| 1 | A novel peptide, selected from phage display library of random peptides, can efficiently target into human breast cancer cell显示文摘To develop a targeting vector for breast cancer biotherapy, MDA-MB-231 cell, a human breast cancer cell line, was co-cultured with pC89 (9 aa) phage display library of random peptides. In multiple inde- pendent peptide-presenting phage screening trials, subtilisin was used as a protease to inactivate ex- tra-cellular phages. The internalized phages were collected by cell lysising and amplified in E. coli XLI-Blue. Through five rounds of selection, the peptide-presenting phages which could be internalized in MDA-MB-231 cells were isolated. A comparison was made between internalization capacities of pep- tide-presenting phages isolated from MDA-MB-231 cells and RGD-integrin binding phage by cocultur- ing them with other human tumor cell lines and normal cells. The nucleotide sequences of isolated peptide-presenting phages were then determined by DNA sequencing. To uncover whether phage coat protein or amino acid order was required for the character of the peptide to MDA-MB-231 cells, three peptides were synthesized. They are CASPSGALRSC, ASPSGALRS and CGVIFDHSVPC (the shifted sequence of CASPSGALRSC), and after coculturing them with different cell lines, their targeting ca- pacities to MDA-MB-231 cells were detected. These data suggested that the internalization process was highly selective, and capable of capturing a specific peptide from parent peptide variants. Moreover, the targeting internalization event of peptides was an amino acid sequence dependent manner. | 2 | A review on application of composite truss bridges composed of hollow structural section members显示文摘Composite truss with hollow structural section(HSS) members is deemed as the structure applicable to large-span and heavily-loaded bridges. To promote the application of composite truss bridge with HSS members in China, this paper described its structural characteristics and technology in details. Besides, not only were 32 typical design cases of composite truss bridges with HSS members collected, but also the corresponding historical development was summed up. Comparisons on structural components, characteristics and engineering applications were made among composite truss bridges with HSS members in different structural forms. Then, the analysis on the characteristics of composite truss bridges with concrete-filled steel tubular(CFST) chords was conducted, and the challenges on the aspects of complicated joints and interfacial debonding were pointed out. To address these problems, the concrete-filled rectangular hollow section(CFRHS)stiffened with PBLs(Perfobond Ribs) was proposed to improve the reliability of confinement effect in CFST member and force transfer in joints. The advantages on the mechanical properties of CFRHS structure stiffened using PBLs were systematically expounded with respect to the behaviors of steel plate, interface, member and joint. | 3 | Reuse of Fenton sludge as an iron source for NiFe_2O_4 synthesis and its application in the Fenton-based process显示文摘The potentially hazardous iron-containing sludge from the Fenton process requires proper treatment and disposal, which often results in high treatment cost. In this study, a novel method for the reuse of Fenton sludge as an iron source for the synthesis of nickel ferrite particles(NiFe_2O_4) is proposed. Through a co-precipitation method followed by sintering at 800°C, magnetic NiFe_2O_4 particles were successfully synthesized, which was confirmed by powder X-ray diffraction(XRD), scanning electronic microscopy(SEM), energy dispersive spectroscopy(EDS), Fourier transform infrared spectroscopy(FT-IR) and Raman spectroscopy. The synthesized NiFe_2O_4 could be used as an efficient catalyst in the heterogeneous Fenton process. In phenol degradation with H_2O_2 or NiFe_2O_4 alone, the phenol removal efficiencies within the reaction time of 330 min were as low as 5.9% ± 0.1% and 13.5% ±0.4%, respectively. However, in the presence of both NiFe_2O_4 and H_2O_2, phenol removal efficiency as high as 95% ± 3.4% could be achieved, indicating the excellent catalytic performance of NiFe_2O_4 in the heterogeneous Fenton process. Notably, a rapid electron exchange between_Ni II and_Fe III ions in the