1The uptake diversity of soil nitrogen nutrients by main plant species in Kobresia humilis alpine meadow on the Qinghai-Tibet Plateau显示文摘We studied the uptake of ammonium,nitrate,and a variety of amino acids by alpine plant species in the Kobresia humilis alpine meadow ecosystem in situ.We examined the extent of niche separation in uptake of N source by different plant species in alpine communities,and investigated the contribution of symbiotically fixed N to the total N in alpine meadow.The results are(1) δ 15 N natural abundance values of 13 plant species lie between 2.680‰ and 5.169‰,and the scope is 7.849‰.(2) Leguminous plants,such as Trigonella ruthenica,Gueldenstaedtia diversiffolia,and Oxytyopis ochrocephala,and non-leguminous plant Gentiana straminea uptake low amounts of 15 N labeled ammonium,nitrate,glycine or aspartate in soil.(3) As far as the plant uptake of organic N is concerned,Kobresia humilis,Poa pratensis,and Gentiuna spathutifolta can effectively uptake organic nitrogen,and about 37%-40% of the nitrogen of these species comes from soil organic nitrogen sources(such as glycine and aspartate).Stipa aliena can effectively uptake nitrate,and 60% of its nitrogen comes from soil nitrate.Potentilla anserina,Poa pratensis,and Thalictrum alpinum can effectively absorb ammonium in comparason to other plant species in the meadow,and about 25%-27% of the nitrogen in these plants comes from soil ammonium.(4) The contribution of leguminous fixed N to total N is 7.48%-9.26% in Kobresia humilis alpine meadow.(5) These data show many plant species of alpine meadow may effectively utilize dissolved organic nitrogen such as amino acids,and these plants have diverse ways to uptake soil nitrogen in alpine meadows.Based on the results we can partly explain why there are abundant biodiversities and how plants at alpine habitat utilize the limited soil N sources.WANG WenYing MA YongGui XU Jin WANG HuiChun ZHU JinFu ZHOU HuaKun 2012Science China Earth Sciences2012,55,10:13
2Functional identification of the terpene synthase family involved in diterpenoid alkaloids biosynthesis in Aconitum carmichaelii显示文摘Aconitum carmichaelii is a high-value medicinal herb widely used across China,Japan,and other Asian countries.Aconitine-type diterpene alkaloids(DAs)are the characteristic compounds in Aconitum.Although six transcriptomes,based on short-read next generation sequencing technology,have been reported from the Aconitum species,the terpene synthase(TPS)corresponding to DAs biosynthesis remains unidentified.We apply a combination of Pacbio isoform sequencing and RNA sequencing to provide a comprehensive view of the A.carmichaelii transcriptome.Nineteen TPSs and five alternative splicing isoforms belonging to TPS-b,TPS-c,and TPS-e/f subfamilies were identified.In vitro enzyme reaction analysis functional identified two sesqui-TPSs and twelve di TPSs.Seven of the TPS-c subfamily genes reacted with GGPP to produce the intermediate ent-copalyl diphosphate.Five Ac KSLs separately reacted with ent-CPP to produce ent-kaurene,ent-atiserene,and ent-13-epi-sandaracopimaradie:a new diterpene found in Aconitum.Ac TPSs gene expression in conjunction DAs content analysis in different tissues validated that ent-CPP is the sole precursor to all DAs biosynthesis,with Ac KSL1,Ac KSL2 s and Ac KSL3-1 responsible for C20 atisine and napelline type DAs biosynthesis,respectively.These data clarified the molecular basis for the C20-DAs biosynthetic pathway in A.carmichaelii and pave the way for further exploration of C19-DAs biosynthesis in the Aconitum species.Liuying Mao Baolong Jin Lingli Chen Mei Tian Rui Ma Biwei Yin Haiyan Zhang Juan Guo Jinfu Tang Tong Chen Changjiangsheng Lai Guanghong Cui Luqi Huang 2021Acta Pharmaceutica Sinica B2021,11,10:6
