3晚期非小细胞肺癌免疫检查点抑制剂新型生物标志物:血液PD-L1动态检测显示文摘背景免疫检查点抑制剂(immune checkpoint inhibitors,ICIs)具有良好疗效已成为恶性肿瘤治疗领域备受瞩目的翘楚。然而,仅有小部分患者获得长期临床获益。如何选择生物标志物来筛查ICIs治疗优势人群仍是一大热点。方法收集51例晚期恶性肿瘤患者的肿瘤组织和配对血样本,进行PD-L1表达的相关性分析。40例晚期非小细胞肺癌(non-small cell lung cancer,NSCLC)患者接受ICIs治疗,2个月时检测血液PD-L1(bloodPD-L1,bPD-L1)表达的动态变化,包括PD-L1 mRNA、外泌体PD-L1(exosomalPD-L1,exoPD-L1)蛋白和可溶性PD-L1(solublePD-L1,sPD-L1)。应用R软件计算最佳截断值,用于预测最佳客观缓解(best objective response,b OR)、无进展生存期(progression-free survival,PFS)和总生存期(overall survival,OS)。结果51例不同恶性肿瘤患者中,组织PD-L1(tissuePD-L1,tPD-L1)表达阳性患者的PD-L1mRNA表达水平显著升高。40例接受ICIs治疗的晚期NSCLC患者中,bPD-L1 mRNA上调≥2.04的患者b OR、PFS和OS更佳;21例接受ICIs治疗的晚期NSCLC患者中,exoPD-L1上调≥1.86的患者临床获益更大。sPD-L1动态变化与ICIs疗效无显著相关性。PD-L1 mRNA和exoPD-L1联合检测更有利于预测ICIs治疗疗效。结论bPD-L1表达与tPD-L1表达存在正相关;晚期NSCLC患者ICIs治疗初期PD-L1 mRNA和(或)exoPD-L1表达升高,可作为评估临床获益的新型生物标志物。Qiao YANG Mingjing CHEN Jiaoyang GU Kai NIU Xianlan ZHAO Linpeng ZHENG Zihan XU Yongxin YU Feng LI Lingxin MENG Zhengtang CHEN Wenlei ZHUO Luping ZHANG Jianguo SUN 杨峤(译) 孙建国(译) 2021中国肺癌杂志2021,24,12:10
4Diagnostic value of fluid attenuated inversion recovery versus T_2-weighted image in diffuse axonal injury显示文摘BACKGROUND:At present, the most common examination modality for diffuse axonal injury (DAI) is CT or MRI. However, both methods exhibit low sensitivity in the diagnosis of DAI lesions. OBJECTIVE: To investigate the value of fluid attenuated inversion recovery (FLAIR) in the clinical diagnosis of DAI, and to compare with T2-weighted images. DESIGN, TIME AND SETTING: This prospective study was based on imaging analysis, and was performed in the First Affiliated Hospital of Chongqing Medical University (Chongqing, China) between October 2002 and April 2004. PARTICIPANTS: Sixty-three patients with craniocerebral injury were admitted to the Department of Neurosurgery at the First Affiliated Hospital of Chongqing Medical University, including 50 males and 13 females. The patients were included in the experiment and were divided into DAI (n=24) and non-DAI (n=39) groups, according to the emergent CT findings and clinical manifestations. METHODS: Both groups underwent MR examinations, including axial and sagittal T1 weighted images (TR = 450 ms, TE = 8–9 ms), T2-weighted images (TR = 3 600 ms, TE = 100 ms), and FLAIR (TR = 10 000 ms, TI = 2 500 ms, TE = 40 ms), 8-mm thick and 2-mm wide, using a GE Sigma MRI device. MAIN OUTCOME MEASURES: The DAI diagnostic rate and lesion-detecting rate of T2-weighted images and FLAIR were determined. RESULTS: All 63 patients were included in the final analysis. The DAI diagnosis rates of FLAIR and T2-weighted images were 88% (21/24) and 62% (15/24), respectively, of which the difference was statistically significant (P < 0.05). T2-weighted images and FLAIR detected lesions located in the gray matter-white matter junction in parasagittal areas, the corpus callosum, deep periventricular white matter, basal ganglia, internal capsule, hippocampus, cerebellum, and brain stem, with a detailed amount of 123 and 256, respectively. FLAIR was significantly greater than T2-weighted images (P < 0.01). 5Detailed resume of RNA m^(6)A demethylases显示文摘N6-Methyladenosine(m^(6)A) is the most abundant internal modification in eukaryotic mRNA,playing critical role in various bioprocesses. Like other epigenetic modifications, m^(6)A modification can be catalyzed by the methyltransferase complex and erased dynamically to maintain cells homeostasis. Up to now, only two m^(6)A demethylases have been reported, fat mass and obesity-associated protein(FTO)and alkylation protein AlkB homolog 5(ALKBH5), involving in a wide range of mRNA biological progress, including mRNA shearing, export, metabolism and stability. Furthermore, they participate in many significantly biological signaling pathway, and contribute to the progress and development of cancer along with other diseases. 6Japanese Encephalitis Virus NS1' Protein Antagonizes Interferon Beta Production显示文摘Japanese encephalitis virus(JEV) is a mosquito-borne virus and the major cause of viral encephalitis in Asia. NS1', a52-amino acid C-terminal extension of NS1, is generated with a-1 programmed ribosomal frameshift and is only present in members of the Japanese encephalitis serogroup of flaviviruses. Previous studies demonstrated that NS1' plays a vital role in virulence, but the mechanism is unclear. In this study, an NS1' defected(rG66A) virus was generated. We found that rG66A virus was less virulent than its parent virus(pSA14) in wild-type mice. However, similar mortality caused by the two viruses was observed in an IFNAR knockout mouse model. Moreover, we found that rG66A virus induced a greater type Ⅰ interferon(IFN) response than that by pSA14, and JEV NS1' significantly inhibited the production of IFN-b and IFN-stimulated genes. 