NiFe_2O_4 structure could be beneficial for the Fenton reaction. | 4 | Removal of phosphate from wastewater using alkaline residue显示文摘Alkaline residue(AR) was found to be an efficient adsorbent for phosphate removal from wastewater. The kinetic and equilibrium of phosphate removal were investigated to evaluate the performance of modified alkaline residue. After treatment by NaOH(AR-NaOH), removal performance was significantly improved, while removal performance was almost completely lost after treatment by HCl(AR-HCl). The kinetics of the removal process by all adsorbents was well characterized by the pseudo second-order model. The Langmuir model exhibited the best correlation for AR-HCl, while AR was effectively described by Freundlich model. Both models were well fitted to AR-NaOH. The maximum adsorption capacities calculated from Langmuir equation were in following manner: AR-NaOH > AR > AR-HCl. Phosphate removal by alkaline residue was pH dependent process. Mechanisms for phosphate removal mainly involved adsorption and precipitation, varied with equilibrium pH of solution. For AR-HCl, the acid equilibrium pH(< 6.0) was unfavorable for the formation of Ca-P precipitate, with adsorption as the key mechanism for phosphate removal. In contrast, for AR and ARNaOH, precipitation was the dominant mechanism for phosphate removal, due to the incrase on pH(> 8.0) after phosphate removal. | 6 | Simultaneous pyridine biodegradation and nitrogen removal in an aerobic granular system显示文摘Simultaneous pyridine biodegradation and nitrogen removal were successfully achieved in a sequencing batch reactor(SBR) based on aerobic granules. In a typical SBR cycle, nitritation occurred obviously after the majority of pyridine was removed, while denitrification occurred at early stage of the cycle when oxygen consumption was aggravated. The effect of several key operation parameters, i.e., air flow rate, influent NH_4^+-N concentration,influent p H and pyridine concentration, on nitritation, pyridine degradation and total nitrogen(TN) removal, was systematically investigated. The results indicated that high air flow rate had a positive effect on both pyridine degradation and nitritation but a negative impact of overhigh air flow rate. With the increase of NH_4^+ dosage, both nitritation and TN removal could be severely inhibited. Slightly alkaline condition, i.e., pH 7.0–8.0, was beneficial for both pyridine degradation and nitritation. High pyridine dosage often resulted in the delay of both pyridine degradation and nitritation. Besides, extracellular polymeric substances production was affected by air flow rate, NH_4^+ dosage, pyridine dosage and p H.In addition, high-throughput sequencing analysis demonstrated that Bdellovibrio and Paracoccus were the dominant species in the aerobic granulation system. | 14 | Transcriptome analysis of purple pigment formation in Colocasia esculenta显示文摘Taro(Colocasia esculenta(L.)Schott)is an important crop in Africa,Southeast Asia,and subtropics and is used as a food and medicine.The purple color pigmentation is an appealing character in taro.We sampled taro corms of the cultivar‘Lipu Taro’at four developmental stages,including LPYS1(without purple pigment,50 days of development(DOD)),LPYS2(very few purple pigments,75 DOD),LPYS3(moderate purple pigments,115 DOD)and LPYS4(high purple pigments,205 DOD).The purpose of our study was to identify the key genes underpinning the purple pigmentation in taro based on RNA-sequencing.Through RNA-Seq,6453 differentially expressed transcripts(DETs)were identified between purple and non-purple pigmented samples.We identified 41 and 12 flavonoid and anthocyanin related DETs transcripts,respectively.These DETs were upregulated at LPYS2,LPYS3,and LPYS4 as compared to LPYS1,indicating their positive contribution to the color formation in taro.Moreover,we identified several DETs encoding for transcription factors,including MYB and bHLH,known to be major regulators of structural genes involved in the flavonoid-anthocyanin pathway.Finally,we reported several plant hormones(ethylene,auxin,gibberellin,jasmonic acid,and cytokinin)related DETs,which are predicted to play important roles in the corm coloration.Different regulation of transcripts representing the flavonoid-anthocyanin biosynthesis pathway,plant hormone transduction pathway,and transcription factors may have key roles in purple pigmentation in taro.Our findings will facilitate future research on improving the quality and appeal of taro. | FANGLIAN HE WEIQING DONG SHAOLONG WEI ZUYANG QIU JINGLI HUANG HUIPING JIANG SHIYU HUANG LILI LIU | 2021 | BIOCELL2021,45,3: | 2 |
