1Metastatic human hepatocellular carcinoma models in nude mice and cell line with metastatic potential显示文摘Metastatic human HCC model is needed for the studies on mechanism and intervention of metastatic recurrence. By using orthotopic implantation of histologically intact tissues of 30 surgical specimens, a patient like metastatic model of human HCC in nude mice (LCI-D20)and a Iow metastatic model of human HCC in nude mice LCI-D35 ) have been established. All mice with transplanted LCI-D20 tumors exhibited extremely high metastatic ability including spontaneous metastasis to liver, lungs, lymph nodes and peritoneal seeding.Remarkable difference was also found in expression of some of the invasiveness related genes and growth factors between the LCI-D20 and LCI-D35 tumors. PAI-Iincreased gradually following tumor progression in LCID20 model, and correlated with tumor size and AFP level,Phasic expression of tissue intercellular adhesion molecule-I in this model was also observed. Using corneal micropocket model, it was demonstrated that the vascular response induced by LCI-D20 tumor was stronger than that induced by LCI-D35 tumor. Similar report on metastatic human HCC model in nude mice and human HCC cell line with metastatic potential was rarely found in the literature. 2Broadband diffusion of terahertz waves by multi-bit coding metasurfaces显示文摘The terahertz region is a special region of the electromagnetic spectrum that incorporates the advantages of both microwaves and infrared light waves.In the past decade,metamaterials with effective medium parameters or gradient phases have been studied to control terahertz waves and realize functional devices.Here,we present a new approach to manipulate terahertz waves by using coding metasurfaces that are composed of digital coding elements.We propose a general coding unit based on a Minkowski closed-loop particle that is capable of generating 1-bit coding(with two phase states of 0 and 180°),2-bit coding(with four phase states of 0,90°,180°,and 270°),and multi-bit coding elements in the terahertz frequencies by using different geometric scales.We show that multi-bit coding metasurfaces have strong abilities to control terahertz waves by designing-specific coding sequences.As an application,we demonstrate a new scattering strategy of terahertz waves—broadband and wide-angle diffusion—using a 2-bit coding metasurface with a special coding design and verify it by both numerical simulations and experiments.The presented method opens a new route to reducing the scattering of terahertz waves.Li-Hua Gao Qiang Cheng Jing Yang Shao-Jie Ma Jie Zhao Shuo Liu Hai-Bing Chen Qiong He Wei-Xiang Jiang Hui-Feng Ma Qi-Ye Wen Lan-Ju Liang Biao-Bing Jin Wei-Wei Liu Lei Zhou Jian-Quan Yao Pei-Heng Wu Tie-Jun Cui 2015Light(Science & Applications)2015,4,1:31
3Oridonin induces apoptosis in gastric cancer through Apaf-1,cytochrome c and caspase-3 signaling pathway显示文摘AIM:To investigate the effect and mechanism of oridonin on the gastric cancer cell line HGC-27 in vitro.METHODS:The inhibitory effect of oridonin on HGC-27 cells was detected using the 3-(4,5-dimethylthiazol2-yl)-2,5-diphenyl tetrazolium bromide assay.After treatment with 10 μg/mL oridonin for 24 h and 48 h,the cells were stained with acridine orange/ethidium bromide.The morphologic changes were observed under an inverted fluorescence microscope.DNA fragmen-tation(a hallmark of apoptosis) and lactate dehydrogenase activity were examined using DNA ladder assay and lactate dehydrogenase-release assay.After treated with oridonin(0,1.25,2.5,5 and 10 μg/mL),HGC-27 cells were collected for anexin V-phycoerythrin and 7-amino-actinomycin D double staining and tested by flow cytometric analysis,and oridonin-induced apoptosis in HGC-27 cells was detected.After treatment with oridonin for 24 h,the effects of oridonin on expression of Apaf-1,Bcl-2,Bax,caspase-3 and cytochrome c were also analyzed using reverse-transcript polymerase chain reaction(RT-PCR) and Western blotting.RESULTS:Oridonin significantly inhibited the proliferation of HGC-27 cells in a dose-and time-dependent manner.The inhibition rates of HGC-27 