells after CRA therapy.On the other hand,anti-PD-L1 antibody promoted the infiltration of NK cells to eliminate PD-L1^(high)CD11b^(+)myeloid cells by antibody-dependent cell-mediated cytotoxicity(ADCC)effect after CRA therapy.Both aspects relieved the immunosuppressive microenvironment after CRA therapy.Notably,the wild-type PD-L1 Avelumab(Bavencio),compared to the mutant PD-L1 atezolizumab(Tecentriq),was better at inducing the ADCC effect to target PD-L1^(high)CD11b^(+)myeloid cells.Collectively,our study uncovered the novel insights that CRA showed superior curative effect than MWA in combining with anti-PD-L1 antibody by strengthening CTL/NK cell immune responses,which provided a strong rationale for combining CRA and PD-L1 blockade in the clinical treatment for HCC.Jizhou Tan Ting Liu Wenzhe Fan Jialiang Wei Bowen Zhu Yafang Liu Lingwei Liu Xiaokai Zhang Songling Chen Haibiao Lin Yuanqing Zhang Jiaping Li 2023Acta Pharmaceutica Sinica B2023,13,2:0
10Lactate regulates major zygotic genome activation by H3K18 lactylation in mammals显示文摘Lactate is present at a high level in the microenvironment of mammalian preimplantation embryos in vivo and in vitro.However,its role in preimplantation development is unclear.Here,we report that lactate is highly enriched in the nuclei of early embryos when major zygotic genome activation(ZGA)occurs in humans and mice.The inhibition of its production and uptake results in developmental arrest at the 2-cell stage,major ZGA failure,and loss of lactate-derived H3K18lac,which could be rescued by the addition of Lac-CoA and recapitulated by overexpression of H3K18R mutation.By profiling the landscape of H3K18lac during mouse preimplantation development,we show that H3K18lac is enriched on the promoter regions of most major ZGA genes and correlates with their expressions.In humans,H3K18lac is also enriched in ZGA markers and temporally concomitant with their expressions.Taken together,we profile the landscapes of H3K18lac in mouse and human preimplantation embryos,and demonstrate the important role for H3K18lac in major ZGA,showing that a conserved metabolic mechanism underlies preimplantation development of mammalian embryos.Jingyu Li Weibo Hou Qi Zhao Wei Han Hongdi Cui Songling Xiao Ling Zhu Jiadan Qu Xiaoyu Liu Weitao Cong Jingling Shen Yuzheng Zhao Shaorong Gao Guoning Huang Qingran Kong 2024National Science Review2024,11,2:0
11Intrinsically bioactive and biomimetic nanoparticle-derived therapies alleviate asthma by regulating multiple pathological cells显示文摘Asthma is a serious global public health concern. Airway neutrophilic inflammation is closely related to severe asthma, for which effective and safe therapies remain to be developed. Here we report nanotherapies capable of simultaneously regulating multiple target cells relevant to the pathogenesis of neutrophilic asthma. A nanotherapy LaCD NP based on a cyclic oligosaccharide-derived bioactive material was engineered. LaCD NP effectively accumulated in the injured lungs of asthmatic mice and mainly distributed in neutrophils, macrophages, and airway epithelial cells after intravenous or inhalation delivery, thereby ameliorating asthmatic symptoms and attenuating pulmonary neutrophilic inflammation as well as reducing airway hyperresponsiveness, remodeling, and mucus production. Surface engineering via neutrophil cell membrane further enhanced targeting and therapeutic effects of LaCD NP. Mechanistically, LaCD NP can inhibit the recruitment and activation of neutrophils, especially reducing the neutrophil extracellular traps formation and NLRP3 inflammasome activation in neutrophils. Also, LaCD NP can suppress macrophage-mediated pro-inflammatory responses and prevent airway epithelial cell death and smooth muscle cell proliferation, by mitigating neutrophilic inflammation and its direct effects on relevant cells. Importantly, LaCD NP showed good safety performance. 12Corrosion Performance of Carbon Steel in CO_(2) Aqueous Environment Containing Silty Sand with Different Sizes显示文摘Corrosion performance of carbon steel in CO_2 aqueous environment containing silty sand with different sizes was investigated by immersion tests and electrochemical measurements.Silty sand could form an adsorption layer on steel surface in initial period,and the sand adsorption layer was turned into a mixture film of silty sand with corrosion product in last period.The adsorption layer in 325 mesh condition(large size)had the fewest pores for H_2CO_3 transport,exhibiting the highest cathodic current inhibition.In spite of little corrosion product,the sand adsorption film formed in 325 mesh condition induced the lowest corrosion rate.For 1000 and 5000 mesh silty sand,the sand adsorption layer had some pores for H_2CO_3 transport,leading to low cathodic current inhibition and much matrix dissolution.But the adsorption layer for5000 mesh silty sand(small size)had the largest special surface area to accelerate heterogeneous precipitation of corrosion product FeCO_3.Therefore,the mixture film in 5000 mesh condition was more compact,exhibiting stronger anodic inhibition and lower corrosion rate than those in 1000 mesh condition.Songle Lu Wei Liu Shian Zhang Xiaolong Qi Xiaogang Li Xuemin Wang 2017Acta Metallurgica Sinica(English Letters)2017,30,11:0
13EXPERIMENTAL STUDY OF FLOW FIELDS IN THE VICINITY OF A FILM COOLING HOLE EXIT显示文摘For the first time, an important ingested flow phenomenon was discovered inexperiments at the film cooling hole exit. The trends of 3-D flow fie1ds and the fullnessfactor, Ci, were discussed in detail over a wide range of now parameters and the geometryof fan-shaped holes at this exit plane.
