1Complete chloroplast genome sequence of Magnolia grandiflora and comparative analysis with related species显示文摘Magnolia grandiflora is an important medicinal,ornamental and horticultural plant species.The chloroplast(cp) genome of M.grandiflora was sequenced using a 454 sequencing platform and the genome structure was compared with other related species.The complete cp genome of M.grandiflora was 159623 bp in length and contained a pair of inverted repeats(IR) of 26563 bp separated by large and small single copy(LSC,SSC) regions of 87757 and 18740 bp,respectively.A total of 129 genes were successfully annotated,18 of which included introns.The identity,number and GC content of M.grandiflora cp genes were similar to those of other Magnoliaceae species genomes.Analysis revealed 218 simple sequence repeat(SSR) loci,most composed of A or T,contributing to a bias in base composition.The types and abundances of repeat units in Magnoliaceae species were relatively conserved and these loci will be useful for developing M.grandiflora cp genome vectors.In addition,results indicated that the cp genome size in Magnoliaceae species and the position of the IR border were closely related to the length of the ycf1 gene.Phylogenetic analyses based on 66 shared genes from 30 species using maximum parsimony(MP) and maximum likelihood(ML) methods provided strong support for the phylogenetic position of Magnolia.The availability of the complete cp genome sequence of M.grandiflora provides valuable information for breeding of desirable varieties,cp genetic engineering,developing useful molecular markers and phylogenetic analyses in Magnoliaceae.LI XiWen GAO HuanHuan WANG YiTao SONG JingYuan HENRY Robert WU HeZhen HU ZhiGang YAO Hui LUO HongMei LUO Kun PAN HongLin CHEN ShiLin 2013Science China(Life Sciences)2013,56,2:17
2Metal Sealing Performance of Subsea X-tree Wellhead Connector Sealer显示文摘The metal sealing performance of subsea X-tree wellhead connectors is crucial for the safety and reliability of subsea X-trees. In order to establish the theoretical relation between metal sealing ring's contact stress and its structural parameters and working pressure, a mechanical analysis method for double-cone sealing of high pressure vessels is applied in analyzing the metal sealing ring under the condition of preload and operation. As a result, the formula of the unit sealing load for the metal sealing ring under operation with residual preload is shown in this paper, which ensures that the metal sealing ring has an excellent sealing effect and can prevent the metal sealing ring from yielding. Besides, while analyzing the sealing process of the metal sealing ring, the change rule of contact stress and working pressure is concluded here, putting forward that the structural parameters of the metal sealing ring are the major factors affecting the change rule. Finally, the analytical solution through theoretical analysis is compared with the simulation result through finite element analysis in a force feedback experiment, and both are consistent with each other, which fully verifies for the design and calculation theory on metal sealing ring's contact stress and its structural parameters and working pressure deduced in this paper. 3Symmetry breaking of photonic spin-orbit interactions in metasurfaces显示文摘Spin-orbit optical phenomena pertain to the wider class of electromagnetic effects originating from the interaction of the photon spin with the spatial structure and propagation characteristics of an optical wave,mediated by suitable optical media.There are many emerging photonic applications of spin-orbit interactions(SOI)of light,such as control of the optical wave propagation via the spin,enhanced optical manipulation,and generation of structured optical fields.Unfortunately,current applications are based on symmetric SOI,that is,the behaviours of polarized photons with two opposite spins are opposite,leading to the limit of spin-based multiplexers.The symmetry of SOI can be broken in our proposed metasurfaces,consisting of spatially varying birefringence,which can arbitrarily and independently build SOI for two opposite spins without reduction of optical energy usage.We obtain three kinds of dual-functional metasurfaces at visible and infrared wavelengths with high efficiency.Our concept of generation of asymmetric SOI for two spins,using anisotropic metasurfaces,will open new degrees of freedoms for building new types of spin-controlled multifunctional shared-aperture devices for the generation of complex structured optical fields.Fei Zhang Mingbo Pu Jun Luo Honglin Yu Xiangang Luo 2017光电工程2017,44,3:8
