| 2 | Potentiating CD8^(+) T cell antitumor activity by inhibiting PCSK9 to promote LDLR-mediated TCR recycling and signaling显示文摘Metabolic regulation has been proven to play a critical role in T cell antitumor immunity.However,cholesterol metabolism as a key component of this regulation remains largely unexplored.Herein,we found that the low-density lipoprotein receptor(LDLR),which has been previously identified as a transporter for cholesterol,plays a pivotal role in regulating CD8+T cell antitumor activity.Besides the involvement of cholesterol uptake which is mediated by LDLR in T cell priming and clonal expansion,we also found a non-canonical function of LDLR in CD8+T cells:LDLR interacts with the T-cell receptor(TCR)complex and regulates TCR recycling and signaling,thus facilitating the effector function of cytotoxic T-lymphocytes(CTLs).Furthermore,we found that the tumor microenvironment(TME)downregulates CD8+T cell LDLR level and TCR signaling via tumor cell-derived proprotein convertase subtilisin/kexin type 9(PCSK9)which binds to LDLR and prevents the recycling of LDLR and TCR to the plasma membrane thus inhibits the effector function of CTLs.Moreover,genetic deletion or pharmacological inhibition of PCSK9 in tumor cells can enhance the antitumor activity of CD8+T cells by alleviating the suppressive effect on CD8+T cells and consequently inhibit tumor progression.While previously established as a hypercholesterolemia target,this study highlights PCSK9/LDLR as a potential target for cancer immunotherapy as well. | Juanjuan Yuan Ting Cai Xiaojun Zheng Yangzi Ren Jingwen Qi Xiaofei Lu Huihui Chen Huizhen Lin Zijie Chen Mengnan Liu Shangwen He Qijun Chen Siyang Feng Yingjun Wu Zhenhai Zhang Yanqing Ding Wei Yang | 2021 | Protein & Cell2021,12,4: | 12 |

| 7 | Myt1L Promotes Differentiation of Oligodendrocyte Precursor Cells and is Necessary for Remyelination After Lysolecithin-Induced Demyelination显示文摘The differentiation and maturation of oligodendrocyte precursor cells(OPCs) is essential for myelination and remyelination in the CNS. The failure of OPCs to achieve terminal differentiation in demyelinating lesions often results in unsuccessful remyelination in a variety of human demyelinating diseases. However, the molecular mechanisms controlling OPC differentiation under pathological conditions remain largely unknown. Myt1 L(myelin transcription factor 1-like), mainly expressed in neurons,has been associated with intellectual disability, schizophrenia, and depression. In the present study, we found that Myt1 L was expressed in oligodendrocyte lineage cells during myelination and remyelination. The expression level of Myt1 L in neuron/glia antigen 2-positive(NG^(2+))OPCs was significantly higher than that in mature CC1^+ oligodendrocytes. In primary cultured OPCs,overexpression of Myt1 L promoted, while knockdown inhibited OPC differentiation. Moreover, Myt1 L was potently involved in promoting remyelination after lysolecithin-induced demyelination in vivo. Ch IP assays showed that Myt1 L bound to the promoter of Olig1 and transcriptionally regulated Olig1 expression. | 9 | Exogenous Application of Abscisic Acid (ABA)Enhances Chilling Tolerance in Seedlings ofNapier Grass (Pennisetum purpureum Schum.)显示文摘Napier grass, an important forage crop with potentials in multi-purpose applications, is widely grown throughout the tropics and subtropics. Low temperature severely limits its productivity and geographical distribution in temperate regions of the world. In this study, we investigated the effect of exogenous abscisic Acid(ABA) on chilling tolerance of napier grass(Pennisetum purpureum Schum.) seedlings. Seven-day-old napier grass seedlings were cultured in dd H_2O or ABA solution at different concentrations and exposed to 1 ℃ for different time durations. The chilling injury, membrane stability index(MSI) and proline content were estimated from leaf samples. The results showed that there was obvious morphological injury of leaf blighting and restrained growth for the seedlings under chilling stress, but this damage can be largely reduced(by 2/3) when the seedlings were treated by 100 μmol/L ABA in the culture solution, and that the application of exogenous ABA can help to maintain a good stability of leaf cell membrane as expressed by a high MSI value and a low level of proline in leaf cells. These results suggested that exogenous ABA can significantly alleviate chilling injury in napier grass seedlings by maintaining the stability of leaf cell membrane during chilling stress, and that the chilling tolerance was not ensured by a proline accumulation although a passive accumulation of proline was observed in the seedlings under chilling stress. | 10 | Heterogeneously integrated,superconducting silicon-photonic platform for measurementdevice-independent quantum key distribution显示文摘Integrated photonics provides a route to both miniaturization of quantum key distribution(QKD)devices and enhancing their performance.A key element for achieving discrete-variable QKD is a singlephoton detector.It is highly desirable to integrate detectors onto a photonic chip to enable the realization of practical and scalable quantum networks.We realize a heterogeneously integrated,superconducting silicon-photonic chip.Harnessing the unique high-speed