treated with four different concentrations of oridonin for 24 h(1.25,2.5,5 and 10 μg/mL) were 1.78% ± 0.36%,4.96% ± 1.59%,10.35% ± 2.76% and 41.6% ± 4.29%,respectively,which showed a significant difference(P < 0.05).The inhibition rates of HGC-27 treated with oridonin at the four concentrations for 48 h were 14.77% ± 4.21%,21.57% ± 3.75%,30.31% ± 4.91% and 61.19% ± 5.81%,with a significant difference(P < 0.05).The inhibition rates of HGC-27 treated with oridonin for 72 h at the four concentrations were 25.77% ± 4.85%,31.86% ± 3.86%,48.30% ± 4.16% and 81.80% ± 6.72%,with a significant difference(P < 0.05).Cells treated with oridonin showed typical apoptotic features with acridine orange/ethidium bromide staining.After treatment with oridonin,the cells became round,shrank,and developed small buds around the nuclear membrane while forming apoptotic bodies.Lactate dehydrogenase(LDH) release assay showed that after treated with 1.25 μg/mL and 20 μg/mL oridonin for 24 h,LDH release of HGC-27 caused by apoptosis increased from 22.94% ± 3.8% to 52.68% ± 2.4%(P < 0.001).However,the change in the release of LDH caused by necrosis was insignificant,suggesting thatthe major cause of oridonin-induced HGC-27 cell death was apoptosis.Flow cytometric analysis also revealed that oridonin induced significant apoptosis compared with the controls(P < 0.05).And the apoptosis rates of HGC-27 induced by the four different concentrations of oridonin were 5.3% ± 1.02%,12.8% ± 2.53%,28.5% ± 4.23% and 49.6% ± 3.76%,which were in a dose-dependent manner(P < 0.05).After treatment for 24 h,DNA ladder showed that oridonin induced a significant increase in DNA fragmentation in a dosedependent manner.RT-PCR revealed that mRNA expression levels were up-regulated compared with the controls in caspase-3(0.917 ± 0.103 vs 0.357 ± 0.019,P < 0.05),cytochrome c(1.429 ± 0.111 vs 1.002 ± 0.014,P < 0.05),Apaf-1(0.688 ± 0.101 vs 0.242 ± 0.037,P < 0.05) and Bax(0.856 ± 0.101 vs 0.278 ± 0.027,P < 0.05)(P < 0.05),whereas down-regulated in Bcl-2(0.085 ± 0.012 vs 0.175 ± 0.030,P < 0.05).Western blotting analysis also confirmed this result.CONCLUSION:Apoptosis of HGC-27 induced by oridonin may be associated with differential expression of Apaf-1,caspase-3 and cytochrome c,which are highly dependent upon the mitochondrial pathway.Ke-Wang Sun Ying-Yu Ma Tian-Pei Guan Ying-Jie Xia Chang-Ming Shao Le-Gao Chen Ya-Jun Ren Hai-Bo Yao Qiong Yang Xu-Jun He 2012World Journal of Gastroenterology2012,18,48:25
4Effects of aeration method and aeration rate on greenhouse gas emissions during composting of pig feces in pilot scale显示文摘The aim of this study was to uncover ways to mitigate greenhouse gas(GHG) emissions and reduce energy consumption during the composting process. We assessed the effects of different aeration rates(0, 0.18, 0.36, and 0.54 L/(kg dry matter(dm)·min)) and methods(continuous and intermittent) on GHG emissions. Pig feces and corn stalks were mixed at a ratio of 7:1. The composting process lasted for 10 weeks, and the compost was turned approximately every 2 weeks. Results showed that both aeration rate and method significantly affected GHG emissions. Higher aeration rates increased NH3 and N2O losses,but reduced CH4 emissions. The exception is that the CH4 emission of the passive aeration treatment was lower than that of the low aeration rate treatment. Without forced aeration,the CH4 diffusion rates in the center of the piles were very low and part of the CH4 was oxidized in the surface layer. Intermittent aeration reduced NH3 and CH4 losses, but significantly increased N2 O production during the maturing periods. Intermittent aeration increased the nitrification/denitrification alternation and thus enhanced the N2 O production. Forced aeration treatments had higher GHG emission rates than the passive aeration treatment. Forced aeration accelerated the maturing process, but could not improve the quality of the end product. 