3Anomaly detection of hot components in gas turbine based on frequent pattern extraction显示文摘Hot components operate in a high-temperature and high-pressure environment. The occurrence of a fault in hot components leads to high economic losses. In general, exhaust gas temperature(EGT) is used to monitor the performance of hot components.However, during the early stages of a failure, the fault information is weak, and is simultaneously affected by various types of interference, such as the complex working conditions, ambient conditions, gradual performance degradation of the compressors and turbines, and noise. Additionally, inadequate effective information of the gas turbine also restricts the establishment of the detection model. To solve the above problems, this paper proposes an anomaly detection method based on frequent pattern extraction. A frequent pattern model(FPM) is applied to indicate the inherent regularity of change in EGT occurring from different types of interference. In this study, based on a genetic algorithm and support vector machine regression, the relationship model between the EGT and interference was tentatively built. The modeling accuracy was then further improved through the selection of the kernel function and training data. Experiments indicate that the optimal kernel function is linear and that the optimal training data should be balanced in addition to covering the appropriate range of operating conditions and ambient temperature. Furthermore, the thresholds based on the Pauta criterion that is automatically obtained during the modeling process, are used to determine whether hot components are operating abnormally. Moreover, the FPM is compared with the similarity theory, which demonstrates that the FPM can better suppress the effect of the component performance degradation and fuel heat value fluctuation. Finally, the effectiveness of the proposed method is validated on seven months of actual data obtained from a Titan130 gas turbine on an offshore oil platform. 11Classical swine fever virus NS5A protein antagonizes innate immune response by inhibiting the NF-κB signaling显示文摘The NS5A non-structural protein of classical swine fever virus(CSFV)is a multifunctional protein involved in viral genomic replication,protein translation,assembly of infectious virus particles,and regulation of cellular signaling pathways.Previous report showed that NS5A inhibited nuclear factor kappa B(NF-κB)signaling induced by poly(I:C);however,the mechanism involved has not been elucidated.Here,we reported that NS5A directly interacted with NF-κB essential modulator(NEMO),a regulatory subunit of the IκB kinase(IKK)complex,to inhibit the NF-κB signaling pathway.Further investigations showed that the zinc finger domain of NEMO and the aa 126–250 segment of NS5A are essential for the interaction between NEMO and NS5A.Mechanistic analysis revealed that NS5A mediated the proteasomal degradation of NEMO.Ubiquitination assay showed that NS5A induced the K27-linked but not the K48-linked polyubiquitination of NEMO for proteasomal degradation.In addition,NS5A blocked the K63-linked polyubiquitination of NEMO,thus inhibiting IKK phosphorylation,IκBαdegradation,and NF-κB activation.These findings revealed a novel mechanism by which CSFV inhibits host innate immunity,which might guide the drug design against CSFV in the future.Jinfu Sun Jiaying Li Liming Li Haixiao Yu Ping Ma Yingnan Wang Jinqi Zhu Zezhong Feng Changchun Tu 2023Virologica Sinica2023,38,6:0
12Polymorphism of the DNA repair gene XPA and susceptibility to lung cancer显示文摘Objective: To study the relationship between one polymorphism in the promoter of the DNA repair gene XPA and the susceptibility to lung cancer. Methods: Genotypes were determined by the PCR-restriction fragment length polymorphism (PCR-RFLP) method in 310 histologically-confirmed lung cancer cases and 341 age and sex frequency-matched cancer-free controls. Results: The XPA A23G genotype frequencies were 27.1% (AA), 42.9% (AG), and 30.0% (GG) in case patients and 21.1% (AA), 52.8% (AG), and 26.1% (GG) in control subjects. Multivariate logistic regression analysis revealed that individuals carrying at least one 23G variant allele (AG+GG genotypes) had a significantly decreased risk for lung cancer (adjusted OR=0.66; 95%CI=0.44-0.98) compared with the wild-type genotype (23AA). Stratified analysis showed that the protective effect was more evident in subjects with a family history of cancer. 