| 1 | Protein trans-splicing based dual-vector delivery of the coagulation factor Ⅷ gene显示文摘A dual-vector system was explored for the delivery of the coagulation factor VIII gene,using intein-mediated protein trans-splicing as a means to produce intact functional factor VIII post-translationally.A pair of eukaryotic expression vectors,expressing Ssp DnaB intein-fused heavy and light chain genes of B-domain deleted factor VIII (BDD-FVIII),was constructed.With transient co-transfection of the two vectors into 293 and COS-7 cells,the culture supernatants contained (137±23) and (109±22) ng mL–1 spliced BDD-FVIII antigen with an activity of (1.05±0.16) and (0.79±0.23) IU mL–1 for 293 and COS-7 cells,respectively.The spliced BDD-FVIII was also detected in supernatants from a mixture of cells transfected with inteinfused heavy and light chain genes.The spliced BDD-FVIII protein bands from cell lysates were visualized by Western blotting.The data demonstrated that intein could be used to transfer the split factor VIII gene and provided valuable information on factor VIII gene delivery by dual-adeno-associated virus in hemophilia A gene therapy. | ZHU FuXiang,LIU ZeLong,CHI XiaoYan & QU HuiGe Life Science College of Ludong University,Yantai 264025,China | 2010 | Science China(Life Sciences)2010,53,6: | 27 |
| 2 | Variations in carbon isotope ratios of C_3 plants and distribution of C_4 plants along an altitudinal transect on the eastern slope of Mount Gongga显示文摘Variations in carbon isotopic ratios (δ 13C) of C3 plants and distribution of C4 plants were investigated along an altitudinal transect on the eastern slope of Mount Gongga, and the environmental effects on them were discussed. It is shown that plants with C4 photosynthetic pathway mainly occur at altitudes below 2100 m a.s.l., suggesting that the low summer temperature is responsible for the distributional pattern. In addition, δ 13C of C3 plants increases with elevation at the region above 2000 m a.s.l. with the characteristics of humid climate, and the increase rate in δ 13C for C3 plants is about 1.3‰ per kilometer. | 4 | Cross-linked hole transport layers for high-efficiency perovskite tandem solar cells显示文摘Perovskite tandem solar cells have recently received extensive attention due to their promise of achieving power conversion efficiency(PCE)beyond the limits of single-junction cells.However,their performance is still largely constrained by the widebandgap perovskite solar cells which show considerable open-circuit voltage(VOC)losses.Here,we increase the VOCand PCE of wide-bandgap perovskite solar cells by changing the hole transport layer(HTL)from commonly used poly(bis(4-phenyl)(2,4,6-trimethylphenyl)amine)(PTAA)to in-situ cross-linked small molecule N_(4),N_(4)′-di(naphthalen-1-yl)-N_(4),N_(4)′-bis(4-vinylphenyl)biphenyl-4,4′-diamine(VNPB).The stronger interaction and lower trap density at the VNPB/perovskite interface improve the PCE and stability of wide-bandgap perovskite solar cells.By using the cross-linked HTL for front wide-bandgap subcells,PCEs of 24.9%and 25.4%have been achieved in perovskite/perovskite and perovskite/silicon tandem solar cells,respectively.The results demonstrate that cross-linkable small molecules are promising for high-efficiency and cost-effective perovskite tandem photovoltaic devices. | Yurui Wang Shuai Gu Guoliang Liu Liping Zhang Zhou Liu Renxing Lin Ke Xiao Xin Luo Jianhua Shi Junling Du Fanying Meng Ludong Li Zhengxin Liu Hairen Tan | 2021 | Science China Chemistry2021,64,11: | 1 |
| 5 | Salvage treatments for prostate-specific antigen relapse of cT3N0M0 prostatic adenocarcinoma after radical prostatectomy combined with neoadjuvant androgen deprivation显示文摘Objective The aim of the study was to evaluate the efficiency of salvage treatments for prostate specific antigen(PSA)relapse of cT3N0M0 prostatic adenocarcinoma(PCa)after radical prostatectomy(RP)combined with neoadjuvant androgen deprivation(ADT).Methods A total of 332 patients with cT3N0M0 PCa were enrolled in the prospective study and received RP and pelvic lymph node dissection with neoadjuvant ADT for 3 months.All patients with PSA relapse were treated with salvage external beam radiation therapy(RT)and ADT for 6 months.Results The 5-year postoperative PSA relapse rate was 40.96%(136/332).The patients have been divided into the PSA relapse and PSA relapse-free groups in order to compare patient characteristics.The ratio of patients with Gleason score≥8 and positive surgical margin in the PSA relapse group were significantly higher than those of in the PSA relapse-free group(P=0.01).The mean duration between the start of operative treatment and PSA relapse was 31 months.Salvage treatment to all 136 PSA relapse patients led to favorable outcomes.PSA relapse was not observed after salvage treatment by the end of follow-up.The 5-year overall survival rates of the PSA relapse and PSA relapse-free groups were 94.9%and 93.9%,respectively.Conclusion In pursuit of curative treatment,our study showed that RP combined with neoadjuvant ADT is an aggressive multimodality strategy associated with lower PSA relapse and better survival outcomes for stage cT3N0M0 PCa patients.Patients with PSA relapse after RP may benefit from early aggressive salvage RT combined with short-term ADT. | Lufang Zhang Dongliang Pan Ludong Liu Yunjiang Zang Ningchen Li | 2020 | Oncology and Translational Medicine2020,6,6: | 0 |
| 6 | Stochastic bi-resonance in non-isothermal carbon monoxide catalytic oxidation system显示文摘The Pt(110)/CO+O2 system subject to reaction heat, heat conduction and radiative heat transfer is non-isothermal and its temperature varies in time and space. In this paper, taking support temperature (ST) as the control parameter, the effect of the ST fluctuations in the oscillatory dynamics of the non-isothermal Pt(110)/CO+O2 system is numerically studied. It is found that the ST fluctuations may induce stochastic oscillations and the oscillations exhibit stochastic bi-resonance (SBR) with the change of the strength or correlation time of the fluctuations. This result shows that the temperature fluctuations may enhance the chemical reaction oscillations. Moreover, the system can selectively and repeatedly employ the temperature fluctuations to enhance its reaction oscillations.