5Achieving partial nitrification by inhibiting the activity of Nitrospira-like bacteria under high-DO conditions in an intermittent aeration reactor显示文摘It is generally accepted that a low dissolved oxygen(DO) concentration is more beneficial for achieving partial nitrification than high-DO. In this study, partial nitrification was not established under low-DO conditions in an intermittent aeration reactor for treating domestic wastewater. During the operational period of low-DO conditions(DO: 0.3 ±0.14 mg/L), stable complete nitrification was observed. The abundance of Nitrospira-like bacteria, which were the major nitrite-oxidizing bacteria, increased from 1.03 × 10~6to2.64 × 10~6cells/m L. At the end of the low-DO period, the batch tests showed that high-DO concentration(1.5, 2.0 mg/L) could inhibit nitrite oxidation, and enhance ammonia oxidation. After switching to the high-DO period(1.8 ± 0.32 mg/L), partial nitrification was gradually achieved. Nitrospira decreased from 2.64 × 10~6 to 8.85 × 10~5cells/m L. 8Changes in temperature extremes in the Yangtze River Basin, 1962-2011显示文摘Based on daily maximum and minimum temperature observed by the China Meteorological Administration at 115 meteorological stations in the Yangtze River Basin from 1962 to 2011,the methods of linear regression,principal component analysis and correlation analysis are employed to investigate the temporal variability and spatial distribution of temperature extremes.Sixteen indices of extreme temperature are selected.The results are as follows:(1) The occurrence of cold days,cold nights,ice days,frost days and cold spell duration indicator has significantly decreased by –0.84,–2.78,–0.48,–3.29 and –0.67 days per decade,respectively.While the occurrence of warm days,warm nights,summer days,tropical nights,warm spell duration indicator and growing season length shows statistically significant increasing trends at rates of 2.24,2.86,2.93,1.80,0.83 and 2.30 days per decade,respectively.The tendency rate of the coldest day,coldest night,warmest day,warmest night and diurnal temperature range is 0.33,0.47,0.16,0.19 and –0.07℃ per decade,respectively.(2) The magnitudes of changes in cold indices(cold nights,coldest day and coldest night) are obviously greater than those of warm indices(warm nights,warmest day and warmest night).The change ranges of night indices(warm nights and cold nights) are larger than those of day indices(warm days and cold days),which indicates that the change of day and night temperature is asymmetrical.(3) Spatially,the regionally averaged values of cold indices in the upper reaches of the Yangtze River Basin are larger than those in the middle and lower reaches.However,the regionally averaged values of most warm indices(except warm spell duration indicator) and growing season length in the middle and lower reaches are larger than those in the upper reaches.(4) The extreme temperature indices are well correlated with each other except diurnal temperature range.WANG Qiong ZHANG Mingjun WANG Shengjie MA Qian SUN Meiping 2014Journal of Geographical Sciences2014,24,1:14
9Changes in China’s lakes: climate and human impacts显示文摘Lakes have played a critical role in providing water and ecosystem services for people and other organisms in China for millennia.However,accelerating climate change and economic boom have resulted in unprecedented changes in these valuable lakes.Using Landsat images covering the entity of the country,we explored the changes in China’s lakes and the associated driving forces over the last 30 years(i.e.mid-1980 s to 2015).We discovered that China’s lakes have changed with divergent regional trends:in the sparsely populated Tibetan Plateau,lakes are abundant and the lake area has increased dramatically from 38596 to46831 km^2(i.e.increased by 8235 km^2,or 21.3%),whereas,in the densely populated northern and eastern regions,lakes are relatively scarce and the lake area has decreased from 36659 to 33657 km^2(i.e.decreased by 3002 km^2,or 8.2%).In particular,severe lake decreases occurred in the Mongolia-Xinjiang Plateau and the Eastern Plain(-2151 km^2).Statistical analyses indicated that climate was the most important factor controlling lake changes in the Tibetan Plateau,the Yun-Gui Plateau and the Northeast Plain.However,the strength of climatic control on lake changes was low in the Eastern Plain and the Mongolia-Xinjiang Plateau,where human activities,e.g.impoldering,irrigation and mining,have caused serious impacts on lakes.Further lake changes will exacerbate regional imbalances between lake resources and population distribution,and thus may increase the risk of water-resource crises in China.Shengli Tao Jingyun Fang Suhui Ma Qiong Cai Xinyu Xiong Di Tian Xia Zhao Leqi Fang Heng Zhang Jiangling Zhu Shuqing Zhao 2020National Science Review2020,7,1:14
