1Role of sex steroid receptors in pathobiology of hepatocellular carcinoma显示文摘The striking gender disparity observed in the incidence of hepatocellular carcinoma (HCC) suggests an important role of sex hormones in HCC pathogenesis. Though the studies began as early as in 1980s,the precise role of sex hormones and the significance of their receptors in HCC still remain poorly understood and perhaps contribute to current controversies about the potential use of hormonal therapy in HCC. A comprehensive review of the existing literature revealed several shortcomings associated with the studies on estrogen receptor (ER) and androgen receptor (AR) in normal liver and HCC. These shortcomings include the use of less sensitive receptor ligand binding assays and immunohistochemistry studies for ERα alone until 1996 when ERβ isoform was identified. The animal models of HCC utilized for studies were primarily based on chemical-induced hepatocarcinogenesis with less similarity to virus-induced HCC pathogenesis. However,recent in vitro studies in hepatoma cells provide newer insights for hormonal regulation of key cellular processes including interaction of ER and AR with viral proteins. In light of the above facts,there is an urgent need for a detailed investigation of sex hormones and their receptors in normal liver and HCC. In this review,we systematically present the information currently available on androgens,estrogens and their receptors in normal liver and HCC obtained from in vitro,in vivo experimental models and clinical studies. 3Blockchain-Assisted Secure Fine-Grained Searchable Encryption for a Cloud-Based Healthcare Cyber-Physical System显示文摘The concept of sharing of personal health data over cloud storage in a healthcare-cyber physical system has become popular in recent times as it improves access quality.The privacy of health data can only be preserved by keeping it in an encrypted form,but it affects usability and flexibility in terms of effective search.Attribute-based searchable encryption(ABSE)has proven its worth by providing fine-grained searching capabilities in the shared cloud storage.However,it is not practical to apply this scheme to the devices with limited resources and storage capacity because a typical ABSE involves serious computations.In a healthcare cloud-based cyber-physical system(CCPS),the data is often collected by resource-constraint devices;therefore,here also,we cannot directly apply ABSE schemes.In the proposed work,the inherent computational cost of the ABSE scheme is managed by executing the computationally intensive tasks of a typical ABSE scheme on the blockchain network.Thus,it makes the proposed scheme suitable for online storage and retrieval of personal health data in a typical CCPS.With the assistance of blockchain technology,the proposed scheme offers two main benefits.First,it is free from a trusted authority,which makes it genuinely decentralized and free from a single point of failure.Second,it is computationally efficient because the computational load is now distributed among the consensus nodes in the blockchain network.Specifically,the task of initializing the system,which is considered the most computationally intensive,and the task of partial search token generation,which is considered as the most frequent operation,is now the responsibility of the consensus nodes.This eliminates the need of the trusted authority and reduces the burden of data users,respectively.Further,in comparison to existing decentralized fine-grained searchable encryption schemes,the proposed scheme has achieved a significant reduction in storage and computational cost for the secret key associated with users.It has been verified both theoretically and practically in the performance analysis section.Mamta Brij B.Gupta Kuan-Ching Li Victor C.M.Leun Kostas E.Psannis Shingo Yamaguchi 2021IEEE/CAA Journal of Automatica Sinica2021,8,12:5
4Estrogen receptor expression in chronic hepatitis C and hepatocellular carcinoma pathogenesis显示文摘AIM To investigate gender-specific liver estrogen receptor(ER) expression in normal subjects and patients with hepatitis C virus(HCV)-related cirrhosis and hepatocellular carcinoma(HCC).METHODS Liver tissues from normal donors and patients diagnosed with HCV-related cirrhosis and HCV-related HCC were obtained from the NIH Liver Tissue and Cell Distribution System. The expression of ER subtypes, ERα and ERβ, were evaluated by Western blotting and real-time RT-PCR. The subcellular distribution of ERα and ERβ was further determined in nuclear and cytoplasmic tissue lysates along with the expression ofinflammatory [activated NF-κB and IκB-kinase(IKK)] and oncogenic(cyclin D1) markers by Western blotting and immunohistochemistry. The expression of ERα and ERβ was correlated with the expression of activated NF-κB, activated