| 15 | Protective effects and potential underlying mechanisms of sodium copper chlorophyllin against ethanol-induced gastric ulcer in mice显示文摘In study,we aimed to determine the mechanisms underlying the gastroprotective effects of sodium copper chlorophyllin(SCC)against ethanol-induced gastric ulcer injury in mice.First,the gastroprotective effects of SCC against gastric ulcer induced by ethanol were assessed.Then,biochemical,histopathological,immunohistochemistry assays,and western blot analysis were conducted to determine the possible mechanisms of action underlying the effects of SCC.Compared to the effects of omeprazole(OME)in a confirmed mouse model of ethanol-induced gastric ulcer injury,treatment with various doses of SCC resulted in up-regulation of Bcl-2 and down-regulation of the pro-apoptotic protein Bax.Significant decreases in the levels of the malondialdehyde(MDA),myeloperoxidase(MPO),and NO in the gastric tissues were observed.Furthermore,inflammatory cytokine analysis revealed that SCC treatment inhibited the expressions of TNF-αand IL-6,greatly reduced the phosphorylation level of IκB,and repressed the nuclear translocation of NF-κB p65,which demonstrated that SCC inhibited the activation of the NF-κB pathway.The present findings suggest that the protective effects of SCC may be beneficial as a potential preventive and therapeutic agent for gastric ulcer through the NF-κB pathway.Taken together,SCC administration significantly decreased the levels of MPO,NO,and MDA in gastric tissue and exerted a powerful anti-inflammatory activity as demonstrated by reduction in the secretions of proinflammatory mediators such as IL-6 and TNF-α in the serum of mice exposed to ethanol. | Huawei Lv Yan Lin Peigang Liu Weiqing Liang Kemin Wei Jinbao Pu Hongjian Zhang | 2019 | Acta Biochimica et Biophysica Sinica2019,51,9: | 2 |
| 16 | Properties and Acceleration Mechanism of Cement Mortar Added with Low Alkaline Liquid State Setting Accelerator显示文摘Low alkaline liquid state setting accelerator(LSA) for Portland cement was prepared in laboratory from aqueous solution of several inorganic sulfate salts and some organic chemical substances. Properties of cement with addition of LSA relating to its setting time and strength development as well as its resistance to sulfate attack for short and long term exposure were experimentally examined. The experimental results showed that 5%-7% addition of LSA significantly accelerated the initial and final setting of Portland cement in the presence or absence of the blending of mineral admixtures, the initial and final setting time being less than 3 min and 6 min respectively. Meanwhile, the early 1 day curing age compressive strength increased remarkably by 20%, while the late 28 th day curing age compressive strength remained almost unchanged as compared with that of the reference accelerator free cement mortar specimen. Furthermore, mortar specimens of cement added with LSA and exposed to 5% Na2SO4 solution showed their excellent resistance to sulfate attack, with their short and long term curing age resistance coefficient to sulfate attack being around 1.04 to 1.17, all larger than 1.0. XRD analysis on hardened cement paste specimens at very early curing ages of several minutes disclosed the existence of more ettringite in specimens added with LSA than that of the reference specimens, meanwhile SEM observation also revealed the existence of well crystallized ettringite at very early hydration stage, suggesting that the accelerated setting of Portland cement can be attributed to the early and rapid formation of ettringite over the whole cement paste matrix due to the introduction of LSA. | 17 | Study on coupled vibration characteristics of a cylindrical container with multiple elastic annular baffles显示文摘The coupled vibration characteristics of multiple elastic annular baffles of the same inner radius in a partially liquid-filled rigid cylindrical container were studied. The liquid domain was divided into several simple