| 1 | Efficacy and safety of the long-acting fusion inhibitor albuvirtide in antiretroviral-experienced adults with human immunodeficiency virus-1:interim analysis of the randomized,controlled,phase 3,non-inferiority TALENT study显示文摘Background:Albuvirtide is a once-weekly injectable human immunodeficiency virus(HIV)-1 fusion inhibitor.We present interim data for a phase 3 trial assessing the safety and efficacy of albuvirtide plus lopinavir-ritonavir in HIV-1-infected adults already treated with antiretroviral drugs.Methods:We carried out a 48-week,randomized,controlled,open-label non-inferiority trial at 12 sites in China.Adults on the World Health Organization(WHO)-recommended first-line treatment for>6 months with a plasma viral load>1000 copies/mL were enrolled and randomly assigned(1:1)to receive albuvirtide(once weekly)plus ritonavir-boosted lopinavir(ABT group)or the WHO-recommended second-line treatment(NRTI group).The primary endpoint was the proportion of patients with a plasma viral load below 50 copies/mL at 48 weeks.Non-inferiority was prespecified with a margin of 12%.Results:At the time of analysis,week 24 data were available for 83 and 92 patients,and week 48 data were available for 46 and 50 patients in the albuvirtide and NRTI groups,respectively.At 48 weeks,80.4%of patients in the ABT group and 66.0%of those in the NRTI group had HIV-1 RNA levels below 50 copies/mL,meeting the criteria for non-inferiority.For the per-protocol population,the superiority of albuvirtide over NRTI was demonstrated.The frequency of grade 3 to 4 adverse events was similar in the two groups;the most common adverse events were diarrhea,upper respiratory tract infections,and grade 3 to 4 increases in triglyceride concentration.Renal function was significantly more impaired at 12 weeks in the patients of the NRTI group who received tenofovir disoproxil fumarate than in those of the ABT group.Conclusions:The TALENT study is the first phase 3 trial of an injectable long-acting HIV drug.This interim analysis indicates that once-weekly albuvirtide in combination with ritonavir-boosted lopinavir is well tolerated and non-inferior to the WHO-recommended second-line regimen in patients with first-line treatment failure.Trial registration:ClinicalTrials.gov Identifier:NCT02369965;http://gffzz90ed6ce9d87f4798sox6x0bxp0w0w6fvp.ffgz.tsg.suse.edu.cn Clinical Trial Registry No.ChiCTR-TRC-14004276;http://gffzz62a03ee25e6f4a77hox6x0bxp0w0w6fvp.ffgz.tsg.suse.edu.cn/enindex.aspx. | Bin Su Cheng Yao Qing-Xia Zhao Wei-Ping Cai Min Wang Hong-Zhou Lu Yuan-Yuan Chen Li Liu Hui Wang Yun He Yu-Huang Zheng Ling-Hua Li Jin-Feng Chen Jian-Hua Yu Biao Zhu Min Zhao Yong-Tao Sun Wen-Hui Lun Wei Xia Li-Jun Sun Li-Li Dai Tai-Yi Jiang Mei-Xia Wang Qing-Shan Zheng Hai-Yan Peng Yao Wang Rong-Jian Lu Jian-Hua Hu Hui Xing Yi-Ming Shao Dong Xie Tong Zhang Fu-Jie Zhang Hao Wu TALENT Study Team | 2020 | Chinese Medical Journal2020,,24: | 10 |

| 3 | Increased Chondrocyte Apoptosis in Kashin-Beck Disease and Rats Induced by T-2 Toxin and Selenium Deficiency显示文摘Objective To investigate chondrocyte apoptosis and the expression of biochemical markers associated with apoptosis in Kashin-Beck disease(KBD) and in an established T-2 toxin-and selenium(Se) deficiency-induced rat model. Methods Cartilages were collected from the hand phalanges of five patients with KBD and five healthy children. Sprague-Dawley rats were administered a selenium-deficient diet for 4 weeks prior to T-2 toxin exposure. The apoptotic chondrocytes were observed by terminal deoxynucleotidyl transferase d UTP nick end labeling staining. Caspase-3, p53, Bcl-2, and Bax proteins in the cartilages were visualized by immunohistochemistry, their protein levels were determined by Western blotting, and m RNA levels were determined by real-time reverse transcription polymerase chain reaction. Results Increased chondrocyte apoptosis was observed in the cartilages of children with KBD. Increased apoptotic and caspase-3-stained cells were