feature of our optical waveguide-integrated superconducting detector,we perform the first optimal Bell-state measurement(BSM)of time-bin encoded qubits generated from two independent lasers.The optimal BSM enables an increased key rate of measurement-device-independent QKD(MDI-QKD),which is immune to all attacks against the detection system and hence provides the basis for a QKD network with untrusted relays.Together with the timemultiplexed technique,we have enhanced the sifted key rate by almost one order of magnitude.With a 125-MHz clock rate,we obtain a secure key rate of 6.166 kbps over 24.0 dB loss,which is comparable to the state-of-the-art MDI-QKD experimental results with a GHz clock rate.Combined with integrated QKD transmitters,a scalable,chip-based,and cost-effective QKD network should become realizable in the near future. | Xiaodong Zheng Peiyu Zhang Renyou Ge Liangliang Lu Guanglong He Qi Chen Fangchao Qu Labao Zhang Xinlun Cai Yanqing Lu Shining Zhu Peiheng Wu Xiao-Song Ma | 2021 | Advanced Photonics2021,3,5: | 5 |

| 12 | Whole Genome Analyses of Chinese Population and De Novo Assembly of A Northern Han Genome显示文摘To unravel the genetic mechanisms of disease and physiological traits,it requires comprehensive sequencing analysis of large sample size in Chinese populations.Here,we report the primary results of the Chinese Academy of Sciences Precision Medicine Initiative(CASPMI)project launched by the Chinese Academy of Sciences,including the de novo assembly of a northern Han reference genome(NH1.0)and whole genome analyses of 597 healthy people coming from most areas in China.Given the two existing reference genomes for Han Chinese(YH and HX1)were both from the south,we constructed NH1.0,a new reference genome from a northern individual,by combining the sequencing strategies of PacBio,10×Genomics,and Bionano mapping.Using this integrated approach,we obtained an N50 scaffold size of 46.63 Mb for the NH1.0 genome and performed a comparative genome analysis of NH1.0 with YH and HX1.In order to generate a genomic variation map of Chinese populations,we performed the whole-genome sequencing of 597 participants and identified 24.85 million(M)single nucleotide variants(SNVs),3.85 M small indels,and 106,382 structural variations.In the association analysis with collected phenotypes,we found that the T allele of rs1549293 in KAT8 significantly correlated with the waist circumference in northern Han males.Moreover,significant genetic diversity in MTHFR,TCN2,FADS1,and FADS2,which associate with circulating folate,vitamin B12,or lipid metabolism,was observed between northerners and southerners.Especially,for the homocysteine-increasing allele of rs1801133(MTHFR 677T),we hypothesize that there exists a “comfort”zone for a high frequency of 677T between latitudes of 35–45 degree North.Taken together,our results provide a high-quality northern Han reference genome and novel population-specific data sets of genetic variants for use in the personalized and precision medicine. | Zhenglin Du Liang Ma Hongzhu Qu Wei Chen Bing Zhang Xi Lu Weibo Zhai Xin Sheng Yongqiao Sun Wenjie Li Meng Lei Qiuhui Qi Na Yuan Shuo Shi Jingyao Zeng Jinyue Wang Yadong Yang Qi Liu Yaqiang Hong Lili Dong Zhewen Zhang Dong Zou Yanqing Wang Shuhui Song Fan Liu Xiangdong Fang Hua Chen Xin Liu Jingfa Xiao Changqing Zeng | 2019 | Genomics, Proteomics & Bioinformatics2019,17,3: | 4 |
| 13 | Screening and regulatory network analysis of survival-related genes of patients with colorectal cancer显示文摘The purpose of this study was to screen key survival-related genes from patients with colorectal cancer and explore signal transduction network of the involved genes.In a previous study,survival-related genes of patients with colorectal cancer were selected by colorectal cancer-related expression data GSE17538 using the Significance Analysis of Microarrays(SAM3.01)software,and 235 genes related to the survival of patients with colorectal cancer were obtained.Therefore,the following screening and analysis were conducted on these 235 genes in this study.First,the enrichment analysis of transcription factor binding sites was conducted on the 235 genes.Genes with more than seven transcription factor binding sites were screened.Then,these genes and upregulated genes in colorectal cancer were intersected.Finally,survival analysis and regulatory network analysis were conducted on the screened genes.This allowed clarification of the relationship between these genes and the survival of patients with colorectal cancer and the signaling network involving these genes in the cell signal transduction network of colorectal cancer.Through the above analysis,six upregulated genes in colorectal cancer related to the survival of colorectal cancer patients and highly regulated by transcription factors were selected,namely STX2,PODXL,KLK6,GRB10,EHBP1 and CREB5.These genes are involved in signal regulatory networks related to colorectal cancer metastasis-related signaling pathways.Therefore,the survival of patients with colorectal cancer is closely correlated with colorectal cancer metastasis.The six survival-related genes affect the survival of patients by regulating colorectal cancer metastasis-associated signaling pathways. | QI Lu DING YanQing | 2014 | Science China(Life Sciences)2014,57,5: | 4 |