sub-domains so that the liquid velocity potential in each liquid sub-domain was of class C1 with continuous boundary conditions. Based on the superposition principle, the general solution of the liquid velocity potential corresponding to each liquid sub-domain was obtained by means of the method of separation of variables. The coupled modes of the multiple elastic annular baffles were expressed in terms of dry-modal functions. The free surface condition, the interface conditions and coupled vibration conditions were expressed in terms of Fourier series along the liquid height and Bessel series in the radial direction, respectively. Stable and fast numerical computations were investigated by the convergence study. Excellent agreements have been achieved in the comparison of results obtained by the proposed approach with those given by the finite element software ADINA. | 18 | H_(2)S promotes developmental brain angiogenesis via the NOS/NO pathway in zebrafish显示文摘Background Hydrogen sulphide(H_(2)S)is considered as the third member of the gasotransmitter family,along with nitric oxide(NO)and carbon monoxide.H_(2)S has been reported to induce angiogenesis by promoting the growth,migration and tube-like structure formation of endothelial cells.Those studies were conducted in conditions of cell culture,mouse Matrigel plug assay model,rat wound healing model or rat hindlimb ischaemia model.Recent in vivo studies showed the physiological importance of H_(2)S in muscle angiogenesis.However,the importance of endogenous H_(2)S for brain angiogenesis during development remains unknown.We therefore aimed at determining the role of H_(2)S in brain vascular development.Methods and results Both knockdown and knockout of H2S-producing enzymes,cystathionineβ-synthase(cbs)and cystathionineγ-lyase(cth),using morpholino oligonucleotides and clustered regularly interspaced short palindromic repeats/Cas9-mediated mutation,impaired brain vascular development of larval zebrafish.Incubation with the slow-releasing H_(2)S donor GYY4137 alleviated the defects of brain vascular development in cbs and cth morphants.Quantitative analysis of the midbrain vascular network showed that H_(2)S enhances angiogenesis without affecting the topological structure of the brain vasculature.Mechanically,nitric oxide synthase 2a(nos2a)expression and NO production were decreased in both cbs and cth morphants.Overexpression of nos2a by coinjection of cbs or cth MO with full-length zebrafish nos2a mRNA alleviated the brain vascular developmental defects in cbs and cth morphants.Conclusion We conclude that H_(2)S promotes brain developmental angiogenesis via the NOS/NO pathway in zebrafish. | Weiqing Jiang Chen Liu Mingzhu Deng Fei Wang Xiao Ren Yilin Fan Jiulin Du Yonggang Wang | 2021 | Stroke & Vascular Neurology2021,6,2: | 2 |
| 19 | MXene based mechanically and electrically enhanced film for triboelectric nanogenerator显示文摘The development of triboelectric nanogenerator(TENG)technology which can directly convert ambient mechanical energy into electric energy may affect areas from green energy harvesting to emerging wearing electronics.And,the material of triboelectric layer is critical to the mechanical robustness and electrical output characteristics of the TENGs.Herein,a MXene enhanced electret polytetrafluoroethylene(PTFE)film with a high mechanical property and surface charge density is developed.The MXene/PTFE composite film was synthesized by spraying and annealing treatment.With the doping of MXene,the crystallinity of composite film could be tuned,leading to an enhancement in the tensile property of 450%and reducing the wear volume about 80%in the friction test.Furthermore,the as-fabricated TENG with this composite film outputs 397 V of open-circuit voltage,21µA of short-circuit current,and 232 nC of transfer charge quantity,which are 4,6,and 6 times higher than that of the TENG made by pure PTFE film,respectively.Therefore,this work provides a creative strategy to simultaneously improve the mechanical property and electrical performance of the TENGs,which have great potential in improving device stability under a complex mechanical environment. | Yuyu Gao Guoxu Liu Tianzhao Bu Yaoyao Liu Youchao Qi Yanting Xie Shaohang Xu Weili Deng Weiqing Yang Chi Zhang | 2021 | Nano Research2021,14,12: | 2 |
