| 4 | HBB-deficient Macaca fascicularis monkey presents with human β-thalassemia显示文摘Dear Editor,β-Thalassemia is a common severe genetic disease caused by mutations in HBB and affects approximately 1.5% of the global population (Origa, 2017). In southern China, the carrier rate of β-thalassemia is as high as 6.43%, creating a high socio-economic burden (Xiong et al., 2010). In adult humans, there are three types of hemoglobin: HbA1 (~97%), HbA2 (~2%) and HbF (~1%). HbA1 (α2β2) is composed of two a-globin and two β-globi n sub units en coded by HBA and HBB, respectively;HbF (α2β2)is made up of two α-globin subunits and two β-globin sub units en coded by HBG. Mutations in the coding region or regulatory region of HBB are involved in β-thalassemia pathogenesis. Except for some rare dominant mutations, most HBB mutations are recessive (Origa, 2017). Depending on the mutation type, the β-globin level will either be reduced or completely depleted, resulting in α-globin accumulation and precipitation. These α-globin precipitates lead to red blood cell death, resulting in anemia and tissue damage, and even death in thalassemia major patients. Blood transfusions can help slow disease progression but lead to iron overload, ultimately resulting in iron toxicity. Bone marrow transfer is the only cure in the clinic and is available only to a small percentage of patients with human leukocyte antigervmatched donors. Recently, gene therapy and gene editing therapy have shown great promise in curing β-thalassemia (Glaser et al., 2015;Thompson et al., 2018). However, no appropriate animal models are available for evaluating the safety and efficacy of such advanced therapeutic strategies in vivo.β-thalassemia mice are the sole animal model available for research. However, substantial differences have been reported between the types and expressi on patter ns of human and mouse globins (McColl and Vadolas, 2016). Moreover, mice contain no fetal globin gene equivalent, and homozygous mutations of HBB in mouse for early models of β-thalassemia major or Cooley anemia are all embryonic lethal (Huo et al., 2009). Recently, significant phenotype and physiology differences have been reported between SIRT6- null mice and the non-human primate model (Zhang et al., 2018). | 7 | Diagnostic Accuracy of Three-Dimensional Whole-Heart Magnetic Resonance Angiography to Detect Coronary Artery Disease with Invasive Coronary Angiography as a Reference:A Meta-Analysis显示文摘Objective:We aimed to evaluate the diagnostic performance of three-dimensional whole-heart magnetic resonance coronary angiography(MRCA)in detecting coronary artery disease(CAD)with invasive coronary angiography as the reference standard.Methods:We searched PubMed and Embase for studies evaluating the diagnostic performance of three-dimensional whole-heart MRCA for the diagnosis of CAD with invasive coronary angiography as the reference standard.The bivariate mixed-effects regression model was applied to synthesize available data.The clinical utility of whole-heart MRCA was calculated by the posttest probability based on Bayes’s theorem.Results:Eighteen studies were included,of which 16 provided data at the artery level.Patient-based analysis revealed a pooled sensitivity of 0.90(95%confi dence interval[CI]0.87–0.93)and specifi city of 0.79(95%CI 0.73–0.84),while the pooled estimates were 0.86(95%CI 0.82–0.89)and 0.89(95%CI 0.84–0.92),respectively,at the artery level.The areas under the summary receiver operating characteristic curve were 0.93(95%CI 0.90–0.95)and 0.92(95%CI 0.90–0.94)at the patient and artery levels,respectively.With a pretest probability of 50%,the patients’posttest probabilities of CAD were 81%for positive results and 11%for negative results.Conclusions:Whole-heart MRCA can be an alternative noninvasive method for diagnosis and assessment of CAD. | Shiqin Yu Chen Cui Minjie Lu Shihua Zhao | 2020 | Cardiovascular Innovations and Applications2020,,1: | 2 |

| 16 | Numerical simulation and data assimilation of the water-energy cycle over semiarid northeastern China显示文摘The default fractional vegetation cover and terrain height were replaced by the estimated fractional vegetation cover, which was calculated by the Normalized Difference Vegetation Index(NDVI) of Earth Observing System Moderate-Resolution Imaging Spectroradiometer(EOS-MODIS) and the Digital Elevation Model of the Shuttle Radar Topography Mission(SRTM) system. The near-surface meteorological elements over northeastern China were assimilated into the three-dimensional variational data assimilation system(3DVar) module in the Weather Research and Forecasting(WRF) model. The structure and daily variations of air temperature, humidity, wind and energy fields over northeastern China were simulated using the WRF model. Four groups of numerical experiments were performed, and the simulation results were analyzed of latent heat flux, sensible heat flux, and their relationships with changes in the surface energy flux due to soil moisture and precipitation over different surfaces. The simulations were compared with observations of the stations Tongyu, Naiman, Jinzhou, and Miyun from June to August, 2009. The results showed that the WRF model achieves high-quality simulations of the diurnal characteristics of the surface layer temperature, wind direction, net radiation, sensible heat flux, and latent heat flux over semiarid northeastern China in the summer. The simulated near-surface temperature, relative humidity, and wind speed were improved in the data assimilation case(Case 2) compared with control case(Case 1). The simulated sensible heat fluxes and surface heat fluxes were improved by the land surface parameterization case(Case 3) and the combined case(Case 4). The simulated temporal variations in soil moisture over the northeastern arid areas agree well with observations in Case 4, but the simulated precipitation should be improved in the WRF model. This study could improve the land surface parameters by utilizing remote sensing data and could further improve atmospheric elements with a data assimilation system. This work provides an effective attempt at combining multi-source data with different spatial and temporal scales into numerical simulations.