observed in the cartilages of rats fed with normal and Se-deficient diets plus T-2 toxin exposure compared to those in rats fed with normal and Se-deficient diets. Caspase-3, p53, and Bax proteins and m RNA levels were higher, whereas Bcl-2 levels were lower in rats fed with normal or Se-deficiency diets supplemented with T-2 toxin than the corresponding levels in rats fed with normal diet. | 4 | Recent Advancements on Photothermal Conversion and Antibacterial Applications over MXenes-Based Materials显示文摘The pernicious bacterial proliferation and emergence of super-resistant bacteria have already posed a great threat to public health,which drives researchers to develop antibiotic-free strategies to eradicate these fierce microbes.Although enormous achievements have already been achieved,it remains an arduous challenge to realize efficient sterilization to cut off the drug resistance generation.Recently,photothermal therapy(PTT)has emerged as a promising solution to efficiently damage the integrity of pathogenic bacteria based on hyperthermia beyond their tolerance.Until now,numerous photothermal agents have been studied for antimicrobial PTT.Among them,MXenes(a type of two-dimensional transition metal carbides or nitrides)are extensively investigated as one of the most promising candidates due to their high aspect ratio,atomic-thin thickness,excellent photothermal performance,low cytotoxicity,and ultrahigh dispersibility in aqueous systems.Besides,the enormous application scenarios using their antibacterial properties can be tailored via elaborated designs of MXenes-based materials.In this review,the synthetic approaches and textural properties of MXenes have been systematically presented first,and then the photothermal properties and sterilization mechanisms using MXenes-based materials are documented.Subsequently,recent progress in diverse fields making use of the photothermal and antibacterial performances of MXenes-based materials are well summarized to reveal the potential applications of these materials for various purposes,including in vitro and in vivo sterilization,solar water evaporation and purification,and flexible antibacterial fabrics.Last but not least,the current challenges and future perspectives are discussed to provide theoretical guidance for the fabrication of efficient antimicrobial systems using MXenes. | Shuyan Hao Hecheng Han Zhengyi Yang Mengting Chen Yanyan Jiang Guixia Lu Lun Dong Hongling Wen Hui Li Jiurong Liu Lili Wu Zhou Wang Fenglong Wang | 2022 | Nano-Micro Letters2022,14,11: | 4 |
| 5 | Assessment of circulating tumor DNA in cerebrospinal fluid by whole exome sequencing to detect genomic alterations of glioblastoma显示文摘Background:Cerebrospinal fluid(CSF)has been demonstrated as a better source of circulating tumor DNA(ctDNA)than plasma for brain tumors.However,it is unclear whether whole exome sequencing(WES)is qualified for detection of ctDNA in CSF.The aim of this study was to determine if assessment of ctDNA in CSF by WES is a feasible approach to detect genomic alterations of glioblastoma.Methods:CSFs of ten glioblastoma patients were collected pre-operatively at the Department of Neurosurgery,Sun Yat-sen University Cancer Center.ctDNA in CSF and genome DNA in the resected tumor were extracted and subjected to WES.The identified glioblastoma-associated mutations from ctDNA in CSF and genome DNA in the resected tumor were compared.Results:Due to the ctDNA in CSF was unqualified for exome sequencing for one patient,nine patients were included into the final analysis.More glioblastoma-associated mutations tended to be detected in CSF compared with the corresponding tumor tissue samples(3.56±0.75 vs.2.22±0.32,P=0.097),while the statistical significance was limited by the small sample size.The average mutation frequencies were similar in CSF and tumor tissue samples(74.1%±6.0%vs.73.8%±6.0%,P=0.924).The R132H mutation of isocitrate dehydrogenase 1 and the G34V mutation of H3 histone,family 3A(H3F3A)which had been reported in the pathological diagnoses were also detected from ctDNA in CSF by WES.Patients who received temozolomide chemotherapy previously or those whose tumor involved subventricular zone tended to harbor more mutations in their CSF.Conclusion:Assessment of ctDNA in CSF by WES is a feasible approach to detect genomic alterations of glioblastoma,which may provide useful information for the decision of treatment strategy. | Hao Duan Ji-Long Hu Zheng-He Chen Jue-Hui Li Zhen-Qiang He Zhen-Ning Wang Guan-Hua Zhang Xiao-Yu Guo Lun Liang Yong-Gao Mou | 2020 | Chinese Medical Journal2020,,12: | 3 |