| 14 | Multiple Spatial Scale Analysis of the Niche Characteristics of the Rhododendron dauricum Plant Communities in Northeast China显示文摘This study aims to verify the concept of niches at multiple spatial scales in plant communities.To this end,we analyzed the niche characteristic of Rhododendron dauricum plant communities in Northeast China at three spatial scales.At the local scale,we calculated the Importance Value(IV)of species in five communities in the north of the Da Hinggan Mountains.At the intermediate scale,we examined five communities in their entirety,calculated the niche breadth of the species,and integrated niche overlap and interspecific association to analyze interspecific relationships.Further,the generalized additive model(GAM)was used to analyze the impact of topography and soil factors on niche characteristics.At the regional scale,we analyzed the geographical distribution of dominant species of R.dauricum plant communities in Northeast China and used principal component analysis(PCA)to analyze the impact of geographical and climate factors on species distribution.The results show that at the local scale,the IV of the species in each community varies widely.At the intermediate scale,species with a wide niche breadth tend to have a high value for IV.Larix gmelinii,Betula platyphylla,R.dauricum,Ledum palustre,and Vaccinium vitis-idaea had a relatively wide niche breadth and a high niche overlap,and the interspecific associations were almost all positive.Elevation and soil nutrients were the most dominant environmental factors.At the regional scale,species with a wide niche breadth tend to have a wide range of distribution,and temperature and precipitation were the most dominant environmental factors.This study suggests that the niche characteristics at three scales are both related and different.Niche characteristics at the local scale were various and labile,and niche characteristics at the intermediate and regional scales were relatively regular.These results show some degree of consistency with previous studies from an evolutionary perspective.The action mechanisms of these communities are related to differences in the dominant environmental factors.In addition,the integration of niche overlap and interspecific association determine interspecific relationships more accurately. | LU Yupeng CHEN Wei YAO Jing HUANG Yanqing ZHANG Yue LIU Huanchu HE Xingyuan | 2020 | Chinese Geographical Science2020,30,4: | 4 |

| 16 | Potential antitumor mechanisms of phenothiazine drugs显示文摘In this study,three kinds of phenothiazine drugs were analyzed to explore their potential antitumor mechanisms.First,target proteins that could interact with chlorpromazine,fluphenazine and trifluoperazine were predicted.Then,the target proteins of the three drugs were intersected.Cell signaling pathway enrichment and related disease enrichment were conducted for the intersected proteins to extract the enrichment categories associated with tumors.By regulation network analysis of the protein interactions,the mechanisms of action of these target proteins in tumor tissue were clarified,thus confirming the potential antitumor mechanisms of the phenothiazine drugs.The final results of cell signaling pathway enrichment and related disease enrichment showed that the categories with the highest score were all found in tumors.Target proteins belonging to the tumor category included signaling pathway members such as Wnt,MAPK and retinoic acid receptor.Moreover,another target protein,MAPK8,could indirectly act on target proteins CDK2,IGF1R,GSK3B,RARA,FGFR2 and MAPK10,thereby affecting tumor cell division and proliferation.Therefore,phenothiazine drugs may have potential antitumor effects,and tumorassociated target proteins play important roles in the process of cell signaling transduction cascades. | QI Lu DING YanQing | 2013 | Science China(Life Sciences)2013,56,11: | 3 |

| 20 | Partial anammox achieved in full scale biofilm process for typical domestic wastewater treatment显示文摘The slow initiation of anammox for treating typical domestic wastewater and the relatively high footprint of wastewater treatment infrastructures are major concerns for practical wastewater treatment systems.Herein,a 300 m^(3)/d hybrid biofilm reactor(HBR)process was developed and operated with a short hydraulic retention time(HRT)of 8 h.The analysis of the bacterial community demonstrated that anammox were enriched in the anoxic zone of the HBR process.The percentage abundance of Candidatus Brocadia in the total bacterial community of the anoxic zone increased from 0 at Day 1 to 0.33%at Day 130 and then to 2.89%at Day 213.Based upon the activity of anammox bacteria,the removal of ammonia nitrogen(NH_(4)^(+)-N)in the anoxic zone was approximately 15%.This showed that the nitrogen transformation pathway was enhanced in the HBR system through partial anammox process in the anoxic zone.The final effluent contained 12 mg/L chemical oxygen demand(COD),0.662 mg/L NH_(4)^(+)-N,7.2 mg/L total nitrogen(TN),and 6 mg/L SS,indicating the effectiveness of the HBR process for treating real domestic wastewater. | Feng Hou Ting Zhang Yongzhen Peng Xiaoxin Cao Hongtao Pang Yanqing Shao Xianchun Lu Ju Yuan Xi Chen Jin Zhang | 2022 | Frontiers of Environmental Science & Engineering2022,16,3: | 2 |