13Raltitrexed as a synergistic hyperthermia chemotherapy drug screened in patient-derived colorectal cancer organoids显示文摘Objective:Organoids have recently been used as in vitro models to screen chemotherapy drugs in combination with hyperthermia treatment in colorectal cancer.Our research aimed to establish a library of patient-derived colorectal cancer organoids to evaluate synergism between chemotherapy drugs and hyperthermia;validate an index of the hyperthermia chemotherapy sensitization enhancement ratio(HCSER)to identify the chemotherapeutics most enhanced by hyperthermia;and recommend chemotherapy drugs for hyperthermic intraperitoneal treatment.Methods:Organoids were grown from cells extracted from colorectal cancer patient samples or colorectal cancer cell lines.Cells from both sources were encapsulated in 3 D Matrigel droplets,which were formulated in microfluidics and phase-transferred into identical cell-laden Matrigel microspheres.The microspheres were seeded in 96-well plates,with each well containing a single microsphere that developed into an organoid after 7 days.The organoids were used to evaluate the efficacy of chemotherapy drugs at both 37℃ as a control and 43℃ for 90 min to examine hyperthermia synergism.Cell viability was counted with 10%CCK8.Results:We successfully established a library of colorectal cancer organoids from 22 patient parental tumors.We examined the hyperthermia synergism of 7 commonly used hyperthermic intraperitoneal chemotherapy drugs.In 11 of the 22 patient organoids,raltitrexed had significant hyperthermia synergism,which was indexed as the highest HCSER score within each patient group.Conclusions:Our results primarily demonstrated the use of patient-derived colorectal cancer organoids as in vitro models to evaluate hyperthermia synergistic chemotherapeutics.We found that hyperthermia enhanced the effect of raltitrexed the most among the common anti-colorectal cancer drugs.Lisi Zeng Quanxing Liao Haoran Zhao Shengwei Jiang Xianzi Yang Hongsheng Tang Qingjun He Xiansheng Yang Shuxian Fang Jinfu He Weiwen Cui Laiqiang Huang Shaohua Ma Shuzhong Cui 2021Cancer Biology & Medicine2021,18,3:0
14Identification of (-)-bornyl diphosphate synthase from Blumea balsamifera and its application for (-)-borneol biosynthesis in Saccharomyces cerevisiae显示文摘Borneol is a precious monoterpenoid with two chiral structures,(-)-borneol and(+)-borneol.Bornyl diphosphate synthase is the key enzyme in the borneol biosynthesis pathway.Many(+)-bornyl diphosphate synthases have been reported,but no(-)-bornyl diphosphate synthases have been identified.Blumea balsamifera leaves are rich in borneol,almost all of which is(-)-borneol.In this study,we identified a high-efficiency(-)-bornyl diphosphate synthase(BbTPS3)from B.balsamifera that converts geranyl diphosphate(GPP)to(-)-bornyl diphosphate,which is then converted to(-)-borneol after dephosphorylation in vitro.BbTPS3 exhibited a K m value of 4.93±1.38μM for GPP,and the corresponding k cat value was 1.49 s−1.Multiple strategies were applied to obtain a high-yielding(-)-borneol producing yeast strain.A codon-optimized BbTPS3 protein was introduced into the GPP high-yield strain MD,and the resulting MD-B1 strain produced 1.24 mg⋅L^(-1)(-)-borneol.After truncating the N-terminus of BbTPS3 and adding a Kozak sequence,the(-)-borneol yield was further improved by 4-fold to 4.87 mg⋅L^(-1).Moreover,the(-)-borneol yield was improved by expressing the fusion protein module of ERG20 F96W-N127W-YRSQI-t14-BbTPS3K2,resulting in a final yield of 12.68 mg⋅L^(-1) in shake flasks and 148.59 mg⋅L^(-1) in a 5-L bioreactor.This work is the first reported attempt to produce(-)-borneol by microbial fermentation.Rui Ma Ping Su Qing Ma Juan Guo Suiqing Chen Baolong Jin Haiyan Zhang Jinfu Tang Tao Zhou Chenghong Xiao Guanghong Cui Luqi Huang 2022Synthetic and Systems Biotechnology2022,7,1:0