| 6 | Atmospheric isoprene and monoterpenes in a typical urban area of Beijing: Pollution characterization, chemical reactivity and source identification显示文摘Continuous observation of isoprene, α-pinene and β-pinene was carried out in a typical urban area of Beijing from March 2014 to February 2015, using an AirmoV OC online analyzer. Based on the analysis of the ambient level and variation characteristics of isoprene, α-pinene and β-pinene, the chemical reactivity was studied, and their sources were identified. Results showed that the concentrations of isoprene, α-pinene and β-pinene in the urban area of Beijing were lower than those in richly vegetated areas; the concentrations of isoprene were at a moderate level compared with those of previous studies of Beijing. Concentrations of isoprene, α-pinene and β-pinene showed different seasonal, monthly, daily and diurnal variations, and all of the three species showed higher level at night than those in the daytime as a whole, the variations of isoprene, α-pinene and β-pinene mainly influenced by emission of sources, photochemical reaction, and meteorological parameters. Isoprene was the largest contributor to the total OFP values than α-pinene and β-pinene. α-Pinene was the largest contributor to the total SOAFP values than isoprene and β-pinene in autumn, while isoprene was the largest one in other seasons. Isoprene, α-pinene and β-pinene were derived mainly from biological sources; and α-pinene level were also affected by industrial sources. | 14 | Structure and species composition of tree stands on verges and slopes along a major highway in Hong Kong显示文摘Arboricultural research focusing on transport land use was lacking in Hong Kong.Some highway slopes were registered in the Systematic Identification of Maintenance Responsibility of Slopes in the Territory(SIMAR),abbreviated as SIMAR slopes.We aimed to analyze patterns in the structure and species composition of the tree stock along a highway in Hong Kong and examined how a slope registration system could help explain the characteristics of urban forests.The 53 slopes and 52 verges along San Tin Highway,Hong Kong were randomly selected.The trees on each slope and verge were collectively sampled as a tree stand.Six variables,namely tree abundance,species richness,maximum tree height,Shannon-Wiener diversity,Simpson's dominance,and Pielou's evenness were measured for each stand.In addition,a limited visual tree risk assessment was performed.The 7,209 trees in 23 species were recorded.Species richness was low,ranging from one to eight species per stand.SIMAR and non-SIMAR slopes were compared.SIMAR slopes had significantly higher species richness,diversity,evenness but lower dominance,with mean difference of 1.41 species,0.17,0.17 and-0.28respectively.SIMAR slopes were associated with lower tree risk rating.When training regression models,boosting as an ensemble method arbitrarily raised the explanatory power and the predictive accuracy of some models.Slope height,length,angle and area could be significant predictors of the biodiversity-related variables.Future research can sample more habitat characteristics related to the structure and species composition of slopes and verges which were important components of urban forestry. | Louis Shing Him Lee Hao Zhang Kathy Tze Kwun Ng Shun Cheong Lo Alan Siu Lun Yu | 2023 | Journal of Forestry Research2023,34,5: | 0 |
