2Temperature-sensitive polymeric nanogels encapsulating withβ-cyclodextrin and ICG complex for high-resolution deep-tissue ultrasound-switchable fluorescence imaging显示文摘One of the thorny problems currently impeding the applications of the fluorescence imaging technique is the poor spatial resolution in deep tissue.Ultrasound-switchable fluorescence(USF)imaging is a novel imaging tool that has recently been explored to possibly surmount the above-mentioned bottleneck.Herein,αβ-cyclodextrin/indocyanine green(ICG)complex-encapsulated poly(N-isopropylacrylamide)(PNIPAM)nanogel was synthesized and studied for ex vivo/in vivo deep tissue/high-resolution near infrared USF(NIR-USF)imaging.To be specific,our results revealed that the average diameter of the as-prepared nanogels was significantly decreased to-32 nm from-335 nm compared to the reported ICG-PNIPAM nanoparticles.Additionally,the excitation/emission characteristics of the ICG itself in present nanogels were almost completely retained,and the resultant nanogel exhibited high physiological stability and positive biocompatibility.In particular,the signal-to-noise ratio of the USF image for the PNIPAM/P-cyclodextrin/ICG nanogel(33.01±2.42 dB)was prominently higher than that of the ICG-PNIPAM nanoparticles(18.73±0.33 dB)in 1.5-cm-thick chicken breast tissues.The NIR-USF imaging in 3.5-cm-thick chicken breast tissues was achieved using this new probe.The e x v iv o NIR-USF imaging of the mouse liver was also successfully obtained.Animal experiments showed that the present nanogels were able to be effectively accumulated into U87 tumor-bearing mice via enhanced permeability and retention effects,and the high-resolution NIR-USF imaging of in v ivo tumor was efficiently acquired.The metabolism and in vivo biodistribution of the nanogels were evaluated.Overall,the results suggest that the current nanogel is a highly promising NIR-USF probe for deep tissue and high-resolution USF imaging.Ruilin Liu Tingfeng Yao Yang Liu Shuai Yu Liqin Ren Yi Hong Kytai T.Nguyen Baohong Yuan 2020Nano Research2020,13,4:1

7In vivo tumor ultrasound-switchable fluorescence imaging via intravenous injections of size-controlled thermosensitive nanoparticles显示文摘Near-infrared fluorescence imaging has emerged as a noninvasive,inexpensive,and ionizing-radiation-free monitoring tool for assessing tumor growth and treatment efficacy.In particular,ultrasound switchable fluorescence(USF)imaging has been explored with improved imaging sensitivity and spatial resolution in centimeter-deep tissues.This study achieved the size control of polymer-based and indocyanine green(ICG)encapsulated USF contrast agents,capable of accumulating in the tumor after intravenous injections.These nanoprobes varied in size from 58 to 321 nm.The bioimaging profiles demonstrated that the proposed nanoparticles can efficiently eliminate the background light from normal tissue and show a tumor-specific fluorescence enhancement in the BxPC-3 tumor-bearing mice models possibly via the enhanced permeability and retention effect.In vivo tumor USF imaging further demonstrated that these nanoprobes can effectively be switched“ON”with enhanced fluorescence in response to a focused ultrasound stimulation in the tumor microenvironment,contributing to the high-resolution USF images.Therefore,our findings suggest that ICG-encapsulated nanoparticles are good candidates for USF imaging of tumors in live animals,indicating their great potential in optical tumor imaging in deep tissue.Liqin Ren Yang Liu Tingfeng Yao Kytai TNguyen Baohong Yuan 2023Nano Research2023,16,1:0

9Analysis of the record-breaking August 2021 rainfall over the Greenland Ice Sheet显示文摘Rainfall was witnessed for the first time at the highest area of the Greenland Ice Sheet on 14 August,2021.The thermodynamic mechanisms supporting the rainfall are revealed by ERA5 reanalysis,in-situ and satellite data.We find that a strong southward intrusion of the polar vortex favored the maintenance of a deep cyclone over Baffin Island and an amplification of anticyclonic circulation over the southeastern ice sheet,which pumped warm and moist air toward Greenland from anomalously warm waters south of Greenland.Across a wide swath of the ice sheet,atmospheric uplift maintained above-melting and rainfall conditions via condensation and enhanced downward infrared irradiance.Without the low-level liquid clouds,the spatial extent and duration of the rainfall would have been smaller.Over the ice sheet topographic summit,the air temperature from the ground to 250 hPa level was~2℃higher than the previous record set on 12 July,2012.Such events may occur more frequently with the decreased temperature contrast between the Arctic and the mid-latitude regions that drives highly amplified jet streams.Thus,this extreme event serves as a harbinger of a more likely wet surface condition across all elevations of the ice sheet.DOU Tingfeng XIE Zuowei Jason E.BOX YANG Qing YANG Yifan TENG Shiwen XU Gaojie LIU Chao LI Xichen Derek HOUTZ GONG Xun DU Zhiheng DING Minghu YU Yongqiang XIAO Cunde 2023Advances in Polar Science2023,34,3:0
10Distinct and shared endothermic strategies in the heat producing tissues of tuna and other teleosts显示文摘Although most fishes are ectothermic,some,including tuna and billfish,achieve endothermy through specialized heat producing tissues that are modified muscles.How these heat producing tissues evolved,and whether they share convergent molecular mechanisms,remain unresolved.Here,we generated a high-quality genome from the mackerel tuna(Euthynnus affinis)and investigated the heat producing tissues of this fish by single-nucleus and bulk RNA sequencing.Compared with other teleosts,tuna-specific genetic variation is strongly associated with muscle differentiation.Single-nucleus RNA-seq revealed a high proportion of specific slow skeletal muscle cell subtypes in the heat producing tissues of tuna.Marker genes of this cell subtype are associated with the relative sliding of actin and myosin,suggesting that tuna endothermy is mainly based on shivering thermogenesis.In contrast,cross-species transcriptome analysis indicated that endothermy in billfish relies mainly on nonshivering thermogenesis.Nevertheless,the heat producing tissues of the different species do share some tissue-specific genes,including vascular-related and mitochondrial genes.Overall,although tunas and billfishes differ in their thermogenic strategies,they share similar expression patterns in some respects,highlighting the complexity of convergent evolution.Baosheng Wu Xueli Gao Mingling Hu Jing Hu Tianming Lan Tingfeng Xue Wenjie Xu Chenglong Zhu Yuan Yuan Jiangmin Zheng Tao Qin Peidong Xin Ye Li Li Gong Chenguang Feng Shunping He Huan Liu Haimeng Li Qing Wang Zhenhua Ma Qiang Qiu Kun Wang 2023Science China(Life Sciences)2023,66,11:0