10Investigations on Nitrate Pollution of Soil, Groundwater and Vegetable from Three Typical Farmlands in Beijing Region, China显示文摘The aim of this study was to determine the nitrate pollution status of soil, groundwater, and vegetable from three typical farmlands (croplands, vegetable fields, and orchards) in Beijing region. During the investigation, hundreds of the soil, groundwater, and vegetable samples from three typical farmlands were collected and analyzed. In addition, attributes of all samples were recorded for data analysis. The results showed that nitrate was substantially accumulated in soil profiles, while the soil nitrate concentrations of vegetable fields and orchards were higher than those of croplands. Nitrate concentration in 0-30 cm soil of vegetable field and orchard were 3.8 and 1.2 times of that of cropland, respectively. Nitrate content of groundwater in vegetable field was 13.8 mg L-1 (with the over-standard ratio 44.8%), which was 2.8 folds of that in cropland. Nitrate concentration of groundwater under orchard was 9.3 mg L-1 (with the over-standard ratio 23.5%), which was 1.9 folds of that in cropland. High concentrations of the nitrate in vegetables were detected, particularly green leafy vegetables ranked first with 2 685.5 mg kg-1, followed by rhizome vegetables, cabbages, and fruit vegetables. The nitrate over-standard ratios of rhizome vegetables, green leafy vegetables, fruit vegetables, and cabbages were 80.9, 37.9, 29.7, and 2.2%, respectively. 12Enhanced absorption and inhibited metabolism of emodin by 2,3,5,4'-tetrahydroxystilbene-2-O-β-D-glucopyranoside: Possible mechanisms for Polygoni Multiflori Radix-induced liver injury显示文摘Polygoni Multiflori Radix(PMR) has been commonly used as a tonic in China for centuries. However, PMR-associated hepatotoxicity is becoming a safety issue. In our previous in vivo study, an interaction between stilbenes and anthraquinones has been discovered and a hypothesis is proposed that the interaction between stilbene glucoside-enriching fraction and emodin may contribute to the side effects of PMR. To further support our previous in vivo results in rats, the present in vitro study was designed to evaluate the effects of 2, 3, 5, 4'-tetrahydroxystilbene-2-O-β-D-glucopyranoside(TSG) on the cellular absorption and human liver microsome metabolism of emodin. The obtained results indicated that the absorption of emodin in Caco-2 cells was enhanced and the metabolism of emodin in human liver microsomes was inhibited after TSG treatment. The effects of the transport inhibitors on the cellular emodin accumulation were also examined. Western blot assay suggested that the depressed metabolism of emodin could be attributed to the down-regulation of UDP-glucuronosyltransferases(UGTs) 1A8, 1A10, and 2B7. 13Epidermal growth factor receptor (EGFR) T790M mutation identified in plasma indicates failure sites and predicts clinical prognosis in non-small cell lung cancer progression during first-generation tyrosine kinase inhibitor therapy: a prospective observational study显示文摘Introduction:Plasma circulating tumor DNA(ctDNA)is an ideal approach to detecting the epidermal growth factor receptor(EGFR)T790M mutation,which is a major mechanism of resistance to first-generation EGFR-tyrosine kinase inhibitor(TKI)therapy.The present study aimed to explore the association of ctDNA-identified T790M mutation with disease failure sites and clinical prognosis in non-small cell lung cancer(NSCLC)patients.Methods:Patients who progressed on first-generation TKIs were categorized into failure site groups of chest limited(CF),brain limited(BF)and other(OF).Amplification