7Neuroprotective effect of heat shock protein 70 Inhibition of Tau protein expression显示文摘BACKGROUND: Previous studies have shown that heat shock protein 70 (HSP70) hasneuroprotective effects by decreasing phosphorylation of Tau protein, thereby reducing theexpression of Tau protein and proper aggregation.OBJECTIVE: To observe and verify expressional changes of HSP70 and Tau in retinal ganglioncells following stretch injury to the right optic nerve in rats, and to determine the effect of heat stresspretreatment on HSP70 and Tau protein expressions.DESIGN, TIME AND SETTING: A randomized, controlled, animal experiment was performed at theLaboratory of Neurology Research Institute of the First Affiliated Hospital of Chongqing MedicalUniversity from March to June 2006.MATERIALS: Instant SABC immunohistochemistry kit, as well as mouse anti-HSP70 and rabbitanti-Tau polyclonal antibodies, were purchased from Wuhan Boster Bioengineering Limited, China.METHODS: A total of 57 male, Wistar rats were randomly assigned to 4 groups: control (n = 3);150-180 g stretch force was induced in the right optic nerves in stretch-only group (n = 18) toestablish optic nerve stretch injury model; heat stress was applied to 18 animals in heat-stresstreatment group; 18 rats in the heat-stress pretreatment plus stretch group were subjected toidentical stretch injury as stretch-only group after 24-hour heat-stress pretreatment. According tosacrifice time, the groups were assigned to 6 subgroups at different time points of 4, 8, and 16 hours,and 1,3, and 5 days, with 3 rats in each subgroup. No treatment was performed in the control groupexcept anesthesia.MAIN OUTCOME MEASURES: Morphological changes of optic nerves and retinal ganglion cellsfollowing stretch injury were observed by light microscopy following hematoxylin-eosin staining.HSP70 and Tau protein expression levels were observed in retinal ganglial cells from each groupusing im munohistochemistry.RESULTS: (1) Compared with the control group, morphological axonal and retinal ganglial cellchanges, as well as a decreased number of retinal ganglial cells, were identified in the stretch-onlygroup (P< 0.01). Pathological damage in optical nerve and retinal ganglial cells were not remarkablein the heat-stress pretreatment plus stretch group, with no statistical difference in the number ofretinal ganglial cells compared with the control group (P > 0.05). (2) Compared with the control groupsignificantly increased HSP70 expression in retinal ganglial cells occurred in the stretch-only,heat-stress treatment, and heat-stress pretreatment plus stretch groups (P < 0.05 or P < 0.01). Thepeak of HSP70 expression was earlier in the heat-stress pretreatment plus stretch group comparedwith the stretch-only and heat-stress treatment groups, and was expressed over a longer period oftime compared with the heat-stress treatment group. Compared with the control group, Tauexpression in the retinal ganglial cells rapidly increased 4-16 hours following stretch injury in thestretch-only group (P < 0.05 or P < 0.01), and obviously decreased in the heat-stress pretreatmentplus stretch group (P < 0.05 or P < 0.01).CONCLUSION: Tau expression increased following stretch injury, with an earlier expression peakthan HSP70, which indicated that stretch injury-induced HSP70 expression was not strong or quickenough to sufficiently protect the nerve. 9High-efficient removal of tetracycline in water via porous magnetic Ce/Fe photocomposite under visible light显示文摘Tetracycline is a typical antibiotic commonly used in various industries which is eco-toxic and quickly causes bacterial resistance.Therefo re,studying the efficient removal of tetracycline is necessary to protect the water environment.Herein,a novel Ce/Fe nanoparticle composite(1CCFO)was prepared by the sol-gel method and its removal effects of tetracycline under visible light were performed.The relationship between physicochemical properties of catalyst and photocatalytic degradation effects of tetracycline was analyzed based on a series of characterizations data such as X-ray diffraction(XRD),Raman spectro scopy,a vibrating sample magnetometer(VSM),scanning electron microscopy(SEM),Brunauer-Emmett-Teller(BET)method,X-ray photoelectron spectroscopy(XPS),and ultraviolet-visible(UV-Vis)spectroscopy.The results show that Ce/Fe photocatalyst possesses a large specific surface area,good visible light response,abundant oxygen vacancies and excellent redox performance,exhibiting good adsorption capacity,remarkable catalytic performance and stability.The optimal conditions for tetracycline removal were explored through orthogonal experiments.About 88%of tetracycline can be photodegraded in 1 h under optimal conditions.The possible decomposition pathways,main reactive oxygen species and suitable mechanism of the photocatalytic system were studied by liquid chromatography-mass spectro metry(LC-MS),an electron spin resonance(ESR)spectrometer and free radical quenching experiments.The results show that 1CCFO has an efficient degradation effect on tetracycline under visible light,which provides a feasible method to improve the performance of 1CCFO.Weihang Han Jingxuan Shou Yifan Yang Liangchen Chen Luping Zhang Yutong Chen Xuewei Tu Dan Jin Shijie Zhang Yurong Chang Hui Zheng 2023Journal of Rare Earths2023,41,10:0