IKK and cyclin D1 by Spearman's correlation. RESULTS Both ER subtypes were expressed in normal livers but male livers showed significantly higher expression of ERα than females(P < 0.05). We observed significantly higher m RNA expression of ERα in HCV-related HCC liver tissues as compared to normals(P < 0.05) and ERβ in livers of HCV-related cirrhosis and HCV-related HCC subjects(P < 0.05). At the protein level, there was a significantly higher expression of nuclear ERα in livers of HCV-related HCC patients and nuclear ERβ in HCV-related cirrhosis patients as compared to normals(P < 0.05). Furthermore, we observed a significantly higher expression of phosphorylated NF-κB and cyclin D1 in diseased livers(P < 0.05). There was a positive correlation between the expression of nuclear ER subtypes and nuclear cyclin D1 and a negative correlation between cytoplasmic ER subtypes and cytoplasmic phosphorylated IKK in HCV-related HCC livers. These findings suggest that dysregulated expression of ER subtypes following chronic HCVinfection may contribute to the progression of HCVrelated cirrhosis to HCV-related HCC.CONCLUSION Gender differences were observed in ERα expression in normal livers. 5Development and organisation of gonadal steroidogenesis in bony fishes - A review显示文摘Gonadal steroidogenesis is pivotal to synchronize various reproductive stages including sexual development,growth and maturation.In all vertebrates including teleost,steroidogenesis is triggered by the mobilization of cholesterol by steroidogenic acute regulatory protein from outer to inner mitochondrial membrane.Thereafter,the entire process occurs in endoplasmic reticulum or mitochondria wherein various steroidogenic enzyme genes play a crucial role.The onset of steroidogenesis during sexual development in teleost is essentially regulated by differential expression of several transcription as well as steroidogenesis-related factors.More specifically,the role of dmrt,sox9,sox3,other sox forms,ad4bp/sf-1,wt-1,foxl2,ftz-f1,gata4,gsdf,Activator protein-1,fgfs and growth factors and steroidogenic enzymes such as cytochrome P450aromatase(cyp19a1),hydroxysteroid 3β-dehydrogenase(hsd3b),hydroxysteroid 17β-dehydrogenase(hsd17b)have been well characterised.Recently,the role of pax2,THO complex(thoc),pentraxin(ptx)and few signalling molecules like wnt4/5 regulating teleostean steroidogenesis has been reported.In females,cyp19a1 appears to be critical as it converts androgens to estrogens during ovarian differentiation which suggests that estradiol-17βis indispensable for the event.Unlike females,males do not depend on testosterone for testicular determination,yet has a major role along with 11-ketotestosterone in testicular development and growth when compared to early testicular differentiation.In males,onset of sex determining or testis-related genes seems most essential.Considering these,the regulation of steroidogenesis is virtually critical at later stages.In view of this,several steroidogenic motifs pertaining to transcriptional regulation have been analysed in teleost,yet far below than the reports on mammals.In this context,the regulatory influence on HHG axis plays a critical role in teleost.Further,multiple gonadotropin-releasing hormones and duality of gonadotropins play a differential role in gonadal function.As for steroidogenic pathway,the synthesis of sex steroids predominantly usesΔ4,however,Δ5 pathway was also evident.The next aspect is shift in steroidogenesis that usually occurs in the maturing follicles during final oocyte maturation,yet such a mechanism is not clear in males.The review highlights the interactions of steroidogenic enzyme gene regulation in terms of HHG axis and other important transcription factors that are involved in the regulatory pathways as well as the influence of environmental and dietary factors by comparing both sexes to present a holistic view on steroidogenesis onset and organisation during gametogenesis in teleost.Swathi Tenugu Akanksha Pranoty Sajwan-Khatri Mamta Balasubramanian Senthilkumaran 2021Aquaculture and Fisheries2021,6,3:2
6镍铁氰化物纳米粒子的合成及其多相催化无溶剂条件下苯甲醇氧化的潜能(英文)显示文摘Nickel hexacyanoferrate nanoparticles were synthesized and characterized using elemental analysis, thermal analysis, infrared spectroscopy, and X-ray diffraction. A FE-SEM image of the nickel hexacyanoferrate showed that it consists of nearly spherical particles with sizes ranging from 30 to 70 nm. The synthesized material was found to be a heterogeneous catalyst useful for the solvent-free oxidation of benzyl alcohol with H2O2 as an oxidant. A 36% conversion of benzyl alcohol to benzaldehyde was achieved under optimized reaction conditions using specific parameters such as the amount of catalyst, the temperature, the benzyl alcohol to H2O2 molar ratio, and the reaction time.Shah R.