refractory mutation system(ARMS)and droplet digital PCR(ddPCR)were used to identify the T790M mutation in ctDNA.Prognosis was analyzed with Kaplan-Meier methods.Results:Overall concordance between the two methods was 78.3%.According to both ARMS and ddPCR,patients in the OF group had a significantly higher rate of T790M mutation than did patients in the BF and CF groups(P<0.001),and a significantly higher T790M mutation rate was also observed in OF-group patients than in those in the CF and BF groups(P<0.001).AZD9291 was found to be an excellent treatment option and yielded the longest survival for T790M+patients in all groups who had progressed on EGFR-TKIs;for other treatments,the prognosis of T790M−patient subgroups varied.Conclusions:The present study demonstrates that T790M mutation in ctDNA is associated with failure sites for NSCLC patients after EGFR-TKI therapy and indicates that both failure site and T790M mutational status greatly influ-ence treatment selection and prognosis.Shirong Zhang Lucheng Zhu Bing Xia Enguo Chen Qiong Zhao Xiaochen Zhang Xueqin Chen Xufeng Chen Shenglin Ma 2018Cancer Communications2018,38,1:12
14Science Letters:IGFBP7 plays a potential tumor suppressor role against colorectal carcinogenesis with its expression associated with DNA hypomethylation of exon 1显示文摘Insulin-like growth factor binding-protein-7 (IGFBP7) was obtained from our previous colonic adenocarcinoma (CRC) and normal mucosa suppression subtraction hybridization (SSH) cDNA libraries. By RT-PCR and immunohistochemistry, we found that IGFBP7 was overexpressed in CRC tissue compared to normal tissue. However, our in vitro experiments performed in 10 CRC cell lines showed that IGFBP7 expressed only in SW480 and Caco2 cell lines, which implied an underlying reversible regulatory mechanism. Using methylation-specific PCR (MSP) and bisulfite sodium PCR (BSP), we found that its expression was associated with DNA hypomethylation of exon1. This was further supported by the in vitro study which showed restored IGFBP7 expression after demethylation agent 5-aza-2’-deoxycytidine treatment. Correlation analysis between IGFBP7 expression and prognosis indicated that overexpression of IGFBP7 in CRC tissue correlated with favourable survival. Investigation of the func-tional role of IGFBP7 through transfection studies showed that IGFBP7 protein could inhibit growth rate, decrease colony for-mation activity, and induce apoptosis in RKO and SW620 cells, suggesting it a potential tumor suppressor protein in colorectal carcinogenesis. 16Microstructure evolution of AZ91 alloy processed by a combination method of equal channel angular pressing and rolling显示文摘In the present work,AZ91 alloy was successfully processed by equal channel angular pressing(ECAP)for up to 16 passes and rolling(R)for multiple passes with a total reduction of 75%in addition to a combination method with ECAP plus rolling(ECAP+R).The effects of various processes(ECAP,R and ECAP+R)on microstructure evolution were analyzed and the influence of ECAP process on the rolling performance was examined.The result shows that ECAP contributed to a homogenous grain structure and formed a texture with higher Schmidt factors that was easy for rolling.A plate with smoother surface and reduced edge cracks was observed in the ECAP+R process than in the single R process.Although the microstructure of the alloy was similar after ECAP+R and R process,the sample of ECAP+R was more refined and had stronger second phase precipitation than the sample of R,which resulted in better rolling characteristics,along with the external surfaces.Qiong Xu Aibin Ma Yuhua Li Jiapeng Sun Yuchun Yuan Jinghua Jiang Chaoying Ni 2020Journal of Magnesium and Alloys2020,8,1:10
