1The role of bone marrow-derived cells in the origin of liver cancer revealed by single-cell sequencing显示文摘Objective:Epithelial cancers often originate from progenitor cells,while the origin of hepatocellular carcinoma(HCC)is still controversial.HCC,one of the deadliest cancers,is closely linked with liver injuries and chronic inflammation,which trigger massive infiltration of bone marrow-derived cells(BMDCs)during liver repair.Methods:To address the possible roles of BMDCs in HCC origination,we established a diethylnitrosamine(DEN)-induced HCC model in bone marrow transplanted mice.Immunohistochemistry and frozen tissue immunofluorescence were used to verify DENinduced HCC in the pathology of the disease.The cellular origin of DEN-induced HCC was further studied by single cell sequencing,single-cell nested PCR,and immunofluorescence-fluorescence in situ hybridization.Results:Studies by using single cell sequencing and biochemical analysis revealed that HCC cells in these mice were coming from donor mice BMDCs,and not from recipient mice.Furthermore,the copy numbers of mouse orthologs of several HCC-related genes previously reported in human HCC were also altered in our mouse model.DEN-induced HCCs exhibited a similar histological phenotype and genomic profile as human HCCs.Conclusions:These results suggested that BMDCs are an important origin of HCC,which provide important clues to HCC prevention,detection,and treatments.Lu Chen Xianfu Yi Piao Guo Hua Guo Ziye Chen Chunyu Hou Lisha Qi Yongrong Wang Chengwen Li Peng Liu Yucun Liu Yuanfu Xu Ning Zhang 2020Cancer Biology & Medicine2020,17,1:4
2Sensitivity of P300 auditory event-related potentials for assessing cognitive impairment in elderly type 2 diabetic patients显示文摘BACKGROUND:In previous studies, cognitive function in elderly type 2 diabetic patients was evaluated by psychometric tests. These studies have confirmed that P300 event-related potential is an objective way of assessing cognitive function. OBJECTIVE: To analyze the objectivity of P300 for assessment of cognitive function in elderly type 2 diabetic patients. DESIGN, TIME AND SETTING: This case-control experiment was performed at the Department of Endocrinology of the Fourth Affiliated Hospital, Guangxi Medical University from January 2004 to December 2006. PARTICIPANTS: Seventy-two patients (38 males and 34 females) with type 2 diabetes mellitus were enrolled in this study. The patients were divided according to those with diabetes alone (diabetes alone group) (n=38) and those with diabetes and cerebral ischemia (diabetes and cerebral ischemia group) (n=34). A further 31 healthy individuals (16 males and 15 females), who received health examinations over the same period, were included as normal controls (normal control group). METHODS: All subjects were assessed by Mini-Mental State Examination (MMSE). Abnormalities in cognitive functions were identified by analyzing the auditory P300 event-related potentials. MAIN OUTCOME MEASURES: Auditory event-related potentials and MMSE scores. Multiple linear regression analysis was conducted using the 'enter method' with the 72 elderly patients with type 2 diabetes mellitus. P3 latency, P3 amplitude and N2 latency served as dependent variables. Age, sex, education, course of the disease, glycosylated hemoglobin, and ischemic brain damage were used as independent variables. RESULTS: No significant difference in scores of MMSE was detected between the diabetes alone and normal control groups (P > 0.05). MMSE score was significantly lower in the diabetes and cerebral ischemia group (P < 0.01) than in the normal control group. N2 and P3 latencies of auditory event-related potential were significantly longer, and P3 amplitude was significantly lower in the diabetes alone and diabetes and cerebral ischemia groups (P < 0.01) than in the normal control group. N2 and P3 latencies were significantly longer in the diabetes and cerebral ischemia group than in the diabetes alone group (P < 0.01), but amplitude was not significantly different. N2 and P3 latencies were negatively correlated with MMSE score in elderly type 2 diabetic patients (r=–0.421, –0.604; both P < 0.01). P3 amplitude was positively related to the score of MMSE (r =0.517; P < 0.01). P3 latency was positively associated with age, course of disease, glycosylated hemoglobin and ischemic brain damage in elderly type 2 diabetic patients (t=2.186 to 3.490; all P < 0.05). P3 amplitude was negatively correlated with age, course of disease and glycosylated hemoglobin (t=–2.220, –2.491, and –2.024, respectively; all P < 0.05). N2 latency was positively correlated with age, course of disease and ischemic brain damage (t=2.946, 2.511, and 2.331, respectively; P < 0.05). CONCLUSION: The course of disease, glycosylated hemoglobin and ischemic brain damage are key influential factors for cognitive impairment in elderly type 2 diabetic patients. 6Lung Nodule Detection Based on YOLOv3 Deep Learning with Limited Datasets显示文摘The early symptom of lung tumor is always appeared as nodule on CT scans,among which 30%to 40%are malignant according to statistics studies.Therefore,early detection and classification of lung nodules are crucial to the treatment of lung cancer.With the increasing prevalence of lung cancer,large amount of CT images waiting for diagnosis are huge burdens to doctors who may missed or false detect abnormalities due to fatigue.Methods:In this study,we propose a novel lung nodule detection method based on YOLOv3 deep learning algorithm with only one preprocessing step is needed.In order to overcome the problem of less training data when starting a new study of Computer Aided Diagnosis(CAD),we firstly pick up a small number of diseased regions to simulate a limited datasets training procedure:5 nodule patterns are selected and deformed into 110 nodules by random geometric transformation before fusing into 10 normal lung CT images using Poisson image editing.According to the experimental results,the Poisson fusion method achieves a detection rate of about 65.24%for testing 100 new images.Secondly,419 slices from common database RIDER are used to train and test our YOLOv3 network.The time of lung nodule detection by YOLOv3 is shortened by 2–3 times compared with the mainstream algorithm,with the detection accuracy rate of 95.17%.Finally,the configuration of YOLOv3 is optimized by the learning data sets.The results show that YOLOv3 has the advantages of high speed and high accuracy in lung nodule detection,and it can access a large amount of CT image data within a short time to meet the huge demand of clinical practice.In addition,the use of Poisson image editing algorithms to generate data sets can reduce the need for raw training data and improve the training efficiency.Zhaohui Bu Xuejun Zhang Jianxiang Lu Huan Lao Chan Liang Xianfu Xu Yini Wei Hongjie Zeng 2022Molecular & Cellular Biomechanics2022,19,1:0
7Positivity and stability of timescale-type linear singular systems with time delays显示文摘This paper investigates positivity and stability problems of timescale-type delayed linear singular systems(LSSs).The existing results put an extremely strict constraint on the time-delay function.By introducing a novel function,this constraint is successfully removed,which generalizes the scope of the considered systems.Then,some necessary and sufficient criteria are proposed for the positivity of LSSs with bounded and infinite time-varying delays.Finally,the exponential(asymptotical)stability of LSSs with bounded(infinite)time-varying delays is analyzed.The derived results are also applicable to timescale-type differential-difference systems(DDSs).Compared with the existing stability criteria of DDSs with bounded time-varying delays,the strict limit on the parameter related to the convergence rate is eliminated.Hence,the conservatism of the existing results can be reduced.Moreover,when investigating stability of DDSs with infinite time-varying delays,this paper proposes a less conservative stability theorem.To illustrate the validity of the derived results,an example is presented regarding LSSs with bounded and infinite time-varying delays.Xiaodong LU Haitao LI Xianfu ZHANG 2022Science China(Information Sciences)2022,65,12:0