4Autoantigen Microarray for High-throughput Autoantibody Profiling in Systemic Lupus Erythematosus显示文摘Systemic lupus erythematosus(SLE) is a complex autoimmune disease characterized by the production of autoantibodies to a broad range of self-antigens. Profiling the autoantibody repertoire using array-based technology has emerged as a powerful tool for the identification of biomarkers in SLE and other autoimmune diseases. Proteomic microarray has the capacity to hold large number of self-antigens on a solid surface and serve as a high-throughput screening method for the determination of autoantibody specificities. The autoantigen arrays carrying a wide variety of self-antigens, such as cell nuclear components(nucleic acids and associated proteins), cytoplasmic proteins, phospholipid proteins, cell matrix proteins, mucosal/secreted proteins, glomeruli, and other tissue-specific proteins, have been used for screening of autoantibody specificities associated with different manifestations of SLE. Arrays containing synthetic peptides and molecular modified proteins are also being utilized for identification of autoantibodies targeting to special antigenic epitopes. Different isotypes of autoantibodies, including Ig G, Ig M, Ig A, and Ig E, as well as other Ig subtypes, can be detected simultaneously with multi-color labeled secondary antibodies. 5Simultaneous determination of brominated phenols in soils显示文摘Brominated phenols(BPs),a widely used group of emerging flame retardants,are important environmental contaminants and exhibit endocrine disrupting potential.Method for simultaneous determination of tetrabromobisphenol A(TBBPA),tribromophenol(TBP),dibromophenols(DBPs)and monobromophenols(MBPs)in soils using gas chromatography-mass spectrometry analysis(GC/MS)was successfully developed.Cleanup methods for soil extracts including several solid-phase extraction cartridges and diferent elution solvents were compared and optimized.Florisil cartridge with dichloromethane as the elution reagent was selected for sample cleanup owing to its high and reproducible recoveries of the target analytes in soils.Derivatization conditions were tested and the optimal conditions were obtained with 20μL silylation reagent at room temperature.The chromatographic separation was optimized with diferent columns and DB-XLB column was selected for its excellent separation of the analytes.The limits of detection for the target compounds were from 0.04 to 0.19 ng/g.Mean recoveries of the compounds from spiked soils exceeded 84%with a good reproducibility,excepting that the recovery of 2-bromophenol was relatively poor(lower than 55%)due to its instability.The developed method was applied to the determination of the BPs in the soils collected from e-waste sites.The contents of BPs in the soils were at ng/g levels with TBBPA and TBP the most frequently detected.To our knowledge,this is the first report for the simultaneous determination of TBBPA,TBP,DBPs and MBPs in soils.Wei Han Sen Wang Honglin Huang Lei Luo Shuzhen Zhang 2013Journal of Environmental Sciences2013,25,11:5

8Layer-by-Layer Assembled Bacterial Cellulose/Graphene Oxide Hydrogels with Extremely Enhanced Mechanical Properties显示文摘Uniform dispersion of two-dimensional(2 D) graphene materials in polymer matrices remains challenging. In this work, a novel layer-by-layer assembly strategy was developed to prepare a sophisticated nanostructure with highly dispersed 2 D graphene oxide in a three-dimensional matrix consisting of onedimensional bacterial cellulose(BC) nanofibers. This method is a breakthrough, with respect to the conventional static culture method for BC that involves multiple in situ layer-by-layer assembly steps at the interface between previously grown BC and the culture medium. In the as-prepared BC/GO nanocomposites, the GO nanosheets are mechanically bundled and chemically bonded with BC nanofibers via hydrogen bonding,forming an intriguing nanostructure. The sophisticated nanostructure of the BC/GO leads to greatly enhanced mechanical properties compared to those of bare BC. 12Human umbilical cord mesenchymal stem cells for psoriasis:a phase 1/2a,single-arm study显示文摘Psoriasis is a common,chronic immune-mediated systemic disease that had no effective and durable treatment.Mesenchymal stem cells(MSCs)have immunomodulatory properties.Therefore,we performed a phase 1/2a,single-arm clinical trial to evaluate the safety and efficacy of human umbilical cord-derived MSCs(UMSCs)in the treatment of psoriasis and to preliminarily explore the possible mechanisms.Seventeen patients with psoriasis were enrolled and received UMsC infusions.Adverse events,laboratory parameters,PASl,and PGA were analyzed.We did not observe obvious side effects during the treatment and 6-month follow-up.A total of 47.1%(8/17)of the psoriasis patients had at least 40%improvement in the PASl score,and 17.6%(3/17)had no sign of disease or minimal disease based on the PGA score.And the efficiency was 25%(2/8)for males and 66.7%(6/9)for females.After UMSC transplantation(UMSCT),the frequencies of Tregs and CD4^(+)memory T cells were significantly increased,and the frequencies of T helper(Th)17 and CD4^(+)naive T cells were significantly decreased in peripheral blood(PB)of psoriasis patients.And all responders showed significant increases in Tregs and CD4^(+)memory T cells,and significant decreases in Th17 cells and serum IL-17 level after UMsCT.And baseline level of Tregs in responders were significantly lower than those in nonresponders.In conclusion,allogeneic UMSCT is safe and partially effective in psoriasis patients,and level of Tregs may be used as a potent biomarker to predict the clinical efficacy of UMSCT.Trial registration Clinical Trials NCT03765957.Lamei Cheng Siqi Wang Cong Peng Xiao Zou Chao Yang Hua Mei Chuang Li Xian Su Na Xiao Qi Ouyang Mi Zhang Qiaolin Wang Yan Luo Minxue Shen Qun Qin Honglin Wang Wu Zhu Guangxiu Lu Ge Lin Yehong Kuang Xiang Chen 2022Signal Transduction and Targeted Therapy2022,7,9:2
