2Spinal Cord Stimulation Frequency Influences the Hemodynamic Response in Patients with Disorders of Consciousness显示文摘Spinal cord stimulation(SCS) is a promising technique for treating disorders of consciousness(DOCs).However, differences in the spatio-temporal responsiveness of the brain under varied SCS parameters remain unclear. In this pilot study, functional near-infrared spectroscopy was used to measure the hemodynamic responses of 10 DOC patients to different SCS frequencies(5 Hz, 10 Hz, 50 Hz, 70 Hz, and 100 Hz). In the prefrontal cortex, a key area in consciousness circuits, we found significantly increased hemodynamic responses at 70 Hz and 100 Hz, and significantly different hemodynamic responses between 50 Hz and 70 Hz/100 Hz. In addition, the functional connectivity between prefrontal and occipital areas was significantly improved with SCS at 70 Hz. 3Recombinant human PDCD5 protein enhances chemosensitivities of hematologic malignancies显示文摘PDCD5 is a novel apoptosis-related gene. Overexpression of PDCD5 facilitates apoptosis in various tumor cells. Here, we investigated the roles of recombinant human PDCD5 (rhPDCD5) protein in chemosensitivities of hematologic malignancies. The rhPDCD5 increased the in vitro cytotoxicity of daunorubicin (DNR), adriamycin (ADM) and As2O3 in three hematologic tumor cell lines. In vivo studies showed that DNR combined with rhPDCD5 significantly suppressed tumor growth of U937 xenograft nude mice compared with DNR alone. 4Identification of serotonin 2A receptor as a novel HCV entry factor by a chemical biology strategy显示文摘Hepatitis C virus(HCV)is a leading cause of liver disease worldwide.Although several HCV protease/polymerase inhibitors were recently approved by U.S.FDA,the combination of antivirals targeting multiple processes of HCV lifecycle would optimize anti-HCV therapy and against potential drug-resistanee.Viral entry is an essential target step for antiviral development,but FDA-approved HCV entry inhibitor remains exclusive.Here we identify serotonin 2A receptor(5-HT2aR)is a HCV entry factor amendable to therapeutic intervention by a chemical biology strategy.The silencing of 5-HT2aR and clinically available 5-HT2aR antagonist suppress cell culture-derived HCV(HCVcc)in different liver cells and primary human hepatocytes at late endocytosis process.The mechanism is related to regulate the correct plasma membrane localization of claudin 1(CLDN1).Moreover,phenoxybenzamine(PBZ),an FDAapproved 5-HT2aR antagonist,inhibits all major HCV genotypes in vitro and displays synergy in combination with clinical used anti-HCV drugs.The impact of PBZ on HCV genotype 2a is documented in immune-competent humanized transgenic mice.Our results not only expand the understanding of HCV entry,but also present a promising target for the invention of HCV entry inhibitor.Lin Cao Jizheng Chen Yaxin Wang Yuting Yang Jie Qing Zihe Rao Xinwen Chen Zhiyong Lou 2019Protein & Cell2019,10,3:2

6Wellbore drift flow relation suitable for full flow pattern domain and full dip range显示文摘Aiming at the simulation of multi-phase flow in the wellbore during the processes of gas kick and well killing of complex-structure wells(e.g.,directional wells,extended reach wells,etc.),a database including 3561 groups of experimental data from 32 different data sources is established.Considering the effects of fluid viscosity,pipe size,interfacial tension,fluid density,pipe inclination and other factors on multi-phase flow parameters,a new gas-liquid two-phase drift flow relation suitable for the full flow pattern and full dip range is established.The distribution coefficient and gas drift velocity models with a pipe inclination range of-90°–90°are established by means of theoretical analysis and data-driven.Compared with three existing models,the proposed models have the highest prediction accuracy and most stable performance.Using a well killing case with the backpressure method in the field,the applicability of the proposed model under the flow conditions with a pipe inclination range of-90°–80°is verified.The errors of the calculated shut in casing pressure,initial back casing pressure and casing pressure when adjusting the displacement are 2.58%,3.43%,5.35%,respectively.The calculated results of the model are in good agreement with the field backpressure data.LOU Wenqiang WANG Zhiyuan LI Pengfei SUN Xiaohui SUN Baojiang LIU Yaxin SUN Dalin 2022Petroleum Exploration and Development2022,49,3:1

14Dingxin recipe alleviates atherosclerosis injury in Apo E-knockout mice via downregulation of visfatin expression and inhibition of the visfatin-induced inflammatory response显示文摘OBJECTIVE:To further elucidate the mechanism underlying the anti-atherosclerotic effect of Dingxin recipe(DXR).METHODS:Fifty 6-week-old male Apo E^-/-mice were randomly divided into the following groups:model,simvastatin(5 mg·kg^-1·d^-1),DXR low-dose(9.30 g·kg^-1·d^-1),DXR middle-dose(18.59 g·kg^-1·d^-1)and DXR high-dose(37.18 g·kg^-1·d^-1)(n=10).Ten male C57BL/6J mice were used as the control group.All Apo E^-/-mice were fed a high-fat diet(HFD)and the control mice received a common diet.After HFD for 12 weeks,the mice were treated with DXR or simvastatin for another 12 weeks.The expression of inflammatory cytokines and visfatin was determined in serum and atherosclerotic lesions by enzyme-linked immunosorbent assay.Visfatin expression was also assessed in aortic atherosclerotic plaques.Cultured vessel endothelial cells(VECs)were pretreated with DXR sera prior to visfatin.The effects of DXR were analyzed to elucidate its protective mechanism against visfatin-induced inflammation in VECs.RESULTS:DXR regulated blood lipids and reduced tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),intercellular adhesion molecules-1(ICAM-1),vascular cell adhesion molecule-1(VCAM-1)and visfatin expression in Apo E^-/-mice,particularly at the higher doses.The areas of atherosclerotic lesions in the DXR groups were significantly smaller than those in the model group.DXR alleviated visfatin-induced VEC injury via downregulation of TNF-α,IL-6,ICAM-1 and VCAM-1 through mitogen-activated protein kinase pathways.CONCLUSION:DXR alleviated atherosclerosis injury via downregulation of visfatin expression and inhibition of the visfatin-induced inflammatory response in VECs.Cui Xiaobing Huang Zhiyong Lou Linjie Cheng Saibo Zhang Yu Zhang Yaxin Jia Yuhua Zhou Fenghua 2020Journal of Traditional Chinese Medicine2020,40,6:1