17Phytoavailability of Copper, Zinc and Cadmium in Sewage Sludge-Amended Calcareous Soils显示文摘The toxicity of trace elements (TEs), such as copper (Cu), zinc (Zn), and cadmium (Cd), often restrict land application of sewage sludge (SS) and there was little information about soil-plant transfer of TEs in SS from field experiments in China. In this study pot and field experiments were carried out for 2 years to investigate the phytoavailability of TEs in calcareous soils amended with SS. The results of the pot experiment showed that the phytoavailability of Zn and Cu in the SS was equal to 53.4%-80.9% and 54.8%-91.1% of corresponding water-soluble metal salts, respectively. The results from the field experiment showed that the contents of total Zn, Cu, and Cd in the soils increased linearly with SS application rates. With increasing SS application rates, the contents of Zn and Cu in the wheat grains initially increased and then reached a plateau, while there was no significant change of Cd content in the maize grains. The bioconcentration factors of the metals in the grains of wheat and maize were found to be in the order of Zn > Cu > Cd, but for the straw the order was Cd > Cu > Zn. It was also found that wheat grains could accumulate more metals compared with maize grains. 18Dynamically controlling terahertz wavefronts with cascaded metasurfaces显示文摘Dynamically controlling terahertz(THz)wavefronts in a designable fashion is highly desired in practice.However,available methods working at microwave frequencies do not work well in the THz regime due to lacking suitable tunable elements with submicrometer sizes.Here,instead of locally controlling individual meta-atoms in a THz metasurface,we show that rotating different layers(each exhibiting a particular phase profile)in a cascaded metadevice at different speeds can dynamically change the effective Jonesmatrix property of the whole device,thus enabling extraordinary manipulations on the wavefront and polarization characteristics of a THz beam impinging on the device.After illustrating our strategy based on model calculations,we experimentally demonstrate two proof-of-concept metadevices,each consisting of two carefully designed all-silicon transmissive metasurfaces exhibiting different phase profiles.Rotating two metasurfaces inside the fabricated devices at different speeds,we experimentally demonstrate that the first metadevice can efficiently redirect a normally incident THz beam to scan over a wide solid-angle range,while the second one can dynamically manipulate both the wavefront and polarization of a THz beam.Our results pave the way to achieving dynamic control of THz beams,which is useful in many applications,such as THz radar,and bio-and chemical sensing and imaging.Xiaodong Cai Rong Tang Haoyang Zhou Qiushi Li Shaojie Ma Dongyi Wang Tong Liu Xiaohui Ling Wei Tan Qiong He Shiyi Xiao Lei Zhou 2021Advanced Photonics2021,3,3:8

20Crosstalk-free achromatic full Stokes imaging polarimetry metasurface enabled by polarization-dependent phase optimization显示文摘Imaging polarimetry is one of the most widely used analytical technologies for object detection and analysis.To date,most metasurface-based polarimetry techniques are severely limited by narrow operating bandwidths and inevitable crosstalk,leading to detrimental effects on imaging quality and measurement accuracy.Here,we propose a crosstalkfree broadband achromatic full Stokes imaging polarimeter consisting of polarization-sensitive dielectric metalenses,implemented by the principle of polarization-dependent phase optimization.Compared with the single-polarization optimization method,the average crosstalk has been reduced over three times under incident light with arbitrary polarization ranging from 9μm to 12μm,which guarantees the measurement of the polarization state more precisely.The experimental results indicate that the designed polarization-sensitive metalenses can effectively eliminate the chromatic aberration with polarization selectivity and negligible crosstalk.The measured average relative errors are 7.08%,8.62%,7.15%,and 7.59%at 9.3,9.6,10.3,and 10.6μm,respectively.Simultaneously,the broadband full polarization imaging capability of the device is also verified.This work is expected to have potential applications in wavefront detection,remote sensing,light-field imaging,and so forth.Yaxin Zhang Mingbo Pu Jinjin Jin Xinjian Lu Yinghui Guo Jixiang Cai Fei Zhang Yingli Ha Qiong He Mingfeng Xu Xiong Li Xiaoliang Ma Xiangang Luo 2022Opto-Electronic Advances2022,5,11:6