15香菇引起的小肠梗阻两例报道显示文摘The shiitake mushroom(Lentinula edodes),known as Xiang-gu in China,has been an important component of Asian cuisine for hundreds of years.Although not easily digestible,there are few reports of them causing bowel obstruction.We present two cases of small bowel obstruction due to a shiitake mushroomrequiring surgical intervention.Two patients who did not have any teeth and did not use dentures presented with intestinal bowel obstruction and were referred to the Emergency Department of our hospital after eating a meal including shiitake mushrooms without cutting.The first patient underwent an emergency laparotomy and a semental small bowel resection and the other underwent laparoscopic small bowel incision for removal of a foreign body.The causes of the small bowel obstruction for the two patients were uncut shiitake mushrooms in the small bowel.The two patients recovered uneventfully post-operatively.Jinfu Tan Kaitao Yuan Jidong Zuo Weigang Dai Yujie Yuan Weidong Feng Min Tan Jinping Ma 2019Gastroenterology Report2019,7,4:0
16Functional characterization of CYP81C16 involved in the tanshinone biosynthetic pathway in Salvia miltiorrhiza显示文摘Danshen,the dried roots and rhizomes of Salvia miltiorrhiza Bunge(S.miltiorrhiza),is widely used in the treatment of cardiovascular and cerebrovascular diseases.Tanshinones,the bioactive compounds from Danshen,exhibit a wide spectrum of pharmacological properties,suggesting their potential for future therapeutic applications.Tanshinone biosynthesis is a complex process involving at least six P450 enzymes that have been identified and characterized,most of which belong to the CYP76 and CYP71 families.In this study,CYP81C16,a member of the CYP71 clan,was identified in S.miltiorrhiza.An in vitro assay revealed that it could catalyze the hydroxylation of four para-quinone-type tanshinones,namely neocryptotanshinone,deoxyneocryptotanshinone,and danshenxinkuns A and B.SmCYP81C16 emerged as a potential broad-spectrum oxidase targeting the C-18 position of para-quinone-type tanshinones with an impressive relative conversion rate exceeding 90%.Kinetic evaluations and in vivo assays underscored its highest affinity towards neocryptotanshinone among the tested substrates.The overexpression of SmCYP81C16 promoted the accumulation of(iso)tanshinone in hairy root lines.The characterization of SmCYP81C16 in this study accentuates its potential as a pivotal tool in the biotechnological production of tanshinones,either through microbial or plant metabolic engineering.REN Li LUO Linglong HU Zhimin MA Ying WANG Jian CHENG Yatian JIN Baolong CHEN Tong TANG Jinfu CUI Guanghong GUO Juan HUANG Luqi 2023Chinese Journal of Natural Medicines2023,21,12:0
17Data driven analysis reveals prognostic genes and immunological targets in human sepsis-associated acute kidney injury显示文摘BACKGROUND:The molecular mechanism of sepsis-associated acute kidney injury(SA-AKI)is unclear.We analyzed co-differentially expressed genes(co-DEGs)to elucidate the underlying mechanism and intervention targets of SA-AKI.METHODS:The microarray datasets GSE65682,GSE30718,and GSE174220 were downloaded from the Gene Expression Omnibus(GEO)database.We identified the co-DEGs and constructed a gene co-expression network to screen the hub genes.We analyzed immune correlations and disease correlations and performed functional annotation of the hub genes.We also performed single-cell and microenvironment analyses and investigated the enrichment pathways and the main transcription factors.Finally,we conducted a correlation analysis to evaluate the role of the hub genes.RESULTS:Interleukin 32(IL32)was identified as the hub gene in SA-AKI,and the main enriched signaling pathways were associated with hemopoiesis,cellular response to cytokine stimulus,inflammatory response,and regulation of kidney development.Additionally,IL32 was significantly associated with mortality in SA-AKI patients.Monocytes,macrophages,T cells,and NK cells were closely related to IL32 and were involved in the immune microenvironment in SA-AKI patients.IL32 expression increased significantly in the kidney of septic mouse.Toll-like receptor 2(TLR2)was significantly and negatively correlated with IL32.CONCLUSION:IL32 is the key gene involved in SA-AKI and is significantly associated with prognosis.TLR2 and relevant immune cells are closely related to key genes.Qing Zhao Jinfu Ma Jianguo Xiao Zhe Feng Hui Liu 2024World Journal of Emergency Medicine2024,15,2:0
